This study compared the physicochemical properties of soybean curd residue and black soybean curd residue produced by hot air-drying and freeze-drying. Regardless of drying method, the crude protein, crude ash, crude fiber contents, pH, L, a, b color values and water soluble index were higher in soybean curd residue, whereas total polyphenol contents and antioxidant activity were higher in black soybean curd residue. Significant differences in water absorption index, oil absorption capacity and emulsion activity were observed between soybean curd residue and black soybean curd residue in freeze-drying. On the other hand, the emulsion stability was not significant difference in both hot-air drying and freeze-drying. The crude protein and crude fiber contents of soybean curd residue were not significant difference between hot-air drying and freeze-drying. Freeze-drying resulted in higher crude ash contents, pH, water absorption index, water soluble index, oil absorption capacity, emulsion activity and emulsion stability than hot-air drying. Hot-air drying have caused significantly higher water contents, water activity, total polyphenol contents and antioxidant activity in soybean curd residue than freeze-drying. In conclusion, soybean type and drying methods affect the physicochemical and quality characteristics of soybean curd residue, which could be important factors in the manufacture of processed foods.

In order to endow with new bioactive functionality from demineralized bone particle (DBP) as natural source to poly(L-lactide) (PLA) synthetic biodegradable polymer, porous DBP/PLA as natural/synthetic composite scaffolds were prepared and compared by means of the emulsion freeze drying and solvent casting/salt leaching methods for the possibility of the application of tissue engineered bone and cartilage. For the emulsion freeze drying method, it was observed that the pore size decreased in the order of 79$\mu\textrm{m}$ (PLA control) > 47$\mu\textrm{m}$ (20% of DBP) > 23 $\mu\textrm{m}$ (40% of DBP) > 15$\mu\textrm{m}$ (80% of DBP). Porosities as well as specific pore areas decreased with increasing the amount of DBR. It can be explained that DBP acts like emulsifier resulting in stabilizing water droplet in emulsion. For the solvent casting/salt leaching method, a uniform distribution of well interconnected pores from the surface to core region were observed the pore size of 80 ∼70 $\mu\textrm{m}$ independent with DBP amount. Porosities as well as specific pore areas also were almost same. For pore size distribution by the mercury intrusion porosimeter analysis between the two methods, the pore size distribution of the emulsion freeze drying method was broader than that of the solvent casting/salt leaching method due to the mechanism of emulsion formation. Scaffolds of PLA alone, DBP/PLA of 40 and 80%, and DBP powder were implanted on the back of athymic nude mouse to observe the effect of DBP on the induction of cells proliferation by hematoxylin and eosin staining for 8 weeks. It was observed that the effect of DBP/PLA scaffolds on bone induction are stronger than PLA scaffolds, even though the bone induction effect of DBP/PLA scaffold might be lowered than only DBP powder, that is to say, in the order of DBP only > DBP/PLA scaffolds of 40 and 80% DBP > PLA scaffolds only for osteoinduction activity. In conclusion, it seems that DBP plays an important role for bone induction in DBP/PLA scaffolds for the application of tissue engineering area.
몇 가지 키토산 비드들을 W/O 에멀젼법과 capillary extrusion법에 의해 제조하여, SEM 사진을 통해 표면 구조와 표면적의 차이를 관찰하였고, BET법으로 표면적을 측정하였으며, 피크린산 적정에 의해 유효 아민함량을 측정하여 상기 결과들과의 상관관계를 알아 보았다. W/O 에멀젼법에 의해 제조한 비드의 경우 capillary extrusion법에 의해 제조된 비드들보다 입자의 크기가 작았으며, 반대로 표면적, 유효 아민함량은 높게 나타났다. 또한 비드의 표면적, 유효 아민함량, 겉보기 밀도(apparent density) 및 기계적 강도(hardness)는 제조시 건조방법에 따라 매우 큰 편차를 나타내었다. Hardness 측정결과 solvent dehydration법에 의해 건조된 비드들이 동결건조법에 의해 건조된 비드들에 비해 높은 강도를 나타내었다. 또한 겉보기 밀도에 있어서도 solvent dehydration한 비드들이 동결건조한 비드들에 비해 높은 겉보기 밀도를 보였다. SEM 사진 분석결과 solvent dehydration법에 의한 비드들은 매우 작은 pore들이 비드의 표면에 무수히 존재한 반면 동결건조한 비드에서는 벌집모양의 pore들이 존재하고 있음을 알 수 있었다. 피크린산 적정결과 solvent dehydration한 비드의 경우 동결건조한 비드보다 약 10배의 높은 유효 아민함량을 나타내었으며 비드의 표면적이 클수록 유효 아민함랑은 높게 나타났다. 또한 fluorescamine을 반응시킨 후 형광현미경으로 관찰하여 비드의 표면과 내부의 pore 주위에 노출된 1차 아민의 존재를 정성적으로 확인할 수 있었다.
