The pollution in closing water can be caused by not only artificial factor like sewage but also natural factor like elution from sediment. In this study we analyzed Sediment Oxygen Demand (SOD) for verification of sediment purification effect and sediment elution experiment as well as general items like COD, TN, TP, SS to complement and assess the effect of sediment and water quality. The experiment result showed that the release rate of OSD system were 4 times and 3 times as large as control for P and Fe respectively. SOD for operated OSD system and control were $12.18gO_2{\cdot}m^{-2}{\cdot}d^{-1}$ and $47.95gO_2{\cdot}m^{-2}{\cdot}d^{-1}$. From water qualities analyzed by COD, TN, TP, SS, chlorophyll-a, the removal efficiency increase of TN, TP, chlorophyll-a and COD were about 10~20%, 40~50% and 10% respectively. In conclusion, OSD can contribute to improvement of both the waterbody and the sediment environment effectively.
A description of simple, isocratic and precise reversed phase HPLC methods is given for simultaneous quantification of pholcodine and ephedrine hydrochloride together with either carbinoxamine maleate or terfenadine in antitussive-antihistaminic oral pharmaceutical formulations. Separations were carried out on X-Terra and symmetry shield C18 column $(250\;{\times}\;4.6\;mm,\;5\;{\mu}m)$. The used isocratic elution systems were either $0.02\;M\;KH_2PO_4-acetonitrile$ in the ratio of 75 : 25 and pH adjusted to 7.70 with orthophosphoric acid or sodium hydroxide, for syrup (method A), or 0.02 octanesulphonic acid sodium salt solution-acetonitrile-acetic acid in the ratio of 75 : 25 : 0.5 for suspension (method B). The elution of both mixtures was achieved with a flow rate of 1 ml/min. Detection was carried out by UV absorbance at wavelengths of 220 and 250 nm for syrup and suspension, respectively. The quantification of the components in synthetic mixtures and actual syrup and suspension were calculated using the internal standard technique with metoclopramide HCl and codeine phosphate as internal standards (IS), respectively. The methods, for both mixtures, were validated and met all the requirements for the quality control analysis recommended by FDA and ICH.
The present study was performed to investigate the excellent microbial anticaries substance, aminoglycoside antibiotic, which is more effective than chlorhexidine for the treatment of dental caries. The aminoglycoside antibiotic against Streptococcus mutans JC-2 from a novel alkaliphilic Bacillus alkalophilshaggy JY-827 exhibited no significant difference at the treatment concentration of $2.5{\times}10^{-7}M$, however, it inhibited the activity of the Streptococcus mutans glucosyltransferase by 70.2% and 99.8% at the concentrations of $2.5{\times}10^{-7}$M\;and\;2.5{\times}10^{-6}M$, respectively. Lineweaver-Burk plot of the inhibitory aminoglycoside antibiotic showed competitive inhibition, with $K_i$ value of $6.4{\times}10^{-6}$ M. The aminoglycoside antibiotic did not show any cytotoxicity against human gingival cells. To evaluate the industrial applicability of the aminoglycoside antibiotic, a toothpaste containing this substance was prepared and tested on the extracted human teeth. The inhibitory rate of tooth calcification and calcium ion elution by the aminoglycoside antibiotic were 50% and 2.5 times, respectively. These results suggested that the aminoglycoside antibiotic from Bacillus alkalophilshaggy JY-827 is an effective agent against dental caries.
