Taxol Production in Taxus spp. Cell Culture : Semisynthesis of Taxol from Baccatin III in Cell Cultures of Taxus baccta Pendula (주목 세포배양에 의한 Taxol 생산 : 유럽 주목의 세포배양에서 추출한 Baccatin III로부터 Taxol의 반합성)

  • Anticancer agent taxol was synthesized from baccatin III which was extracted from cell cultures of European yew, Taxus baccata Pendula. Callus and suspension cultures of T. baccata Pendula showed the formation baccatin III. The content of baccatin III in cell cultures reached 0.015% (w/w) on dry weight basis. The semisynthetic approaches were made with baccatin III and taxol side chain. To prepare taxol side chain, (-)-N-((S)-2-hydroxy-1-phenylethyl) benzamide was synthesized first from (S)-(+)-phenylglycine. Then (-)-N-((IS,2S)-2-hydroxy-1-phenyl-3-butenyl) benzamide was synthesized with vinyl magnesium bromide. The synthesis of (2R, 3S)-(-)-2-(1-ethoxyethoxy)-3-phenyl-3-(phenylmethanamido) propanoic acid with RuCl3, catalyst was the final step to prepare taxol side chain. The semisynthetic reactions yielded 0.0002% taxol, 0.0005% 7-epi-10-deacetyltaxol, and unidentified taxanes on dry weight basis. It is suggested that the semisynthesis of taxol from baccatin III could be an alternative source of taxol and related taxanes.

High-Level Expression of Aspergillus ficuum Acetyl Xylan Esterase Gene in Pichia pastoris, (Pichia pastoris에서 Aspergillus ficuum 유래 Acetyl Xylan Esterase 유전자의 과발현)

  • Acetyl xylan esterase gene (AXE) from Aspergillus ficuum was cloned and its Pichia expression plasmid, pPICZ$\alpha$C-AXE (4.6 kb), was constructed, in which the AXE gene was under the control of the AOXI promoter and connected downstream of mating factor u-1 signal sequence. The plasmid linearized by Sacl was integrated into the 5'AOXI region of the chromosomal DNA of P. pastoris. In the flask batch culture of P. pastoris transformant on methanol medium, the cell concentration and total AXEase activity reached at 6.0 g-dry cell weight/1 and 77 unit/ml after 36 h cultivation, respectively. In the fed-batch culture employing the optimized methanol and histidine feeding strategy, the cell concentration and total AXEase activity were significantly increased to about 97 g-dry cell weight/1 and 930 unit/ml. Most of AXEase activity (>90%) was found in the extracellular medium and the majority of extracellular protein (>80%) was AXEase enzyme (33.5 kDa). This result means that about 9.8 g/1 of AXEase protein was produced in the extracellular medium.

Effects of Sucrose Concentration on the Production of hGM-CSF in Transgenic Plant Cell Suspension Culture (형질전환된 식물세포에서 Sucrose 농도가 hGM-CSF 생산에 및 미치는 영향)

  • The effects of sucrose concentration on the secretion of hGM-CSF, total protein and protease into the medium were investigated in transgenic tobacco cells. The dry cell weight (11.22 g/L), hGM-CSF (181.53 $\mu\textrm{g}$/L) and total protein (66.8 mg/L) were detected as highest at 30 g/L sucrose and protease activity (2660 U/L) was highest at 120 g/L sucrose after 5-day culture. However after 10-day culture, the maximum dry cell weight (28.36 g/L) was found at 60 g/L sucrose while the maximum hGM-CSF (95 $\mu\textrm{g}$/L) was appeared at 150 g/L sucrose. The total protein and protease activity was secreted as 52.28mg/L and 3430 U/L, respectively in the same culture.

Influence of Thickness of Styrofoam-Panel on Tobacco Seedling Growth in Newly Developed Floating System (새로 개발된 부상형 육묘에 있어서 스티로포움 판의 두께가 연초 묘 생육에 미치는 영향)

  • The inluence of styrofoam-panel thickness of floating frame on the seedling growth of flue-cured tobacco(Nicotiana tabacum L.) in the newly developed direct-seed float system was investigated . Floating frame used instead of styrofoam-tray consisted of styrofoam-panel (106×108cm) for floating, eight holes(1.Sx2S.0 cm) to uniform the water, and nutrition supply far seedling and water and nutrition absorption cloth placed on the styrofoam-panel. Each floating-frame may lay eight plastic-trays on it. Eight grades of styrofoam-panel thickness from 26 mm to 46mm and two kinds of media were used in the greenhouse system. Dry cells were found from the styrofoam-panel thickness of 43mm in carbonized chaff, compost and original soil rate of L3-3(v/v) media, and of 37mm in peat and perlite rate of 8-2(v/v) media. The thinner styrofoam-panel produced more tender and succulent seedling with the more trunk and the less root weight. When considering the appearance of dry cell, seedling growth, producing healthy tobacco seedlings, and utilization of styrofoam-panel for two kinds of medias the ideal styrofoam-panel thickness were suggested to be around 34mm for flue-cured tobacco reeling production in the newly developed direct-seeding float system.   • It was reported that the digestion ratio of chlorella was low because it had a low metabolic rate in body. Generally, the thickness of a cell membrane of it is $200-250\;{\AA}$, the weight of it is approximatly 13% of the total weight of a dry cell. And it is composed of protein, lipid, hemicellulose and ash etc. So, in order to elevate the digestion ratio of chlorella in body, we experimented the crude treatment methods of chlorella. The results obtained in this experiment are summarized as follows : 1. The digestion ratios calculated from ordinary N- balance method were 83.05% for 10% chl. (b) plus diastase group ; 81.25% for 10% chl. (b) plus amylase group, and 79.23% for 10% chl. (b), 58.55% for 10% chl. (a). 2. Biological values from this method were 80.25% for 10% chl. (b) plus diastase group, and 60% for 10% for chlorella(a).

