사료 공급율 및 공급횟수가 저수온기에 사육된 넙치 미성어의 성장 및 체조성에 미치는 영향 (Effects of Feeding Rate and Feeding Frequency on Growth and Body Composition of Sub-adult Flounder Paralichthys olivaceus in Suboptimal Water Temperature)

  • Two 15-week feeding trials were conducted to investigate feeding rate and feeding frequency for growth of sub-adult flounder in suboptimal water temperature ($12^{\circ}C$). In the first experiment, duplicated groups of fish (initial mean weight of 279 g) were fed a extruded pellet once a day at the different feeding rates of 0.1, 0.15, 0.2, 0.3% body weight per day (BW/d), and satiation (0.32% BW/d). Weight gain and feed efficiency were increased significantly (P<0.05) with increase in feeding rates from 0.1 to 0.3% BW/d, but there were no significant differences in weight gain and feed efficiency of fish fed the diet between 0.3% BW/d and to satiation. Crude protein content of dorsal muscle of fish fed the 0.1% BW/d diet was significantly (P<0.05) lower than that of the other groups. In the second experiment, duplicated groups of fish (initial mean weight of 281 g) were fed to apparent satiation with a extruded pellet at three different feeding frequencies (one meal every two days, one meal a day and two meals a day). Weight gain, feed efficiency and condition factor of fish fed one meal every two days were significantly (P<0.05) lower than those of fish fed one meal and two meals a day, but there were no significant differences in weight gain, feed efficiency and condition factor of fish fed between one meal and two meals a day. Based on the results of this study, optimum feeding frequency is one meal a day with satiation feeding for growth and feed efficiency of sub-adult flounder (270-350 g) in suboptimal water temperature ($12^{\circ}C$).

Effects of the Dietary Moisture Levels and Feeding Rate on the Growth and Gastric Evacuation of Young Olive Flounder Paralichthys olivaceus

  • This study determined the effects of the dietary moisture level and feeding rate on the growth and gastric evacuation of young olive flounder. Four experimental diets with different moisture levels (9%, 21%, 30%, and 40%) were prepared through the addition of water to the commercial extruded pellet. Three replicate groups of fish (initial weight: $106{\pm}1.4\;g$) were fed diets containing 9%, 21%, 30%, and 40% moisture to satiation or a moisture level of 9% and 30% at a restricted feeding rate (95% of satiation) for 15 weeks. The mean water temperature was $22{\pm}1.6^{\circ}C$ during the feeding trial. Gastric evacuation rates were determined post-feeding. The dietary moisture levels did not significantly affect weight gain, but the weight of the fish receiving 9% and 30% moisture diets to 95% satiation were significantly lower than those of the fish fed 9-40% moisture diets to 100% satiation (P<0.05). The feed efficiency, protein efficiency ratio, viscerosomatic index, and survival were not significantly affected by the dietary moisture levels and feeding rates. The daily feed intake of the fish fed to 100% satiation did not significantly differ among the treatment groups. The stomach contents that peaked within 3 h of feeding gradually decreased, and the stomachs of fish were completely evacuated within 18 h. The contents of the intestine peaked at 3-12 h post-feeding, and then declined with the intestine being mostly evacuated at 30 h. The moisture of the stomach contents reached approximately 70% within 3 h post-feeding and gradually increased to approximately 75% within 12 h. No considerable differences were observed in the gastric evacuation and moisture levels of the stomach contents in the fish fed the different diets. The results of this study suggest that the gastric evacuation of olive flounder was not affected by the dietary moisture level and that the addition of water into the diet displayed no beneficial effects on the growth of young olive flounder.

흰쥐에서 Divalent Metal Transporter 1의 조직내 분포와 Iron에 의한 조절 (Tissue Distribution of Divalent Metal Transporter 1 and Regulation by Dietary Iron in Rats)

