Vibrio vulnificus, a Gram-negative bacterium found in estuarine waters, is responsible for over 95% of all seafood-related deaths in the United States. As a result of a temperature downshift to 5$^{\circ}C$, this organism enters the viable but nonculturable (VBNC) state. Changes in the membrane fatty acid (FA) composition of V. vulnificus may be a contributing factor to the ability of this organism to enter into and survive in the VBNC state. This hypothesis was tested by incubating the organism at 5$^{\circ}C$ in arti-ficial sea water and analyzing the cells' FAs during the initial hours of temperature and nutrient down-shift. Prior to downshift, the predominant FAs were 16:0, 16:1 and 18:0. During the first four hours of downshift, statistically significant changes occurred in 15:0, 16:1, 16:0, 17:0, and 18:0. These results indicate that changes in FA composition occur prior to entry of V. vulnificus into the VBNC state, suggesting that the ability to maintain membrane fluidity may be a factor in this physiological response. Cells in which fatty acid synthesis was inhibited did not survive, indicating that active fatty acid metab-olism is essential for entry of cells into the VBNC state.

The propagation characteristics of high power microwave pulse in an air-breakdown environment are examined. The maximum electron density produced by microwave air-breakdown is limited to $10^6cm^{-3}$ by the tail-erosion effect. Inorder to increase the electron density, the scheme using two pulses intersecting at a desired height is considered. Increasing the carrier frequency, it is shown that microwave pulse can be transferred without the serious erosion in the numerical simulation. This result is useful for the above scheme. Also, an experiment is conducted to show the tail-erosion effect and confirm that a rapidly generated lossy plasma can cause spectral breaking and frequency shift of a high-power microwave pulse. The experimental results are presented by comparing the frequency spectrum of an incident pulse with that of the pulse transmitted through a self-induced air-breakdown environment. The experimental results show that the amount of frequency upshift is co-related with the ionization rate, whereas that of frequency downshift is correlated with the energy losses from the pulse in the self-generated plasma.
Ingestion of shellfish-associated Vibrio parahaemolyticus is the primary cause of potentially severe gas-troenteritis in many countries. However, only Kanagawa phenomenon (hemolysin) positive (KP$\^$+/) strains of V. parahaemolyticus are isolated from patients, whereas >99% of strains isolated from the environment do not produce this hemolysin (i.e. are KP$\^$-/). The reasons for these differences are not known. Following a temperature downshift, Vibrio parahaemolyticus enters the viable but noncultur-able (VBNC) state wherein cells maintain viability but cannot be cultured on routine microbiological media. We speculated that KP$\^$+/ and KP$\^$-/ strains may respond differently to the temperature and salinity conditions of seawater by entering into this state which might account for the low numbers of cul-turable KP$\^$+/ strains isolated from estuarine waters. The response of eleven KP$\^$+/ and KP$\^$-/ strains of V. parahaemolyticus following exposure to a nutrient and temperature downshift in different salinities, similar to conditions encountered in their environment, was examined. The strains included those from which the KP$\^$+/ genes had been selectively removed or added. Our results indicated that the ability to produce hemolysin did not affect entrance into the VBNC state. Further, VBNC cells of both biotypes could be restored to the culturable state following an overnight temperature upshift.

Compressed Sensing(CS) is the theory that can recover signals which are sampled below the Nyquist sampling rate to original analog signals. In this paper, we propose the estimation algorithm of ultrasonic attenuation coefficients in the frequency domain using CS. While most estimation algorithms transform the time-domain signals into the frequency-domain using the Fourier transform, the proposed method directly utilize the spectral information in the recovery process by the basis matrix without the completely recovered signals in the time domain. We apply three transform bases for sparsifying and estimate the attenuation coefficients using the Centroid Downshift method with Dual-reference diffraction compensation technique. The estimation accuracy and execution time are compared for each basis matrix. Computer simulation results show that the DCT basis matrix exhibits less than 0.35% estimation error for the compressive ratio of 50% and about 6% average error for the compressive ratio of 70%. The proposed method which directly extracts frequency information from the CS signals can be extended to estimating for other ultrasonic parameters in the Quantitative Ultrasound (QUS) Analysis.
Low-temperature adaptation and protection for environmental stresses were studied in the gram-negative soil bacterium Bradyrhizobium japonicum 61A101c. B. japonicum was more resistant to alcohol, $H_2O_2$, heat and freezing following a pretreatment at $4^{\circ}C$, resulting in approximately 10 to 1,000 folds increased survival compared to mid-exponential-phase cells grown at an optimal temperature at $28^{\circ}C$. This phenomena relate to the cold shock protein expressed when cells are exposed to a downshift in temperature. To confirm the presence of cold shock protein genes in B. japonicum, a PCR strategy was employed using a degenerate primer set, which successfully amplified a putative csp gene fragment. Sequence analysis of the PCR product(200bp) revealed csp-like sequences that were up to 96% identical to csp gene of S. typhimurium.

