치주질환 심도에 따른 치은조직내의 Superoxide Dismutase와 Catalase의 활성변화에 관한 연구 (ALTERATION OF SUPEROXIDE DISMUTASE-AND CATALASE-ACTIVITY IN HUMAN GINGIVAL TISSUES BY THE PERIODONTAL DISEASE SEVERITY)

  • It has been believed that antioxidant enzymes such as CuZn- and Mn-superoxide dismutase and catalase protect the tissue from damage resulting from the oxygen derived free radicals($O_2\;^-$, $H_2O_2$ and OH ). The purpose of this study was to investigate the relationship between activity of antioxidant enzymes including CuZn- and Mn- superoxide dismutase and catalase and inflammatory periodontal disease and periodontal parameters. For this study, the patients were classified into normal, gingivitis, adult periodontitis and rapidly progressive periodontitis, and then their papillary bleeding index(PBI) and probing depth were checked. Gingival tissues were surgically obtained from the patients during periodontal surgery, extraction, and clinical crown lengthening procedure. The activity of CuZn- and Mn- superoxide dismutase and catalase in the gingival tissues was measured by using UV-spectrophotometer by the same methods as Crapo et al. And Aebi did, respectively. The results were as follows : 1. CuZn- and Mn- and total-superoxide dismutase activity were significantly low in rapidly progressive periodontitis group in comparison to normal group (P<0.05). 2. In comparison of the antioxidant enzyme activity according to papillary bleeding index group(PBI), Mn-superoxide dismutase activity only was significantly lower in PBI 2, 3, and 4 groups than PBI 0 group(P<0.05). 3. Superoxide dismutase activity failed to show any significant difference according to probing depth. But significant]y high catalase activity was shown in deep pocket group (${\ge}7mm$)(P<0.05). In conclusion, these results suggest that the activity of Mn-superoxide dismutase among the antioxidant enzymes may reflect the inflammatory status of gingival tissue and that the decreased activity of superoxide dismutase may be one of responsibe factors for progression of rapidly progressive periodontitis.

고압산소환경이 신생 및 성숙백서의 폐 superoxide dismutase 활성도에 미치는 영향에 관한 실험적 연구 (Effect of Hyperbaric Oxygenation on Pulmonary Superoxide Dismutase Activity in Neonatal and Adult Rats)

  • To investigate the effect of hyperbaric oxygenation on superoxide dismutase activity, neonatal rats (7-10 days old) and adult rats (approximately 100 days old) were continuously exposed to hyperbaric oxygen environment of 2.4ATA for 8 hours and their superoxide dismutase activity were measured. Neonatal rats, all survived through exposure, showed significant increases in the pulmonary superoxide dismutase activity at immediately and 24 hours after exposure. Adult rats, whose 8 hour survival rates were 14%, did not show any significant increase in the activity of pulmonary superoxide dismutase as compared to the control adult rats. These findings are indicating that increased tolerance to oxygen toxicity in neonatal animals during exposure may be attributed to the increase in activity of superoxide dismutase in neonatal rats.

폐흡충 성충 Cu, Sn-Superoxide Dismutase의 정제 및 생화학적 특성 (Purification and Characterizatlon of a Cu, Zn-Superoxide Dismutase from Adult Paragonimus westermani)

  • 폐흡충 성충의 인산완충액 추출액을 원심분리하여 만든 세포질 현탁액을 조효소(조효소)로 사용하여 Xanthine- zanthine oxidase system으로 superoxide digmutase 환성을 측정한 결과 비활성도(비활성도: specific activity)는 4.3 units/mg이었다. 이 효소의 활성도를 30∼85% ammonium sulfate 침전 및 DEAE-Trisacryl M anion exchange chromatography와 Sephadex G-100 column chromatography를 통과시키면서 측정하는 방법으로 superoxide dismutase를 정제하고, 정제한 효소의 생화학적 특성을 관찰하여 다음과 같은 결과를 얻었다. 1. 정제과정을 통해 세포질 내 superoxide dismutase를 150배 정제하였다. Sephadex G-100 gel filtration에 의해 계산한 이 효소의 분자량은 34kDa이었고 환원성 SDS-PAGE상 subunit가 17 kDa이었다. 따라서 이 효소는 subunit 두개로 구성된 중항체로 판단하였다. 3. 이 효소는 1 mM 이상의 cyanide 농도에서는 효소 황성이 100% 억제되었고, 5mM 농도의 azide에서는 11.1%, 10mM azide에서는 22.2% 그 환성이 각각 억제되었다. 3. 이 효소는 완충액의 pH가 10.0일 때 효소 황성이 5.4배 증가되었다. 이상의 결과로 폐흡충의 세포질 내에 존재하는 superoxide dismutase는 구리와 아연을 함유한 superoxide dismutase라고 판단할 수 있었다.

