When organic loading rate is less than 1.0 kg/m$^3$ㆍd on site, it is achieved good removal rate independent of changes of loading rate.

자연계로 유출된 유류 및 유기용매 등 비수용성액체는 토양공극사이에 존재하며 서서히 용해되면서 지속적인 수질오염원으로 작용한다. 본 연구는 유리로 제작된 다공성매체 실험장치를 이용하여 토양 공극사이에 위치한 비수용성액체의 용해현상을 시각화하였고, 이미지 분석기술을 이용하여 용해속도를 정량화하였다. 다공성매체 공극사이 TCE는 물의 유량을 빠르게 할수록 용해속도가 증가하는 것으로 나타났다. 이는 TCE로 오염된 지역의 강우량이 많거나, 지하수 유속이 빠른 지역일 경우 토양내 TCE 제거는 용이하게 일어날 수 있지만, 용해된 TCE의 지하수 유입으로 증가한다는 점을 시사한다. 다공성매체 공극사이 TCE의 현미경 이미지 관측결과, 물의 유량이 빠른 경우 역시 TCE blob 수가 급격히 감소함을 확인하였다. 물의 흐름이 용이한 우선흐름경로에 존재하고 있는 TCE는 물과의 용이한 접촉으로 용해가 빠르게 진행되었다. 반면 정체구역에 존재하는 TCE의 용해속도는 매우 느리게 나타났다. 현장 토양에서도 동일한 현상이 일어날 것이므로 지하수 조사결과 TCE의 검출이 계속된다면, 토양 공극사이에 여전히 TCE가 존재하고 있음을 암시하므로 이에 대한 대책이 강구되어야 한다.
The effects of ammonium ion on cell growth kinetics, monoclonal antibody productivity, and cell metabolism of hybridoma cells were investigated. The mouse-mouse hybridoma cell line VlIIH-8 producing mouse IgG2a was used as a model system. Ammonium ion showed an inhibitory effect on cell growth and monoclonal antibody production. New immobilized adsorbents were developed for the reduction of the inhibitory effect of ammonium ion. The ammonium ion selective zeolite, Phillipsite-Gismondine was entrapped in calcium alginate bead or in dialysis membrane and applied to the hybridoma cell culture system for the in situ removal of ammonium ion from culture media. The effects of ammonium the both serum supplemented and serum free media on the cell growth were studied by applying immobilized adsorbents of calcium alginate bead type. The results demonstrated a substantial enhancement in cell growth. Applying immobilized adsorbents of dialysis membrane type to serum supplemented media also resulted in the stimulation of cell growth, cell viability and monoclonal antibody production.

Staining profiles and bacterial morphology were compared in Xanthomonas citri pv. citri by a transmission electron microscopy. Four types of negative staining regimes were employed depending on culture media and heavy metal stains. The bacterial cells grown on LB agar media often appeared clustered on the supporting film. Meanwhile, individual bacterial cells could be readily found on the preparations from LB broth media. Typical rod-shaped cells (ca. $1\;{\mu}m$ in length) and their flagella were observed in either 2% uranyl acetate (UA) or 2% neutralized potassium phosphotungstate (PTA) staining. The UA-stained bacteria often showed relatively intact cell morphology and rather positively stained cells with a thin electron-dense stain depth around bacteria. The PTA-stained bacteria were characterized by the wrinkled cell surface where the stain was entrapped in grooves. In addition, distinct electron-dense stain depth was evident around the PTA-stained preparations. Numerous fimbriae could be mostly observed from the PTA-stained preparations of the two culture media, but not from the UA-stained preparations.
Progestrone bioconversion by immobilized Aspergillus phoenicis was studied. Progesterone was converted into 11$\alpha$-hydroxyprogesterone and 3-minor byproducts. Whole cells of A. phoenicis were immobillized by enreappment with calcium-alginate, K-carrageenan, or polyacrylamide. Of these materials tested, cell immobilized in $Ca^{2+}$ -alginate gels showed the highest activity for 11$\alpha$-hydroxylation of progesterone. In the case of mycelia immobilized in $Ca^{2+}$-alginate, futher progressing hydroxylation of 11$\alpha$-hydroxyprogesterone was greatly reduced. Spores of A. phoenicis which were immobillized with $Ca^{2+}$-alginate and germinatedin situ for 25 hours showed higher 11$\alpha$-hydroxylase activity than those of entrapped whole mycelia and maintained initial enzyme activity for all 8 times of repeated use. After 16 times of reuse, the activity was declined 30% or more. When culture media and $Zn^{2+}$ were introduced into the reaction media, the activity of the immobilized mycelia which had been lowered due to many times of reuse was effectively reactivated.
Quorum quenching (QQ) bacteria entrapped in a polymeric composite hydrogel (QQ medium) have been successfully applied in membrane bioreactors (MBRs) for effective biofouling control. However, in order to bring QQ technology closer to practice, the physical strength and lifetime of QQ media should be improved. In this study, enforcement of physical strength, as well as an extension of the lifetime of a previously reported QQ bacteria entrapping hollow cylinder (QQ-HC), was sought by adding a dehydration procedure following the cross-linking of the polymeric hydrogel by inorganic compounds like $Ca^{2+}$ and boric acid. Such prepared medium demonstrated enhanced physical strength possibly through an increased degree of physical cross-linking. As a result, a longer lifetime of QQ-HCs was confirmed, which led to improved biofouling mitigation performance of QQ-HC in an MBR. Furthermore, QQ-HCs stored under dehydrated condition showed higher QQ activity when the storage time lasted more than 90 days owing to enhanced cell viability. In addition, the dormant QQ activity after the dehydration step could be easily restored through reactivation with real wastewater, and the reduced weight of the dehydrated media is expected to make handling and transportation of QQ media highly convenient and economical in practice.