  • Iron (Fe) is an essential metal in biological processes, which maintains a homeostasis in the human body. Divalent metal transporter 1 (DMT1) has been known as an iron transporting membrane protein, which is involved in the uptake Fe at the apical portion of intestinal epithelium, and may transport Fe across the membrane of acidified endosome in peripheral tissues. In this study, we studied the tissue distribution of DMT1 in the Fe supplemented (FeS) diet fed rats, and the regulation of DMT1 expression by depleting body Fe. Sprague-Dawley rats were divided into two groups, and fed FeS (120 mg Fe/kg) diet or Fe deficient (FeD, 2∼6 mg Fe/kg) diet for 4 weeks. The evaluation of body Fe status was monitored by measuring sFe, UIBC and tissue Fe concentration. Additionally, DMT1 mRNA levels were analyzed in the peripheral tissues by using the quantitative real time RT-PCR method. In the FeS diet fed rats, the tissue Fe was maintained at a relatively high level, and DMT1 was eventually expressed in all tissues studied. DMT1 was highly expressed in the testis, kidney and spleen, while a moderate levels of DMT1 expression was detected in the brain, liver and heart. In the digestive system, the highest level of DMT1 was found in the duodenum. Feeding the FeD diet caused a reduced body weight gain and depletion of body Fe with finding of decreased sFe, increased UIBC and decreased tissue Fe concentration. The depletion of body Fe upregulated DMT1 expression in the peripheral tissue. The expression of DMT1 was very sensitive to the body Fe depletion in the small intestine, especially in the duodenum, showing dramatically higher levels in the FeD rats than those of the FeS group. In the FeD diet fed animals, the expression of DMT1 was low significantly in other tissues compared with the duodenum. The expression of DMT1, however, was 60∼120% higher in the testis, kidney and spleen, and 30∼50% higher in the lung, liver and heart, compared to the FeS diet fed rats. In summary, DMT1 expression was ubiquitous in mammalian tissue, and the level of expression was the organ-dependent. The expression of DMT1 in peripheral tissues was upregulated by depletion of body Fe. Duodenum was the most sensitive tissue among organs studied during Fe depletion, and expressed the greatest level of DMT1, while other tissues were less higher than in duodenum. This study supports that DMT1 plays a role in maintaining the body Fe level through intestinal uptake as well as homeostasis of Fe in the peripheral tissue.

호주산(産) 밀의 제분(製粉) 특성(特性)과 밀가루의 물리화학적(物理化學的) 성질(性質)에 관(關)한 연구 (Milling Property of Australian Wheats and Physicochemical Properties of the Flours)

  • 호주산(産) 밀 6종류(種類)에 대(對)한 제분특성(製粉特性)을 $B{\ddot{u}}hler$ test mill로 조사(調査)하고 우리나라 재래종(在來種) 수원 219호와 비교(比較)하였다. 또한 이들로부터 얻어진 밀가루(patent flour)에 대한 반죽형성능력 및 전분(澱粉)호화성질을 Farinograph, Mixograph, Amylograph, 침강시험(沈降試驗) 및 Pelshenke 시험(試驗)등을 통하여 조사(調査)하였다. 호주산(産) 밀의 제분율(製粉率)은 $59{\sim}66%$ 범위(範圍)에 속하였다. 제분율(製粉率)은 천립중(千粒重)과 밀접(密接)한 관계(關係)를 나타내었으며 liter 중(重)과는 유의적 상관관계(相關關係)를 나타내지 않았다. 이들 밀로부터 얻어진 밀가루의 단백질 함량(含量)은 밀의 종류(種類)에 따라 14.47%에서부터 6.59%로 크게 상이(相異)하였으며 반죽의 물성(物性)도 밀의 종류(種類)에 따라 큰 차이(差異)를 보였다. Australian Prime Hard와 Australian Hard는 강력한 gluten 형성능력을 가진 반면 Australian Standard White와 Australian Soft Wheat는 매우 약(弱)한 gluten 형성능력(形成能力)을 보였다. 한편 Australian Standard White와 Western Australian Wheat는 대단히 높은 호화점도를 가지는 반면 South Australian Wheat는 이례적으로 낮은 호화점도를 보였다.

가금폐기물 처리를 위한 세균유래 케라틴 분해효소의 특성 (Characteristics of Bacteria-Originated Keratinase for Feather Waste Treatment)

  • Keratin wastes are generated in excess of million tons per year worldwide and biodegradation of keratin by microorganisms possessing keratinase activity can be used as an alternative tool to prevent environmental pollution. For practical use of keratinase, its physicochemical properties should be investigated in detail. In this study, we investigated characteristics of keratinase produced by Xanthomonas sp. P5 which is isolated from rhizospheric soil of soybean. The level of keratinase produced by the strain P5 increased with time and reached its maximum (10.6 U/ml) at 3 days. The production of soluble protein had the same tendency as the production of keratinase. Optimal temperature and pH of keratinase were $40^{\circ}C-45^{\circ}C$ and pH 9, respectively. The enzyme showed broad temperature and pH stabilities. Thermostability profile showed that the enzyme retained 94.6%-100% of the original activity after 1 h treatment at $10^{\circ}C-40^{\circ}C$. After treatment for 1 h at pH 6-10, 89.2%-100% of the activity was remained. At pH 11, 71.6% of the original activity was retained after 1 h treatment. Although the strain P5 did not degrade human hair, it degraded duck feather and chicken feather. These results indicate that keratinase from Xanthomonas sp. P5 could be not only used to upgrade the nutritional value of feather hydrolysate but also useful in situ biodegradation of feather.

