건강 에틸아세테이트 분획의 예쁜꼬마선충 내의 항산화 효과 (Anti-oxidative Activity of Ethyl acetate Fraction of the Dried Ginger in Caenorhabditis elegans)

  • Ginger (Zingiber officnale Roscoe, Zingiberaceae), which is a well-known food seasoning, has been used as a traditional medicine for the treatment of gastrointestinal disorder, vomiting and cough in Korea, China and Japan. Ethanol extract from the dried ginger (DG) was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions the ethyl acetate soluble fraction (EDG) showed the most potent DPPH radical scavenging and superoxide quenching activities. To know the effect of antioxidant activities of EDG, we tested the activities of superoxide dismutase (SOD) and catalase together with oxidative stress tolerance and intracellular ROS level in Caenorhabditis elegans. To investigate whether EDG-mediated increased stress tolerance was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain including CF1553. Consequently, EDG elevated SOD and catalase activities of C. elegans, reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, EDG-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.

중국산 생강의 올레오레진 제조를 위한 극초단파 추출조건 설정 (Pre-establishment of Microwave-Assisted Extraction Conditions for Chinese Ginger Oleoresins)

  • 중국산 건생강으로부터 올레오레진을 추출하고자 극초단파를 이용하여 몇 가지 기초실험을 실시하였다. 극초단파(2,450 MHz, 50 W) 추출에서 원료의 입자크기는 60 mesh 내외, 시료와 용매의 비는 1:10 (g/mL) 범위가 적당한 것으로 나타났다. 에탄올 가용성 고형분은 에탄올농도가 낮을수록 증가하였으나 추출물의 전자공여능은 에탄올 25%에서 가장 높게 나타났고, 추출물의 총페놀 함량은 에탄올 농도 50% 이상에서 급속히 증가되었다. 건생강 올레오레진의 성분들은 추출시간에 따라 함량이 증가하였으나 7분 이상의 극초단파 처리는 효과적이지 않았다. 따라서 극초단파를 이용하여 생강으로부터 고품질의 올레오레진을 추출하기 위해서는 추출공정 중 에찬올 농도에 따른 추출특성과 최적화 연구가 필요하다고 생각된다.

건조채소(乾燥菜蔬)의 수분측정방법(水分測定方法) 비교(比較) (Comparisons of Measurement Methods of the Moisture Content of Dried Vegetables)

  • Measurement methods of moisture content were compared with 7 dried vegetables (red pepper, onion, green onion, garlic, ginger, carrot and radish). The moisture contents of dried vegetables having different moisture contents were determined by atmospheric oven drying, infrared balance, vacuum oven and Karl Fisher methods. Vacuum oven and Karl Fisher methods gave the relatively agreed results and considered to give the accurate moisture content. Atmospheric oven drying and infrared balance methods resulted in higher moisture content than methods mentioned above, because of the thermal decomposition of solid. Calibration of the moisture data of atmospheric oven drying method into the vacuum oven data was undertaken. The thermodecomposable solid fraction was high in onion, radish, green onion and carrot, and was in the range of 8.0-11.7% of the total solid in these products.

건강 Ethyl acetate 분획의 선충에 대한 수명연장 효과 (Longevity Property of Ethyl acetate Fraction of Dried Zingiber officinale in Caenorhabditis elegans)

  • This study was performed to evaluate the lifespan extending effects of the dried ginger (Zingiber officnale Roscoe, Zingiberaceae). Ethanol extract of the dried ginger was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions, and lifespan-extending effects of the fractions were investigated using Caenorhabditis elegans model system. Ethyl acetate-soluble fraction showed the most potent longevity property. Furthermore, ethyl acetate-soluble fraction elevated thermal stress tolerance, and increased expression of stress resistance protein. Further studies were performed to investigate several aging-related factors such as reproduction, food intake, growth and movement of C. elegans. The results revealed that there were no significant changes in aging-related factors including reproduction, food intake and growth, however, ethyl acetate-soluble fraction treatment led to an up-regulation of locomotory ability of aged worms, suggesting ethyl acetate-soluble fraction affected healthspan as well as lifespan of nematode.

