Diurnal Variation in Endogenous Gibberellin Levels of Rice Shoots

  • Diurnal changes in levels of endogenous gibberellins(GAs) were investigated in three rice cultivars i.e. Sangjubyeo, Shingeumobyeo(photo-neutral) and Chucheongbyeo(photosensitive). The rice cultivars were grown under a 12-hr photoperiod and endogenous GA levels were assayed by gas chromatography-mass spectrometry(GC-MS-SIM) every 3 h for 24 h. The endogenous bioactive $GA_1$ and its immediate precursor $GA_{20}$ contents were significantly different in both photosensitive and photo-neutral rice cultivars, though less pronounced differences were observed for endogenous $GA_{12},\;GA_{53},\;GA_{19}$, and $GA_8$ levels with in the three rice cultivars. The levels of bioactive $GA_1$ and its immediate precursor $GA_{20}$ were significantly higher in Chucheongbyeo than in the other two cultivars. In Chucheongbyeo, the $GA_1$ contents increased significantly from 11.00 to 17.00 o'clock, thus indicating a correlation with light. In Shingeumobyeo, $GA_1$ contents slightly increased during morning hours, while a similar hike in $GA_1$ contents was observed for Sangjubyeo during evening hours. $GA_{19}$ was found to be the most abundant GA form in the three rice cultivars. Our results suggested that GA production in rice depends upon the response potential of rice cultivars and that light positively correlated to GA production in photosensitive rice cultivar.

Gibberellins-Producing Rhizobacteria Increase Endogenous Gibberellins Content and Promote Growth of Red Peppers

  • The growth of red pepper plants was enhanced by treatment with the rhizobacterium, Bacillus cereus MJ-1. Red pepper shoots showed a 1.38-fold increase in fresh weight (fw) and roots showed a 1.28-fold fw gain. This plant growth-promoting rhizobacterium (PGPR) has been reported to produce gibberellins (GAs). Other GAs-producing rhizobacteria, Bacillus macroides CJ-29 and Bacillus pumilus CJ-69, also enhanced the fw of the plants. They were less effective than B. cereus MJ-1, though. The endogenous GAs content of pepper shoots inoculated with MJ-1 was also higher than in shoots inoculated with CJ-29 or CJ-69. When inoculated with MJ-1, bacterial colonization rate of the roots was higher than that of roots inoculated with CJ-29 or CJ-69. These results support the idea that the plant growth-promoting effect of the bacteria also positively related with the efficiency of root colonization by the bacteria. In addition, we identified the major endogenous GAs of the red pepper as originating from both the early C-13 hydroxylation and the early non C-13 hydroxylation pathways, with the latter being the predominant pathway of GA biosynthesis in red pepper shoots.

Growth and Gibberellins level of Two Rice Cultivars as Influenced by Different Nitrogen Containing Compounds

  • Seedlings of two rice cultivars i.e. cv. Daesanbyeo and cv. Dongjinbyeo were analyzed for growth and endogenous gibberellins (GAs) in response to nitrogen nutrition applied in the forms of $KNO_3$, $(NH_4)_2SO_4$ and $NH_4NO_3$. All the growth parameters showed an increase in N applied treatments and their magnitudes of increase were different depending on different nitrogen fertilizer forms. The endogenous GAs contents were increased with N application but differentially affected by various N-forms in both rice cultivars. In cv. Daesanbyeo, maximum amount of bioactive $GA_1$ was recorded for $(NH_4)_2SO_4$, while maximum amount of bioactive $GA_1$ in cv. Dongjinbyeo was observed in $NH_4NO_3$ applied treatments. In both rice cultivars, $KNO_3$ applied rice plants contained least $GA_1$ contents. Also, $GA_{19}$ was the most abundant GA found in rice seedlings whereas $GA_4$ was absent at seedling stage. Our study indicated that different rice cultivars showed different responses for the same fertilizer depending upon the response potential of each cultivar and the pertinent physiological responses to changes of endogenous GAs in rice cultivars, which were comparatively lesser in magnitude.

