A Simple Method for Cat Bone Marrow-derived Mesenchymal Stem Cell Harvesting

  • Bone marrow (BM) cell harvesting is a crucial element in the isolation of mesenchymal stem cells (MSCs). A simple method for harvesting cat BM cells is described. The results show that a large number of BM cells can rapidly be harvested from the cat by this simple procedure. MSCs prepared by density-gradient method were spindle-shaped morphology with bipolar or polygonal cell bodies and strongly positive for CD9 and CD44 and negative for CD18 and CD45-like. They were capable of differentiation to adipocytic and osteocytic phenotypes when exposed to appropriate induction media. The advantages of this method are its rapidity, simplicity, low invasiveness, and low donor attrition and good outcome.

  • On January 6, 2010, two months earlier than normal breeding season, a red fox vixen was implanted with synthetic GnRH analogue, Deslorelin. Blood was sampled every 2~3 days from the day of implant to identifying spermatozoa on stains of epithelial cells. Estradiol and progesterone were examined. Even though the vixen was in non-breeding season, she was mated by a male fox. Pregnancy was confirmed by canine pregnancy detection kit that detect relaxin released from placenta. Four healthy pups were born on March 9, 2010. This is the first report showing synthetic GnRH can activate ovarian function and lead to fertile estrus of red fox in non-breeding season.

Plant Regeneration of Soybean Cultivars via Somatic Embryogenesis

  • Effective plant regeneration from immature cotyledons of soybean [Glycine max (L.) Merr.] cultivars was achieved via somatic embryogenesis. Somatic embryogenesis was performed with the cotyledons of immature embryos 14-20 d after flowering. Immature cotyledons of cv. Whangkeum were placed abaxial or adaxial side down on modified MS medium containing 20mg/L 2,4-D. The greatest number of somatic embryos, 1.2 per cotyledon, was produced from those of 4.0-4.9 mm in length which had been placed abaxial side down. Among cvs. Pecking, Whangkeum and Baekwoon, Pecking had the highest embryo induction efficiency with 4.3 somatic embryos per cotyledon in 20mg/L 2,4-D treatment and with 1.0 embryo per cotyledon in 8mg/L NAA treatment. Germinable globular somatic embryos were induced with the highest efficiency, 27.6%, in 20mg/L 2,4-D and were proliferated efficiently on liquid medium containing 10mg/L 2,4-D. The globular somatic embryos developed into germinable mature somatic embryos on medium containing 10 $\mu$M CoCl2, 9% sucrose, and 0.5% activated charcoal. These mature somatic embryos germinated on hormone-free mediu. After transfer to the soil, regenerated plants with seeds were obtained.

  • To establish a protocol of estrus induction and synchronization in European mouflon, we performed artificial insemination using frozen-thawed semen and exogenous hormones. CIDR was inserted into vaginas of four mouflons for 16 days. A day before removal of CIDR, PG 600 was injected intramuscularly. $PGF_2{\alpha}$ was injected when removing CIDR. Artificial insemination was cervically conducted with injecting LHRH 48 hours after CIDR withdrawal. Even though no pregnancy was confirmed, estrous signs were notified like open cervix, congestion of vaginal wall and discharge of cervical mucus. Further research in the wild sheep would be needed for development of artificial breeding methods and advancing sustainability of domestic zoos.

Development of a Protocol for Somatic Embryogenesis of Cnidium officinale M akino

  • This study was conducted to develop a somatic embryogenesis protocol for the Cnidium officinale Makino difficult to seed propagation. The immature flowers were used as explants. The concentration of a 2,4-D 1.0mg/L was found to be optimal concentration for induction of embryogenic callus and somatic embryos. Addition of 0.3mg/L, 0.5mg/L and 1.0mg/L to the embryo germination medium promoted somatic embryo germination. Among four concentrations, GA3 1.0mg/L were superior to others. Shoots were transferred to hormone-free MS medium after 2 months of culture in the dark. We obtained an optimized protocol for the regeneration of C. officinale.