흑생강(Kaempferia parviflora)의 항응고 및 혈소판 응집저해 활성 (Anti-coagulation and Anti-platelet Aggregation Activities of Black Ginger (Kaempferia parviflora))

  • 흑생강은 생강과 초본과 뿌리식물로 태국 및 라오스에서는 크라차이담으로 불리며, 뿌리는 향신료와 차로 이용되어 왔다. 말린 뿌리는 위장장애, 통풍, 이질, 알러지 치료 및 강장용으로 전통적으로 이용되어 왔으며, 최근에는 항비만, 항산화, 항염증, 혈전 용해 활성 등의 다양한 유용 생리활성이 보고되어 있다. 본 연구에서는 흑생강의 혈액순환 개선 활성을 확인하기 위해, 흑생강 지하부의 ethanol 추출물 및 열수 추출물을 조제하고 각각 이의 순차적 유기용매 분획물인 hexane 분획물, ethylacetate (EA) 분획물, butanol 분획물 및 물 잔류물을 조제하여 각각의 성분을 분석하였고, 혈전 생성과 관련된 항응고 활성, 혈소판 응집저해 활성 및 적혈구 용혈활성을 평가하였다. 그 결과, ethanol 추출물의 EA 분획물 5 mg/ml 농도에서 TT, PT, aPTT를 각각 1.22, 1.49 및 15배 이상 연장시켜 강력한 항응고 활성을 나타냄을 확인하였으며, ethanol 및 열수 추출물의 EA 분획물 모두에서 동량의 아스피린보다 강력한 혈소판 응집저해 활성을 확인하였다. 또한 상기 EA 분획물들은 0.5 mg/ml 농도까지 인간 적혈구에 대한 용혈활성을 나타내지 않았다. 본 연구 결과는 흑생강의 혈전 생성 억제 활성에 대한 최초의 보고이며, 흑생강의 EA 분획물이 인간 적혈구 용혈활성 없이 강력한 항응고 및 혈소판 응집저해 활성을 나타내어 신규의 항혈전제로 이용 가능함을 제시하고 있다.

Physicochemical Properties of Repetitive Heat-treated Ginger and Its Quantitative Conversion of Gingerol to Shogaol

  • Ginger was steamed at $121^{\circ}C$ and $1.5lb/in^2$ for 30 min, dried at $60^{\circ}C$ for 12 h, and each step was repeated nine times. During processing, the lightness ($L^*$ value) and yellowness ($b^*$ value) decreased from $85.65{\pm}0.33$ and $26.99{\pm}0.20$ in the non-treated ginger to $56.91{\pm}0.25$ and $16.69{\pm}0.06$ in ginger treated for the ninth treatment. On the other hand, redness ($a^*$ value) increased from $-1.51{\pm}0.03$ to $7.34{\pm}0.08$ on the eight treatment and then decreased to $7.21{\pm}0.04$ on the ninth theatment. The contents of 6-gingerol decreased from $3.257{\pm}0.067mg/g$ in the non-treated ginger to $0.567{\pm}0.036mg/g$ on the theatment, whereas the contents of 6-shogaol increased from $1.299{\pm}0.050mg/g$ to $2.999{\pm}0.089mg/g$ on the sixth treatment and decreased to $2.099{\pm}0.039$ on the ninth treatment. The contents of 10-gingerol decreased slightly from $1.106{\pm}0.125mg/g$ to $0.806{\pm}0.026mg/g$. Unlike the 6- and 10-gingerol, the contents of 8-gingerol did not change greatly, with values between $0.916{\pm}0.005mg/g$ and $1.106{\pm}0.005mg/g$ being observed during processing. The tyrosinase inhibitory activities were increased from $43.42{\pm}11.45%$ in the non-treated ginger to 100% on the sixth treatment and then decreased to $51.98{\pm}7.36%$ on the theatment. The antioxidative activity was retained during processing.

생강 클로로포름 분획의 활성화된 뇌신경교세포(腦神經膠細胞)에서 염증반응 억제효과 (Chloroform Fraction of Zingiberis Rhizoma Recens Modulates the Production of Inflammatory Mediators in LPS-stimulated BV2 Microglial Cells)

  • Objectives : The root of Zingiber officinale ROSC. (Zingiberis Rhizoma Recens; Ginger) has been widely used as one of folk remedies and food materials in many traditional preparations. Ginger is known as an effective appetite enhancer and anti-inflammatory agent. This study was performed to investigate the effect of ginger chloroform fraction (GCF) in microglia which play a central role on brain inflammation in neurodegenerative diseases. Methods : Dried ginger was extracted with 80% methanol, and then fractionated with chloroform. BV2 mouse microglial cells were cultured with different concentrations of GCF and then stimulated with LPS (1 ${\mu}g/m{\ell}$) at indicated times. The cell toxicity of GCF was determined by MTT assay. The concentrations of NO, PGE2 and cytokines were measured by Griess assay and enzyme-linked immunosorbant assay. The mRNA and protein expressions of iNOS, COX-2 and cytokines were determined by RT-PCR and Western blotting. The phosphorylation of three MAPKs (p38 MAPK, ERK1/2 and JNK) and $NF-{\kappa}B$ activation were determined by Western blotting. Results : GCF significantly inhibited LPS-induced production of inflammatory mediators, NO, $PGE_2$ and proinflammatory cytokines ($TNF-{\alpha}$ and $IL-1{\beta}$) in a dose-dependent manner. GCF attenuated LPS-induced expression of mRNA and protein of inflammatory enzymes, iNOS, COX-2 and proinflammatory cytokines through suppressing the phosphorylation of ERK1/2 and p38 MAPK and the activation of p65 $NF-{\kappa}B$ in BV2 cells. Conclusions : This study suggests that GCF may have an anti-inflammatory property through suppressing the inflammatory mediator production released by activated microglia after the brain injury.

