Analysis of Facilitied Transport through Fixed Site Carrier Membranes

  • A simple mathematical model for gacilitated mass transport with a fixed site carrier membrane was derived by assuming an instantaneous, microscopic concentration (activity) fluctuation, The concentration fluctuation, developed due to reversible chemical reaction between carrier and solute, could acuse the higher chemical potential gradient and the facilitated transport. For mathematical formulation, an analogy was employed between the mass transfer for the facilitated transport with fixed site carrier membrane and the electron transfer in a parallel resistor-capacitor (RC) circuit. For the single RC model, it was assumed that a single capacitor represented the total carrier and a solute could not inter-diffuse between matrix and carrier, allowing only two diffusional pathways, This assumption was relaxed by adopting a serial combination of the parallel RC circuit. Here, a solute diffuses in two elements (matrix or carrier) can exchange its pathway, exhibiting four diffusional pathways. The current models were examined against experimental data and the agreement was exceptional.

Effects of Several Medicinal Plants Extract on Survival Rate, Chlorophyll Contents and Photosynthetic Electron Transport Activity of Liverwort Photoautotrophic Cultured Cell (약용식물 추출액이 우산이끼 자가관양배양세포의 생존율, 엽록소함량 및 광합성전자전달 활성에 미치는 영향)

  • The effects of allelochemicals from medicinal plants have been studied as photo-synthetic inhibitor for photoautotrophic(PA) cultured cells. The extracts from 9 plant species were used for measuring the physiological effects on the liverwort cultured cell in following areas; germination inhibition, chlorophyll contents, hill activity, cell viability, photosynthetic oxygen evolution,and protein pattern changes on SDS PAGE. Germination inhibitions were detected in all plant after treating with 10% extract. Especially, treatment with 10% extract from Pulsatilla koreana and Aconitum carmichael inhibited germinations completely. Chlorophyll fornation was inhibited completely by treating PA cells with extract of Pulsatilla koreana, whose effect was similar to that of DCMU 10-3M, inhibitor for photosynthetic electron trans-fer. The treatment with extract from Pulsatilla koreana on PA cell showed the highest hill activity and the lowest cell viability among extracts studied. Oxygen releasing has been decreased down to 14-77% after treating with extracts from Pinellia ternata, Araliacont inentaila, Pulsatilla koreana and Vitex rotundifolia. Especially, 60$\mu$l of Pulsatilla koreana extract into 2ml mixture of PA cell inhibit-ed oxygen release up to 50%. Protein bands on SDS-PAGE, 14kD, 31kD, 41kD, 53kD, and 73kD, were not detected after treating Pulsatilla koreana extract on PA cells.

  • Quantifying rates of microbial processes under subsurface conditions is difficult, and is most commonly approximated by laboratory studies using aquifer materials. In this study a single-well, 'push-pull' test method is adapted for the in situ determination of denitrification rates in groundwater aquifers. The rates of stepwise reduction of nitrate to nitrite, nitrous oxide, and molecular nitrogen were determined by performing a series of push-pull tests at an experimental well field of Korea University. A single Transport Test, one Biostimulation Test, and four Activity Tests were conducted for this study. Transport tests are conducted to evaluate the mobility of solutes used in subsequent tests. These included bromide (a conservative tracer), fumarate (a carbon and/or source), and nitrate (an electron acceptor). At this site, extraction phase breakthrough curves for all solutes were similar, indicating apparent conservative transport of the solutes prior to biostimulation. Biostimulation tests were conducted to stimulate the activity of indigenous heterotrophic denitrifyinc microorganisms. Biostimulation was detected by the simultaneous production of carbon dioxide and nitrite after each injection. Activity tests were conducted to quantify rates of nitrate, nitrite, and nitrous oxide reduction. Estimated zero-order degradation rates decreased in the order nitrate '||'&'||'gt; nitrite '||'&'||'gt; nitrous oxide. The series of push-pull tests developed and field tested in this study should prove useful for conducting rapid, low-cost feasibi1ity assessments for in situ denitrification in nitrate-contaminated aquifers.