Penicillium sp. 유래 Inulinase의 정제 및 특성 (Purification and Characterization of Inulinase from Penicillium sp.)

  • 열안정성 inulinase를 분비하는 Penicillium sp.를 돼지 감자 서식지의 토양으로부터 분리하여 이 균주의 inulinase를 50%-80% 염석, DEAE-Sephacel column chromatography, Toyopearl HW 65 F column chromatography에 의해 정제하여 inulinase 활성을 가진 단일 band를 얻었다. 이 band의 단백질을 polya-cryamide gel electrophoresis 후 추출하여 inulinase 활성을 가지는 것으로 확인되었다. 이 단백질의 분자량은 SDS-PAGE에 의해 약 77,000 dalton으로 추정되었고, 최종 정제도는 53.3배이었다. 정제된 효소의 효소학적 성질을 조사한 결과, 최적온도는 약 6$0^{\circ}C$이었고, 열 안정성은 30-5$0^{\circ}C$에서 비교적 안정하였다. pH 4에서 가장 높은 활성을 나타냈으며, pH 4-5에서는 비교적 안정했고 염기성에서는 아주 낮은 활성을 나타내었다. 금속이온과 다른 화학물질의 영향을 조사한 결과 1mM의 $MnCl_2$$CaCl_2$에 의해 활성이 증가했으며 특히, $MnCl_2$는 1.7배까지 활성을 증가시켰다. 그러나, 1mM의 $CuCl_2$,$HgCl_2$와 1mM EDTA 시에는 활성이 저해되었다. TLC분석결과 모든 산물이 monosaccharide였으므로 이 효소는 exo-acting inu-linase로 추정되었고, inulin에 대한 이 효소의 $K_M$치는 $2.2\times10^{-3}$이었다. 이 효소의 결정된 N-terminal 아미노산 배열은 $NH_2$-X-Glu-Tyr-Thr-Glu-Lys/Leu-Tyr-Arg-Pro이다.

국내 시판 엿기름의 품질 특성 분석 (Comparison of the Quality Characteristics of Commercial Barley Malts in Korea)

  • As basic research for optimal barley cultivars selection and technical development for quality maintenance, a total of 21 commercial malt products (for making 'Sikhye', a traditional Korean sweet drink) were collected from the Korean market. And then we analyzed the component of the barley malt products as well as conducted comparative analysis on enzyme activity and quality characteristics of the commercial barley malt products. Of the 21 barley malt products, 12 were made from 100 % barley. The result of analyzing general components of barley malts turned out different level of components; moisture 4.91~11.99%, lipid 1.71~2.48%, protein 7.80~11.97%, carbohydrate 73.64~82.24%, total starch 5.50~8.22%, reducing sugar 3.64~14.44%. As a result of measuring enzyme activity of barley malts, there was a wide range of activity difference by the product; diastatic power $36.80{\sim}94.30^{\circ}$, ${\alpha}$-amylase activity 18.17~186.50 unit/g, ${\beta}$-amylase activity 6.53~25.05 unit/g. The results of this study would be used as basic data for optimal cultivars selection to produce barley malts and technical development for quality maintenance.

Quality evaluation of local brand rice in rice exporting countries

  • This study evaluated the grain quality of local brand rice from 6 major rice exporting countries which are anticipated to export their rice to Korea. Recently, with the end of the postponement of rice import tariffs in 2014, Korea is in a very easy environment to import foreign rice. Therefore, the quality evaluation of local brand rice in those countries is needed to secure the quality competitiveness of Korean rice, also to protect the rice industry in Korea. We provided total 38 local brand rice from USA(7), China(16), Australia(4), Thailand(3), Vietnam(5) and India(3), and 2 imported brand rice through MMA from USA and China to find out the status of the grain quality for each country. For the quality evaluation, we analyzed the physicochemical properties, milling and palatability-related characteristics. The amylose content on country average ranged from 24.4(India)~16.2%(Thailand). The protein content was 6.66% by overall average, and was higher in order of India(7.86), Australia(6.80), Vietnam(6.61), Thailand(6.59), China(6.28), USA(5.82). In Toyo glossiness value, it ranged from 75.7~45.2, and the figures in USA and China were the highest level. The head rice ratio ranged from 95.2~72.4%, and the figures in Thailand, USA and China were distinctly high. When we analyze the palatability of boiled brand rice with Chucheongbyeo as check variety by expert panelists, several rice brands from USA and China showed equal or better scores in shape, smell, taste, stickiness, texture and overall score, although most rice brands showed a tendency to decrease significantly in the taste characteristics compared with Chucheongbyeo. From the above results, it can be seen that the grain quality of USA and China rice is very competitive when imported into Korea. The results will provide basic information for the quality control of foreign rice which will be imported into Korea in the near future, also for the quality information which could be applied on the development of high-quality Korean rice varieties. Continuous monitoring about the foreign brand rice is advisable to improve the quality competitiveness of Korean rice.