건조방법에 따른 생강분말의 흡습특성과 예측모델에 관한 연구 (Absorption Characteristics and Prediction Model of Ginger Powder by Different Drying Methods)

  • 열풍, 진공 및 동결건조한 생강분말의 흡습특성을 조사하고 저장 중 수분함량 예측모델식을 수립하였다. 단분자층 수분함량은 BET식보다 GAB식이 더 높은 유의성을 보였으며 등온흡습곡선은 sigmoid 형태로서 건조방법에 따른 차이는 없었으나 낮은 온도에서 평형수분함량이 높았다. 건조방법에 따른 흡습엔탈피는 큰 차이가 없었으나 수분활성도가 증가함에 따라 낮은 흡습에너지를 보였다. 등온흡습곡선의 적합도는 Caurie 모델이 $R^2\;0.99$ 이상으로 높은 적합도를 나타내었고 그 외 Henderson, Bradley 모델의 순으로 높은 적합도를 나타내었으며, 편차도 Caurie 모델이 $1.2{\sim}5.4%$로 가장 낮았다. 수분함량 예측을 위해 각 독립변수의 최적함수로 시간은 ln 함수, 수분활성도와 온도는 일차식을 선정하였으며 수분함량 예측모델식은 건조방법에 관계없이 시간과 수분활성도, 온도로 수립한 모델식의 적합도가 높았다.

수종한국상용식품중(數種韓國常用食品中)의 Tryptophan 및 Available lysine 함량(含量)에 대(對)하여 (A Study on the Contents of Tryptophan and Available Lysine in Korean Foods)

  • In order to observe the contents of some essential amino acids in Korean foods, total and free tryptophan, and available lysine in fifty kinds of Korean foods were analyzed by the sfectrophotometry. The results obtained are summarized as follows : 1) The tryptophan contents per 100g of soybean, wet green laver and skim milk were over 250 mg, and the contents Per g nitrogen in ginger, dried persimmon and chestnut were over 150 mg. 2) The free tryptophan contents per 100 g of soybean, small red bean, dried persimmon, ginseng stem were over 100 mg, ana the contents per g nitrogen in dried persimmon was over 150 mg. 3) The available lysine contents Per 100g of soybean, wet green laver and skim milk, fish(auchovy), dried yeast, casein and silkworm pupa were over 1000 mg, and the contents per g nitrogen in potato, perilla(wild sesame), red pepper, sausage and skim milk were over 300mg. 4) The contents of tryptophan and available lysine in soybean, green laver and skim milk were higher than in other samples. 5) In general, the contents of tryptophan and available lysine were abundant in seaweeds.

김치 첨가가 수프의 맛에 미치는 영향 (Effects of Kimchi Addition on the Sensory Quality of Soup)

  • 김치가 첨가된 수프를 제조하기 위하여 1$0^{\circ}C$에서 10~15일 숙성된 김치를 동결 건조하여 수프 제조에 사용하였다. 수프는 밀가루를 17$0^{\circ}C$의 오븐 속에서 2시간 동안 볶아 갈색을 낸 후 1,000 g을 계량하여 버터 140 g을 혼합하고 고운 체에 내려 쇠고기와 생강, 마늘, 양파 등의 야채와 수분이 있는 재료는 후라이팬에 볶아 익힌 후 수분을 제거하여 김치수프를 제조하였다. 관능검사의 결과에서는 대조군에서 버터향이 고소하고 부드러운 맛에 영향을 나타내었으나 전반적으로 김치 분말 20%를 첨가한 시험구에서 맛, 색, 향의 기호도가 최적점을 나타내었으므로, 김치수프를 만드는데는 김치를 1$0^{\circ}C$에 저장하면서 2주정도 경과한 김치를 동결건조 한 다음 분말화하여 건조 김치채와 분말 20% 첨가하는 것이 가장 적당하였다.