Streptomyces subrutilus P5가 생산하는 철 함유 superoxide dismutase의 분비 (Secretion of the iron containing superoxide dismutase of Streptomyces subrutilus P5)

  • 본 연구에서는 Streptomyces subrutilus P5의 생장과 세포내 외 철 함유 superoxide dismutase 활성을 비교 분석하여 철함유 superoxide dismutase의 분비 시점을 확인하고 분자 수준에서 이 효소의 분비에 관여하는 유전정보를 확인하고자 하였다. Streptomyces subrutilus P5의 균체 생장은 건체 중량을 측정하여 결정하였다. Glucose는 log phase에서 급격히 소모되어 24시간 후에 이르러 완전히 고갈되었다. 세포내의 철 함유 superoxide dismutase는 배양 후 3시간에 나타나며 세포외 철 함유 superoxide dismutase는 배양 후 7.5시간부터 나타난다. 따라서 superoxide dismutase는 용균에 의해서가 아니라 능동적인 분비기작에 의해서 세포 외로 분비된 것으로 추측할 수 있다. Streptomyces subrutilus P5의 sodF에는 signal peptide 유전정보가 존재하지 않았다. 그러나 sodF의 상류지역에서 다른 세균의 type III 분비단백질 유전자와 유사한 type III 분비상자가 발견되었다. Streptomyces 균주에서 type III 분비단백질이 존재할 가능성이 있음을 처음으로 제시하였다.

오이 추출물에 존재하는 Superoxide Dismutase의 열안정성 (Thermostability of Superoxide Dismutase from Cucumber(Cucumis sativa))

  • The superoxide dismutase(SOD) in peeled pericarp of cucumber was most stable at pH 8.0 and relatively stabe between pH 5.0 and 9.0. The enzyme was stable up to 6$0^{\circ}C$ and retained 12% by heat treatment at 10$0^{\circ}C$ for 5 min. At pH 2.0, the peeled pericarp enzyme activity was decreased to 10% by incubation for 3 hrs. However, the enzyme activity was increased above 25% after incubating the enzyme at pH 7.0 for 6 hrs. Retention of SOD activity in cucumber by various heating methods was also measured. The residual SOD activities of peeled pericarp and whole cucumber was estimated to be 25% and 27% after blanching(2 min), respectively. The skin enzyme retained 53% of its activity after steaming (3 min). When the peeled pericarp enzyme was incubated at 4$^{\circ}C$ for 20 days, the enzyme activity remained about 81%. However, when the enzyme incubated at 3$0^{\circ}C$ for 20 days, the peeled pericarp enzyme activity decreased to 17% of its original activity. The enzyme activity of peeled pericarp cucumber was not changed after exhaustive dialysis for 3 days, which indicated that the SOD activity in cucumber seems to have molecular weight above 12,000.

항산화제인 Cu(II)-DIPS와 재조합 인간 수퍼옥사이드 디스뮤타제의 비교 (Comparison of Cu(II)-DIPS and Human Recombinant Superoxide Dismutase, an Antioxidant)

  • The superoxide dismutase (SOD) mimetic activity of copper complex of 3,5-disopropylsalicylic acid (Cu(II)-DIPS) was tested and compared to those of human recombinant SOD (hrSOD) and its conjugate form with polyethyleneglycol (PEG) using fer- ricytochrome c reduction assay. Stability constant of Cu(II)-DIPS was measured po- tentiometrically using SCOGS2 program. In the presence of 10 g/L albumin, Cu(II)-DIPS lost most of its SOD mimetic activity. HrSOD was modified with polyethylene glycol (PEG) of M.W. 5000. These conjugates have markedly prolonged plasma half-lives of enzymatic activity (15.5 hr) compared to native hrSOD (5 min). In summary, efficient SOD mimetics should be stable enough not to dissociate in blood by serum protein. HrSOD could have longer half-life by conjugation with inert PEG for sustained SOD effect.

  • In order to study the antioxidative action of the effectient garlic components, peroxidase and superoxide dismutase activity were compared through the in vitro and in vivo experiments. RESULTS : 1. Observing the effects on peroxidase activity of efficient components in vitro, garlic oil, alliin and ethanol fraction showed effective, which was similar to the trend of TBA value, peroxide value and induction time for the first period of lipoperoxide formation in vitro. 2. In vivo experiment with peroxidase activity, the ethanol fraction and garlic oil were effective when intraperitoneally administered as well as orally administered. 3. Considering the superoxide disutase activity in vitro, the garlic oil, alliin and ethanol fraction were effective in efficient components. But non-kaolin fraction inhibited the activity on the contrary. 4. In terms of the efefcts on superoxide dismutase activity in vivo, alliin and garlic oil wee effective in intraperitoneal adminstraton and the ethanol fraction and alliin in oral administration.

