The infection of irido-like virus in cultured turbot (양식산 터봇, Scophthalmus maximus에서의 irido-like virus 감염)

  • The high mortality of cultured juvenile turbot, Scophthalmus maximus occurred in Gochang on June, 2003. The diseased fish was lethargic with reduced feed intake. Grossly, these fish showed pale body, abdominal extension and exophthalmia. The dominant internal gross features of diseased fish were severely enlarged spleen, pale gills and/ or liver. Diseased fish histologically showed basophilic enlarged cells in the kidney, spleen, gill, heart, stomach, intestine, liver, pancreas and adipose tissue. Transmission electron microscopy (TEM) reveled hexagonal virions in the cytoplasm of necrotic cells. The viral particles lead a central electron-dense core and an electron translucent zone, and were 136-159 nm in diameter. These results suggest that the virus belonging to the iridoviridae was responsible for the mortality of cultured juvenile turbot.

Changes in bound water and microstructure during consolidation creep of Guilin red clay

  • Creep of soils has a significant impact on mechanical properties. The one-dimensional consolidation creep test, thermal analysis test, scanning electron microscope (SEM) test, and mercury compression test were performed on Guilin red clay to study the changes in bound water and microstructure during the creep process of Guilin red clay. According to the results of the tests, only free and weakly bound water is discharged during the creep of Guilin red clay. When the consolidation pressure p is in the 12.5-400.0 kPa range, it is primarily the discharge of free water; when the consolidation pressure p is in the 800.0-1600.0 kPa range, the weakly bound water is converted to free water and discharged. After consolidation creep, the microstructure of soil changes from granular overhead contact structure to flat sheet-like stacking structure, with a decrease in the number of large and medium pores, an increase in the number of small and micro pores, and a decrease in the fractal dimension of pores. The creep process of red clay is the discharge of weakly bound water as well as the compression of large pores into small pores and the transition of soil particles from loose to dense.

Effects of Precursor Co-Precipitation Temperature on the Properties of LiNi1/3Co1/3Mn1/3O2 Powders (전구체 공침 온도가 LiNi1/3Co1/3Mn1/3O2 분말의 특성에 미치는 영향)

  • $Ni_{1/3}Co_{1/3}Mn_{1/3}(OH)_2$ powders have been synthesized in a continuously stirred tank reactor via a co-precipitation reaction between aqueous metal sulfates and NaOH using $NH_4OH$ as a chelating agent. The co-precipitation temperature is varied in the range of $30-80^{\circ}C$. Calcination of the prepared precursors with $Li_2CO_3$ for 8 h at $1000^{\circ}C$ in air results in Li $Ni_{1/3}Co_{1/3}Mn_{1/3}O_2$ powders. Two kinds of obtained powders have been characterized by X-ray diffraction (XRD), scanning electron microscopy, particle size analyzer, and tap density measurements. The co-precipitation temperature does not differentiate the XRD patterns of precursors as well as their final powders. Precursor powders are spherical and dense, consisting of numerous acicular or flaky primary particles. The precursors obtained at 70 and $80^{\circ}C$ possess bigger primary particles having more irregular shapes than those at lower temperatures. This is related to the lower tap density measured for the former. The final powders show a similar tendency in terms of primary particle shape and tap density. Electrochemical characterization shows that the initial charge/discharge capacities and cycle life of final powders from the precursors obtained at 70 and $80^{\circ}C$ are inferior to those at $50^{\circ}C$. It is concluded that the optimum co-precipitation temperature is around $50^{\circ}C$.

  • A new RNA virus isolated from abnormally swimming behavior has caused mortalities in salmonid fish (Oh et al., 1995 a), A reverse transcriptase (RTase) activity of the virus was determined by using poly r(A) : oligo d(T) as templete : primer. This RTase activity was associated with virus particles of buoyant density of 1.16 g/ml. The virus particles in sucrose fractions were enveloped and were about 85 nm diameter with central electron-dense core. The brain and kidney samples of artificially infected fish showed RTase activity. Virus particle associated proteins about 120, 80, 65, 61, 48, 42, 35, 30, 25, 19, and 15 kDa were observed when examined by polyacrylamide gel electrophoresis analysis. This study showed the presence of a new retrovirus in salmonid fish, which tentatively called RVS (Retrovirus of salmonid).

