Scaffolds of tissue engineering should be biocompatible and biodegradable for cell attachment, proliferation and differentiation. In the various scaffold fabrication, 3D printing technique can make the three dimensional scaffold with interconnected pores for cell ingrowth. Polycaprolactone (PCL) is biodegradable polyester with a low melting temperature and has been approved by the Food and Drug Administration (FDA). In this study, PCL scaffold was fabricated by 3D bioprinting system and surface modification of PCL scaffold was controlled by NaOH treatment. Morphological change and wetability of NaOH-treated scaffold were observed by SEM and contact angle measurement system. The remnant of PCL treated with NaOH was measured by ATR-FTIR. In vitro study of scaffolds was evaluated with WST-1 and ALP activity assay. NaOH treatment of PCL scaffolds increased surface roughness, hydrophilicity, cell proliferation and osteogenic differentiation. These results indicate that NaOH-treated PCL scaffold made by 3D bioprinting has tissue engineered potential for the development of biocompatible material.
Dye-sensitized solar cells (DSSC) show great promise as an inexpensive alternative to conventional p-n junction solar cells. Investigations into the various factors influencing the photovoltaic efficiency have recently been intensified. The conventional absorber electrode in DSSC is composed of compacted or sintered $TiO_2$ nanopowder that carries an anchored organic dye. The absorbance of incident light in the DSC is realized by specifically engineered dye molecules placed on the semiconductor electrode surface ($TiO_2$). The dye absorbs light at wavelengths up to about 920nm, the energy of the exited state of the molecule should be about 1.35eV above the electronic ground state corresponding to the ideal band gap of a single band gap solar cell. The dye molecules ar adhered onto the nanostrutured $TiO_2$ electrode by immersing the sintered electrode into a dye solution, typically 3mM in alcohol, for a long enough period to fully impregnate the electrode. However, the concentrations of the dye is slightly changed due to the evaporation of the alcohol. The dye is more expensive than other materials in DSSC and related to the efficiency of DSSC. Therefore, the concentrations of the dye should be carefully measured. In this study, we investigated to the dye loading on fired $TiO_2$ powder as a function of temperature by the TG-DTA and the dye solution by UV-visible spectroscopy after the impregnation process. The dye loading is related to the porosity of the nanostructured $TiO_2$ electrode.

In lithium-ion batteries, separator membrane's, main role is to physically isolate a cathode and an anode while maintaining rapid transport of ionic charge carriers during the passage of electric current. As far as battery safety is concerned, the electrical isolation of electrodes is most crucial since unexpected short-circuits across the membrane induces hot spots where thermal runaway may break out. Internal short-circuits are generally believed to occur by protrusions on the electrode surface either by unavoidable deposits of metallic impurities or by dendritic lithium growth during battery operation. Another cause is shrinkage of the separator membrane when exposed to heat. If separator membrane can be engineered to prevent the internal short-circuit, it will not be difficult to improve lithium-ion batteries' safety. Commonly the separators employed in lithium-ion batteries are made of polyethylene (PE) and/or polypropylene (PP). These materials have terrible limitations in preventing the fore-mentioned internal short-circuit between electrodes due to their poor dimensional stability and mechanical strength. In this study we have developed a novel separator membrane that possesses very high thermal and mechanical stability. The cells employing this separator provided noticeable safety improvement in the various abuse tests.

