Purpose : To investigate the Presence of adaptive response by low dose radiation in murine tumors in relation to radiation induced apoptosis as well as related mechanism. Materials and Methods : Syngeneic murine tumors, OCa-I and HCa-l, were given 0.05 Gy pretreatment followed by therapeutic dose of 25 Gy radiation. Induction of apoptosis was analyzed for each treatment group. Regulating molecules of apoptosis, p53, Bcl-2, Bax, Bcl-X, were also analyzed by Western blotting. Results : In 0.05 Gy pretreatment group of OCa-I, 25 Gy-induced apoptosis per 1000 cells was 229, which was estimated at $30\%$ lower level than the expected (p<0.05). In contrast, this reduction in radiation induced apoptosis was not seen in HCa-l. In the expression of apoptosis regulating molecules, p53 increased in both tumors in response to radiation. Bcl-2 and Bax did not show significant change in both tumors however, the expression of Bcl-2 surpassed that of Bax in 0.05 Gy pretreatment group of OCa-l. Bcl-X was not expressed in OCa-l. In HCa-l, Bcl-X showed increased expression even with 0.05 Gy. Conclusion : Adaptive response by low dose radiation Is shown in one murine tumor, OCa-l, in relation to radiation induced apoptosis. Apoptosis regulating molecules including Bcl-2/Bax and Bcl-X, appear to related. This study shows an evidence that adaptive response is present, but not a generalized phenomenon in vivo.
Background: Rapid advances in neuroendocrine immunology have established the concept of bidirectional communication between the immune and neuroendocrine systems. Capsaicin suppresses the immune function by destroying substance P acting as mediatior of neuroendocrine immune system. Methods and Results: In this study, effect of capsaicin on mature murine lymphocyte functions and lymphoid tissue morphology was examined. Formally, capsaicin showed the strong cytotoxic effect on splenocyte over $10{\mu}g/ml$ concentration in citro. And proliferation and Th1-cytokine expression of splenic cells in mice that received high dose of capsaicin ($100{\mu}g/mouse$) were significantly diminished. However, low dose of capsaicin treatment did not influence these responses in vivo($1{\mu}g/mouse$) and in vitro (under $5{\mu}g/ml$). And the morphology of spleen and lymph nodes after capsaicin treatment was observed. In the spleen of mice injected with high dose of capsaicin (100, $200{\mu}g/mouse$), the size of white pulp was significantly decreased and the length of red pulp was increased, Moreover, vascularity index was diminished in a dose dependent manner. Conclusion: These results implies that immunosuppressive effect of capsaicin is associated with cytotoxic activity on lymphocyte, Th1-cytokine down-regulation and lymphoid tissue abnormalization, and this report is expected to give a hand to the study for the mechanism of action of neurotoxin of the immune system.
Sulfated fucans are known to have both anti-thrombotic and anti-coagulant activities. In this study, the variation in platelet aggregation and anti-coagulant activities was investigated in vitro with regard to administered dose, molecular weight distribution, sulfate content, and sugar composition in two algal fucoidans from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu). The anti-coagulant activity largely correlated with sulfate content and with molecular weight distribution in a dose-dependent manner. However, both fucoidans demonstrated inhibitory responses to ADP-induced platelet aggregation in dose- and structure-dependent manners that contrasted with the anti-coagulant activity. Neither molecular weight distribution nor sulfate content greatly affected platelet-aggregation inhibition (PA-inhibition) by the fucoidan fractions, whereas anti-coagulant activity was sensitive to these structural factors. Interestingly, an E. bicyclis fucoidan fraction exhibited almost complete PA-inhibition at a treatment dose of 500 mg/mL while retaining weak anti-coagulant activity. In conclusion, these observations suggest that fucoidan may be a useful anti-thrombotic or anti-platelet agent in various arterial thrombotic disorders, including post-vascular intervention with controlled bleeding complications, due to its anti-coagulant modulating activity.

