Optimization for Enzymatic Hydrolysis of Mannitol (만니톨의 효소 가수분해 반응 조건 최적화)

  • This study aimed to investigate the enzymatic hydrolysis of mannitol using Viscozyme$^{(R)}$ L, Celluclast$^{(R)}$ 1.5 L, Saczyme$^{(R)}$, Novozym$^{(R)}$, Fungamyl$^{(R)}$ 800 L, Driselase$^{(R)}$ Basidiomycetes sp., and Alginate Lyase, and to optimize of reaction conditions for production of reducing sugar. Response surface methodology (RSM) based on central composite rotatable design was used to study effects of the independent variables such as enzyme (1-9% v/w), reaction time (10-30 h), pH (3.0-7.0) and reaction temperature ($30-70^{\circ}C$) on production of reducing sugar from mannitol. The coefficient of determination ($R^2$) of $Y_1$ (yield of reducing sugar by Viscozyme$^{(R)}$ L) and $Y_3$ (yield of reducing sugar by Saczyme$^{(R)}$) for the dependent variable regression equation was analyzed as 0.985 and 0.814. And the p-value of $Y_1$ and $Y_3$ showing 0.000 and 0.001 within 1% (p < 0.01), respectively, was very significant. The optimum conditions for production of reducing sugar with Viscozyme$^{(R)}$ L were 9.0 % (v/w) amount of enzyme, 30.0 hours of reaction time, pH 4.5 and $30.0^{\circ}C$ of reaction temperature, and those with Saczyme$^{(R)}$ were 9.0% (v/w) of amount of enzyme dosage, 30.0 h of reaction time, pH 7.0 and $30.0^{\circ}C$ of reaction temperature, consequently, the predicted reducing sugar yields were 22.5 and 27.9 mg/g-mannitol, respectively.

  • In the era of building engineering the intensification of Self Compacting Concrete (SCC) is world-shattering magnetism. It has lot of rewards over ordinary concrete i.e., enrichment in production, cutback in manpower, brilliant retort to load and vibration along with improved durability. In the present study, the mechanical strength of CM-2 (SCC containing 10% of rice husk ash (RHA) as cement replacement and 600 grams of glass fibers per cubic meter) was investigated at various dosages of cement replacement by fly ash (FA) and GGBS. A total of 17 SCC mixtures including two control SCC mixtures (CM-1 and CM-2) were developed for investigating fresh and hardened properties in which, ten ternary cementitious blends of SCC by blending OPC+RHA+FA, OPC+RHA+GGBS and five quaternary cementitious blends (OPC+RHA+FA+GGBS) at different replacement dosages of FA and GGBS were developed with reference to CM-2. For constant water-cement ratio (0.42) and dosage of SP (2.5%), the addition of glass fibers (600 grams/m3) in CM-1 i.e., CM-2 shows lower workability but higher mechanical strength. While fly ash based ternary blends (OPC+RHA+FA) show better workability but lower mechanical strength as FA content increases in comparison to GGBS based ternary blends (OPC+RHA+GGBS) on increasing GGBS content. The pattern for mixtures appeared to exhibit higher workablity as that of the concentration of FA+GGBS rises in quaternary blends (OPC+RHA+FA+GGBS). A decrease in compressive strength at 7-days was noticed with an increase in the percentage of FA and GGBS as cement replacement in ternary and quaternary blended mixtures with respect to CM-2. The highest 28-days compressive strength (41.92 MPa) was observed for mix QM-3 and the lowest (33.18 MPa) for mix QM-5.

  • Ginsing polysaccharide, known to have an immune regulating effect, was administered to 23 randomly selected healthy male subjects with a mean age of 23 years in accordance with an IRB approval. Then, these subjects underwent physico-chemical tests and serum proteome was analyzed from the blood sample taken from these subjects. Analyses of proteome involved image analysis, protein sections and protein identification in sequence after two-dimensional electrophoresis was carried out. During the physico-chemical test, 4 subjects were excluded from the study. In the proteome analysis, identified were 5 spots such as SP40, 40, Cytokeratin 9, hypothetical protein LOC544932, Apolipoprotein E ,similar to Human albumin, which showed differences in the amount of protein expression. In conclusion, changes of 5 proteins were remarkable before and after administration of ginsing polysaccharides. In certain cases, hepatic and renal slight injury occurred. Thus, further clinical study on dosage regimen would be necessary for securing the basis for concentration-dependent effectiveness and safety.