본 연구는 인천광역시에 위치한 A 유수지의 효율적인 수환경 개선 방안을 모색하기 위해 수행되었다. 유수지 수질 및 저니질의 개선을 위해서 약품 응집과 해수 응집, 펜턴 처리 실험 등을 수행하였다. 실험 결과 해수 응집을 위한 최적 pH는 11이었으며, 해수 응집에 의해 SS, TP 항목 모두 높은 제거효율을 얻을 수 있었다. 저니토의 오염물질 제거를 위해서는 펜턴 산화 처리를 수행하였다. 저니토를 직접 펜턴 처리하는 경우와 용출 후 펜턴 처리하는 경우에 대한 처리효과를 비교한 결과, 제거-COD/$H_2O_2$는 단순 펜턴 처리하는 경우 0.55인 반면 용출 후 펜턴 처리하는 경우 0.69를 나타내어 용출 후에 산화 처리하는 방법이 처리효율 면에서 약 25% 보다 우수한 것으로 나타났다. 이들 두 처리로 유수지 수질을 호소수질기준 VI등급(매우 나쁨)에서 III등급(보통)의 수준으로 개선할 수 있었다.
We investigated the most effective pre-treatment processes and LC/MS/MS condition for microcystins analysis. With a step-by-step pre-treatment, efficiencies of several established methods were compared. At the level of cell burst, sonication method was found to be the most efficient. As a mycrocystins first extraction solvent, 5% acetic acid showed the highest efficiency. An isolation and recovery rate of mycrocystins of ODS Sep-Pak $C_{18}$ cartridge was higher than HLB SPE cartridge. As a final elution solvent from cartridge, 100% MeOH had a better efficiency than others. Using a LC/MS/MS, effective analytical methods were established. C18 reverse column was used and gradient elution was performed with using acetonitrile, 0.1% formic acid as a mobile phase. We analysed to 0.8 mL/min flow rate fit to the $5{\mu}m$ particle size column and $55^{\circ}C$ housing temperature. The validity of established analytical method was evaluated that MDL as average $0.050{\pm}0.014{\mu}g/L$ and LOQ as average $0.160{\pm}0.045{\mu}g/L$ had a good sensitivity over 40 magnification rather than $2{\mu}g/L$ detection limit of HPLC.

A new method to extract gold from pyrite roster cinder, which combines ultra-fine-grinding with resin-in-pulp, has been studied in this paper. Compared with traditional leaching technology, it can short leaching time, avoid complex filter process, lower sodium cyanide consumption and increase gold recovery by 35%. During leaching, aluminium oxide ball was used as stirred medium, hydrogen peroxide as leaching aid and sodium hexametaphosphate as grinding aid. With the high efficiency and chemistry effect of ultra-fine-grinding, the leaching process was developed and the gold leaching rate may reach 88%. With AM-2 Б resin as abosorber and sulfocarbamide (TU) as eluent, gold was recovered from cyanide pulp by resin-in-pulp. AM-2 Б resin has good adsorbability in cyanide solution(pH=10). It was easy to elude gold from the loaded resin with 0.1㏖/L cholhydric acid and 1㏖/L sulfocabamide. The effect of contact time, temperature and acidity etc. on the gold absorption had been examined with static methods. The results showed that the adsorption and desorption of gold could both reach over 98%. The effects of flow rate of solution on dynamic adsorption and elution of gold had been examined with dynamic methods. Breakthrough curve and elution curve had been drawn in this paper. A mild condition was determined through a number of experiments: leaching time 2 hours, liquid solid ratio 4:1, sodium cyanide 3kg/t, hydrogen peroxide 0.05%, sodium hexametaphosphate 0.05%; adsorption time 30 minutes, temperature 10-3$0^{\circ}C$, resin($m\ell$) solid(g) ratio 1:10, eluent resin ratio 10-20:1, velocity of eluent $1.5m\ell$/min. Under the mild condition, the gold recovery may reach 85%.