A study of the immunogenicity and reactogenicity of two doses of lot H(10, 20 mcg), two doses of lot L (20, 40 mcg) of the Smith Kline-RIT recombinant DNA yeast-derived hepatitis B vaccine and a 20-mcg dose of the Merck Sharp and Dohme plasma-derived hepatitis B vaccine was conducted in young adults under randomized, double-blind conditions. Immunization was carried out according to a 0-, 1-, and 6-month vaccination schedule. Results indicated that the yeast-derived hepatitis B vaccine was well tolerated and immunogenic. Reactogenicity to both yeast- and plasma-derived vaccines was mild in severity and low in incidence with no significant differences appearing between the study groups. One month after the third dose, the yeast-derived vaccines induced a high degree of soroconversion ranging between 95.0% and 100%. The response was not lot or dose-dependent. The administration of the plasma-derived vaccine resulted in anti-HBs geometric mean titres statistically signifirantly higher than those elicited by the different yeast-derived hepatitis B vaccines one month after the third dose of vaccine but the difference was not large enough to be of great clinical significance.
Purpose: To evaluate the clinical outcomes of symptomatic bone lesions in patients with multiple myeloma (MM) who received local radiotherapy (LRT). Materials and Methods: Fifty-one patients with 87 symptomatic bone lesions treated via LRT were analyzed. LRT was delivered at a median total dose of 21 Gy (range, 12 to 40 Gy) in a median of 7 fractions (range, 4 to 20 fractions). The clinical outcomes of LRT and the factors affecting treatment response were assessed. Results: After a median follow-up time of 66.7 weeks, symptom relief was achieved for 85 of 87 lesions (97.7%). The median time to symptom relief was 7 days from the start of LRT (range, 1 to 67 days). The duration of in-field failure-free survival ranged from 1.1 to 450.9 weeks (median, 66.7 weeks). The radiation dose or use of previous and concurrent chemotherapy was not significantly associated with in-field failure for LRT (p = 0.354, 0.758, and 0.758, respectively). Conclusion: Symptomatic bone lesions in patients with MM can be successfully treated with LRT. A higher radiation dose or the use of concurrent chemotherapy may not influence the in-field disease control. A relatively low radiation dose could achieve remission of symptoms in patients with MM.
We investigated the combinatorial effects of different doses of dietary soy isoflavones (SI) and fructooligosaccharide (FOS) in a rat model of colon cancer. We hypothesized that increased bioavailability of SI metabolites due to dietary FOS may increase production of bioactive equol and affect colon carcinogenesis in a dose-dependent manner. Sprague-Dawley male rats were injected with 12-dimethylhydrazine (DMH) and were providec experimental diets that contained 0, 10, 50, 150, or 500 mg SI per kg of diet and 6% FOS for 12 weeks. The number of aberrant crypt foci (ACF) and the expression of cyclooxygenase-2 (COX-2) in colonic tissues were significantly decreased in the 6% FOS-fed groups compared to the control group. Gut transit time and fecal pH were significantly lower, and fecal concentrations of bifidobacteria were increased with 6% FOS. However, dietary SI supplementation in combination with 6% dietary FOS did not affect ACF formation or COX-2 expression. Plasma equol concentrations were dose-dependently increased by supplementation of SI up to 500 mg/kg of diet. In conclusion, SI supplementation up to 500 mg/kg of diet appeared to have no additive beneficial effects in rats with chemically-induced colon cancer that were fed 6% FOS, although plasma equol was dose-dependently increased.
Dose response for the nuscheduled DNA synthesis induced by various concentration of MMS was dose dependent and directly proportional to dose increased. Time dependence of unscheduled DNA synthesis was continued up to 4 hours, with the peak appearing 2$\\sim$3 hours after treatment with MMS and $^3 H$-thymidine labeling. Single treatment with BUdR or IUdR does not induce unscheduled DNA synthesis. BUdR and IUdR greatly enhanced MMS-induced unscheduled DNA synthesis, but the dose responses were different from that of single treatment with MMS. IUdR was found to be more effective sensitizer on MMS-induced unscheduled DNA synthesis.