Photoreactivation of the Oxygen Evolving Center in TIB-treated Chloroplasts of Spinach (TIB로 처리된 시금치의 엽록체에서 산소발생계의 광재활성화)

  • In Tris-iso-butanol (TIB; Tris buffer pH 8.8 and 1% iso-butanol)-treated chloroplasts, oxygen evolving activity was more inhibited than Tris-treated chloroplasts, but restored highly by 2,6-dichlorophenol-indophenol (DCPIP) and photoreactivation. To understand the mechanism of this results of TIB in photosynthetic electron transport, system, oxygen consumption and evolution of PS I and PS II were measured and protein of the chloroplasts was analysed. In Tris- and TIB-treated chloroplasts, oxygen evolving activity was increased according to the light intensity. Under 48 W·m-2 light intensity, the oxygen evolving activity in both chloroplasts were similar but as the light intensity was increased, TIB-treated chloroplasts showed higher activity. Under 240 W·m-2 light intensity, TIB-treated chloroplasts showed about 25% higher oxygen evolving activity than Tris-treated chloroplasts. Oxygen evolving activity was increased after photoreactivation in both Tris-treated and TIB-treated chloroplasts. Addition of NH4Cl increased the activity in both chloroplasts but in TIB-treated chloroplasts the increase was 30% higher than that in Tris-treated chloroplasts. In PS I, oxygen evolving activity was not inhibited by both treatments whereas in PS II, significant difference was observed between two treatments. Addition of Mn2+ and Ca2+ enhanced oxygen evolution in both Tris- and TIB-treated chloroplasts. Though enhancement was higher in TIB-treated chloroplasts. No difference was observed n protein analysis of the two thylakoid membrane.

The other was that the different concentrations of ${Zn}^{2+}$was applied in each growth medium. Then, it was not added into the chloroplasts suspensions isolated from the plants. PS II activity in both way of the treatments was more severely inhibited than PS I by the increment of ${Zn}^{2+}$ concentration. The photophosphorylation activity measured by pH measurement was gradually decreased with the increase of ${Zn}^{2+}$concentration in both ways, too. However, it was shown that M $n^{2+}$ could be near fully overcome the inhibitory effect of ${Zn}^{2+}$in PS II, and $Mg^{2+}$ could also reduce the Z $n^{2+}$ inhibition in the photophosphorylation. In the low concentrations of $Mg^{2+}$ (3 to 5$\times$10$^{-3}$ M) in the suspension, ${Zn}^{2+}$(2$\times$10$^{-5}$ M) could increase the activity of photophosphorylation. As compares to other cations, Z $n^{2+}$ caused less inhibitory effect on the photophosphorylation activity than Cu, Cd, but more than Pb and Ni. Influence of Charge Transport of Pt-CdSe-Pt Nanodumbbells and Pt Nanoparticles/GaN on Catalytic Activity of CO Oxidation

  • Among multicomponent nanostructures, hybrid nanocatalysts consisting of metal nanoparticle-semiconductor junctions offer an interesting platform to study the role of metal-oxide interfaces and hot electron flows in heterogeneous catalysis. In this study, we report that hot carriers generated upon photon absorption significantly impact the catalytic activity of CO oxidation. We found that Pt-CdSe-Pt nanodumbbells exhibited a higher turnover frequency by a factor of two during irradiation by light with energy higher than the bandgap of CdSe, while the turnover rate on bare Pt nanoparticles didn't depend on light irradiation. We also found that Pt nanoparticles deposited on a GaN substrate under light irradiation exhibit changes in catalytic activity of CO oxidation that depends on the type of doping of the GaN. We suppose that hot electrons are generated upon the absorption of photons by the semiconducting nanorods or substrates, whereafter the hot electrons are injected into the Pt nanoparticles, resulting in the change in catalytic activity. We discuss the possible mechanism for how hot carrier flows generated during light irradiation affect the catalytic activity of CO oxidation.

