Occurrence of a Hybrid Between Taenia saginata and Taenia asiatica Tapeworms in Cambodia

  • Human infection with Taenia asiatica or a hybrid between Taenia saginata and T. asiatica has not been reported in Cambodia. We detected for the first time a hybrid form between T. saginata and T. asiatica in Preah Vihear Province, Cambodia. An adult tapeworm specimen, i.e., 75 cm long strobila without scolex, was expelled from a 27-year-old man after praziquantel medication and purging. It was morphologically indistinguishable between T. saginata and T. asiatica. Several proglottids were molecularly analyzed to confirm the tapeworm species. The mitochondrial gene encoding cytochrome c oxidase subunit 1 (cox1) and nuclear genes encoding elongation factor-1α (ef1) and ezrin-radixin-moesin (ERM)-like protein (elp) were sequenced, and a single-allele analysis was performed to confirm the haploid genotype. The results revealed that our sample showed a discrepancy between the mitochondrial and 2 nuclear genes. It possessed homozygous sequences typical of T. saginata at cox1 and ef1 loci. However, it was heterozygous at the elp locus, with 1 allele in T. asiatica (elpA) and 1 in T. saginata (elpC), which indicates that it is a hybrid between T. saginata and T. asiatica. The present results confirmed the presence of a hybrid between T. saginata and T. asiatica in Cambodia and strongly suggest the existence of also 'pure' T. asiatica in Cambodia.

Effect of Tempering Treatment on Mechanical Properties of Ausformed Martensite in Fe-30% Ni-0.35%C Alloy (Fe-30%Ni-0.35%C 합금에서 Ausformed Martensite의 기계적 성질에 미치는 Tempering처리의 영향)

  • In order to investigate the effect of tempring treatment on the mechanical properties of ausformed martensite in Fe-30%Ni-0.35%C alloy, the hardness, yield strength and elongation were examined by tensile test. 1. The strength of deformed austenite in Fe-30%Ni-0.35%C alloy was increased due to the work hardening induced from the dislocation density increased during deformation. The strength of ausformed martensite was increased because of defects inherited from deformed austenite by martensitic transformation. 2. The ductility of ausformed martensite was shown a nearly constant values independent of deformation degrees because of the interaction of multiple factors such as increased retained austenite, formation of void and decrement of twin in ausformed martensite. 3. The strength of ausformed martensite by tempering treatment was shown a little decrement up to $340^{\circ}C$, especially showed remarkable softening resistance in higher deformation degrees. 4. Virgin martensite and ausformed martensite were shown a maximum yield strength by clustering in tempering at $100^{\circ}C$ and above $100^{\circ}C$, yield strength was very small decreased due to the decrement of solute carbon by the destruction of clustering. 5. The decomposition of retained austenite was not shown up to $450^{\circ}C$ in ausformed martensite with tempering treatment, and the matrix was rapidly softening because of the decomposition of martensite and the formation of reversed austenite with tempering above $400^{\circ}C$.

High-frequency regeneration by stem disc culture in selected clones of Populus euramericana

  • An efficient regeneration protocol for stem disc culture of Populus euramericana, which is important species for bioenergy resource in agroforestry, was established. The number of explants that were obtained and the number of explants that regenerated varied with the genotypes. However, in all the genotypes, stem disc culture produced more regenerated shoots than did in axillary bud culture. A comparison of the effects of cytokinin type and concentration on shoot regeneration in different explants (i.e., petiole, leaf, and root segments of P. euramericana) revealed that a concentration of $0.002mg\;l^{-1}$ thidiazuron (TDZ) used on petiole segments resulted in the greatest shoot regeneration (95.83%). The hormonal requirements for the greatest shoot regeneration in the three explant types varied. Different concentrations of $AgNO_3$ and $CoCl_2$ were added separately to the medium to stop the yellowing and subsequent necrosis of the regenerated shoots. Lower concentrations (3 and $5mg\;l^{-1}$) of these compounds improved shoot regeneration and elongation, compared with the control. The in vitro-regenerated shoots were transferred to rooting medium and subsequently acclimatized. The highly efficient regeneration system of P. euramericana reported here can be used for mass propagation of this recalcitrant for regeneration, economically important tree species.