Tissue engineered bone (TEB) can replace an autogenous bone graft requiring an secondary operation site as well as avoid complications like inflammation or infection from xenogenic or synthetic bone graft. Adult mesenchymal stem cells (MSC) for TEB are considered to have various ranges of differentiation capacity or multipotency by the donor site and age. This study examined the effect of age on proliferation capacity, differentiation capacity and bone morphogenetic protein-2 (BMP-2) responsiveness of human bone marrow stromal cells (hBMSC) according to the age. In addition, to evaluate the effect on enhancement for osteoblast differentiation, the hBMSC were treated with Trichostatin A (TSA) and 5-Azacitidine (5-AZC) which was HDAC inhibitors and methyltransferase inhibitors respectively affecting chromatin remodeling temporarily and reversibly. The young and old group of hBMSC obtained from the iliac crest from total 9 healthy patients, showed similar proliferation capacity. Cell surface markers such as CD34, CD45, CD90 and CD105 showed uniform expression regardless of age. However, the young group showed more prominent transdifferentiation capacity with adipogenic differentiation. The osteoblast differentiation capacity or BMP responsiveness was low and similar between young and old group. TSA and 5-AZC showed potential for enhancing the BMP effect on osteoblast differentiation by increasing the expression level of osteogenic master gene, such as DLX5, ALP. More study will be needed to determine the positive effect of the reversible function of HDAC inhibitors or methyltransferase inhibitors on enhancing the low osteoblast differentiation capacity of hBMSC.
Purpose: The aim of this review is to introduce a novel bone-graft material for hard-tissue regeneration based on the calcium phosphate glass(CPG). Materials and Methods: CPG was synthesized by melting and subsequent quenching process in the system of CaO-$CaF_2-P_2O_5$-MgO-ZnO having a much lower Ca/P ratio than that of conventional calcium phosphates such as HA or TCP. The biodegradability and bioactivity were performed. Effects on the proliferation, calcification and mineralization of osteoblast-like cells were examined in vitro. Influence in new bone and cementum formations was investigated in vivo using calvarial defects of Sprague-Dawley rats as well as 1-wall intrabony defect of beagle dogs. The application to the tissue-engineered macroporous scaffold and in vitro and in vivo tests was explored. Results: The extent of dissolution decreased with increasing Ca/P ratio. Exposure to either simulated body fluid or fetal bovine serum caused precipitation on the surface. The calcification and mineralization of osteoblast-like cells were enhanced by CPG. CPG promoted new bone and cementum formation in the calvarial defect of Sprague-Dawley rats after 8 weeks. The macroporous scaffolds can be fabricated with $500{\sim}800{\mu}m$ of pore size and a three-dimensionally interconnected open pore system. The stem cells were seeded continuously proliferated in CPG scaffold. Extracellular matrix and the osteocalcin were observed at the $2^{nd}$ days and $4^{th}$ week. A significant difference in new bone and cementum formations was observed in vivo (p<0.05). Conclusion: The novel calcium phosphate glass may play an integral role as potential biomaterial for regeneration of new bone and cementum.
Ligand-receptor clustering event is the most important step in leukocyte adhesion and spreading on endothelial cells. Intercellular adhesion molecule-1 (ICAM-1) has been shown to enhance leukocyte adhesion, but the molecular event during the process of adhesion is unclear. To visualize the dynamics of ICAM-1 movement during adhesion, we have engineered stable Chinese hamster ovary cell lines expressing ICAM-1 fused to a green fluorescent protein (IC1_GFP/CHO) and examined them under the fluorescence microscopy. The transfection of IC1_GFP alone in these cells was sufficient to support the adhesion of K562 cells that express $\alpha$L$\beta$2 (LFA-1) integrin stimulated by CBR LFA-1/2 mAb. This phenomenon was mediated by ICAM-1-LFA-1 interactions, as an mAb that specifically inhibits ICAM-1-LFA-1 interaction (RRl/l) completely abolished the adhesion of LFA-1* cells to IC1_ GFP/CHO cells. We found that the characteristic of adhesion was followed almost immediately (~10 min) by the rapid accumulation of ICAM-1 on CHO cells at a tight interface between the two cells. Interestingly, ICI_GFP/CHO cells projected plasma membrane and encircled approximately half surface of LFA-1+ cells, as defined by confocal microscopy. This unusual phenomenon was also confirmed on HUVEC after adhesion of LFA-1* cells. Neither cytochalasin D nor 2,3-butanedione 2-monoxime an inhibitor of myosin light chain kinase blocked LFA-1-mediated ICAM-1 clustering, indicating that actin cytoskeleton and myosin-dependent contractility are not necessary for ICAM-1 clustering. Taken together, we suggest that leukocyte adhesion to endothelial cells induces specialized form of ICAM-1 clustering that is distinct from immunological synapse mediated by T cell interaction with antigen presenting cells.