Flocculation of Red Tide Organisms in Sea Water by Using an Ignited Oyster Shell Powder and Loess Combination (소성굴패각분말과 황토의 동시 사용에 의한 적조생물의 응집)

  • This study determined the optimum dosage for coagulation reactions of red tide organisms (RTO) using a combination of ignited oyster shell powder (10sp) and loess and examined the electrokinetic and rheological characteristics of their flocs. Two kinds of RTO, Cylindrotheca closterium and Skeletonema costatum, were sampled in Masan Bay and cultured in the laboratory. Coagulation experiments were conducted using various concentrations of IOSP, loess, IOSP+1oess, RTO, and a jar tester RTO cell numbers were counted for both the supernatant and RTO culture solution. The removal rates increased rapidly with increasing IOSP concentrations up to 50 mg/L and loess concentrations up to 800 mg/L. A removal rate of $100\%$ was reached at 400 mg/L of IOSP and 6,400 mg/L of loess. The highest increment $(16.7\%)$ of the rates of coagulation reaction occurred using both IOSP and loess (50+200 mg/L) in comparison with IOSP alone. The rate of coagulation reaction using both IOSP and loess (50+200 mg/L), $90.6\%,$ was similar to employing either IOSP of 150 mg/L or loess of 3,200 mg/L. All of the coagulation liquids for RTO, IOSP (200 mg/L), loess (200 ma/L), and IOSP+1oess (200+200 mg/L) revealed non-Newtonian fluid properties and therefore their shear rate vs. shear stress curves were non-linear. The coagulation liquids revealed elastic body properties at a lower shear rate increasing in the following order: RTO, IOSP (200 mg/L), loess (200 mg/L), and IOSP+1oess (200+200 mg/L. IOSP+1oess (200+200 mg/L) especially demonstrated plastic flow properties at a lower shear rate.

  • This paper investigates the fundamental properties and shrinkage characteristics of low shrinkage high performance concrete(LSHPC), using mock-up specimens. According to the test results, the most suitable mix proportions of LSHPC need a higher dosage of SP agent and AE agent, in order to obtain the target of slump flow and air content. This is due to reduce fluidity and air content respectively. It also presented earlier setting time than control concrete by 6 hours and exhibited compressive strength of 60MPa at age 28 days. Autogenous shrinkage of LSHPC was the half of the value of control concrete. Drying shrinkage of center section of LSHPC showed similar tendency with autogenous shrinkage, because of no internal moisture movement, while surface section had larger drying shrinkage. The specimen with embedded reinforcing bar had smaller deformation owing to confinement of reinforcing bar.

First Infestation Case of Sarcoptic Mange from a Pet Rabbit Oryctolagus Cuniculus in Republic of Korea

  • In November 2019 a 5-month-old mixed-breed rabbit presented to Chungbuk National University Veterinary Teaching Hospital, Cheongju-si, Chungbuk, Republic of Korea (Korea) with symptoms comprising pruritus, crusts on skin, poor appetite and reduced defecation. The rabbit was purchased 2 months prior from a pet shop located in a big market, and that the symptoms were first observed about 2 weeks prior to the hospital visit. Physical examination revealed that the patient had crust formation and alopecia on the nose together with lesions on the digits. A skin scraping test was performed using mineral oil and a high density of mites was observed by microscopy. Each mite showed a round, tortoiselike body with 4 comparatively short pairs of legs. The anus was located at the terminal unlike with suspected pathogen, Notoedres cati. Based on morphological characteristics, we identified the mite as Sarcoptes sp. Ivermectin was administered weekly by subcutaneous injection at a dosage of 0.4 mg/kg, and 4 weeks of follow-up study revealed the patient was fully recovered. And no more mites were detected from the case. This is the first case report of sarcoptic mange in a pet rabbit in Korea.

  • Two vibrio sp. strains were isolated from disease catfish(silurus asotus). The present isolates were identified as Vibrio ordalii based on their biological and biochemical characteristics ; they were positive for acid production from glucose, Iactose, maltose, sucrose and salicine, while negative for arabinose, galactose, inocitol and xylose. They are named KL-1 and KL-2, KL-1 and KL-2 strains were similar to physiological characteristics ; growth was observed at pH 5 to 10 and in 0% to 6.0% NaCl. Two strains did not growth at a concentration above 7.0% NaCl and pH10. This bacterium was infected into health catfish hypodermically. Such injection was found to induce haemorrhagic ulcers very similar to those observed in naturally infected fish. At 24h post-infection, the red spot developed around the injection site and grew bigger to from a red sport area. At 120h post-infection, the muscle ncerosis was extended near the ventral fin. The seventy percent lethal dosage was appeared to water temperature at $25^{\circ}C$. Two strains were tested for drug senistiveity by plate method. KL-1 and KL-2 strains were sensitive to GM. K, N, S and SxT, but resistant to CF, L and VA.
