  • The production of inulooligosaccharides from inulin by a dual endoinulinase system of Pseudomonas sp and Xanthomonas sp. was investigated the optimum conditions for a dual endoinulinase reaction were as follows : pH,5.8; temperature, $50^{\circ}C$; substrate concentration, 50 g/l; enzyme ratio, 3:1 as Xanthomonas endoinulinase to Pseudomonas endoinulinase. Under optimum conditions, the maximum yield of oligosaccharides was 90.5% in total sugar basis by dual endoinulinase system

  • Phytase increases the availability of phosphate and trace elements by hydrolyzing the phosphomonoester bond in phytate present in animal feed. It is also an important enzyme from an environmental perspective because it not only promotes the growth of livestocks but also prevents phosphorus contamination released into the environment. Here we present a novel phytase derived from Turicimonas muris, TmPhy, which has distinctive structure and properties compared to other previously known phytases. TmPhy gene expressed in the Pichia system was confirmed to be 41 kDa in size and was used in purified form to evaluate optimal conditions for maximum activity. TmPhy has a dual optimum pH at pH3 and pH6.8 and exhibited the highest activity at 70℃. However, the heat tolerance of the wildtype was not satisfactory for feed application. Therefore, random mutation, disulfide bond introduction, and N-terminal mutation were performed to improve the thermostability of the TmPhy. Random mutation resulted in TmPhyM with about 45% improvement in stability at 60℃. Through further improvements, a total of three mutants were screened and their heat tolerance was evaluated. As a result, we obtained TmPhyMD1 with 46.5% residual activity, TmPhyMD2 with 74.1%, and TmPhyMD3 with 66.8% at 80℃ heat treatment without significant loss of or with increased activity.

Continuous Production of Fructooligosaccharides from Sucrose by a Dual Immobilized Enzyme System of Fructosyltransferase and Glucose Isomerase (과당전이효소와 포도당 이성화 효소의 고정화 혼합효소계에 의한 설탕으로부터 프락토올리고당의 연속생산)

  • Continuous production of fructooligosaccharides from sucrose by a dual immobilized enzyme system of fructosyltransferase and glucose isomerase was studied in a column reactor. The optimal temperature and pH of the immobilized fructosyltransferase were $65^{\circ}C$ and 5.5, respectively. The activity of glucose isomerase was favorable as temperature and pH were increased within the ranges examined. However, both the immobilized enzymes were thermally unstable over $5^{\circ}C$, suggesting that long-term operation of the dual immobilized enzyme column should be carried out below $50^{\circ}C$. The optimum packing ratio of fructosyltransferase to glucose isomerase was found to be around 5/3. Under the optimized reaction conditions, the dual enzyme column was successfully operated for 40 days without any loss of initial enzyme activities, yielding 66% of fructooligosaccharides. Furthermore, the relative sweetness of fructooligosaccharides produced by a dual emzyme system was enhanced by 6% compared with that of fructosyltransferase alone.

  • A biosensor was prepared with dual cathode electrode and immobilized enzyme membrane. A nylon net was used for the immobilization of glucose oxidase and alcohol oxidase. The immobilized enzymes were placed on the surface of the electrode which was prepared with one anode and two cathodes as an oxygen electrode. The determination of components by the biosensor was based on the consumption of dissolved oxygen. The optimum condition of this system was 0.1 M potassium phosphate buffer solution, pH 7.5 at $35^{\circ}C$. Glucose and ethanol in takju were simultaneously determined by the biosensor. Comparing with UV-spectrophotometer and gas chromatograph for cross checking, there was a good correlation between the biosensor and the conventional methods. Biosensor with dual cathode electrode required no clarification or pretreatments. It was used for simultaneous determination of glucose and ethanol during the fermentation of takju.

The Production and Enzymatic Properties of Extracellular Chitinase from Pseudomonas stutzeri YPL-1, as a Biocontrol Agent

  • An antagonistic bacterium Pseudomonas stutzeri YPL-1 liberated extracellular chitinase and $\beta$-1,3-glucanase which are key enzymes in the decomposition of fungal hyphal walls. The lytic enzymes caused abnormal swelling and retreating at the hyphal tips of plant pathogenic fungus Fusarium solani in a dual culture. Scanning electron microscopy revealed the hyphal degradation of F. solani in the regions interacting with P. stutzeri YPL-1. The production of chitinase and properties of a crude preparation of the enzyme from P. stutzeri YPL-1 were investigated. Peak of the chitinase activity was detected after 4 hr of cultivation. The enzyme had optimum temperature and pH of 50$^{\circ}C$ and pH 5.3, respectively. The enzyme was stable in the pH range of 3.5 to 6.0 up to 50$^{\circ}C$. The enzyme was significantly inhibited by metal compounds such as $HgCl_2$, but was stimulated by $CoCl_2$. P. stutzeri YPL-1 produced high levels of the enzyme after 84 hr of incubation. Among the tested carbon sources, chitin was the most effective for the enzyme production, at the concentration level of 3%. As a source of nitrogen, peptone was the best for the enzyme production, at the concentration level of 4%. The maximum amount of enzyme was produced by cultivating the bacterium at a medium of initial pH 6.8.