  • This paper investigates the applicability of newly developed Cu-Al-Mn shape memory alloy (SMA) bars to retrofitting of historical masonry constructions by performing quasi-static tests of half-scale brick walls subjected to cyclic out-of-plane flexure. Problems associated with conventional steel reinforcing bars lie in pinching, or degradation of stiffness and strength under cyclic loading, and in their inability to restrain residual deformations in structures during and after intense earthquakes. This paper attempts to resolve the problems by applying newly developed Cu-Al-Mn SMA bars, characterized by large recovery strain, low material cost, and high machinability, as partial replacements for steel bars. Three types of brick wall specimens, unreinforced, steel reinforced, and SMA reinforced specimens are prepared. The specimens are subjected to quasi-static cyclic loading up to rotation angle enough to cause yielding of reinforcing bars. Corresponding nonlinear finite element models are developed to simulate the experimental observations. It was found from the experimental and numerical results that both the steel reinforced and SMA reinforced specimens showed substantial increment in strength and ductility as compared to the unreinforced specimen. The steel reinforced specimen showed pinching and significant residual elongation in reinforcing bars while the SMA reinforced specimen did not. Both the experimental and numerical observations demonstrate the superiority of Cu-Al-Mn SMA bars to conventional steel reinforcing bars in retrofitting historical masonry constructions.

  • This experiment was carried out to determine the effect of harvesting dates on the dry matter yield and quality of "Kool grazer" winter rye(Seca1e cereale L.) as a soilage crop at the Forage Experimental Field of College of Agrlc. and Life Sciences, SNU, Suweon. Four-day harvasting interval was allocated to measure the growth, yield and quality of rye from April 14 to May 18, 1987. Rye was rapidly grown as 3 cm/day and the growth was linearly recorded from the elongation to flowering stage. The dry matter and in vitro digestible dry matter(1VDDM) yields per ha were linearly increased up to 0.26 and 0.19 tonlday from April 14 to May 4 and April 14 to 29, respectively, but major breaks in yield increases occurred at the 50% heading stage, than yields increased. The highest crude protein yield was observed at the first heading stage of April 29, then decreased. Acid detergent fiber (ADF) and crude protein(CP) contents of rye at the heading stage of May 5 were 35 and 17%, respectively and the nutritive value of rye in terms of ADF and CP contents was evaluated as the I st grade hay, but neutral detergent fiber(NDF) content of rye was 47 % at the boot stage ofApril 22 and evaluated as the 2nd grade hay in terms of NDF content. The results indicate that the first heading stage or the end of April would be the most suitable harvesting time for early maturing winter rye as a soilage crop in terms of yield and quality in the middle northwestern coast rcgion of Korea. of Korea.

Effect of UV Radiation on Early Growth of Korean Rice Cultivars(Oryza sativa L.)

  • The concerns on the crop damage by ultraviolet (UV) radiations is increasing owing to the decrease of their absorbing stratospheric ozone in the tropospheric. Cultivar differences on early growth of UV radiation among five Korean rice cultivars, four japonica types and one Tongil type (indica-japonica cross hybrid), were studied. Pot-seeded rice plants were grown under four different radiation conditions, i.e., visible radiation only, visible radiation with supplemented with high or low dose of UV-B (280~320 nm in wavelength) and UV-C (less than 280 nm in wavelength). The inhibitory degree on plant height, shoot and root weight and length of leaf blade and leaf sheath were determined at 40 days after seeding. UV-C showed the most severe inhibitory effect on the degree of biomass gain and leaf growth in most cultivars examined, followed by high UV-B and low UV-B. Among the cultivars used, the Kuemobyeo was the most sensitive cultivar and had not repair or showed resistance ability to continued irradiation of UV radiation. However, Janganbyeo and Jaekeon showed different responses that the elongation of leaf blades was promoted on 2nd and 3rd leaves and inhibited on 4th and 5th leaves but this inhibitory degree was reduced on 6 th and 7th leaves. Such tendency on leaf growth means that both cultivars had low sensitivity and most resistant ability to continued irradiation of UV radiation. While Tongil showed different response to enhanced UV radiation, ie., low UV-B promoted leaf growth but the inhibitory was severely increased by continued irradiation of high UV-B and UV-C, which means that Tongil had high threshold of UV radiation for response as an inhibitory light of plant growth. The results of this study indicate that the differences on sensitivity or resistant to the effects of UV radiation were existed among Korean rice cultivars.

Effects of Dried Days on Properties of Seawater and Freshwater Flooded CSPE in NPPs

  • Accelerated thermal aging of chlorosulfonated polyethylene (CSPE) was performed for 0 days, 80.82 days, and 161.63 days at 100℃, which is equivalent to 0 y, 40 y, and 80 y of aging, respectively, at 50℃. After freshwater flooding, the volume electrical resistivity of CSPE was highest after 180 days of drying, and its insulating property recovered when dried for more than 300 days. The dielectric constant of the CSPE was not measured after seawater flooding. The dielectric constant of the accelerated thermally aged CSPE was higher after freshwater flooding than that before seawater flooding. The bright, open pores of CSPE were converted into dark, closed pores after seawater flooding, and the dark, closed pores of the accelerated thermally aged CSPE samples were partly converted into bright, open pores after freshwater flooding. The apparent density of CSPE increased slightly whereas its elongation at break (EAB) decreased until 80 y of accelerated thermal aging before seawater flooding. The peak binding energies of oxygen in the non-accelerated and accelerated thermally aged CSPE for 40 y and 80 y were shifted by more than 1.0 eV after seawater and freshwater flooding. The CH2 content in the non-accelerated and accelerated thermally aged CSPE for 40 y and 80 y after seawater flooding for 5 days was lower than that before seawater flooding whereas atoms such as Cl, O, Pb, Al, Si, Sb, and S that are related to conducting ions such as Na+, Cl-, Mg2+, SO4 2-, and K+ were relatively increased.

