Changes in Allergenicity and Quality of Nuruk during Fermentation (전통 누룩 발효과정 중 품질 및 항원성 변화)

  • Wheat is the most widely cultivated cereal and an important source of dietary protein worldwide. Wheat allergy, defined as an adverse immunologic reaction to wheat, encompasses a broad spectrum of disorders with different pathomechanisms and clinical manifestation. The Nuruk, a traditional Korean Koji for brewing, was made with wheat flour and fermenting microbes such as bacteria, yeast and mold. The strains grown on Nuruk secrete various enzymes as amylase and protease. By the activation of such enzymes, starch and proteins in Nuruk are hydrolyzed to sugar and amino acid. Therefore, it is supposed to reduce allergic proteins in wheat. To study quality properties and degradation degree of allergenicity in Nuruk by fermentation, we investigated the changes of general ingredients and allergenicity in Nuruk during fermentation. Moisture contents was decreased from 24.2% to 13.6% during fermentation. Crude lipid and protein contents were gradually increased during fermentation. After 15 days of fermentation, reducing sugar and total sugar contents were reached its maximum level, and they were 27.45% and 39.00%, respectively. Acid and neutral protease activity were significantly increased during fermentation, but alkaline protease activity was not detected. ${\alpha}$-amylase activity was gradually increased and showed maximum level about 2,833.00 U/g after 15 days of fermentation. Glucoamylase activity was the highest level about 497.9 U/g after 10 days of fermentation. The increase of these proteolytic and saccharogenic enzyme activities will provide efficient condition for production of rice wine. Also, protein fractions were isolated from Nuruk, and degradation of these proteins during fermentation were confirmed by SDS-PAGE. IgE immunoblotting using patient's sera with wheat allergy was performed to confirm allergenic protein in Nuruk. These results as fermentation of Nuruk will provide a useful tool for developing safer wheat products to prevent wheat allergy.

Feasibility of Phytoremediation for Metal-Contaminated Abandoned Mining Area (광산 인근 토양의 중금속 오염에 따른 식물정화기술의 적용성 탐색)

  • This study was carried out to provide information for the present status of soil pollution near abandoned old-zinc mining area through analysis of bound form and 0.1 N-HCl extractable concentrations of heavy metals in soils and plants. Feasibility of endemic plants for phytoremediation was evaluated by the investigation of vegetation in soils. Cd contents of the selected samples near old-zinc mining soils ranged from 0.2 to $42mg\;kg^{-1}$. Nonagricultural soils near the mining area contained great amounts of Zn, Pb, Cd, and Cu than the paddy and upland soils. Some Korean wild plants, Artemisia princeps, Artemisia montana, Erigeron canadensis, and Pueraria thunbergiana, were found to grow vigorously in the studied area. Among them, Artemisia princeps was selected as a possible phytoremediator for cleaning heavy metal contaminated soils. Artemisia princeps contained about 43 and $52mg\;kg^{-1}$ of Cd in their root and shoot as dry weight, respectively. Average contents of Cd in the rhizosphere soil, $15.68mg\;kg^{-1}$, was slightly higher than the soil-root interface soils, $14.1mg\;kg^{-1}$. Sequential extraction of Cd contaminated soils showed that average $2.4mg\;kg^{-1}$ (about 7%) of cadmium existed as exchangeable form and the average amounts increased as follows : adsorbed < organically bound < exchangeable << oxide carbonate << sulfide residual fractions. Amendment of organic by-product fertilizer in metal-contaminated soils promoted the growth of roots significantly as compared with the other treatments containing chemical fertilizer.

Fractionation and Potential Mobility of Heavy Metals in Tailings and Paddy Soils near Abandoned Metalliferous Mines (폐광산 광미와 논토양의 중금속 분획화 및 잠재적인 이동도)

