Antimutagenic Effects of Juices from the Peppers in Salmonella Assay System

  • The antimutagenic effects of juices from green pepper(GP), red pepper leaf(RPL), red pepper(RP) and sweet pepper(SP) were examined by the Ames method using Salmonella typhimurium TA100. The juice supernatants of GP, RPL and RP showed antimutagenic activities against afltoxin B1(AFB1) in Salmonella typhimurium TA 100. The juice supermatants of GP and RPL also exhibited the inhibitory effects(p<0.05) to the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitro-quinoline-1-oxide(4-NQO). The juice of RP showed antimutagenic activities against indirect mutagen of AFB1, however, the activity was reduced at higher concentration(5.0%), furthermore, as the adding concentration of sample increased to 5.0%, it exhibited slight comutagenicith on direct mutagen of MNNG. Theantimutagenic activities of GP and RPL juices were reduced significantly after heating at 100℃ for 20min, supposing that the antimutagenic compound(s) in the juices were heat labile.

먹물버섯 자실체 및 균사체 추출물의 돌연변이 억제효과 (Antimutagenic Effect of the fruiting Body and the Mycelia Extracts of Coprinus comatus)

  • The inhibitory effect of Coprinus comatus on the mutagenicity in Salmonella assay system and SOS chromotest were studied. In Ames test, the ethanol and water extracts and the cultured mycelia fractions of Coprinus comatus did not show any mutagenicity, but the Coprinus comatus ethanol extracts showed inhibitory effects of 8 0∼90% on the mutagenicity induced by indirect mutagen of benzo(a)pyrene(B(a)P) and aflatoxin B1(AFB1) in Salmonella typhimurium TA98 and TA100. The antimutagenic effect increased with increasing concentration of the ethanol extract toward N methyl N' nitro N nitrosoguanidine(MNNG). However, the water extracts inhibited about 40∼50% against direct and indirect mutagen. The cultured mycelial filtrate of Coprinus comatus, the fractionⅡ, showed antimutagenic effect of 90% on MNNG and 25∼50% on B(a)P and AFB1. In SOS chromotest, the ethanol extracts of Coprinus comatus showed antimutagenic effect of 65∼81% on SOS function induced by 4 NQO, and the cultured mycelia fractionⅡ showed low inhibitory effect of 20∼50%.

Toxicogenomic Study to Identify Potential New Mechanistic Markers on Direct-Acting Mutagens in Human Hepatocytes (THLE-3)

  • Exposure to DNA-damaging agents can elicit a variety of stress-related responses that may alter the expression of genes associated with numerous biological pathways. We used 19 k whole human genome chip to detect gene expression profiles and potential signature genes in human normal hepatocytes (THLE-3) by treatment of five direct acting mutagens, furylfuramide (AF-2), N-nitroso-N-methylurea (MNU), methylmethanesulfonate (MMS), 4-nitroquinoline-N-oxide (4-NQO) and 2-nitrofluorene (2NF) of the $IC_{20}$ concentration for 3 h. Fifty one up-regulated common genes and 45 down-regulated common genes above 1.5-fold by five direct-acting mutagens were identified by clustering analysis. Many of these changed genes have some association with apoptosis, control of cell cycle, regulation of transcription and signal transduction. Genes related to these functions, as TP73L, E2F5, MST016, SOX5, MAFB, LIF, SII3, TFIIS, EMR1, CYTL1, CX3CR1 and RHOH are up-regulated. Down-regulated genes are ALOX15B, xs155, IFITM1, BATF, VAV2, CD79A, DCDC2, TNFSF8 and KOX8. We suggest that gene expression profiling on mutagens by toxicogenomic analysis affords promising opportunities to reveal potential new mechanistic markers of genotoxicity.

강황(Curcuma longa L.)으로부터 초임계 유체 추출한 curcumin의 생리활성 (Physiological Effects of Curcumin Extracted by Supercritical Fluid from Turmeric (Curcuma longa L.))

  • Curcumin의 추출효율은 에탄을 추출방법이 가장 높았고, 항산화효과는 에탄올, 초임계, 열수 추출물 순으로 나타났다. Ames의 mutagenicity test를 실시한 결과 각각의 추출물은 돌연변이원이 없는 것으로 확인되었고, TA98에 직접변이원 2-NF 처리시 초임계 추출물의 저해율이 20.1%로 에탄올과 열수 추출물에 비하여 가장 높은 항돌연변이 효과를 나타냈다. 또한 간접 변이원 2-acetamidofluorene($10\;{\mu}L/plate$; Sigma Co.; 2-AF)에 대한 항돌연변이 효과를 알아본 결과 2-AF로 유도된 TA98은 초임계 추출물을 0.1 mL/plate로 처리시 항돌연변이의 효과는 12.2%였으며, 알코올 추출물과 열수 추출물의 경우는 항돌연변이 효과가 나타나지 않았다. Nitrite의 분해능의 경우에 있어서도 에탄올 및 열수 추출물에 비하여 초임계 추출물이 높은 분해율을 나타냈으며 pH 1.2에서 가장 높은 분해율을 보였다. 이와 같이 초임계를 이용해 추출한 curcumin은 항산화효과, nitrite 분해능, 항돌연변이효과 등에 대한 생리활성 기능을 보였으며, 기존의 에탄올이나 열수 추출법에 비하여 유사하거나 더 높은 생리활성 효과가 나타나기도 했다.