The objective of this study was to investigate microbial removal using layered double hydroxides (LDHs) and iron (hydr)oxides (IHs) immobilized onto granular media. Column experiments were performed using calcium alginate beads (CA beads), LDHs entrapped in CA beads (LDH beads), quartz sand (QS), iron hydroxide-coated sand (IHCS) and hematite-coated sand (HCS). Microbial breakthrough curves were obtained by monitoring the effluent, with the percentage of microbial removal and collector efficiency then quantified from these curves. The results showed that the LDH beads were ineffective for the removal of the negatively-charged microbes (27.7% at 1 mM solution), even though the positively-charged LDHs were contained on the beads. The above could be related to the immobilization method, where LDH powders were immobilized inside CA beads with nano-sized pores (about 10 nm); therefore, micro-sized microbes (E. coli = 1.21 ${\mu}m$) could not diffuse through the pores to come into contact with the LDHs in the beads, but adhere only to the exterior surface of the beads via polymeric interaction. IHCS was the most effective in the microbial removal (86.0% at 1 mM solution), which could be attributed to the iron hydroxide coated onto the exterior surface of QS had a positive surface charge and, therefore, effectively attracted the negatively-charged microbes via electrostatic interactions. Meanwhile, HCS was far less effective (35.6% at 1 mM solution) than IHCS because the hematite coated onto the external surface of QS is a crystallized iron oxide with a negative surface charge. This study has helped to improve our knowledge on the potential application of functional granular media for microbial removal.
A series of experiments using transparent micromodels with an artificial pore network etched on glass plates was performed to investigate the effects of flow rate on the migration and distribution of resident wetting porewater (deionized water) and injecting non-wetting fluid (n-hexane). Multicolored images transformed from real RGB images were used to distinguish n-hexane from porewater and pore structure. Hexane flooding followed by immiscible displacement with porewater, migration through capillary fingering, preferential flow and bypassing were observed during injection experiments. The areal displacement efficiency increases as the injection of n-hexane continues until the equilibrium reaches. Experimental results showed that the areal displacement efficiency at equilibrium increases as the flow rate increases. Close observation reveals that preferential flowpaths through larger pore bodies and throats and clusters of entrapped porewater were frequently created at lower flow rate. At higher flow rate, randomly oriented forward and lateral flowpaths of n-hexane displaces more porewater at an efficiency close to stable displacement. It may resulted from that the pore pressure of n-hexane, at higher flow rate, increases fast enough to overcome capillary pressure acting on smaller pore throats as well larger ones. Experimental results in this study may provide fundamental information on migration and distribution of immiscible fluids in subsurface porous media.