Diversity of Anther-derived Plants in Hot pepper (Capsicum annuum L.) (고추 약배양 후대식물체의 다양성)

  • Anthers of three cross combinations of hot-pepper(Capsicum annuum L.) were cultured on Dumas De Vaulx medium supplemented with some growth regulators. The embryo production efficiency and the diversity for agronomic traits in $A_2$ lines were investigated. The embryo production frequencies of hybrid combinations were ranged from 16.4% to 43.4%, the highest embryo induction combination was DGSH $\times$ C-NH with 43.4% embryogenic efficiency. Among total 275 $A_2$ lines, phenotypic variants were found in six lines, 2.1% variant frequency. The diversity of $A_2$ lines derived from anther culture was different according to the cross combinations. Fruit color was within parental range, no transgressive variation was observed. However leaf color showed transgressive variation. In fruit length, fruit width and fruit weight, one C-HC $\times$ DGSH and DGSH $\times$ C-NH showed great diversity compared with doner parents while Cheokjo 1 $\times$ C-NH crossed with Cheokjo 1 with big fruit shape showed small diversity. Stem length to 1st branch was relatively similer to or longer than donor parents. Stem thick exhibited remarkable diversity. Node number to 1st branch distributed alomost within the range of donor parents in C-HC $\times$ DGSH combination, however great transgressive variations were observed in DGSH $\times$ C-NH and Cheokjo 1 $\times$ C-NH combinations.

Effects of Estrus Synchronization and Ovulation Induction Methods on Hormone Concentrations and Pregnancy Rate in Artificial Insemination of Riding Horses (말 인공수정에서 발정동기화와 배란유도 방법이 호르몬 농도와 임신율에 미치는 효과)

  • Reproductive research such as artificial insemination and embryo transfer is necessary to produce high-quality riding horses. In this study, we investigated the effects of estrus synchronization and ovulation induction methods, which can be considered the basis of artificial insemination in horses, on the hormone concentration and artificial insemination pregnancy rate of mares. For the purpose of synchronization of estrus in horses, Cidr-plus insertion method, Regumate feeding method, and 150mg progesterone + 10mg estradiol mixed administration method were used. In the Cidr-plus insertion method and the Regumate feeding method, the progesterone concentration reached the appropriate level for ovulation induction on the 8th day of administration. The mixed administration method of 150mg progesterone + 10mg estradiol maintained the progesterone concentration at an appropriate level immediately after administration. With the administration of PGF2a and hCG, progesterone concentration decreased rapidly, making ovulation induction possible. As a result of comparing the pregnancy rate between natural estrus and estrus synchronization, the pregnancy rate was found to be higher in estrus synchronization and ovulation induction. From the results of this study, it is insufficient to judge the effect of the pregnancy rate due to the small number of tests, but in terms of usability, estrus synchronization and ovulation induction were useful. Therefore, it is expected to contribute to improving the efficiency of future roadster production.

Somatic embryogenesis induction in four cassava landraces in East Java, Indonesia

  • Manihot esculenta Crantz, commonly known as cassava, is a staple aliment that is a significant source of revenue for farmers. The embryogenic callus is crucial in the genetic engineering of various crop species, including cassava. Four cultivar cassava landraces from East Java were assessed for their ability to produce friable embryogenic callus (FEC) for protoplast isolation. In this study, four cassava cultivars; (Kaspro, Kuning, Gajah, and Gendruwo); were used to obtain FEC, which involved the culture of immature leaf lobes (ILLs) and apical buds (ABs) media containing MS supplemented with 33 μM picloram and 2 μM of CuSO4 (M1) or MS supplemented with 50 μM 2,4-D and 2 μM CuSO4 (M2). The highest FEC induction efficiency ranged from 72% to 57%, and the highest FEC number ranged from 4.7 to 3.7 with AB explants in media containing MS + 33 μM pilocram and 2 μM CuSO4 (M1). On the other hand, the efficiency of somatic embryogenesis induction ranged from 67% to 53%, and the number ranged from 4.4 to 3.4. The efficiencies of FEC induction ranged from 48% to 42%, and the number ranged from 3.1 to 2.6 with AB explants in media containing MS + 50 μM 2,4-D and 2 μM CuSO4 (M2); the efficiency of FEC induction ranged from 56% to 50%, and the value ranged from 3.6 to 2.4 with ILL explants. The FEC induction of the Gendruwo cultivar, which was examined using AB and ILL explants, demonstrated the lowest efficiency. Nevertheless, all four cultivars showed the ability to generate FEC, even though their effectiveness differed depending on the explant genotype and the applied media.