  • The misuse of anabolic androgenic steroids is of particular concern in sports and society. Thus, it is of great importance to discriminate endogenous steroids such as testosterone or testosterone prohormones from their chemically identical synthetic copies. In this study, gas chromatography-combustion/isotope ratio mass spectrometric (GC-C/IRMS) method has been developed and validated for discriminating the origin of anabolic androgenic steroids. The method involves the solid-phase extraction, enzymatic hydrolysis with ${\beta}$-glucuronidase, HPLC-fractionation for the cleanup and analysis by GC-C/IRMS. The difference(${\Delta}^{13}C$) of urinary ${\delta}^{13}C$ values between synthetic analogues and endogenous reference compounds (ERC) by GC-C/IRMS was used to elucidate the origin of steroids, and intra- and inter-day precision, specificity and isotope fractionation were evaluated. The present GC-C/IRMS method combined with HPLC cleanup was accurate and reproducible enough to be successfully applied to the test of urine sample from suspected anabolic steroid abusers.

  • Sulfur dioxide (SO2), a novel endogenous gas signaling molecule, is involved in the regulation of cardiac function. Exerting a key role in progression of hyperthyroidism-induced cardiomyopathy (HTC), myocardial fibrosis is mainly caused by myocardial apoptosis, leading to poor treatment outcomes and prognoses. This study aimed to investigate the effect of SO2 on the hyperthyroidism-induced myocardial fibrosis and the underlying regulatory mechanisms. Elisa, Masson staining, Western-Blot, transmission electron microscope, and immunofluorescence were employed to evaluate the myocardial interstitial collagen deposition, endoplasmic reticulum stress (ERS), apoptosis, changes in endogenous SO2, and Hippo pathways from in vitro and in vivo experiments. The study results indicated that the hyperthyroidism-induced myocardial fibrosis was accompanied by decreased cardiac function, and down-regulated ERS, apoptosis, and endogenous SO2-producing enzyme aspartate aminotransferase (AAT)1/2 in cardiac myocytes. In contrast, exogenous SO2 donors improved cardiac function, reduced myocardial interstitial collagen deposition, up-regulated AAT1/2, antagonized ERS and apoptosis, and inhibited excessive activation of Hippo pathway in hyperthyroid rats. In conclusion, the results herein suggested that SO2 inhibited the overactivation of the Hippo pathway, antagonized ERS and apoptosis, and alleviated myocardial fibrosis in hyperthyroid rats. Therefore, this study was expected to identify intervention targets and new strategies for prevention and treatment of HTC.

Effect of Growth Retardant BX-112 on Growth, Floral Initiation, and Endogenous GA Levels in Sorghum

  • To define the relations between endogenous GA levels and growth and flowering in short-day plant sorghum, growth retardant BX-112 was applied to two sorghum genotypes, wild-type and phytochrome B mutant (phyB-1), which grows faster and flowers earlier than the wild-type. BX-112 and $GA_3$ were applied as a soil drench, and plant height, culm length, and date to floral initiation were investigated. Endogenous GAs contents were measured with GC-MS-SIM. BX-112 treatments inhibited shoot growth in both genotypes and drastically reduced $GA_1$ and $GA_8$ levels. With increasing BX-112 concentrations, $GA_1$ concentrations declined linearly, but caused the accumulation of intermediates from $GA_12$ to $GA_20$. This result implies that $GA_1$ is the major active endogenous GA in shoot elongation in a short day plant sorghum. The inhibition of plant growth in both of wild type and phyB-1 by BX-112 was very similar, while BX-112 effects on floral initiation in two types of plants differed significantly. Floral initiation of phyB-1 was not affected by BX-1l2, but that of wild-type was delayed as BX-1l2 concentration increased. Because BX-112 treatment causes accumulation of biosynthetic intermediates between synthetic pathway from $GA_12$ to $GA_20$ and because phyB-1 is altered in GA metabolism in this same region of the early C13-hydroxylation pathway, BX-112 may fail to block flowering of phyB-1.

