Effect of dark incubation in germination of indirect date palm somatic embryos and conversion into plantlets

  • All studies on date palm somatic embryogenesis have focused on germination in the presence of light while neglecting germination in darkness, which mimics the germination process of zygotic embryos within seeds. To improve the date palm micropropagation protocol, we investigated the effects of light and darkness incubation on the germination of indirect date palm somatic embryos and their subsequent conversion into plantlets. Darkness incubation emerged as a pivotal factor in the germination of indirect date palm somatic embryos and their successful conversion into plantlets. Darkness incubation significantly decreased the time required for the conversion of indirect somatic embryos into plantlets, halving the duration from 24 weeks to only 12 weeks. The micropropagation protocol was modified, consolidating the previous two distinct stages of germination and elongation under light incubation into a single stage under darkness incubation. These findings modified the protocol and significantly reduced the overall duration of the date palm micropropagation protocol.

  • This research was performed to develop seminal propagation method of Saururus chinensis (Lour.) Baill. by conducting a rigorous germination study. Well-selected seeds were dry-stored at 4 ± 1.0℃ during the experiment. To study dormancy type, non-stored seeds were analyzed by embryo observation, germination test and detecting for any difficulties in seed coat to absorb moisture. Then to improve germination, seeds were submerged for 24 hours in a solution of varying concentrations containing one of plant growth regulators and minerals. According to research, fleshly matured seeds had an undifferentiated embryo and had a low germination rate below 5%. In addition, water submersion led to moisture absorption, embryo in the seeds grew and germinated so it was deemed morphophysiological dormant seeds. Percent germination (PG) and germination energy (GE) was greatly improved by soaking in plant growth regulators and minerals for 24 hours. Especially, 500 ㎎/L GA3 treatment resulted in the highest GE as 46.1%. KNO3 meaningfully improved PG (54.3∼57.7%) at 10∼20 mM but effect of minerals on germination acceleration as GE were negatively impacted in all concentrations.

Dormancy type and Germination Characteristics of Seeds of Lonicera chrytsantha Turcz. Ex Ledeb (Caprifoliaceae) (각시괴불나무 종자의 휴면유형과 발아특성)

  • This study was conducted to investigate seed dormancy type and germination characteristics of Lonicera chrysantha. In imbibition test, the seeds imbibed water readily. Therefore, the seeds did not have physical dormancy (PY). The seeds have undeveloped embryo at seed dispersal and, then the E:S ratio (embryo/seed ratio) increased more than about 46% just before germination. In temperature experiments (5, 15, 20 and $25^{\circ}C$), the seeds did not germinate during incubation at $5^{\circ}C$. At $20^{\circ}C$, the seeds started to germinate from a week after sowing and the final germination was 86.7% at 2 weeks after sowing. At $15^{\circ}C$, the seeds started to germinate at 2 weeks and the final germination rate was 75.0% at 4 weeks after sowing. At $25^{\circ}C$, the seeds started to germinate at a week and the final germination rate was 48.3% at 19 weeks after sowing. Consequently, Optimum temperature of germination is $20^{\circ}C$ that shown highest final germination rate and shortest mean germination time. The seeds germinated all temperature treatments within 4 weeks except to $5^{\circ}C$. Therefore, it is concluded that seeds of L. chrysantha have only morphological dormancy (MD).

Somatic Embryo Germination and the Related Biochemical Changes of Liriodendron tulipifera by Bioreactor Immersion Time (생물반응기 내 침지시간에 따른 백합나무 체세포배 발아 및 생화학적 변화)

  • To determine physical and physiological factors for Liriodendron tulipifera L. somatic embryo germination, temporary immersion bioreactor (TIB) system was investigated. It was designed to immerse liquid media with plantlets so that it was able to adjust the immersion time. Immersion of 120 minutes every 4 hours and 60 minutes every 4 hours was found to be effective in germination (91.64%, 85.67%, respectively). However, hyperhydricity of the plantlets was higher in short immersion time (15 minutes every 6 hours) and long immersion time (120 minutes every 4 hours) (51.61%, 34.28%, respectively). Immersion of 60 minutes every 4 hours showed the lowest hyperhydric plantlets, and also it showed the lowest activities of abscisic acid (ABA), superoxide dismutase (SOD), and catalase. The overall results implied that immersion time of media affected germination and growth of somatic embryo, and it was able to make use of germination and growth of L. tulipifera somatic embryos.

Protein Synthesis during Somatic Embryo Development and Artificial Seed Germination of Apium graveolens L. after Abscisic Acid or Cold Treatment (쎌러리(Apium graveolens L.)의 체세포배 발생 및 인공종자 발아에 있어서 앱시스산 및 저온처리에 의한 단백질 합성)

  • To understand the molecular mechanism of hardening process in somatic embryo development and artificial seed germination in celery (Apium graveolens L.), the changes of protein synthesis by ABA or cold teatment at early globular stage were examined. Protein content and nitrate reductase activity in ABA- or cold-treated somatic embryo and seedlings were higher than that in unheated ones. The protein content and nitrate reductase activity were more prominent in somatic embryos than in seedlings. From two-dimensional electrophoresis, several protein spots specific to ABA or cold treatment were identified: 30 KD, 32 KD, 171 KD and 205 KD at heart-shaped stage; and 29 KD, 33 KD, 37 KD, 38 KD, 41 KD, 55 KD, 66 KD, and 110 KD at cotyledonary stage were the most specifically synthesized However the synthesis of certain polypeptides were repressed at heart-shaped or cotyledonary stage: 42 KD, 44 KD, 59 KD, 64 KD, 101 KD, 104 KD, and 190 KD at heart-shaped stage; and 29 KD and 116 KD at cotyledonary stage. The protein pattern changes by ABA or cold treatment occurred simultaneously and mainly in acid-soluble proteins during somatic embryo development and artificial seed germination. Therefore it is suggested that the metabolic changes for adaptation to environmental change occur during somatic embryo development and the germination and growth of seedling from embryo.