  • Most of the tailings have been left without any management in their mines and have become the main source of serious environmental problems in nearby groundwater, stream and cultivated lands. To compare fractionation and potential mobility of heavy metals in tailings and paddy soils near abandoned 10-metalliferous mines in Korea, the distribution and chemical fractions of heavy metal and their mobility in relation to chemical compositions were investigated. The pollution index of heavy metal in mine tailing calculated with the permissible levels were in the order Cheongyang>Dogok>Beutdeun>Baegwoul mine, which were considered sufficient to raise environmental problems. The rates of 0.1M-HCl extractable Cd, Cu, Pb, Zn, and Ni to total content in paddy soils were 49.1, 50.7, 26.8, 18.4 and 2.9%, respectively, and their rates of heavy metals in paddy soils were higher than that of mine tailing. Dominant chemical forms of heavy metals in tailings were sulfide and residual form (63-91%), specially, the exchangeable portion of Cd (21%) was relatively higher than that of other metals in paddy soils. The mobility factor of heavy metals in tailings and paddy soils was in the order Cd>Zn>Cu>Pb, and the mobility factor in tailing varied considerably among the mines. The potential mobility of heavy metals in tailings showed significant positive correlation with water-soluble $Al^{3+}$ and $Fe^{3+}$ contents, while in paddy soils, it correlated negatively with soil pH values.

Biological Activities and Bioactive Compounds in the Extract of Acer tegmentosum Maxim. Stem (산겨릅나무 줄기추출물의 생리활성 및 유효성분 분리)

  • Acer tegmentosum (Acereaceae) has been used a source of traditional medicines for the treatment of hepatic disorders in Korea. This research was conducted to determine biofunctional activities of A. tegmentosum stem extract and to identify its bioactive components. Methanolic extract from A. tegmentosum stem was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Two compounds were isolated by using an ODS column chromatography from ethyl acetate soluble fraction shown to the strongest antioxidant activity ($RC_{50}=3.15\;{\mu}g/m{\ell}$) among the fractions. The isolated compounds were analyzed by $^1H$ and $^{13}C$ NMR, IR, UV/VIS, MS spectrum data and identified as catechin, ${\rho}-Hydroxyphenethyl$ alcohol $1-O-{\beta}-_D-(6'-O-galloyl)-glucopyranoside$. The compounds have shown strong antioxidant activity, with similar activity to BHA ($RC_{50}=2\;{\mu}g/m{\ell}$). Especially, ${\rho}-Hydroxyphenethyl$ alcohol 1-O-{\beta}-_D-(6'-O-galloyl)-glucopyranoside$ was shown strong anti-lipid peroxidative activity. However, the compounds were not shown antimicrobial activities. In antimicrobial activity assays, ethyl acetate soluble fraction was effective to bacterial inhibition, such as Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125\;{\mu}g/m{\ell}$. Otherwise, antifungal activity against Candida albicans was shown in n-hexane soluble fraction exhibiting $63\;{\mu}g/m{\ell}$ of minimum inhibitory concentration. In anticomplementary activity assays, water soluble fraction was the most effective exhibiting 24% inhibitory activity.

Free Radical Scavenging Effect and Extraction Condition of Ethanol Extracts and Fractions of Wild Grape Seed (Vitis coignetiea) (머루종자의 적정 추출조건 및 추출 분획물의 Free Radical 소거능 효과)

  • Wild grape is a traditional medicine plant in north-eastern part of Asia and has been known to have healing properties for various illnesses. This study was to determine the optimum extraction condition and antioxidant activity of ethanol extracts of wild grape (V. coignetiae) seed. Also, organic solvent fractions of hexane, chloroform, ethyl acetate and butanol were obtained from the ethanol extract of wild grape seed at different temperatures. Total ethanol extraction yield of wild grape seed ranged from $4\%\;to\;12\%$ depending on the ethanol concentration, extraction temperature and time condition. The highest extraction yield of $11.9\%$ was obtained at $90\%$ ethanol condition for 12 hour at $70^{\circ}C$. However, the strongest free radical scavenging effect $(RC_{50})$ with $20.93\mu g/mL$ was observed in $70\%$ ethanol extract of wild grape seed extracted for 6 hour at $70^{\circ}C,\;while\;RC_{50}\;with\;40.42$\mu g/mL$ was observed in $90\%$ ethanol extract for 12 hour at $70^{\circ}C$. Antioxidant activity of ethanol extracts of wild grape seed increased as total phenol contents increased. Among each fraction obtained from organic solvents, ethyl acetate fraction was found to have the strongest $RC_{50}\;(8.6\mu g/mL)$ and 636.77 mg GAE/g phenol contents .

