Studies on the Pregnancy Diagnosis from Monoclonal Antigen of Progesterone (Progesterone Monoclonal Antigen에 의한 임신진단에 관한 연구)

  • This study was carried out to evaluate the ability of clinical application of pregnancy diagnosis based upon the determinatin of progesterone in milk, utilizing a chymosin inhibitor labelled with progesterone and monoclonal antibody to progesterone, and its compared with progesterone concentrations in the milk were assayed by radioimmunoassay. 1. The progesterone concentration of the pregnant cows (2.07$\pm$0.54ng/ml) were significantly higher than those of non-pregnant cows (1.04$\pm$0.19 ng/ml), and thereafter began to increase and maintained high levels. 2. During 20 to 22 days after artificial insemination, the accuracy of pregnancy diagnosis from monoclonal antigen of progesterone were 92.9% for non-pregnant cows, and 88.5% for pregnant cows. 3. During 20 to 22 days after artificial inseminatin, the accuracy of pregnancy diagnosis from milk progesterone concentrations were 92.9% for non-pregnant cows(<3.4ng/ml), and 92.3% for pregnant cows( 4.0ng/ml). The average overall accuracy of pregnancy prediction for pregnant and non-pregnant cows were 92.6%. 4. Accordingly, the pregnancy diagnosis from monoclonal antigen of progesterone is thought to be recommendable because this early diagnostic means are simple with accurate result.

  • Peritoneal pregnancy is an implantation in the peritoneal cavity exclusive of tubal, ovarian, or intra-ligamentary implantations. This is a rare obstetric complication with high maternal mortality and even higher perinatal mortality, and secondary type was most common. Risk factors for peritoneal pregnancy are previous history of extrauterine pregnancy or tubal surgery pelvic post-inflammatory status or presence of an intrauterine device. As it is a life-threatening condition, expectant management carries a risk of sudden life-threatening intra-abdominal bleeding and a generally poor fetal prognosis. So, when it is recognized, immediate termination of pregnancy is usually recommended. Early diagnosis of peritoneal pregnancy is difficult, but is important by their life threatening progress course to patients. Recently, we experienced primary peritoneal pregnancy which meets both the original and modified criteria. In this paper, we reported the case of early diagnosed and successfully treated peritoneal pregnancy despite of their diagnosis was incidentally.

  • Progesterone levels in the blood plasma or skim milk of cows are considered to be very useful indicator fur the detection of estrus cycle and early pregnancy diagnosis. During 13 to 14 days after estrus the level of progesterone in plasma or skim milk were not different between the inseminated arid non-inseminated cows. In the pregnant cows the peak level of progesterone reached on 14th day after artificial insemination (AI), but in the absence of conceptus the level declines after the 14th day slowly, and then very rapidly towards the basal level after the 17th day. This low level persists about 4 days, including those of estrus and ovulation a highly characteristic pattern which differs so markedly from that in the pregnant cows. Progesterone levels in blood plasma or skim milk can provided a reliable diagnosis of early pregnancy and monitoring ovarian activity in cows. The mean $\pm$ standard deviation of milk urea nitrogen(MUN) and protein concentration in the cows at 9 herds were 17.7$\pm$2.35 mg/dL and 3.2$\pm$0.17%, respectively. The days of nonpregnant after parturition was shorter in the cows in which the lower level of MUN than higher level of MUN concentration.

  • This stduy was carried out to evaluate the ability of clinical application of pregnancy diagnosis based upon the determination of progesterone in serum, utilizing EIA-kit of progesterone concentrations in the serum were assayed by radioimmunoassay. 1. The progesterone concentrations of the pregnant cows(2.40$\pm$0.34ng/ml) were significantly higher than those of non-pregnant cows(1.03$\pm$0.09ng/ml), and thereafter began to increase and maintained high levels. 2. During 20 to 22 days after artificial insemination, the accuracy of pregnancy diagnosis from EIA-kit of progesterone were 95.0% for non-pregnant cows, and 92.3% for pregnant cows. 3. During 20 to 22 days after artificial insemination, the accuracy of pregnancy diagnosis from serum progesterone concentrations were 100% for non-pregnant cows(<1.4ng/ml), and 96.2% for pregnant cows( 2.0ng/ml). The average overall accuracy of prediction for pregnant and non-pregnant cows were 98.1%. 4. Accordingly, the pregnancy diagnosis from EIA-kit of progesterone is thought to be recommendable because this early diagnostic means are simple with accurate results.

  • Combined intrauterine and extrauterine pregnancy is rare, occuring 1 in 30,000 pregnancies. The early diagnosis of combined pregnancy is very difficult, and so there is a higher maternal morbidity and fetal loss. Recently the incidence of combined pregnancy is increased. We experienced a case of combined intrauterine and right tubal pregnancy, and this case was presented with a brief review of the literatures.

Changes in Serum Concentration of Progesterone and Estrone Sulphate during Gestation in Korean Native Goats (한국재래산양의 임신기간중 혈중 Progesterone 및 Estrone Sulphate 농도의 변화)

  • This study was conducted to find out the changes of progesterone and estrone sulphate concentration in serum of Korean native goats throughout gestation, and to apply the serum levels of the hormones to pregnancy diagnosis. Serum concentration of progesterone and estrone sulphate were assayed by radioimmunoassay. Serum progesterone concentration was similar to its luteal phase values during early pregnancy and remained at the high level continually at 20∼140days and decreased rapidly at the day of parturition. Serum estrone sulphate concentration showed to increase markedly at 40∼50dyas gestation and steadily increased to the maximum of 7.13ng/ml at 140days, but declined sharply at the day of parturition. The accuracy of pregnancy diagnosis by the measurement of serum progesterone at 20∼24days after mating was 85.7∼92.3% and that of non-pregnancy diagnosis was 100%, when the serum progesterone levels higher and lower than 3.0ng/ml were supposed to indicate pregnancy and non-pregnancy, respectively. The accuracy of pregnancy diagnosis by the mearsurement of serum estrone sulphate was found to be nearly 100% since 50days after mating, when the serum levels of estrone sulphate higher then 0.5ng/ml were diagnosed to be pregnant. The optimal sampling time for pregnancy diagnosis was considered to be at 50 days after mating or to be later. It appears that estrone sulphate values above 7.0ng/ml at any time in gestation are highly indicative of twin. But there was found no significant difference(P<0.05) in serum estrone sulphate concentration and number of kids between does with single and twin kids.