Studies on the Effect of Silicon Nutrition on Plant Growth, Mineral Contents and Endogenous Bioactive Gibberellins of Three Rice Cultivars

  • Silicon is one of the key elements for healthy growth and development in rice crops. We analyzed the effect of silicon(Si) on some growth parameters, plant mineral contents, and bioactive gibberellins in three rice cultivars. Silicon was applied at the rates of 0 kg/0.1ha(control), 40 kg/0.1ha, and 80 kg/0.1ha throughout the course of experiment. Plant growth parameters were enhanced by the application of elevated Si, though plant height and culm length were more favorably affected than the respective dry weights. The plant mineral contents analyzed also increased in treatments where Si was applied without potassium, demonstrating that Si application promotes the absorption of these minerals in rice crops. The endogenous gibberellins measured in our study showed that $GA_1$ is the only bioactive GA form present in rice seedlings. The endogenous $GA_1$ and its precursor $GA_{20}$ contents increased after Si application. However, this increase in endogenous $GA_1$ and $GA_{20}$ contents, and plant growth parameters were different according to the rice cultivars. Our results indicate that Si is a beneficial element in rice nutrition and that different cultivars of Oryza sativa show differential responses to Si nutrition in terms of their growth and development.

Efficiency analysis in the presence of network effect with DEA method (네트워크 효과를 고려한 천연가스산업의 기술적 효율성 분석)

  • This study takes an issue of efficiency analysis in the presence of network effect utilizing the DEA (Data Envelopment Analysis) framework. Network effect has important policy implication for the regulation of local monopolies which undertake their business through physical network, such as electricity, natural gas, local telephony, etc. If the difference in spatial condition between companies is not controlled properly, the performance comparison and associated incentive regulation bear significant bias. In this study, we propose a methodology to measure the true managerial or technical efficiency apart from efficiency difference accruing from the difference in spatial condition. A series of modified DEA efficiency models are combined to investigate the extent of exogenous and endogenous efficiency component in the Korean natural gas distribution companies. Empirical results show that the network effect plays significant role in determining superficial performance difference.

Studies on resveratrol and its metabolite in human urine by GC/MS (GC/MS를 이용한 요 중 resveratrol과 그 대사체에 관한 연구)

  • This study was conducted to define metabolite of the resveratrol by gas chromatography- time-offlight mass spectrometric detection. From these results, we suppose that the structure of metabolite is the result of reduction of double-bond attached by two-phenyl groups. Also, validity of method for determining metabolite of resveratrol and endogenous steroids was tested. The recoveries ranged from 96.47 to 114.74%, and intraand inter-day precision ranged 11.40 - 10.87% and 1.10 - 10.93%, accuracy ranged 80.03 - 119.92% and 80.02 - 119.56%, respectively. Resveratrol and endogenous steroids had correlation coefficients above or equal to 0.996. The method was successfully validated for the determination of resveratrol and endogenous steroids. Urinary samples from volunteers dosed resveratrol were analyzed to confirm a correlation resveratrol and its metabolite. From these results, the highest level of resveratrol and its metabolite was excreted in 10 - 15 hr more slowly than common drug, and conversion rate of metabolite was higher in woman than that in man. In addition, endogenous steroids were shown same the highest level of 10 - 15 hr. For estrone and estradiol, sensitivity was relatively higher in female than in man. And there were no significant changes of excretion patterns in the other endogenous steroids. Thus, we assumed that activation of resveratrol has impact on woman than man.

Changes in Endogenous Gibberellin Contents during Bulb Development Period in the Cold-type Cultivar of Garlic (Allium sativum L.) of Korea (한지형 마늘의 인경 발육 과정에서 내생 지베렐린류의 함량변화)

  • This study was performed to investigate the role of phytohormones in the bulbing of garlic in order to assess the yield and quality. The effect on endogenous plant hormones such as gibberellin (GA) content was also examined during growth stage i.e. clove differentiation to bulbing in garlic. More than 18 gibberellins in garlic were identified with extensive gas chromatograph-mass spectrometry-selected ion monitoring (GC-MS-SIM) quantitative analysis. The results showed that GAs were biosynthesized by both non C-13 hydroxylation pathway (NCH) and early C-13 hydroxylation pathway (ECH) in garlic plant. It was also revealed that NCH pathway leading to synthesis of bioactive $GA_4$ was the more prominent GA biosynthesis pathway than ECH pathway in which bioactive $GA_1$ was synthesized. Total GAs level was gradually increased from clove differentiation to bulbing and later decreased, which portrays the active role of GA in differentiation. The biosynthesis ratio of bioactive $GA_4$ and $GA_1$ concentration was similar to that of total GAs content, which was closely related with bulb development in garlic.
