Biosynthesis and Control of Keratinase in Recalcitrant Feather-Degrading Bacillus megaterium F7-1

  • This study was performed to investigate the nutritional conditions controlling keratinase activity in Bacillus megaterium F7-1. B. megaterium F7-1 produced keratinase using chicken feather as a sole source of carbon, nitrogen and sulfur. Addition of the feather medium with glucose enhanced keratinase production (68.9 U/ml), compared to control without glucose (63.2 U/ml). The synthesis of keratinase was repressed by addition of $NH_4Cl$ in B. megaterium F7-1. The highest keratinase production (70.9 U/ml) was obtained with the feather medium containing glucose and $MgSO_4{\cdot}7H_2O$. Keratinase was produced in the absence of feather (4.9 U/ml), indicating its constitutive synthesis. Feather degradation resulted in free SH group formation. B. megaterium F7-1 effectively degraded chicken feather meal (86%), whereas duck feather, human nail, human hair and sheep wool displayed relatively low degradation rates (8-34%).

Isolation and Characterization of Terephthalic Acid-degrading Bacteria (Terephthalic Acid 분해 세균의 분리 및 특성)

  • A bacterial strain, designated T116, degrading terephthalic acid (TPA) was isolated from the soil around Taegu industrial area into which dye works wastewater flow. The isolate was identified as pseudomonas sp. based on its morphological and physiological characteristics. Degradation of TPA by the strain T116 was confirmed with UV scanning and HPLC. About 90% and 98% of TPA were degraded after 36 and 60 hours, respectively, during the culture in a liquid medium containing 0.1% TPA. Addition of KH2PO4 at a final concentration of 100ppm enhanced the chemical oxygen demand (COD) removal rate about 50% from dye works wastewater by Pseudomonas sp. T116. Optimum pH and temperature for COD reduction from wastewater were 7.0 and 3$0^{\circ}C$, respectively. The bacterium was applied to the continuous culture for the treatment of dye works wastewater whose TPA concentration and CODMn were 2,200ppm and 1,620ppm, respectively. It was observed that 90-95% of COD was eliminated after 4 days culture in the continuous culture with a retention time of 37 or 47 hours.

Super-Resolution Iris Image Restoration using Single Image for Iris Recognition

  • Iris recognition is a biometric technique which uses unique iris patterns between the pupil and sclera. The advantage of iris recognition lies in high recognition accuracy; however, for good performance, it requires the diameter of the iris to be greater than 200 pixels in an input image. So, a conventional iris system uses a camera with a costly and bulky zoom lens. To overcome this problem, we propose a new method to restore a low resolution iris image into a high resolution image using a single image. This study has three novelties compared to previous works: (i) To obtain a high resolution iris image, we only use a single iris image. This can solve the problems of conventional restoration methods with multiple images, which need considerable processing time for image capturing and registration. (ii) By using bilinear interpolation and a constrained least squares (CLS) filter based on the degradation model, we obtain a high resolution iris image with high recognition performance at fast speed. (iii) We select the optimized parameters of the CLS filter and degradation model according to the zoom factor of the image in terms of recognition accuracy. Experimental results showed that the accuracy of iris recognition was enhanced using the proposed method.

Biological Removal of Explosive 2,4,6-Trinitrotoluene by Stenotrophomonas sp. OK-5 in Bench-scale Bioreactors

  • The biological removal of 2,4,6-trinitrotoluene (TNT) was studied in a bench-scale bioreactor using a bacterial culture of strain OK-5 originally Isolated from soil samples contaminated with TNT. The TNT was completely removed within 4 days of incubation in a 2.5 L bench-scale bioreactor containing a newly developed medium. The TNT was catabolized in the presence of different supplemented carbons. Only minimal growth was observed in the killed controls and cultures that only received TNT during the incubation period. This catabolism was affected by the concentration ratio of the substrate to the biomass. The addition of various nitrogen sources produced a delayed effect for the TNT degradation. Tween 80 enhanced the degradation of TNT under these conditions. Two metabolic intermediates were detected and identified as 2-amino-4, 6-dinitrotoluene and 4-amino-2, 6-dinitrotoluene based on HPLC and GC-MS analyses, respectively. Strain OK-5 was characterized using the BIOLOG system and fatty acid profile produced by a microbial identification system equipped with a Hewlett Packard HP 5890 II gas chromatograph. As such, the bacterium was identified as a Stenotrophomonas species and designated as Stenotrophomonas sp. OK-5.

  • Lead free $(Na_{0.5}K_{0.47}Cd_x)NbO_3$ ceramics (x=0.015-0.005) were prepared by the conventional mixed oxide route. The effects of CdO addition on the sintering, dielectric and piezoelectric properties of the samples were examined. The addition of CdO greatly enhanced the sinterability of ceramics and the sample has a lower dielectric loss. The $k_p\;and\;Q_m$ of the samples with 0.02 mol CdO addition are 0.35 and 125, respectively, The $(Na,K)NbO_3$ ceramics show a large degradation of $k_p$ due to the deliquescence when exposed to humidity, while the $(Na,K,Cd)NbO_3$ ceramics have no a significant degradation of it.