  • The purposes of this study were to examine effect by Solid Matrix Priming(SMP) ; method far enhanced rapid and uniform seedling emergence in tobacco seeds. The results were as follows. No enhancement of germination rate was by SMP treatment. In KF114 during 9 days treatment and in KB108 during 7 days treatment, germinative energy was highest, and T50 and mean germination time were the shortest. Cells in embryo and hypocotyl were divided and extended during SMP treatment. The contents of fatty acid were increased in SMP treatment. The changes of glucose content were little.

Rapid in vitro Germination of Zygotic Embryos via Endosperm Removal in Eleutherococcus senticosus

  • Eleutherococcus senticosus (also called Acanthopanax senticosus), belonging to Araliaceae family, has been used as an important medicinal woody plant. Mature seeds of Eleutherococcus senticosus have rudimentary (extremely immature) zygotic embryos and require a long-term stratification for about 18 months to induce germination. Here, through the methods of endosperm removal and other exogenous treatments, we investigated the factors for inducing rudimentary embryos by in vitro culture, Rudimentary zygotic embryos in seeds were at globular to heart-shaped stage at about $250{\mu}m$ in length just after harvest of fruits. When the seeds without testa were cultured on 1/2 MS (Murashige and Skoog 1962) medium, they did not germinate regardless of medium and sucrose concentrations but the removal of endosperm tissue markedly stimulated the growth of rudimentary zygotic embryos. The embryo reached ear-lier maturation, once when the endosperm surrounding the rudimentary embryos was removed. Rudimentary zygotic embryos developed cotyledons within 3 weeks of culture after endosperm emoval. However, post-mature zygotic embryos failed to germinate though they were morphologically normal, indicating another dormancy of embryos. $GA_3\;(2.0\;\cal{mg/L})$ and/or charcoal ($0.2\%$) treatment rapidly enhanced the germination of zygotic embryos. These results suggest that E. senticosus seeds have double dormancy; i. e. morphological rudimentary dormancy influenced by surrounding endosperm and physiological dormancy after post-maturation of zygotic embryos. Based on the above findings, we established the rapid germination of rudimentary zygotic embryos by in vitro culture of excised seeds with endosperm removal and $GA_3$ treatment.

  • The effects of temperature, prechilling, chemicals such as $GA_3$, IAA, kinetin and $KNO_3$ on the germinability of Foeniculum vulgare seed of medicinal plant were examined. In Foeniculum vulgare, the germination rate appeared to be around 50~60% or more in general, showing no differences in germination rate with different temperatures, promoting substances, physical or chemical treatments, and prechilling treatments. The observation of embryo under stereoscopic microscope for Foeniculum vulgare in Umbelliferae showed that seeds with or without embryo was almost the same in number. This result suggests that the lower rate of germination in this species is caused by embryolessness of seeds. The straight-shaped embryos as well as Y-shaped embryos were also observed. Foeniculum vulgare of medicinal plants in Umbelliferae were observed under scanning electron microscope, and did not show any opening problem near micropyle area. Final count should be made on 7th day of germination test.

Effect of Gibberellin and Light on Germination of Seeds in Codonopsis lanceolata Benth

  • Seed of Codonopsis lanceolata exhibits low germination due to impermeable seed coat. Prolonged seed dormancy can be overcome by the application of gibberellins, as it promotes growth of the embryo and weakens the structures surrounding of embryo. The effects of photoperiod, sugar and gibberellin concentration were investigated at constant temperature for 12 days and 22 days in vitro and invivo conditions respectively. The rate of germination of seeds of Codonopsis lanceolata in wet filter paper in both complete dark and light treatments was significantly lower than that of seed treated with $GA_3$. It clearly indicates that there is significant coat imposed dormancy in the seed of Codonopsis lanceolata. The rate of germination in vivo condition was lower than that of the in vitro condition supplemented with $GA_3$ Thus, the physical dormancy due to impermeable seed coat and low level of endogenous gibberellins in the seed was the cause of poor germination rate in Codonopsis lanceolata.

Effect of Red Light on Changes of Embryo Tissue of Barley during Germination (맥아제조시(麥芽製造時) 적색광(赤色光) 희사(熙射)에 의한 배유조직(胚乳組織)의 변화(變化))

  • This study was carried out to investigate the effect of the red light on embryo tissue of barley during germination. The solubility of starch in endosperm of germinated barley was different between dark and red treatments at the 3rd day of germination, but was increased by the red light from the 4th day of germination. Blue value of the starch in the germinated barley decreased rapidly from 0.42 at the 1st day to 0.13 at the 6th day in the dark, and same tendency was found in the red light, but blue value was lower in the red light than in the dark. Aleurone cell wall was swollen much faster in the red light than in the dark during germination. The cell wall was broken down more greatly in the red light than in the dark at the 5th day of germination.