젖소 유방염에 대한 오존가스의 치료효과 (Therapeutic Effect of Ozone Gas on Bovine Mastitis)

  • 오존가스의 젖소 유방염에 대한 치료효과를 구명할 목적으로 본 실험을 수행하였다. 만성유방염에 이환된 비유우 18분방을 대상으로 하였다 실험분방은 대조군(항생제 투여군: norfloxacin 10g/tube, 2회/일, 3일간: 5분방), 실험군 I (오존가스 0.1 ppm, 250m1, 2회/일, 7일간: 5분방) 및 실험군 II(오존가스 1 ppm, 50m1, 2회/일, 3일간: 8분방)로 각각 나누어 공시하였다. 우유 중 체세포수, 혈액충 백혈구수, 호중구수, 림프구수, 호중구/림프구(N/L)비 및 혈청 단백량의 변화를 처치 전 및 처치 후 7일에 각각 비교 검토하였다. 그 결과, 실험군 1에서는 오존가스 주입 후 7일에 주입 전에 비하여 우유 중 체세포수가 감소하였으나 대조군 보다 높은 수치를 나타내었다. 실험군 II에서는 오존가스 주입 후 7일에 주입 전에 비하여 현저한 감소소견을 나타내었으며(p<0.05), 대조군 보다 낮은 수치를 나타내었다. 그러나 대조군 및 실험군 간의 유의성은 인정되지 않았다 또한 말초혈액 백혈구수, 호중구수, 림프구수, N/L비 및 혈청 단백량의 변화에서는 대조군 및 실험군에 있어서 각각 유의한 변화를 나타내지 않았다. 이상의 결과를 종합해 볼 때, 오존가스는 젖소 유방염의 치료에 유효할 것으로 판단된다.

사료내 산패 지질 및 비타민 E 첨가가 넙치 치어의 성장 및 체조성에 미치는 영향 (Effect of Dietary Oxidized Squid Liver oil and DL-${\alpha}$-Tocopherol Level on Growth and Body Composition of Juvenile Flounder (Paralichthys olivaceus))

  • This study was conducted to investigate the effect of dietary oxidized oil and ${\alpha}$-tocopherol level on growth and body composition of juvenile flounder. To prepare oxidized diets, squid liver oil was oxidized by aeration at $25^{\circ}C$ for 30 days. The six diets were prepared to contain 6% fresh or oxidized squid liver oil as the lipid sources in combination with three levels of ${\alpha}$-tocopheryl acetate at 0, 80 and 800 mg/kg diet. Triplicate groups of fish ($3.9{\pm}0.1$) were fed to apparent satiation twice a day for 8 weeks. Survival was not significantly different among treatments. Weight gain, feed efficiency, daily feed intake, protein efficiency ratio and condition factor of fish fed the fresh oil diets were significantly higher than those of fish fed the oxidized oil diets (P<0.05). The increase of the vitamin E level in diets did not result in any significant improvement on growth performance of fish fed both oil diets. The vitamin E content of the liver and dorsal muscle increased with increasing dietary vitamin E level at both oil diet groups. A decreasing trend in vitamin E content of the tissues was observed in fish fed the oxidized oil diets at the same dietary vitamin E level. Significantly higher moisture content and lower crude lipid content were observed in the whole body of fish fed the oxidized oil diets than fish fed the fresh oil diets (P<0.05). Dietary lipid source affected the fatty acid content of the whole body; higher contents of saturated and monoenoic fatty acids, and lower n-3 HUFA contents such as 20:5n-3 and 22:6n-3 were observed in fish fed the oxidized oil diets than those of fish fed fresh oil diets. The results of this study suggest that the dietary oxidized oil may impair the growth performance, and an increase in ${\alpha}$-tocopheryl acetate supplementation have no beneficial effect on growth and feed efficiency of juvenile flounder.