  • Various early pregnancy diagnostic methods have been developed in order to improve the reproductive efficiency in cow, mare, mule, sow, sheep, goat, dog, cat, rabbit, buffalo, camel, elephant, monkey, deer, lion, coipus and guinea pig. These methods include abdominal swelling, abdominal palpation, esturs cylce detection, Lupin test, gonadotropin assay, colostrum injection test, sperm motility assessment, cervical mucus viscosity test, Kaber chromagens method, estrogen test, A Scheim-Zond다 test, spectrophotometric detection of estrogen in urine and feces, boric acid crystraline formation test in urine, oxytocin injection test, diamino-oxidase test, PMSG HA test, behaviour test, Simolus iodine detection test, detection of tryptophane in urine, x-ray method, Cuboni and Lunaas method, vaginal biopsy method, Friedmann Schneider diagnostic method, electrode method, barium chloride detection method, ECG, Doptone method, ultrasound method, ultrasound scanning method, LDH method, rectal palpation method, CL palpation method, radioautography, serum creatine test, serum globulin test, chlormadine method, CAP method, Medata Do, pp.ers method, body fluid test, Plasma oCS detection method, ERIA, LHRH method, negative latex cogulation test and oestrone sulphate detection method. The most reliable methods with high a, pp.icability to farm animals such as sheep, mare, sow and cow are rectal palpation, ultrasound method and hormonal assay in blood and milk. However, they require complicated laboratory works for the early diagnosis of pregnancy and in most cases, the simple and economical methods which are described up to now need a long period of time after conception. Generally, it is possible to detect pregnancy after one estrus cycle, even though it varies depending on the species of animals. For improvement of the reproductive efficiency, it is required to develop a more accurate, economical, simple and early detectable method. It is anticipated that the result of a study on the detection method of EPF(early pregnancy factor) would be a, pp.icable to various animals within 6 hours after conception.

  • Plasma progesterone concentrations were measured in 24 Korean native cows by a radioimmunoassay at 0, 6 and 20 days after insemination. As a result, 23 cows (95.8%) were judged to be in estrus with ovulation and one cow (4.2%) was judged to be in estrus without ovulation. Plasma progesterone concentrations at 20 days after insemination were $5.11{\pm}1.33$ (mean${\pm}$SD, range 3.20~7.86) ng/ml in pregnant cows and $0.35{\pm}0.10$ (range 0.30~0.50) ng/ml in nonpregnant cows. Of the 20 cows, 18 cows were confirmed the pregnant by rectal palpation at 60 days after insemination. The accuracy of the pregnancy diagnosis based on plasma progesterone concentration was 90 % for positive case and 100% for negative case.

  • Most progesterone enzyme immunoassays(EIA) are used liquid-phase double-antibody or single-antibody seperation. These methods consume considerable time and reagents because of the requirements for several washing and centrifugation steps involving the reactants. Because of these several problems, we were prompted to develop an effective enzyme-linked immunosorbent assay(ELISA) system that would be equal or superior to RIA for assay of progesterone. The results were obtained as follows. 1. Cross reaction of the progesterone antiserum with other steroids determined was shown with progesterone(100%), $11{\alpha}$-deoxycorti-costerone(2.271%), but the other steroids were shown below 0.9%. 2. Standard curve for progesterone ELISA was shown available difference according to progesterone concentration from 0 to 1,000pg/ml. 3. The lower limit of sensitivity was 0.2pg/well 4. Progesterone concentration was 1.6ng/ml for before parturition, and that was below 0.5ng/ml for after parturition. This development enzyme-linked immunosorbent assay for progesterone can be detected pregnancy diagnosis in cattle, and also applicable 10 research on physiological function including such as reproductive disorders.

Identification of plasma miRNA biomarkers for pregnancy detection in dairy cattle

  • A pregnancy diagnosis is an important standard for control of livestock's reproduction in paricular dairy cattle. High reproductive performance in dairy animals is a essential condition to realize of high life-time production. Pregnancy diagnosis is crucial to shortening the calving interval by enabling the farmer to identify open animals so as to treat or re-breed them at the earliest opportunity. MicroRNAs are short RNA molecules which are critically involved in regulating gene expression during both health and disease. This study is sought to establish the feasible of circulating miRNAs as biomarkers of early pregnancy in cattle. We applied Illumina small-RNA sequencing to profile miRNAs in plasma samples collected from 12 non-pregnant cows ("open" cows: samples were collected before insemination (non-pregnant state) and after pregnancy check at the indicated time points) on weeks 0, 4, 8, 12 and 16. Using small RNA sequencing we identified a total of 115 miRNAs that were differentially expressed weeks 16 relative to non-pregnancy ("open" cows). Weeks 8, 12 and 16 of pregnancy commonly showed a distinct increase in circulating levels of miR-221 and miR-320a. Through genome-wide analyses we have successfully profiled plasma miRNA populations associated with pregnancy in cattle. Their application in the field of reproductive biology has opened up opportunities for research communities to look for pregnancy biomarker molecules in dairy cattle.