RNA Production from the Hydrolyzate of Sliced and Dried Sweet Potatoes. (절간고구마를 이용한 RNA 생산에 관한 연구)

  • During an extensive screening tests of yeasts for their RNA formation, it was found that Cryptococcus laurentii had especially high RNA content and high dry cell weight, when hydrolyzate of sliced and dried sweet potatoes was used as a carbon source. Growth conditions of this strain were examined, and the most desirable results were obtained at 48 hours of cultivation on a reciprocal shaker at 3$0^{\circ}C$ with initial pH 6.0. Under the above conditions, the RNA content and yield of dry cells were investigated using various media compositions. Ammonium sulfate 0.40%, peptone 0.6 %, and yeast extract 0.4% were appeared to be favorable as a nitrogen sources. The optimum concentrations of K $H_2$P $O_4$, M $n^{++}$, C $O^{++}$ were 0.05 %, 0.1 %, and 0.001 %, respectively. Ca-pantothenate, 400$\mu\textrm{g}$/$m\ell$, showed relatively favorable effects as a growth factor. The maximum RNA content obtained in this study was 16.8 % of the total dry cell weight.t.t.

Production of a Platelet Aggregation Inhibitor, Salmosin, by High Cell Density Fermentation of Recombinant Escherichia coli

  • Optimal conditions for a high cell density fermentation were investigated in a recombinant Escherichia coli producing salmosin, a platelet aggregation inhibitor. The optimized carbon and nitrogen sources were glycerol 10 g/l, yeast extract 30 g/l, and bacto-tryptone 10 g/l, yielding the dry cell weight (DCW) of 10.61 g/l in a 500 ml flask culture. The late-stage induction with 1% L-arabinose in a 5 l jar fermentor showed the highest DCW of 65.70 g/l after 27 h of the fed-batch fermentation. Around 2,200 mg/l of the protein was expressed as an inclusion body that was then refolded to obtain the active salmosin of 96 mg/l. We also confirmed the inhibitory activity against platelet aggregation of the active salmosin from the high cell density fermentation.

Production of Lactic Acid from Cheese Whey by Repeated Batch and Continuous Cultures

  • This study is concerned with development of efficient culture methods for lactic acid fermentation of Lactobacillus sp. RKY2. The cell-recycle repeated batch fermentation using cheese whey and corn steep liquor as raw materials was tried in order to further enhance the productivity of lactic acid. In addition, fermentation efficiencies could be considerably enhanced by cell-recycle continuous culture. Through the cell-recycle repeated batch fermentation, lactic acid productivity was maximized to 6.34 $g/L{\cdot}h,$ which corresponded to 6.2 times higher value than that of the batch fermentation. During the cell-recycle continuous fermentation, the last dry cell weight at the end of fermentation could be increased to 25.3 g/L.

Continuous Fermentationof L-Lysine by Immobilized Corynebacterium glutamicum (Corynebacterium glutamicum 고정화균체에 의한 L-라이신 연속발효)

  • For the improvement of L-lysine productivity, development of the continuous fermentation system by a bioreactor assembly was attempted. Primarily, optimal conditions on the whole cell immobilization of Corynebacterium glutamicum ATCC21514 were studied and 76.2% of immobilization ratio was obtained when the cells were entrapped with 4% k-carrageenan showing 4.0kg gel strength. A bioreactor system was set up using the immobilized cells was applied for the continuous production of L-lysine. The results obtained under the optimum conditions were compared with those of the batchwise fermentation. Experimental results obtained from 14 day continuous fermentation showed 36.7% of sugar conversion to L-lysine while the productivity of L-lysine was disclosed as 4.96mg/ml mg-dry cell weight /hr which is 2.5times and 4.1 times higher than those of the batchwise fermentation by the intact cells and by the immobilized cells, respectively.

The Combined Effects of Carbon Dioxide Concentration and Irradiation on Growth of the Green Alga Haematococcus pluvialis

  • The biological fixation of carbon dioxide using microalgae have many advantages over chemicals and remove carbon dioxide simultaneously. A ketocarotenoid astaxanthin is hyper-accumulated in the green freshwater microalga, Haematococcus pluvialis. In the present study, the combine effects of carbon dioxide concentration and light intensity on the growth of H. pluvilais were investigated. The carbon dioxide concentration above 10% caused a severe inhibition and around 5% is optimal for growth. Adaptation to high concentration of carbon dioxide enhanced the $CO_2$ tolerance. Specific growth rate calculated differently based upon cell number or dry weight because of the distinctive life cycle patterns of H. pluvialis : small-sized motile green cell and thick cell walled red cyst cell. Based on the light dependence of H. pluvialis, internally illuminated air-lift photobioreactor was designed and operated. Gradual increase of light supply gave more active growth and more effective productivity of astaxanthin than constant light supply.
