Cancer Stem Cells and Response to Therapy

  • The cancer stem cell (CSC) model states that cancers are organized in cellular hierarchies, which explains the functional heterogeneity often seen in tumors. Like normal tissue stem cells, CSCs are capable of self-renewal, either by symmetric or asymmetric cell division, and have the exclusive ability to reproduce malignant tumors indefinitely. Current systemic cancer therapies frequently fail to eliminate advanced tumors, which may be due to their inability to effectively target CSC populations. It has been shown that embryonic pathways such as Wnt, Hedgehog, and Notch control self-renewal and cell fate decisions of stem cells and progenitor cells. These are evolutionary conserved pathways, involved in CSC maintenance. Targeting these pathways may be effective in eradicating CSCs and preventing chemotherapy or radiotherapy resistance.

Natural Products Targeting Wnt/β-catenin Signaling Pathway

  • The canonical Wnt/β-catenin signaling pathways play an important role in the embryonic development, cell proliferation, differentiation, and adhesion. Therefore, the abnormal activation and repression have been associated with uncontrolled homeostasis in human tissues. In particular, the activation of Wnt signaling is highly correlated with a diverse of diseases including cancer. On this regard, a strategy for targeting Wnt/β-catenin signaling has been employed in the discovery and development of antitumor agents. Herein, the evolution of Wnt signaling and the Wnt inhibitors derived from natural products were briefly summarized in the drug discovery of anticancer agents.

Isolation and Localization of New Germination-related Sequences from Wheat Embryos

  • Subtractive library hybridization was used to isolate the cDNA clones that corresponded to the transcripts that were specifically up-regulated during wheat embryo germination. The clones with numbers 5, 6, 7, 8, 24, and 26 appeared to be more abundant in germinating wheat embryos. Among the isolated clones, we identified four new members of the wheat "germin" gene family. We also identified two novel sequences which exhibited distinct germination up-regulation, and displayed characteristic spatial patterns of expression. One of these, represented by clone pSB10, was principally expressed in the root tissue of germinating embryos. The second was represented by the pSB7 clone and was expressed in both the root and shoot primordia of the embryonic axis, as well as within the coleoptile.

폐국균증을 동반한 내엽형 폐격리증 -1례 보고- (Aspergillosis within an Intralobar Sequestration -A case report-)

  • 폐격리증이란 기형적 동맥에 의해 영양공급을 받는 대생 폐조직의 일부를 일컫는 용어이다. 이는 두 가지로 대별되며 -외협형과 내엽형- 다른 임상양상을 보인다. 본 과에서는 폐국균증을 동반한 내엽형 폐격리증 1례를 경험하였다. 환자는 32세 여자로 특별한 증상은 없었으며, 우연히 낭종성 병변을 지닌 폐종물이 발견되었다. 수술시야에서 하폐인대의 비정상적 동맥을 보어 폐격리증으로 진단하고, 우하엽 절제술을 시행하였다.

  • Recombinant human pBrathyriod hormone related peptide (1-341 (rhPTHrP) has been known to stimulate bone resorption in intact bone tissue culture system. Osteoclast has been known as a primary responsible cell for bone resorption. To examine the effect of rhPTHrP on this cell, we employed disaggregated rat osteodast culture. As a result, we found that rhPTHrP sisnificBntly elevates both the number and total area of resorbed pits in this culture. On the other hand, the conflicting results between disagsregated rat osteoc13st culture and Ca2+-deficient hen osteoclast culture system have been a big obstacle for the progress of bone research. To verify the differences between rat 3nd chick osteoclast system, we performed the same experiment using chick embryonic osteoclast. Since the similar results were obtained from the disaggregated chick osteoclast culture, the discrepancy between chick and rat osteoclast culture study seemed to be due to the difference in culture method, rather due to the species-difference.

Structure and Tissue Distribution of a Trinucleotide-Repeat-containing Gene (cag-3) Expressed Specifically in the Mouse Brain

  • Using in silico approaches and RACE we cloned a full length trinucleotide (CAG) repeat-containing cDNA (cag-3). The cDNA is 2478 bp long and the deduced polypeptide consists of 140 amino acids of which 73 are glutamines. The genomic sequence spans approximately 79 kb on mouse chromosome 7 and the gene is composed of four exons. Standard and real-time PCR analyses of several mouse tissues showed that the gene is exclusively expressed in the brain and is not detected in embryonic stages. Within the brain, it is expressed throughout the forebrain region with predominant expression in the hypothalamus and olfactory bulb and very low levels in the mid- and hindbrain.

  • Cyclopia is a congenital ocular abnormality in which two orbits are fused together due to an arrest in the normal process of embryonic tissue development. Reported here is a case of cyclopia in a calf observed from the uterus of cow slaughtered in an abattoir in Korea on March 24, 2004. The calf possessed a spherical-shaped head and only one centrally placed eye. The one eyeball showed complete eyelashes on the defective eyelids. The nasal region and the anterior nares were absent along with a dorsally curved mandible extended beyond the defective maxillae.

Wnt signaling in cartilage development and degeneration

  • The Wnt signaling network, which is composed of Wnt ligands, receptors, antagonists, and intracellular signaling molecules, has emerged as a powerful regulator of cell fate, proliferation, and function in multicellular organisms. Over the past two decades, the critical role of Wnt signaling in embryonic cartilage and bone development has been well established, and much has been learnt regarding the role of Wnt signaling in chondrogenesis and cartilage development. However, relatively little is known about the role of Wnt signaling in adult articular cartilage and degenerative cartilage tissue. This review will briefly summarize recent advances in Wnt regulation of chondrogenesis and hypertrophic maturation of chondrocytes, and review data concerning the role of Wnt signaling in the maintenance and degeneration of articular chondrocytes and cartilage.

폐격리증 2례 보고 (Pulmonary Sequestration - Report of 2 cases -)

  • Pulmonary sequestration is applied to the congenital malformation characterized by an area of embryonic lung tissue that derives its blood supply from an anomalous systemic artery. Two cases of pulmonary sequestration were treated at the department of thoracic and cardiovascular surgery, college of medicine, Hallym University. One case was extralobar type, associated with the pneumothorax due to rupture of bulla. The other was intralobar type with symptom of massive and recurrent hemoptysis. The supplying arteries of both cases arose from the thoracic aorta. The venous return of the extralobar type was systemic into the hemiazygos vein, and that of the intralobar type was normal into the inferior pulmonary vein. Treatment for the former was resection of the sequestrated lung, and that for the latter was lobectomy of the left lower lobe. With the brief review of literature, we report the cases.

Role of Gremlin-1 in Cancer

  • Gremlin-1 (GREM1) has been defined as an antagonist of bone morphogenetic proteins (BMPs), particularly during embryonic development and tissue differentiation. However, recent studies have shown that GREM1 has BMPs-dependent or -independent functions in diverse human diseases. GREM1 plays a key role in the process of organ fibrosis, including lungs, kidneys, and so on. The GREM1-induced fibrosis typically promotes the development of other diseases, such as pulmonary hypertension, renal inflammation, and diabetic nephropathy. More recently, considerable evidence has been reported showing that GREM1 is involved in the promotion and/or progression of tumors in vitro and in vivo. It also performs an oncogenic role in the maintenance of cancer stem cells. Although GREM1 is known to function in a variety of diseases, here we focus on the role of GREM1 in cancer, and suggest GREM1 as a potential therapeutic target in certain types of cancer.