Fusion technology based on the nanotechnology should be introduced to clean technology for the breakthrough advances. Today, nanoparticles, nanotubes and other engineered nanomaterials are already in use in hundreds of everyday consumer products, and these materials are able to move around the human and environmental mediamore readily than larger particles of pollution. Because of their extremely small size and large surface area, nanoparticles are known to be more reactive and toxic than larger particles. Consequently, this feature raised many concerns of significant health, safety and environment. Herein, we reviewed risk assessment for health and environmental hazards of nanomaterials, and then revealed the potential hazardous of nanomaterials.

This study was conducted to predict and evaluate the uncertainty of safety after closure of the second phase surface disposal facility of the Gyeongju intermediate and low level repository in Korea. In this study, four scenarios are developed considering both intact and degraded states of multi-layered covers and disposal containers; also, the fluid flow by a rainfall into the disposal facility is simulated. The rainfall conditions are implemented based on the monthly average data of the past 30 years (1985~2014); the simulation period is 300 years, the management period regulated by institutional provisions. As a result of the evaluation of the basic scenario, in which the integrity of both of the containers and the covers is maintained, it was confirmed that penetration of rainfall does not completely saturate the inside of the disposal facility. It is revealed that the multiple cover layers and concrete containers effectively play the role of barrier against the permeation of rainfall.

Although timber structures have been extensively used in underground temporary supporting system, their actual performance is poorly understood, resulting in potentially conservative and over-engineered design. In this paper, a novel wireless sensor technology, SmartPlank, is introduced to monitor the field performance of timber structures during underground construction. It consists of a wooden beam equipped with a streamlined wireless sensor node, two thin foil strain gauges and two temperature sensors, which enables to measure the strain and temperature at two sides of the beam, and to transmit this information in real-time over an IPv6 (6LowPan) multi-hop wireless mesh network and Internet. Four SmartPlanks were deployed at the London Underground's Tottenham Court Road (TCR) station redevelopment site during the Stair 14 excavation, together with seven relay nodes and a gateway. The monitoring started from August 2013, and will last for one and a half years until the Central Line possession in 2015. This paper reports both the short-term and long-term performances of the monitored timber structures. The grouting effect on the short-term performance of timber structures is highlighted; the grout injection process creates a large downward pressure on the top surface of the SmartPlank. The short and long term earth pressures applied to the monitored structures are estimated from the measured strains, and the estimated values are compared to the design loads.
Selecting an appropriate antigen with optimal immunogenicity and physicochemical properties is a pivotal factor to develop a protein based subunit vaccine. Despite rapid progress in modern molecular cloning and recombinant protein technology, there remains a huge challenge for purifying and using protein antigens rich in hydrophobic domains, such as membrane associated proteins. To overcome current limitations using hydrophobic proteins as vaccine antigens, we adopted in silico analyses which included bioinformatic prediction and sequence-based protein 3D structure modeling, to develop a novel periodontitis subunit vaccine against the outer membrane protein FomA of Fusobacterium nucleatum. To generate an optimal antigen candidate, we predicted hydrophilicity and B cell epitope parameter by querying to web-based databases, and designed a truncated FomA (tFomA) candidate with better solubility and preserved B cell epitopes. The truncated recombinant protein was engineered to expose epitopes on the surface through simulating amino acid sequence-based 3D folding in aqueous environment. The recombinant tFomA was further expressed and purified, and its immunological properties were evaluated. In the mice intranasal vaccination study, tFomA significantly induced antigen-specific IgG and sIgA responses in both systemic and oral-mucosal compartments, respectively. Our results testify that intelligent in silico designing of antigens provide amenable vaccine epitopes from hard-to-manufacture hydrophobic domain rich microbial antigens.
