In this study, we cultured bovine capillary endothelial cells from adrenal cortex and compared these cells with capillary endothelial cells obtained from bovine adrenal medulla on morphological and cytokinetic properties. We demonstrated that bFGF and gelatin matrix were required for the growth of adrenal cortex-derived capillary endothelial cells over middle passage, but not for the growth of adrenal medulla-derived capillary endothelial cells. Also, we showed that the growth of adrenal cortex-derived capillary endothelial cells must be stimulated by bFGF and the gelatin matrix for the measurement of in vitro angiostatin activity. These data indicate that adrenal cortex-derived capillary endothelial cells over middle passage are more suitable than adrenal medulla-derived capillary endothelial cells for in vitro angiogenesis assay.
Histopathological vascular changes in hemorrhagic fever with renal syndrome (HFRS) caused by Hantaan virus include increased vascular permeability, disseminated intravascular coagulation, thrombocytopenia and changes in coagulation activity. Although vascular endothelial cells of main target organs such as kidney infected with Hantaan virus are not damaged but swelling of endothelial cells, perivascular exudates and infiltration of mononuclear cells and fresh interstitial hemorrhages are common. However, the pathogenesis of cell infiltration and hemorrhages around vascular endothelial cells are not well understood. Some endothelial cell molecules or vascular adhesins that acts as adhesion moleulces for leukocyte are expressed on endothelial cells close to site of inflammation. However, whether the expression of endothelial adhesion molecules such as vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule (ICAM-1) and endothelial leukocyte adhesion molecule (ELAM) on vascular endothelial cells are increased by infection with Hantaan virus has not been studied. In this study, the relationship between the expression of VCAM-1, ICAM-1 and ELAM and adhesion of mononuclear cells on endothelial cells of human blood vessels infected with Hantaan virus was investigated. The endothelial cells of umbilical vein was passaged three times in culture medium and the monolayered cells were infected with $10^5\;pfu/ml$ of Hantaan virus grown in Vera E6 cell cultures. The multiplication of virus in cultured endothelial cells was monitored by immunohistochemistry and the expression of adhesion molecules was demonstrated by immunohistochemistry using monoclonal antibodies against VCAM-1, ICAM-1 and ELAM. And in situ hybriditation against ICAM-1 was also performed. The endothelial adhesion molecules, VCAM and ICAM, were expressed after 6 hours postinfection, respectively, and their expressions lasted for 72 hours. Similar expression of VCAM and ICAM appeared on endothelial cells by infection with virus, but the expression of ELAM was not recognized up to 72 hours postinfection. Microscopically, it was noted that many monocuclear cells adhered on endothelial cells infected with viruses. In an electronmicroscopic study, the transendothelial migration of mononuclear cells was observed on monolayered endothelial cells infected with virus. This results suggested that the endothelial adhesion molecules, particulary VCAM and ICAM, might be expressed on endothelial cells by infection with Hantaan virus and these molecules play a key role in the adhesion and extravasation of inflammatory cells around blood vessels.
Synthetic and biosynthetic vascular grafts of small diameter have long been considered to be prone to thrombosis, ultimately leading to the complete graft occlusion. Endothelial cell seeding onto synthetic blood-contacting surfaces has been suggested to be an ideal means to solve this problem. This study described a culture method of bovine endothelial cells and evaluated blood-compatibility and seeding efficiency of cultured endothelial cells. Bovine pulmonary artery endothelial cells were harvested enzymatically and grown to confluence on polystyrene culture flask surfaces using established techniques. The identification of endothelial cells was made through the demonstration of expression of factor VIII R:Ag by immunofluorescent technique. To quantitate the effect of improvement in blood-compatibility of viable endothelial cells, endothelial monolayers were exposed to blood containing $\^$111/In-oxine labeled platelets. Viable endothelial monolayers retained less labeled platelets than control surfaces. The Indium-labeled endothelial cells were seeded onto three different blood-contacting surfaces of Dacron vascular graft immobilized in specially equipped wells and incubated for specific time intervals (t=15, 30, 60, 120 minutes). Longer incubation times showed improved cell adherence in collagen-coated and fibrin-coated Dacron vascular graft groups. However in untreated Dacron grafts, no direct relationship was observed between incubation time and endothelial cell seeding efficiency. This may be due to leakage of endothelial cells through porosity of Dacron grafts in this in-vitro experimental condition.
It is important to coordinated interaction among neurons, astrocytes and endothelial cells to maintain the function of brain. To study their regulatory mechanisms in vitro system, the co-culture system among the isolated cells from brain may be needed. However, the method for purifying brain microvascular endothelial cells (BMEC) far culture have not established yet. In this study, the proper culture methods of mice cells using two different strains, CD1 and C57BL6, to obtain the pure and plentiful endothelial cells were described. The flatted-round forms of CD1 endothelial cells grew on the collagen-IV coating plates, while the purified cells from C57 mice preferred type collagen-I dishes for their growth. Both cells displayed anti-PECAM-1 (CD31) and von Willebrand Factor immune-reactivity. These results indicated that different coating materials not only improve attachment of isolated cells but also promoting growth of cells, suggesting that this method of purifying murine Brain microvascular endothelial cells (BMEC) provides a suitable model to investigate blood-brain-barrier (BBB) properties within neurovascular unit in vitro.