Physical Properties of Biofilm Manufactured from Gelatin of Yellowfin Tuna Thunnus albacares Skin Treated with Acetic Acid (아세트산 처리 황다랑어(Thunnus albacares) 껍질 유래 젤라틴으로 제조한 바이오필름의 물리적 특성)

  • The present study examined the physical properties of biofilms manufactured from yellowfin tuna Thunnus albacares skin gelatin with the aim of developing a biofilm from fisheries by-products to replace mammalian sources. The physical properties of biofilms from yellowfin tuna gelatin were compared with those of biofilms from porcine gelatin. The yellowfin tuna gelatin biofilm exhibited higher tensile strength (69.08 MPa) and greater elongation (14.32%) than did porcine gelatin biofilm (50.50 MPa and 10.21%, respectively). The ${\Delta}E$ and YI (yellowness index) Huntercolor values of yellowfin tuna gelatin biofilm were three-fold and 15-fold higher, respectively, than values for porcine gelatin biofilm. The opacity value of yellowfin tuna gelatin biofilm was higher than that of porcine gelatin biofilm. The stability against water of yellowfin tuna gelatin biofilm was lower than that of porcine gelatin biofilm at pH 3 to pH 11. Thermogravimetric analysis (TGA) indicated that the thermal stability of the biofilms was about $270^{\circ}C$ for porcine gelatin biofilm and about $250^{\circ}C$ for yellowfin tuna gelatin biofilm.

Formation and Properties of Corn Zein Coated ${\kappa}-Carrageenan$ Films (Corn Zein을 코팅한 카라기난 필름의 제조 및 특성)

  • Corn zein coated carrageenan films were manufactured by immersing preformed ${\kappa}-carrageenan$ films into solution of different concentrations of corn zein (CZ) solids (10, 20, and 30% in 95% ethyl alcohol) with polyethylene glycol (20% w/w of CZ), glycerol (24% w/w of CZ). For all types of films, water vapor permeability (WVP), water solubility (WS), swelling ratio (SR), tensile strength (TS), percent elongation at break (E), heat sealing strength (HS), and Hunter color values (L, a, and b) were determined. WVP of corn zein coated ${\kappa}-carrageenan$ films decreased significantly (p<0.05) as the concentration of com zein increase. Coating with corn zein also decreased film WS and SR linearly with the concentration of corn zein. TS of corn zein coated carrageenan films decreased linearly with corn zein concentration. All the com rein coated carrageenan films showed heat sealing properties even though their sealing strength was less than half of corn zein film. Obviously corn zein coating affected color of ${\kappa}-carrageenan$ films, which was mainly caused by increase in yellowness (Hunter b-value).

Expression and Characterization of Polyketide Synthase Module Involved in the Late Step of Cephabacin Biosynthesis from Lysobacter lactamgenus

  • The cephabacins produced by Lysobacter lactamgenus are ${\beta}$-lactam antibiotics composed of a cephem nucleus, an acetate residue, and an oligopeptide side chain. In order to understand the precise implication of the polyketide synthase (PKS) module in the biosynthesis of cephabacin, the genes for its core domains, ${\beta}$-ketoacyl synthase (KS), acyltransferase (AT), and acyl carrier protein (ACP), were amplified and cloned into the pET-32b(+) expression vector. The sfp gene encoding a protein that can modify apo-ACP to its active holo-form was also amplified. The recombinant KS, AT, apo-ACP, and Sfp overproduced in the form of $His_6$-tagged fusion proteins in E. coli BL21(DE3) were purified by nickel-affinity chromatography. Formation of stable peptidyl-S-KS was observed by in vitro acylation of the KS domain with the substrate [L-Ala-L-Ala-L-Ala-L-$^3H$-Arg] tetrapeptide-S-N-acetylcysteamine, which is the evidence for the selective recognition of tetrapeptide produced by nonribosomal peptide synthetase (NRPS) in the NRPS/PKS hybrid. In order to confirm whether malonyl CoA is the extender unit for acetylation of the peptidyl moiety, the AT domain, ACP domain, and Sfp protein were treated with $^{14}C$-malonyl-CoA. The results clearly show that the AT domain is able to recognize the extender unit and decarboxylatively acetylated for the elongation of the tetrapeptide. However, the transfer of the activated acetyl group to the ACP domain was not observed, probably attributed to the improper capability of Sfp to activate apo-ACP to the holo-ACP form.