Role of cytoskeleton in Host Cell Invasion by Intracellular Protozoa Toxoplasma gondii

  • A microfilament-based motility in Toxoplasma gondii (T. gondii) Is involved in host cell invasion, yet the exact mechanism has not yet been determined. Accordingly, the current study examined the localization of actin and tubulin in T gondii using immunofluorescent (IF) and immunogold staining for electron microscopy. Indirect immunofluorescence (IF) staining using anti-actin and anti-tubulin monoclonal antibodies (mAbs) revealed localization of fluorescence on the entire surface of the tachyzoites. The actin in T. gondii was observed by immunogold staining, and the gold particles were seen on the surface, especially at the anterior end and in the cytoplasm of the parasite. However, there were no gold particles in the nucleus, rhoptries, and dense granules. The tubulin in T gondii was located on the surface and in the cytoplasm of the tachyzoites in the extracellular parasite, compared with anterior part of tachyzoites in the intracellular parasite. The antigens of T gondii recognized by anti-actin mAb were 107 kDa, 50 kDa, 48 kDa, and 40 kDa proteins, while those recognized by anti-tubulin mAb were 56 kDa, 52 kDa, and 34 kDa proteins. Tachyzoites of T gondii pretreated with the actin inhibitor, cytochalasin D (20 $\mu\textrm{g}$/ml), and tubulin inhibitor, colchicine (2$\times$10$\^$-6/ M), for 30 min at 37$\^{C}$ were used to infect the isolated mouse macrophages (tachyzo ites:macrophage=2:1). Pretreatment with the inhibitors resulted in lower multiplication of tachyzoites within the macrophages than in the untreated group 18 h post infection (p<0.05). Therefore, the present results suggest that actin and tubulin appear to be involved in the invasion of and multiplication in host cells.

Study on mechanical properties of Yellow River silt solidified by MICP technology

  • With the development of infrastructure, there is a critical shortage of filling materials all over the word. However, a large amount of silt accumulated in the lower reaches of the Yellow River is treated as waste every year, which will cause environmental pollution and waste of resources. Microbial induced calcium carbonate precipitation (MICP) technology, with the advantage of efficient, economical and environmentally friendly protection, is selected to solidify the abandoned Yellow River silt with poor mechanical properties into high-quality filling material in this paper. Based on unconfined compressive strength (UCS) test, determination of calcium carbonate (CaCO3) content and scanning electron microscope (SEM) test, the effects of cementation solution concentration, treatment times and relative density on the solidification effect were studied. The results show that the loose silt particles can be effectively solidified together into filling material with excellent mechanical properties through MICP technology. The concentration of cementation solution have a significant impact on the solidification effect, and the reasonable concentration of cementation solution is 1.5 mol/L. With the increase of treatment times, the pores in the soil are filled with CaCO3, and the UCS of the specimens after 10 times of treatment can reach 2.5 MPa with a relatively high CaCO3 content of 26%. With the improvement of treatment degree, the influence of relative density on the UCS increases gradually. Microscopic analysis revealed that after MICP reinforcement, CaCO3 adhered to the surface of soil particles and cemented with each other to form a dense structure.

THE EFFECTS OF IRRADIATION AND CALCIUM-DEFICIENT DIET ON DENTIN AND CEMENTUM FORMATION OF RAT MOLAR (방사선조사와 저칼슘식이가 백서구치의 상아질과 백악질형성에 미치는 영향에 관한 실험적 연구)