The objective of this study is to investigate the effects of the hemodynamics on the morphological changes of the human endothelial cells due to the blood flow by in vitro experiment and computer simulation. The morphological changes of the endothelial cells due to the t10w shear stress were observed in the laminar t10w chamber as a function of the exposure time. The observed shapes of the endothelial cells are used to the model shapes of the endothelial cells for numerical study and the pressure and the wall shear stress variations around the endothelial cells are calculated from the numerical results. The endothelial cells elongate along the t10w direction and lessen their heights in the flow field to reduce the pressure and the wall shear stress on the surface.
The effects of tumor necrosis factor alpha $(TNF-{\alpha})$ on growth and tubular formation of bovine aortic endothelial cells were examined using an in vitro angiogenesis model system. The growth of endothelial cells was enhanced in a dose-dependent manner when the cells were cultured with $TNF-{\alpha}$ for 3 days, but $TNF-{\alpha}$, at the concentration of 1 nM or higher, produced a growth inhibition of endothelial cells when the cells were cultured for 8 days. The endothelial cells incubated with $TNF-{\alpha}$ for 48-h exhibited a typical morphologic change. Then, they showed a fibroblastoid organization of overlapping, elongated, and spindle-shaped cells. $TNF-{\alpha}$, at the concentration of O. 1 nM or higher, inhibited the tubular formation of vascular endothelial cells in an in vitro anglogenesis model using a 3-dimensional culture system.
To investigate the effects of hydrocortisone on new-born rat cardiac endothelial cells in culture, the endothelial cells were isolated by means of enzyme-cocktail method. The cells were cultivated in Lees modified Dulbeco\ulcorner medium and 10[M or 10[M of hydrocortisone was added to the medium. The cells were harvested or coverglass and processed for thiamin pyrophosphatase reaction and Feulgen reaction. The enzymatic activities of Golgi complex, number of cells and number of large nucleated[more than tetraploid] cells were counted and discussed for their significance. The results were summarized as follows; 1. Hydrocortisone seemed to accelerate the rate of recovery of cardiac endothelial cells from isolation damage. 2. Endothelial cells treated with hydrocortisone revealed strong positive reaction to thiamine pyrophosphatase in early culture and 10 M group had stronger reaction than that of 10 AM group 3. Hydrocortisone had inhibiting effects on endothelial proliferation and the higher the concentration of the reagent was the stronger effects. 4. Hydrocortisone inhibited the appearance of large nucleate cells in endothelial cell population. 5. Hydrocortisone seemed to suppress the nuclear DNA synthesis.
Construction of the stable monolayer of endothelial cells onto physicochemically modified polymeric surFace is one of the appropriate method to develop the small caliber vascular graft with the long-term patency. In this study, we constructed the monolayer of endothelial cells on the fibronectin rind the extracellular matrix-coated polyurethane surface derived from human fibroblast cells. To elucidate the adhesion strength of endothelial cells on the extracellular matrix-coated polyurethane, a laminar flow chamber apparatus was developed to exposure the shear stress on the apical membrane of ondothelial cells. Endothelial cells show the strongest adhesion after two days of seeding onto the fibronectin-coated polyurethane surface, whereas endothelial cells on the extracellular matrix derived from the human flbroblast cells show the minimal doubling time of cellular growth.
Background: Platelets take part in repairing the lesions of endothelial damage. To understand the molecular mechanism of this process, we tested the hypothesis that CD154 expressed on activated platelets stimulates proliferation of human endothelial cells. Methods: The expression levels of CD154 and CD40 on platelets and endothelial cells, respectively, were measured by flow cytometry and confocal microscopy. Function-blocking monoclonal antibody against CD154 was developed after immunization with CD154-transfected L cells. Results: An anti-CD40 agonist antibody and soluble CD154 both induced significant proliferation of endothelial cells. In addition, a function-blocking anti-CD154 antibody inhibited the platelet-induced proliferation of endothelial cells, indicating that the CD154-CD40 pathway is involved in these cellular interactions. An anti-VEGF antibody failed to inhibit the proliferation. This, in addition to the fact that very small amounts of VEGF are released from platelets or endothelial cells, suggests that VEGF does not play an important role in the platelet-stimulated proliferation of endothelial cells. Conclusion: Our results indicate that platelets induce proliferation of endothelial cells by CD154-CD40 interactions independently of VEGF.
We have previously observed that Bambusae Caul is in Liquamen (BCL) stimulates the adipose conversion of 3T3-L1 cells and molecular chaperones were involved in the process of the assembly and replacement of laminin subunits in Bovine aortic endothelial cells(BAEC). Endothelial cells are exposed to continuous shear stress due to the blood flow. Heat shock protens(hsp) are a well-known stress response protein, namely, stress proteins. To investigate effects of BCL on the stress proteins induced by heating in endothelial cells, we have analyzed synthetic amounts of stress proteins in sodium dodecyl sulfate gel electrophoresis under reducing conditions. Under the condition of heating stress, BCL inhibited the synthesis of stress proteins in endothelial cells. These results suggest that BCL may have an important role for expression of stress proteins induced by heating in endothelial cells.