In this paper, the control strategy of ratio changing system for a metal belt CVT adopting primary pressure regulation is developed, and the shirting performance of pressure regulating type CVT with the suggested control strategy is investigated. The control strategy suggested in this study is composed of 2 feedback loop, one is speed ratio feedback and the other is primary pressure feedback. The pressure feedback is adopted to ensure prohibiting a belt slip during transient period in a fast downshift mode. Simulation results show that the system with suggested control strategy gives appropriate response time and tracking Performance for upshift and also gives a proper primary pressure which can prohibit the belt slip. In addition, it is fecund that the given system has an acceptable servo property in tracking the target speed ratio and robustness for the disturbance of line pressure.
In this paper, thrust equations for a rubber belt CVT are derived by considering the geometry and mechanism of the mechanical actuators. In order to solve the thrust equations, an algorithm to calculate the speed ratio is suggested for the given driver speed and load torque based on the actuator characteristic equations and existing formula for the belt thrust forces. Experiments are performed to investigate the driver speed-load torque-speed ratio characteristics at a steady state. The speed and torque efficiencies are measured and used to modify the actuator equations. It is found that the modified equations well predict the steady state characteristics. In addition, the shift dynamic model for a rubber belt CVT is derived experimentally. Simulation results of the CVT shift dynamics are in good accordance with the experiments and it is noted that different coefficients are required to describe the CVT shift dynamics for the upshift and the downshift.
Sporulation of Streptomyces griseus NRRL B-2682 occurs at 26~28 hours during incubation while shaking at 3$0^{\circ}C$ in a defined medium. The time of sporulation is the same when the levels of each nutrient is increased ten times. The levels of the carbon, nitrogen and phosphate source are at a high level when sporulation begins. Sporulation of S. griseus B-2682 is clearly not caused by nutrient deprivation. It appears that a clock mechanism is involved instead. Once spores are germinated, the time of sporulation is programmed. Sporulation of S. griseus is repressed by high levels of casein hydrolysate. A study of the effect of individual amino acids revealed that L-valise when added to the normal growth medium causes an inhibition or repression of sporulation without affecting growth.

Dynamic models of line regulator valve(LRV) and ratio control valve (RCV) are obtained for an electronic controlled CVT. LRV and RCV are operated by variable force solenoid(VFS). Considering the CVT shift dynamics, oil pump's efficiency and saturation characteristics of VFS, simulations are performed and compared with test results. Simulation results are in good agreement with the experiments, which shows the validity of the dynamic models of LRV and RCV obtained. In addition, the effects of the orifice size in the exhaust port of RCV are investigated. Simulation results show that as the orifice size decreases, the residual pressure in the primary actuator increases which insures the large torque transmission capacity, meanwhile the duration time for the downshift increases.

This paper reports a new method to enable the normally-off operation of AlGaN/GaN heterojunction field-effect transistors (HFETs). A capacitor was connected to the gate input node of a normally-on AlGaN/GaN HFET with a Schottky gate where the Schottky gate acted as a clamping diode. The combination of the capacitor and Schottky gate functioned as a clamp circuit to downshift the input signal to enable the normally-off operation. The normally-off operation with a virtual threshold voltage of 5.3 V was successfully demonstrated with excellent dynamic switching characteristics.