Synthesis of CdS with Graphene by CBD(Chemical Bath Deposition) Method and Its Photocatalytic Activity

  • Synthesis of RGO (reduced graphene oxide)-CdS composite material was performed through CBD (chemical bath deposition) method in which graphene oxide served as the support and Cadmium Sulfate Hydrate as the starting material. Graphene-based semiconductor photocatalysts have attracted extensive attention due to their usefulness for environmental and energy applications. The band gap (2.4 eV) of CdS corresponds well with the spectrum of sunlight because the crystalline phase, size, morphology, specic surface area and defects, etc., of CdS can affect its photocatalytic activity. The specific surface structure (morphology) of the photocatalyst can be effective for the suppression of recombination between photogenerated electrons and holes. Graphene (GN) has unique properties such as a high value of Young's modulus, large theoretical specific surface area, excellent thermal conductivity, high mobility of charge carriers, and good optical transmittance. These excellent properties make GN an ideal building block in nanocomposites. It can act as an excellent electron-acceptor/transport material. Therefore, the morphology, structural characterization and crystal structure were observed using various analytical tools, such as X-ray diffraction, scanning electron microscopy, transmission electron microscopy, and Raman spectroscopy. From this analysis, it is shown that CdS particles were well dispersed uniformly in the RGO sheet. Furthermore, the photocatalytic property of the resulting RGO-CdS composite is also discussed in relation to environmental applications such as the photocatalytic degradation of pollutants. It was found that the prepared RGO-CdS nanocomposites exhibited enhanced photocatalytic activity as compared with that of CdS nanoparticles. Therefore, better efficiency of photodegradation was found for water purification applications using RGO-CdS composite.

Influences of Temperature and Light on the Herbicidal Activity of Bleaching Herbicides (Bleaching Herbicides의 제초활성에 영향을 미치는 온도 및 광의 영향)

  • This research was carried out to investigate the influences of temperature and light on the herbicidal activity of oxyfluorfen, oxadiazon and paraquat. Increased temperature from 10 to $35^{\circ}C$ resulted in increase of herbicidal activity in whole plants or leaf discs treated with herbicides. It seemed that temperature affected herbicide penetration into and reaction to the action site rather than appearance process of herbicidal activity (maybe membrane peroxidation after being absorbed. The activity of compounds tested increased with increased light intensity. Paraquat showed similar activities regardless of light qualities but oxyfluorfen and oxadiazon showed the highest activities in blue light spectrum, indicating that they seemed to be closely related to chlorophyll biosynthesis rather than carotenoid biosynthesis or electron transport systems of photosynthesis and respiration.

  • The active sites, intracellular transport, function of cellulase in association with the disintegration of the storage materials of the endosperm cells during seed maturation and after-ripening of Panax ginseng C.A. Meyer seeds were studied by electron microscopy. Cytochemical activities of the cellulase occurred in protein bodies and vesicles of endosperm cells in seed with red seed coat. In after-ripening seed, the activities were strongly found in the cell wall of endosperm near the umbiliform layer and on neighbouring vesicles, so it is assumed that these cells begin to be decomposed. Cellulase activities were initiated before the decomposition of storage materials. But, no activity was observed in the umbiliform layer, so it is suggested that cellulase lose its activity after the completion of lysis process.

  • To investigate the effect of cadmium and cadmium binding protein on the electron transport system and conformational changes of rat kidney mitochondria, various cadmium concentration were treated in vitro and respiration rate, NADH-CoQ reductase activity were measured. Ultrastructural changes at state IV respiration were also observed. CdBP was isolated from the rat liver by Sephadex G-75 column fractionation and treated in vitro with cadmium. Also mitochondrial state IV respiration rate was measured. When cadmium was treated in vitro, state IV respiration and enzyme activity were decreased and ultrastructural transformation of mitochondria from a condensed to an orthodox conformation was inhibited under state IV respiration. In case cadmium and CdBP were treated together, oxygen consumption was more increased than cadmium only. Conformational changes of mitochondria from a condensed to orthodox conformation were also observed. This indicates that CdBP have a protective effect against cadmium toxicity.