The role and characterization of .betha.-1, 3-glucanase in biocontrol of fusarium solani by pseudomonas stutzeri YPL-1

  • An antifungal Pseudomonas stutzeri YPL-1 produced extracellular chitinase and .betha.-1, 3-glucanase that were key enzymes in the decomposition of fungal hyphal walls. These lytic extracellular enzymes markedly inhibited mycelial growth of the phytopathogenic fungus Fusarium solani. A chitinase from P. stutzeri YPL-1 inhibited fungal mycelial growth by 87%, whereas a .betha.-1, 3-glucanase from the bacterium inhibited growth by 53%. Furthermore, co-operative action of the enzymes synergistically inhibited 95% of the fungal growth. The lytic enzymes caused absnormal swelling and retreating on the fungal hyphal walls in a dual cultures. Scanning electron microscopy clearly showed hyphal degradation of F. solani in the regions interacting with P. stutzeri YPL-1. In an in vivo pot test, P. stutzeri YPL-1 proved to have biocontrol ability as a powerful agent in controlling plant disease. Planting of kidney bean (Phaseolus vulgaris L.) seedlings with the bacterial suspension in F. solani-infested soil significantly suppressed the development of fusarial root-rot. The characteristics of a crude preparation of .betha.-1, 3-glucanase produced from P. stutzeri YPL-1 were investigated. The bacterium detected after 2 hr of incubation. The enzyme had optimum temperature and pH of 40.deg.C and pH 5.5, respectively. The enzyme was stable in the pH range of 4.5 to 7.0 and at temperatures below 40.deg.C, with a half-life of 40 min at 60.deg.C.

  • For the simultaneous methane recovery and $CO_2$-stripping, we have been developed dual vent auto circulation bubble lift column reactor, and evaluate optimum conditions for monoethanolamine (MEA) solutions as a $CO_2$ absorbent. At the 5 wt% MEA solution, we investigated the pH change during $CO_2$-stripping and absorption reaction, $CO_2$-stripping rate with reaction time, methane recovery efficiency for various inflow rates of air, $CO_2$-stripping rate for flow liquid over flow height, and $CO_2$-stripping dependency on the temperature of absolvent solutions. The suggested optimum conditions for $CO_2$ recovery with MEA in the dual vent auto circulation bubble lift column reactor were 40 mm over flow liquid height, 1.5 L/min of air inflow rate, and $25^{\circ}C$ of absorbent solution temperature.

Study on the fabrication and the growth mechanism of Bi-2223 superconducting phase by diffusion method (확산법에 의한 Bi-2223 초전도상의 제조 및 성장기구에 관한 연구)

  • According to spread volume of B(BiPbCuO) layer, composition ratio and each stage of sintering process, we studied stability of high Tc superconductor phase and generation and growth movement of superconducting phase. The dual layer composed of SrCaCuO and BiPbCuO compound were prepared to develop the Bi-2223 superconductor[108K] through interaction and diffusion during sintering process. The dual layer samples were sintered at 830.deg. C for 0-210 hours. From the result, the optimum conditions were : spread volume(A:B=1:0.6), sintering time(210h) and composition ratio(A:S $r_{2}$C $a_{2}$C $u_{2}$- $O_{x}$, B:B $i_{1.9}$P $b_{0.5}$C $u_{3}$ $O_{y}$) at 830.deg. C.. C.C.C.

  • The purpose of this study is to investigate the effect of various types of coagulant on dewaterability and settleability of sewage sludge for the application of dewatering process. Cationic organic coagulants and inorganic coagulants of the aluminium base were used; PAC (Poly Aluminium chloride, $Al_2O_3$ 17%) and C-210P (0.2%). After Jar test, PAC 26 mg/L and 0.2% C-210P 55 mg/L was decided as the optimum concentration of the coagulant according to zeta potential measurement. pH, alkalinity and viscosity were measured in all experiments and the data on sedimentation characteristics is analyzed by SDI, SVI sedimentation rate and solid flux. The SRF(Specific Resistance of Filtration) experiment was conducted with the result of single and dual injection system as the dewaterability experiment. As a result, the organic coagulant making large floc has good characteristics of sedimentation and agglutination. Also, it is observed that the organic coagulants injection has a better dewaterability efficiency of coagulants under the condition of the lowest SRF value, followed by dual and inorganic coagulants injection.

Control of Odor Emissions Using Biofiltration: A Case Study of Dimethyl Disulfide

  • A laboratory- scale dual-column biofilter system was used to study the biofiltration of dimethyl disulfide (DMDS). The gas flow rate and DMDS concentration to the biofilter were varied to study their effect on the remov-al of dimethyl disulfide. Operating parameters such as pH, temperature, and water content were monitored during the biofilter operation and necessary precautions were taken to keep these parameters within the acceptable limits. It was observed that the removal efficiency of DMDS was optimal at neutral pH values. After five month op-eration, the neutralization of the filter beds with sodium carbonate became necessary for the optimum operation of the biofilters. The microbial population already present in the compost mixtures was found to be adequate in treat-ing DMDS. The compost mixtures were found to be similar in terms of biofiltration efficiency of DMDS. However, pressure drops observed in the first column compost mixture (compost/ peat mulch) was extremely high, making this compost economically not feasible. The second mixture (compost/bark) provided pressure drops within accept-able limits. A minimum residence time of 30 seconds at the optimal operating conditions appeared to be adequate for achieving high removal efficiencies (>90%).