As a part of studies on acute effects of high dose irradiation the present report was carried out to evaluate the changes of the leukocyte life span in the Newzealand white male rabbits by a single whole body exposure to gamma rays from $^{60}Co$ teleirradiation unit. The exposure was done in dose levels of 100, 300, 550 and 1,000 rad to each experimental group of 10 rabbits. The life span and apparent half survival time of leukocytes, and the elution rate of leukocytes in the circulating blood were measured by McMillan method using $^{51}Cr$. 1. As a critical indicator of radiation hazards of the Newzealand male rabbits, the LD 50/30 and LD 100/30 after whole body exposure was estimated as 550 and 1,000 rads respectively. 2. The life span of leukocyte in the circulation after irradiation was slightly shortened in the 100 rad irradiated group, as compared with the unirradiated control group, but markedly shortened in the 300, 550 and 1,000 rads irradiated group. 3. After irradiation, decrease of leukocyte half survival time in the circulation showed the same pattern as that of leukocytes life span. 4. As the irradiation doses increased, the elution rate of $^{51}Cr$ loss from $^{51}Cr$ tagged leukocytes in the circulation were markedly increased gradually. 5. The life span shortening of leukocytes in the circulation after irradiation seems to occure by two processes of senescence acceleration and early destruction.
실리카 입자를 기공 형성제로 사용하여 물리적 강도와 단백질 결합용량이 높은 다공성 키토산 및 키틴 친화 막을 제조하였다. 키토산 친화 막의 BSA 단백질 결합용량은 최대 21.8mg/mL이었으며, 키틴 친화 막의 lysozyme 효소 결합용량은 최대 26.1mg/mL이었다. 제조된 다공성 키토산 및 키틴 친화 막을 사용하여 단백질 용액의 loading 유량, loading 양 및 농도 변화에 따른 BSA와 lysozyme의 친화 막 여과 크로마토그래피 분리 실험을 수행하였다. 친화 막 여과 크로마토그래피 분리 실험을 통해 얻어진 loading/washing/elution의 단계로 구성된 일련의 크로마토그램으로부터 단백질 용출량과 결합수율을 구하였다. 키토산 및 키틴 친화 막에의 BSA 및 lysozyme 단백질의 결합량과 결합수율은 loading용액의 유량이 작을수록, 주입량 및 농도가 클수록 증가하였다. 이 결과로부터 실리카 입자를 기공 형성제로 사용하여 제조된 다공성 키토산 및 키틴 막은 단백질의 대규모 여과 크로마토그래피 분리를 위한 친화 막으로서 효과적인 활용이 기대된다.
The analytical methods of polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans (PCDD/Fs) for water sample with extremely low concentration was developed to extract large volume and to improve fast chromatography for clean up. Semi-automated solid phase extraction (SPE) system and column coupling chromatography using elution pump was optimized and applied to each processes of treatment plant. Results of disk type SPE indicated that this system was applicable below 40 L of aqueous sample with a flow rate of 0.08 ~ 0.2 L/min. Average recoveries of SPE using labeled sampling spike of $^{37}Cl$-2,3,7,8-TCDD was 97%. Column coupling method resulted in reduced clean up time, solvent volume, increased average recoveries with constant elution rate. The combined methods were applied to the monitoring of drinking water treatment plant. Limits of detection (LOD) of each process were calculated. For example, LOD of raw and treated water ranged 0.094~0.968 pg/L and 0.028~0.364 pg/L, respectively. Combined methods of extraction and cleanup techniques provided fast analysis of PCDD/Fs with high accuracy and low LODs for water samples.
Long-term water treatment performance analysis was conducted for the constructed wetland treating agricultural tailwater. Studied wetland was established in 2001 and operated from June, 2002 to November, 2010. Wetland vegetation cover was shown over 90 % coverage after 2005. According to vegetation development, accumulation of sediment nutrient was observed; Organic Carbon, T-P and T-N. In addition, DO concentration and temperature was decreased in the constructed wetland output. An infiltration rate also significantly decreased due to compaction of wetland soil. A runoff coefficient was increased due to the low infiltration rate after 2005. A T-N, TSS and Chl.a removal rate was maintained constantly. However, the T-P removal rate was slightly decreased along to wetland operation because low DO concentration could increase elution of phosphorus from sediment. After constructing open water, the T-P removal rate was increased. This is because open water could accelerate the reaearation process. Consequently, over three years of vegetation development could be helpful for wetland performance. In addition, DO concentration is important factor to maintain the T-P treatment.