  • The present study was designed to elucidate the effects of the Co-60 γ irradiation and/or calcium­deficient diet on the dentin and cementum formation of rat molar. The pregnant three-week old Sprague­Dawley rats were used for the study. The experimental group was divided into two groups, irradiation/normal diet group and irradiation/calcium-deficient diet group. The control group was non­irradiation/normal diet group. The abdomen of the rats at the 19th day of pregnancy were irradiated with single absorbed dose of 350cGy. The rat pups were sacrificed on the 14th day after delivery and the maxillae including molar tooth germ were taken. The specimens including the 1st molar tooth germ were prepared to make tissue sections for light and transmission electron microscopy. Some of tissue sections for light microscopy were stained immunohistochemically with anti-fibronectin antibody. The results were as follows; 1. The Hertwig's epithelial root sheath cells, which are related to the differentiation of the tooth-forming cells, showed irregular cellular arrangement, decrease of intercellular junctional complex, and decreased immunoreactivity to the fibronectin after irradiation. These were more severe in the irradiation/calcium-deficient diet group. 2. The cementoblasts at the cementum-forming area showed chromatin clumpings after irradiation. The immu noreactivity to the fibronectin was weaken after irradiation, especially irradiation/calcium-deficient diet group. 3. The odontoblasts at the dentin-forming area showed increase of lysosomes in the cytoplasm and destruction of intercellular junctional complex. The irradiation/calcium-deficient diet group showed decrease of number and density of the electron dense particles and a large number of vacuoles scattered in the dentin matrix. The immunoreactivity was weaken.

Multiplication of Infectious Flacherie and Densonucleosis Viruses in the Silkworm, Bombyx mori (가잠의 전염성 연화병 및 농핵병 바이러스 증식에 관한 연구)

  • Flacherie, as one of the most prevalent silkworm diseases, causes severe economic damage to sericultural industry and its pathogens have been proved to be flacherie virus (FV) and densonucleosis virus (DNV). Multiplications of the viruses in the larvae of the silkworm, Bombyx mori, were studied by the sucrose density gradient centrifugation and electron microscopy. The quantitative and qualitative changes of nucleic acids and proteins were investigated from the midgut and hemolymph in the silkworm larvae infected separately with FV and DNV. The histopathological changes of epithelial cells of infected midgut also were examined by an electron microscope. 1. Purified fractions of FV or DNV in a sucrose density gradient centrifugation yielded one homogenous and sharp peak without a shoulder, suggesting no heterogenous materials in the preparation. Electron microscopy also revealed that FV and DNV were spherical particles, 27nm and 21nm in diameter, respectively. 2. Silkworm larvae showed a decrease in body weight on the 6th day and in midgut weight on the 3rd day after inoculation with FV or DNV. 3. DNA content was higher in the midgut when infected with FV or DNV, but the hemolymph of the infected larvae showed no difference during first 6 days after inoculation, after which DNA concentration declined rapidly. 4. RNA synthesis of silkworm larvae infected separately with FV and DNV was stimulated in the midgut, but RNA content was reduced in the hemolymph at the early stage of virus multiplication. At the late stage of virus multiplication, however, it was extremely reduced in both midgut and hemolymph. 5. The concentration of protein in the midgut and hemolymph of silkworm larvae infected separately with FV and DNV showed no difference from that of the healthy larvae at the early stage of virus multiplication, but it was significantly reduced at the late stage of virus multiplication. 6. There was no difference in the electrophoretic patterns of RNAs extracted from the midgut of healthy or virus-infected larvae. 7. The electrophoresis of proteins extracted from the midgut infected with FV or DNV, when carried out on the 1st and 5th day after virus inoculation, showed no difference from that of the healthy larvae. But, there was an additional band with medium motility in the proteins on the 8th day after virus inoculation, while a band with low mobility shown in the proteins of healthy larvae disappeared in the infected larvae. However, a band with high mobility in the healthy larvae was separated into two fractions in the infected larvae. 8. The electrophoretic pattern of hemolymph proteins of the silkworm larvae infected separately with FV and DNV was similar to that of the healthy larvae, but the concentration of hemolymph proteins in the infected larvae was lower than that of the healthy larvae at the late stage. 9. Two types of inclusion bodies were shown by the double staining of pyronin-methyl green in the columnar cell of the midgut on the 8th day after FV inoculation. 10. Electron microscopy of the infected midgut revealed that the 'cytoplasmic wall' of the goblet cell thickened on the 5th day after FV inoculation and several types of the cytopathogenic structures, such as virus$.$specific vesicles, virus particles, linear structures, tubular structures, and high electron-dense matrices were observed in the cytoplasm of the goblet cell. The virus particles were also observed in the microvilli and the structures similar to spherical virus particles were observed around the virus-specific vesicles, suggesting the virus assembly in the cytoplasm. 11. Fluorescence micrograph of the infected midgut stained with acridine orange showed that the nucleus, the site of DNV multiplication in the columnar cell, enlarged on the 5th day after virus inoculation. 12. Electron microscopic examination of DNV infected midgut revealed that the nucleolus of the columnar cell was broken into granules and those granules dispersed into apical region of the nucleus on the 5th day after virus inoculation. On the 8th day after inoculation, it was also observed that the nucleus of the columnar cell was full with the high electron-dense virogenic stroma which were similar to virus particles. These facts suggest that the virogenic stroma were the sites of virus assembly in the process of DNV multiplication.