  • Inflammatory cells may produce active species of oxygen in antimicrobial defense. While such species can directly damage surrounding tissue, their major secondary role may be to mediate important components of the inflammatory response. Superoxide dismutase, antioxidant, have significant anti-inflammatory properties in rheumatoid arthritis, ischemic tissue injury and gastrointestinal disease. Increased oxidative product formation diseases. And superoxide dismutase produced by Porphyromonas Gingivalis is resistant to killing by polymorphonuclear leukocyte. The purpose of this study was to investigate on the effects of superoxide dismutase in 3T3 fibroblast and in experimental gingivitis in the rats. The effect of superoxide dismutase(SOD) to cell morphology and cell activity was measured in cultured mouse 3T3 fibroblast. After experimental gingivitis were induced by lipopolysaccharide(LPb) and bovine serum albumin(BSA), injection of SOD were done. WBC count and histologic findings were observed at 1, 2, 3, and 7 days. The results were as follows; 1. There was a little difference between LPS treated groups and SOD treated groups in 3T3 fibroblast morpholoy. 2. There was no difference between only SOD treated groups (except SOD 150U at 3days) and control in 3T3 fibroblast activity. 3. LPS $0.5{\mu}g/ml$ and SOD treated groups (except 150U) had decreased 3T3 fibroblast activity and no significant difference at 3 days. 4. LPS $5.0{\mu}g/ml$ and SOD treated groups were significantly increased cell activity of 3T3 fibroblast than control group at 1 day(P<0.05). 5. In LPS induced gingivitis, the number of leukocytes in SOD treated was significantly decreased than in saline treated at 1 day(P<0.05). 6. In histopathologic findings of LPS or BSA induced gingivitis, inflammatorycell infiltration in SOD treated groups were less than in saline treated group at 1, 2 and 3 days.

Glycerol이 흰쥐 신장에서의 Malondialdehyde 함량과 Superoxide Dismutase 활성도 및 요중 단백질 배설량과 $N-acetyl-{\beta}-D-glucosaminidase$ 활성도에 미치는 영향 (Effects of Glycerol on the Malondialdehyde Level and Superoxide Dismutase Activity in the Kidney and Urinary Protein Excretion and $N-acetyl-{\beta}-D-glucosaminidase$ Activity of the Rats)

  • In an attempt to dofine the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals and $N-acetyl-{\beta}-D-glucosaminidase$ (NAG) activity, we studied 24-hours urine outflow, 24-hours urinary protein excretion and urinary NAG activity after the injection of glycerol and also we studied malondialdehyde(MDA) level and superoxide dismutase(SOD) activity in the kidney of rats at 24hr after the injection of glycerol. Sprague-Dawley albino rats weighing 240 to 260 gm were injected intramuscularly with a 50% solution of glycerol(2ml/kg, 4ml/kg and 8ml/kg). The group treated with glycerol showed significantly lower urine outflow level and urinary protein excretion level and higher urinary NAG activity after the injection as compared to those of control group. Also the group treated with glycerol showed significantly higher MDA level and lower SOD activity at 24hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity and higher urinary NAG activity is an index of renal tubular cell damage in the glycerol-induced nephrotoxicity.

환경 스트레스에 의한 벼의 Peroxidase, Superoxide dismutase 및 Diterpene cyclase 활성비교 (Comparision of Peroxidase, Superoxide dismutase and Diterpene cyclase Activity under Environmental Stress Conditions in Rice)

  • UV-B와 도열병병원균 접종들의 환경 스트레스에 의한 벼의 POD, SOD및 Diterpene cyclase등의 활성 변화를 조사하여 해 본 결과, 일품벼에서 POD 와 SOD활성이 가장 놓았고, 도열병진전에 따른 PDO활성은 감수성 품종인 일품과 화성은 증가하였으나 저항성 품종인 다산은 감소하였다. 키토산 처리에의한 SOD의 활성 변화는 처리 농도가 높을수록 증가하였다. 키토산 처리후 14일에서의 감수성품종인 화성의 SOD활성은 무처리 식물체에 비하여 높았다. UV-B조사 시간이 길수록 감수성품종인 일폼의 POD활성은 감소하였고, 저항성 품종인 다산은 증가하였다. 특히UV-B의 조사시간에 따른 다산의 POD활성은 차가 없었으나, 화성의 활성은 40분 조사처리시 2배 놓았다 UV-B처리에 비해 p.oryzae를 접종시는 diterpene cyclase의 활성이 놓아졌으며, 병원균 접종후 5일까지 효소활성이 검진적으로 증가하였다.