Solid lipid nanoparticles (SLNs) have been regarded to behave similar to the vegetable oil emulsions because emulsions of lipid melts are formed before lipid droplets being solidified to turn into SLNs. Compared to lipid emulsion, however, it has been more difficult to obtain stable SLNs and needs more extensive considerations on stabilizer and manufacturing process. In the present study, we tried to prepare phosphatidylcholine-based trymyristin (TM) SLNs using high pressure homogenization method and optimize the manufacturing variables such as homogenization pressure, number of homogenization cycles, cooling temperature, co-stabilizer and freeze-drying with cryoprotectants. Nano-sized TM particles could be Prepared using egg Phosphatidylcholine and pegylated phospholipids ($PEG_{2000}$PE) as stabilizers. Based on the optimization study, the dispersion was manufactured by homogenization under the pressure of 100 MPa for more than 5 cycles, and solidifying the intermediately formed lipid melt droplets by dipping in liquid nitrogen followed by thawing at room temperature. In addition, TM SLNs could be freeze-dried and then redispersed easily without significant particle size changes after freeze drying with 10% and 12.5% sucrose or trehalose. The TM SLNs established in this study can be used as delivery system for drugs and cosmetics.
본 연구에서는 세포 배양시 감염방지용으로 널리 쓰이는 겐타마이신 설페이트(GS)가 서방화된 다공성 PLLA 지지체를 유화동결건조방법으로 제조하였다 이들의 물성을 전자현미경 및 수은다공도계로 특성결정하였고, 적심성은 푸른염색 수용액으로 관찰하였으며, 방출거동은 high performance liquid chromatography (HPLC)를 이용하여 측정하였다. GS가 5, 10 및 20%가 포접된 PLLA 지지체의 다공도는 80~90%이었으며 평균다공크기 범위는 30~57 ${\mu}{\textrm}{m}$ 그리고 가장 큰 것으로는 150${\mu}{\textrm}{m}$이상의 것도 관찰되었다. 전체적인 다공도의 모양은 다공과 다공사이의 연결이 양호하고 대부분이 열린 셀 구조를 하고 있는 것으로 나타났다. 대조군에 비해 GS의 농도가 증가함에 따라 PLLA 지지체의 다공도가 감소하는 것으로 보아 GS내의 설페이트 부분은 친수성 역할 그리고 겐타마이신 부분은 소수성 역할을 수행하는 계면활성제의 역할에 기인한 것으로 사료된다. PLLA 지지체의 방출거동은 GS의 농도가 증가함에 따라 방출되는 양이 증가하였고, 적심성 또한 향상되어 세포배양시 긍정적 효과를 끼칠 것으로 예상되었다.
조직의 재생을 위한 지지체재료의 개발은 조직공학 분야의 연구에 있어서 매우 중요한 요인이다. 히알루론산은 조직을 수복하기 위한 체내이식용 구조물로써 널리 사용되고 있는 천연고분자이며, 이를 이용하여 본 연구에서는 히알루론산을 함유한 락타이드-글리콜라이드 공중합체(PLGA) 다공성 지지체를 유화동결 건조법으로 제조하였다. HA-PLGA 지지체는 수은다공도계, 전자현미경 및 물흡수성을 측정하여 특성을 결정하였다. 제조된 HA-PLGA 지지체의 다공도는 약 96.5%, 전체다공면적은 $261\;m^2/g$ 이였으며, 세포가 성장하기에 적합한 환경인 다공사이의 상호 연결이 전자주사현미경을 통해 관찰되었다. 또한 세포의 생존율과 성장률은 MTT(3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide) 방법을 이용하여 분석하였고, 사이토카인 및 수용성 약물의 방출경향을 확인하기 위하여 과립구-대식세포 집락자극인자(GM-CSF)를 지지체에 담지시킨 후 효소결합 면혁 흡착검사(ELISA)를 이용하여 이들의 방출경향을 확인하였다. 천연/합성 하이브리드 담체로서의 HA/PLGA 담체가 PLGA 단독으로 사용하였을 때와 비교하여 볼 때 세포의 증식이 우수하였고, 이는 히알루론산의 우수한 생체적합성 및 수분 보유능력에 기인된 것으로 사료된다.
조직공학적 신경재생 및 파킨슨씨병 등의 시경퇴행성 질환에서의 치료에 이용 목적으로 신경성장인자(nerve growth factor, NGF)를 생분해성 고분자 담체에 NGF를 서방화시키고자 PLA 담체에 함유시켜 유화동결건조법으로 제조하였다. 제조된 NGF의 방출량은 생체외 pH 7.4, 37$^{\circ}C$의 PBS 조건하에서 4주 동안 방출실험 하였으며, 함유된 NGF의 활성을 확인하기 위하여 PC-l2 세포에 직접 배양하여 확인하였다. 제조되어진 PLA 담체는 열린 셀 구조를 가졌으며, 초기 NGF의 함량이 많을수록 방출량도 증가를 보였으며, 제조과정에서의 NGF의 환성을 확인하기 위하여 PC-12 세포를 배양한 결과 신경돌기가 성장하였다. 본 연구는 생분해성 고분자 특정인 확산과 분해에 의해서 생물학적 활성물질인 NGF의 방출을 조절할 수 있으며, 조직공학적으로 서방화되어 3차원적인 신경재생을 가능케 할 것으로 기대된다.