  • Human plasma low density lipoprotein(LDL) is the main carrier for cholesterol, and recent studies suggest the normal LDL can be readily oxidized by free radical and not interact with LDL receptor. Lipoprotein pariticles are consisted of lipid andprotein, and fatty acids of lipoproteins are prone to oxidation. LDL particles readily undergo oxidative modification by copper. From the results, oxidized LDL altered its biological properties. A marked increase in the electrophoretic mobility of LDl on agarose gel indicated that negative surface charge of the LDL particles was increased. Also, the results from the HPLC showed that oxidized LDL was degraded into several polypeptides nonenzymatically. Degradation tests which measured the amount of 5-IAF labelled oxidized LDL were carried out by monocyte and hepatocyte cell culture. Hepatocyte cell culture of modified LDL did not show consistent pattern. However, binding rate of modified LDL with HMDM(human monocyte derived macrophage) was enhanced with oxidation, but was retarded by addition of antioxidants(hyaluronic acid, vitamin A, vitamin E). Also comparisons of oxidized-LDL, acetyl-LDL and MDA-LDL showed significant differences in the chemical properteis and binding affinity to HMDM. Thus, modificaition of normal LDL altered its biological properties.

Degradation of VOC by Photocatalysts and Dark Discharge Hybrid Systems (광촉매와 암방전(dark discharge) 복합 시스템을 이용한 VOC의 분해)

  • The immobilization technique is important to extend the application field of a photocatalyst. Titanium surface was changed into a $TiO_2$ thin film by the anodizing process. The anodized $TiO_2$ had photocatalytic activity, and showed sponge like shape. The photocatalytic degradation of gas phase acetaldehyde and VOCs by anodized titania has been studied in various initial concentrations, humidity and discharge potentials. The reactivity of anodized titania was increased with relative humidity, but excessive humidity led to a decrease of the reaction rate. The electric dark discharge that was combined with photocatalytic reaction enhanced the decomposition rate of the organic compounds. But excessively applied voltage caused corona discharge, which decreased the reaction rate. Optimum relative humidity was 40% and discharge potential was 5 kV under dark discharge region in photocatalytic reaction.

  • In this paper, the reliability (NBTI degradation: ${\Delta}V_{th}$) and device characteristic of nano-scale PMOSFET with plasma nitrided oxide (PNO) is characterized in depth by comparing those with thermally nitrided oxide (TNO). PNO case shows the reduction of gate leakage current and interface state density compared to TNO with no change of the $I_{D.sat}\;vs.\;I_{OFF}$ characteristics. Gate oxide capacitance (Cox) of PNO is larger than TNO and it increases as the N concentration increases in PNO. PNO also shows the improvement of NBTI characteristics because the nitrogen peak layer is located near the $Poly/SiO_2$ interface. However, if the nitrogen concentration in PNO oxide increases, threshold voltage degradation $({\Delta}V_{th})$ becomes more degraded by NBT stress due to the enhanced generation of the fixed oxide charges.

Effect of calcination temperature on photocatalytic activities of Er-TiO2 nanotubes

  • 0.7 mol% Er-TiO2 nanotubes were prepared using a sol-gel derived electrospinning and subsequent calcination at intervals of 50 ℃ from 500 ℃ to 650 ℃ to investigate the effect of calcination temperature on the crystal structure and the photocatalytic activity of methylene blue (MB). X-ray diffraction (XRD) results indicated that Er-TiO2 nanotubes calcined at 500 ℃ were composed of anatase only. However, mixed phases of anatase (51%, 55%, 96%) and rutile (49%, 45%, 4%) were observed for the nanotubes calcined at 550 ℃, 600 ℃ and 650 ℃, respectively. As the calcination temperature rose from 500 ℃ to 600 ℃, the Barrette-Emmett-Teller (BET) surface area and degradation kinetic constant increased from 97.77 ㎡/g to 117.62 ㎡/g and from 1.2 × 10-2min-1 to 1.6 × 10-2 min-1, respectively. The Er-TiO2 nanotubes calcined at 600 ℃ exhibited enhanced MB degradation (87%) compared to that of Er-TiO2 nanofibers (37%) due to the synergic combinations of tailored mixed crystals and larger BET area.

Photocatalytic Membrane for Degradation of Antibiotics: A Review (항생제 분해용 광촉매막: 리뷰)

  • There is evidence that the presence of active pharmaceutical ingredient (APIs) are a danger for aquatic ecosystems and the human health. The presence of APIs such as tetracycline, an antibiotic, in water causes antimicrobial resistance (AMR) in microorganisms inflicting enormous costs on individuals and society. Membranes embedded with catalysts such as TiO2 or bismuth based catalysts degrade and separate the organic effluents from wastewater. The photocatalytic activity of the catalysts can be enhanced with noble metal doping and addition of carbonaceous materials and formation of heterojunction with other semiconductors. The recollection of photoctalysts is possible through the immobilization of the photocatalysts in polymeric membranes. In this review, the degradation of antibiotics in water is discussed.