A STUDY ON THE PHYSICAL PROPERTIES OF GLASS IONOMER CEMENT FOR RESTORATIVE FILLING USING VISIBLE LIGHT POLYMERIZATION (가시광선중합화에 따른 충전용 Glass Ionomer Cement의 물리적 성질에 관한 연구)

  • The aim of this study was to investigate the physical properties of visible light curing Glass Ionomer cement for restorative esthetic filling. The control group was the autopolymerizing GC Fuji II Glass Ionomer cement (2.2: 1 P/L ratio) and the experimental groups were made by following procedure. To induce the polymerization by visible light, the powder of GC Fuji II GI cement and the liquid of Vitrabond for base & liner were mixed in an amalgam capsule with 2.5:1, 3.0:1, 3.5:1 P/L ratio (% wt/wt). After fabrication of specimens, compressive strength, fracture toughness ($K_{IC}$) Scanning Electron Microscope and X-ray Diffraction, water-leachable content, marginal leakage and surface roughness were studied. The results were as follows: 1. Only experimental No. 1 group (visible light curing) showed less compressive strength than control group 1 hour after curing. Strength was increased with aging in all groups, so the compressive strength of light curing groups was no less than that of autopolymerizing group after 3 weeks. 2. Experimental No.3 group (visible light curing) was inferior to No.2 group (visible light curing) in fracture resistance but light curing groups were more resistant to fracture than autopolymerizing group and showed ductile fracture pattern as compared with the brittle fracture pattern of autopolymerizing group. 3. From scanning electron microscopic image, various sized unreacted powder particles, surrounded by silica gel, were embedded in polysalt matrix. Light curing groups showed little crack and more dense unreacted particles than autopolymerizing group. 4. From X-ray diffraction analysis, GC Fuji II Glass Ionomer cement powder and all groups showed glassy appearance but light curing groups seemed to be more intensive in crystaline than autopolymerizing group. S. The most significant dissolution was shown in early setting period in all group. Light curing groups were dissolved less than autopolymerizing group. 6. Marginal leakage was not different significantly in case of cavity margin composed of same tooth structure (ex. only enamel margin, only dentin margin) but much more leakage was shown in dentin/cementum margin than enamel margin. In only case of only enamel margin, light curing groups were superior to autopolymerizing group. 7. All groups showed relatively smooth surface, which irregularity was less than $1{\mu}m$. Light curing groups were smoother than autopolymerizing group.

Morphological Differentiation of Leydig Cells in Human Fetal Testes (사람 태아 고환에서 간질세포 분화의 형태학적 관찰)

  • The Leydig cell found within the interstitium of the testis is important in the spermatogenesis. The differentiation of Leydig cell, even though relatively well known in animals, is not fully elucidated yet in human. In the present study, human fetal testes ($14\sim27$ weeks) obtained from artificially induced abortions legally without gross malformations were studied using light and transmission electron microscopy to make clear the differentiation process of Leydig cells in humans. Leydig cells could be classified as 4 types: fusiform, light, dark, and degenerating cells. The most immature cell was the fusiform cells found frequently at 14 weeks, which seemed to differentiate successively into light and dark cells. Light cells were most frequently found at 17 weeks and dark cells at 24 weeks. Light cells were found to have mitochondria and smooth endoplasmic reticuli (sER) most prominently than any other cell types. The lumen of sER became to be expanded with age. Some electron-dense inclusions were observed in the mitochondrial matrix of the dark cells. Lipid droplets found more in light cells than dark cells were most prominent at 16 weeks and gradually decreased after 20 weeks. Glycogen particles were rich in dark cells. Degenerating cells were most frequently found at 27 weeks. From the results, it is suggested that Leydig cells in human fetal testes undergo similar differentiation process af in animals.