Smoking-attributable Mortality in Korea, 2020: A Meta-analysis of 4 Databases

  • Objectives: Estimating the number of deaths caused by smoking is crucial for developing and evaluating tobacco control and smoking cessation policies. This study aimed to determine smoking-attributable mortality (SAM) in Korea in 2020. Methods: Four large-scale cohorts from Korea were analyzed. A Cox proportional-hazards model was used to determine the hazard ratios (HRs) of smoking-related death. By conducting a meta-analysis of these HRs, the pooled HRs of smoking-related death for 41 diseases were estimated. Population-attributable fractions (PAFs) were calculated based on the smoking prevalence for 1995 in conjunction with the pooled HRs. Subsequently, SAM was derived using the PAF and the number of deaths recorded for each disease in 2020. Results: The pooled HR for all-cause mortality attributable to smoking was 1.73 for current men smokers (95% confidence interval [CI], 1.53 to 1.95) and 1.63 for current women smokers (95% CI, 1.37 to 1.94). Smoking accounted for 33.2% of all-cause deaths in men and 4.6% in women. Additionally, it was a factor in 71.8% of men lung cancer deaths and 11.9% of women lung cancer deaths. In 2020, smoking was responsible for 53 930 men deaths and 6283 women deaths, totaling 60 213 deaths. Conclusions: Cigarette smoking was responsible for a significant number of deaths in Korea in 2020. Monitoring the impact and societal burden of smoking is essential for effective tobacco control and harm prevention policies.

Identification of Novel Extracts of Saccharina japonica with High Deodorant Performance and Antioxidant Activity (우수한 탈취율과 항산화능을 갖는 다시마 추출물의 규명)

  • The purpose of this study was to identify novel extracts of brawn algae with the high deodorant capacity and antioxidant activity. To achieves this, an composition, deodorant performance and free radical scavenging activity were analyzed in powder and water extracts of Ecklonia cava(PEC/EEC), Saccharina japonica(PSJ/ESJ), Eisenia bicyclis(PEB/EEB), Sargassum spp.(PSS/ESS) and Hizikia fusiformis(PHF/EHF) using several analytical methods. Major component of five powders was verified as PEC, phlorotannin; PSJ, alginic acid and glutamic acid; PEB, carotenoid, chlorophyll, phlorotannin, tocopherol and fucoxantin; PSS, ${\beta}$-carotene and polyphenol; PHF, tannin. The highest level of DPPH scavenging activity was detected in EEC($IC_{50}=23.018{\mu}g/ml$), followed by EEB, ESJ, EHF and ESS. The deodorizing performance to ammonia was maintained high level in PEC, PSJ, PEB and PSS, while PHF showed low level in the same condition. But, the deodorizing performance to acetic acid was slightly higher in PSJ and PEC than those of PEB, PSS and PHF. Based on the results of DPPH scavenging activity and deodorizing capacity, we selected fractions of S. japonica as leading extracts and further prepared the 70% acetone extract (AESJ) and 70% ethanol extract (EESJ) from PSJ. The contents of total flavonoid, total phenol and total condensed tannin was significantly increased with 112-209% in AESJ and EESJ compared to those of ESJ. Also, a similar enhancement was observed on the level of DPPH scavenging activity and deodorizing performance against ammonia/acetic acid in AESJ and EESJ. Therefore, these results suggest that AESJ and EESJ contained with high total phenol and total condensed tannin contents may exhibit high deodorant capacity and antioxidant activity as well as has the potential for use as a powerful biomaterial for antibacterial deodorant.

Synthesis and Micellar Characterization of CBABC Type PLGA-PEO-PPO-PEO-PLGA Pentablock Copolymers