  • Chinin is the second most plentiful natural polymer. Currently, chitin and chitosan are manufactured commercially in large scale from crab and shrimp shell as fish processing waste. They is being used in many commercial application because of their various functional properties. Chitosan, in particular, is being evaluated as biomedical materials in a number of food and pharmaceutical industries. Despite their potential abilities, the perfect safety had been demonstrated until now. However, the long-term feeding with chitin was not any negative effect the body weights and serum enzymatic activities in mice. And, in rats supplied with $5\%$ of chitosan diet for 450 days, there was no changes of Ca concentration in blood, bone and other organs except for in muscle. Consequently, there was no direct toxicity of chitin and chitosan against some animals such as rat and mouse by recently reports.

  • The effects of monosodium glutamate (MSG) on the activities if tyrosine hydroxylase (TH), dopamine ${\beta}$-hydroxylase (DBH), tryptophan hydroxylase (TPH) and monoamine oxidase (MAO) in various regions (cerebral cortex, striatum, midbrain, pons and medulla of nat brain have been determined. It was observed that up to 1mM MSGhad no significant effects on the activities of brain tyrosine hydroxylase, dopamine ${\beta}$-hydroxylase, tryptophan hydroxylase and monoamine oxidase in all regions of rat brain. These results indicated that MSG itself exerted no direct effect on the important enzymes synthsizing and metabolizing the monoaminergic neuronal system.

Immunotoxicology Evaluation of New Drugs

  • Drugs can have various adverse effects on the immune system including unintended immun-osuppression, induction of both drug-specific immune responses (including drug allergies) and non-specific immunostimulation (including autoimmune reactions), and direct activation of effector mechanisms (such as histamine release). As a practical matter, the Center for Drug Evaluation (CDER) relies on standard non-clinical toxicology studies to detect unintended immunosuppression. Specific assays using guinea pigs and mice are available to identify drugs that can induce immune-mediated dermal hypersensitivity reactions. Respiratory and systemic hypersensitivity and autoimmune reactions are more difficult to model in non-clinical studies. Unintended nonspecific immunstimulation can be detected in animal studies. CDER is currently developing specific guidance for evaluating potential drug immunotoxicity.

발암물질 노출량 산출 및 암 위해성 평가에 있어서 Biomcrker의 활용 (Application of Biomarkers for the Assessment of Carcinogen Exposure and Cancer Risk)

  • Risk Assessment is an important area in toxicology and the methodology for risk assessment has been developed. Mathematical models used for risk assessment include one-hit multi-hit, two-stage, probit logistic, multistage, and linearized multistage models. For the assessment of exposure dose, environmental monitoring has been applied, but it has limitation to accurately assess exposure level because the levels in the air, water, foods, and soil may vary depending on time of sampling. In addition, humans can be exposed to various sources of exposure and thus it will be impossible to estimate the total level of exposure in humans by environmental monitoring. To eliminate the limitation of environmental monitoring, a direct measurement of toxic materials or modified biomolecules (called biomarkers) associated with the exposure of toxic materials is needed. Here, scientific basis of biomarkers and future direction have been considered for the assessment of carcinogen exposure and cancer risk in humans.

마우스 말초혈액 망상적혈구를 이용한 Mitomycin C의 소핵생성효과 (The micronucleus formation in peripheral blood of mitomycin C-treated mice using supravital staining with acridine orange)

  • In this study, the micronucleus test with peripheral blood using acridine orange coated slides was evaluated in mice treated with mitomycin C(MMC) at doses of 0.5, 1.0 and 1.5 mg/kg body weight. The peripheral bloods were obtained at 0, 24, 48 and 72h after treatment. The frequencies of micronucleated reficulocytes(MNRET) in the MMC-treated groups increased dose-dependently, and showed a peak time at 48h after treatment. We also performed the sex differences of MNRET frequency in 0.5 mg/kg MMC treated group, and we observed no sex differences in this experiment. And we evaluated the usefulness of a direct acting clastogen, N-methyl-N-nitrosourea and a indirect acting clastogen, benzo(a) pyrene as the positive control in this supravital micronucleus test. They also caused a significant increase in MNRET frequencies. These results suggest that the supravital staining micronucleus test using MNRET can be useful tool to evalulate the quantitative and qualitative assessment of genotoxicity in vivo compared to classical in vivo micronucleus test using bone-marrow cells.

Antimutagenic Effects of Linoleic Acid

  • In order to determine the effectiveness of linoleic acid(LA) to inhibit carcinogens/mutagens-induced mu-tagenesis, Ames test using Salmonella typhimurium TA100 and the SOS chromotest using E. Coli PQ37, were carried out. The inhibitory effect of LA(1%) on the Ames mutagenicity test were 98%, 78%and 69% mediated by aflatoxin B₁(AFB₁), N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitroquinoline-1-oxide(4-NQO), respectively. LA exhibited a strong antimutagenic activity aganist indirect mutagen, AFB₁whereas exhibited the same concentration of LA showed weaker inhibitory effects on direct mutagens of MNNG and 4-NQO than that AFB₁. LA also reduced the SOS responses induced by MNNG and 4-NQO significantly. This result showed a possibility that LA can be a protective agent in early step of cancinogenesis.
