An Experimental Study on the Antioxidant Effect of Electroacupuncture at the Waiguan($TE_5$) (외관전침의 항산화 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study is to observe the antioxidant effect of electroaupuncture at TE5 on the rats that were given AAPH(50mg/kg) everyday. Methodes : The Rats were given AAPH(50mg/kg) in abdominal cavity everyday for one week. $TE_5-NR$ group were treated by acupucture on left $TE_5$ for 15min. $TE_5-EA$ group were treated by electroacupucture on left $TE_5$ for 15min. The author observe several changes of rats. First, it is change of rat weight. Second, it is change of Liver index. Third, it is changes of albumin, total bilirubin, LDL-cholesterol, LDH, Glucose, GOT, GPT. Fourth, it is changes of SOD & Catalase activity, Glutathione & NO & MDA concentration. Fifth, it is change of tissue. Results: 1. In the $TE_5-EA$ group, the live index was decreased significant compared with control & holder group. 2. In the $TE_5-EA$ group, the albumin level were increased significant compared with control & holder group, LDL-cholesterol, GOT level were decreased significant compared with control & holder group. 3. In the $TE_5-EA$ group, the SOD activity, Catalase activity were increased significant compared with control group, Glutathione level was increased significant compared with control & holder, sham-EA, $TE_5-NR$ group, NO and MDA concentration were decreased significant compared with control group.

Effects of Electroacupucture on NMDA Receptor-dependent Spinal ERK MAPK Expression in CFA-induced Pain Model (전침에 의한 CFA유발 통증모델의 NMDA 수용체 의존적 ERK MAPK 발현 변화)

  • The present study aims to investigate a possible mechanism of electroacupuncture (EA) in the spinal dorsal horn that may underlie N-methyl-D-aspartate (NMDA) receptor-associated extracellular signal-regulated kinase (ERK) mitogen-activated protein kinase (MAPK) signaling pathways. The hot plate latency of the ipsilateral hindpaw of EA-treated rats was significantly decreased compared with complete Freund's adjuvant (CFA)-injected ones. The expressions of NR1 and NR2B subuint mRNA of NMDA receptor in the whole L4-5 segments are decreased by CFA treatment, but NR2B subunit was significantly recovered by EA treatment. When we detected the expression of ERK, there were no significant difference between normal and CFA-treated rats with EA or NMDA receptor antagonist MK801. But phosphorylated ERK expressions were markedly induced by CFA, but these inductions were significantly modulated by EA treatment. Although hosphorylation of ERK was also arrested by MK801, these inductions of CFA-injected rats was markedly inhibited only by co-treatment with EA and MK801. Phosphorylated cAMP response element-binding protein (CREB), ERK-related transcriptional factor, showed a significant increase in CFA-treated rats and this increase was slightly inhibited by EA and MK801 treatments. But immunoreaction for phosphorylated CREB were significantly increased by CFA treatment in the superficial laminae of the dorsal horn and these inductions were significantly arrested by co-treatment of EA and MK801. Consequently, the hyperalgesia induced by CFA are associated NMDA receptor and EA and MK801 may showed anti-hyperalgesia via same mechanism for inhibition of ERK and CREB phosphorylation in the dorsal horn.

  • Objectives : Acupuncture is expected to have somewhat like the efficacy parallel increasing activity of immune system in Western modem medicine. There, already, are many animal researches on activating effect of acupuncture for the immune system in peripheral organs. So, we carried out this experiment to see whether acupuncture has controlling effect on interleukin-6(IL-6) activity in rat's brain. Methods and Results : We had topical application of IL-6(1U=lpg, $10{\mu}l$) on brain of rat. It reduced afferent sensory transmission to the primary somatosensory(SI) cortex from periphery. Whereas, electrical stimulation(ES, 2Hz, 1.5V, 15min) of ST36(足三里) with application of IL-6 prominently activated afferent sensory transmission. ES of non-acupoint(proximal tail) with IL-6 showed suppression of afferent transmission. ES of ST36 without IL-6 application also exerted facilitation of afferent transmission to the SI cortex. Conclusions : Electoacupuncture(EA) on ST36 has noticeable influences on modulating activation of IL-6 in central nervous system, which do major role in immune system.