  • Poly(lactic-co-glycolic acid) (PLGA) were grafted to both ends of Pluronic$^{(R)}$ F68 ($(EO)_{75}(PO)_{30}(EO)_{75}$) triblock copolymer to produce poly{(lactic acid)$_m$-co-(glycolic acid)$_n$}-b-poly(ethylene oxide)$_{75}$-b-poly(propylene oxide)$_{30}$-b-poly(ethylene oxide)$_{75}$-b-poly{(lactic acid)$_m$-co-(glycolic acid)$_n$} (PLGA-F68-PLGA) pentablock copolymers. Molecular weights of PLGA blocks were controlled and five kinds of pentablock copolymers with different PLGA block lengths were synthesized using in-situ ring-opening polymerization of D,L-lactide and glycolide with tin(II) 2-ethylhexanoate ($Sn(Oct)_2$) catalyst. PLGA-F68-PLGA pentablock copolymers were characterized by $^1H$- and $^{13}C$-NMR, GPC, and TGA. The numbers (2m, 2n) of repeating units for lactic acid and glycolic acid inside PLGA segments were obtained as (48, 17), (90, 23), (125, 40), (180, 59), and (246, 64), with $^1H$-NMR measurement. From NMR data, the resultant molecular weights were determined in the range of 12,700-29,700, which were similar to those obtained from GPC. Polydispersity index was increased in the range of 1.32-1.91 as the content of PLGA blocks increased. TG and DTG thermograms showed discrete degradation traces for PLGA and F68 blocks, which indicate the weight fractions of PLGA blocks in pentablock copolymers can be calculated by TG profile and it is possible to remove PLGA block selectively. Hydrodynamic radius and radius of gyration of pentablock copolymer micelle were obtained in the range of 46-68 nm and 31-49 nm, respectively, in very dilute (i.e. 0.005 wt %) aqueous solution of THF:$H_2O$ = 10:90 by volume at $25^{\circ}C$.

Protein Patterns on a Vaginal Mucus during Spontaneous and Estrus Synchronization using CIDR in Korean Native Cattle (Hanwoo)

  • The aim of the present recent study was to compare the protein patterns in the vaginal mucus of Hanwoo cattles during spontaneous and CIDR induced-estrus. Ten cattles, who had been observed in estrus, received no treatment and served as the group of cattles with normal spontaneous estrus. Thirteen cattles in the CIDR received an CIDR insert on day 14 were removed and cattles were injected GnRH on day 15. Vaginal mucus samples were collected from all cattles at the same time the single AI in cattles with spontaneous estrus and the AI in cattles with induced estrus. Spontaneous and CIDR-induced estrus vaginal mucus samples were analyzed on two different array surfaces: cation-exchange (CM10), anion-exchange (Q10). In addition, using the NaCl solution by which the proteins combined after washing are 0.5, 1 and 2 M, it was fractionated and a protein was collected successively. The results are summarized as follows: 1) Ionic surfaces chemistries (Q10 and CM10) gave the best results in terms of detectable protein peaks, with more than 100 protein peaks in the two fractions and under each condition. 2) Protein mass spectrometer using 11 different proteins in protein identification of 7 were able to determine the protein. List of identified proteins as follows; Ribosome-binding protein 1, GRIP 1-associated protein 1, Katanin p60 ATPase-containing subunit A-like 1, Protein FAM44A, DUF729 domain-containing protein 1, Prolactin precursor, Dihydrofolate erductase. Conclusively, on the basis of this study, protein expression in the vaginal mucus could be used as an indicator for time of estrus manifestation in order to increase conception rates by applying AI at an optional time.

Evaluation on Anti-fungal Activity and Synergy Effects of Essential Oil and Their Constituents from Abies holophylla (전나무 정유의 항진균 효과와 유효성분의 시너지효과 평가)

  • This study was to investigate the antifungal activity of A. holophylla essential oil against dermatophytes, such as Epidermophyton floccosum, Trichophyton mentagrophytes and Trichophyton rubrum, and to determine the potential effective compound as dermatitis treatment. To evaluate the potential antifungal activities of A. holophylla essential oil and its fractions, paper disc diffusion and agar dilution method tested with morphological observation. Also, their major constituents were analyzed by GC/MS. To determine synergic effects of active ingredient from A. holophylla essential oil were carried out by checkerboard microtiter plate testing. The morphological changes of the dermatophytes exposed to active fraction G4 were observed by electron microscopes. As the results, the highest activities were identified in the fraction containing ${\alpha}$-bisabolol. A mixture of ${\alpha}$-bisabolol and bornyl acetate showed the synergy effects, expressing high potential effects. Also, morphological observation using electron microscopes showed a dramatic changes of cell membrane of E. floccosum and T. rubrum exposed to fraction G4 containing ${\alpha}$-bisabolol. In conclusion, A. holophylla essential oil and its constituents were expected to be used as antifungal agent or raw material for dermatitis therapy.
