Antioxidant Activity of Substances Extracted by Alcohol from Chungkookjang Powder (분말청국장에서 알코올로 추출한 물질의 항상화능)

  • It is previously reported that Bacillus licheniformis B1 strain isolated from nature was successfully used for Chungkookjang fermentation. Antioxidant activity of its powder was determined in this study. Sephadex G-75 gel filtration chromatography was performed, far soluble fractions of the powder extracted by distilled water. The soluble fractions were separated into large and small fractions. The substance 1,1-diphenyl-2-picrylhydrazyl (DPPH) was used as an electron acceptor. Antioxidant activity was found in the small fractions. Five% solution of the Chungkookjang powder was the most effective in the extraction of antioxidant substances from the powder. It was proven in this study that strong antioxidant activity still remained in the Chungkookjang powder.

  • Twenty-four Oriental herbal medicines including Platycodi Radix and Scutellariae Radix, etc., which have been used for the cure of hyperlipidemia, coronary heart, disease were evaluated for the 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase inhibition and antioxidant effect on a free radical, 1,1-diphenyl-2-picrylhydrazyl(DPPH) in vitro, and antihyperlipidemic effect on hyperlipidemic rats induced by Triton WR-1339 in vivo. 80% MeOH extract of eight herbs including Scutellariae Radix, Coptidis Rhizoma, Paeoniae Radix Rubra, Moutan Cortex Radicis, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam, Cinnamomi Ramulus and Crataegi Fructus inhibited the HMG-CoA reductase activities and exhibited a radical-trapping action on a stable free radical, DPPH. On Triton WR-1339-induced hyperlipidemic rats, four herbs including Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus showed respectively the significant suppression of serum total cholesterol, triglyceride, phospholipid and LDL-cholesterol levels and serum transaminase(ALT and AST) activities. From these results, it is suggested that each 80% MeOH extract of Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus have effective antihyperlipidemic action against hyperlipidemia.

Antioxidative Effects of Pine (Pinus denstifora) Needle Extracts. (솔잎 추출물의 항산화 효과)

  • Antioxidative activities of pine (Pinus denstifora) needle extracts were tested in vitro experimental models. The concentration of total polyphenolic compound of water extracts from pine needle was 1.61 %. In DPPH ($\alpha$, $\alpha'$-diphenyl-$\beta$-picrylhydrazyl) method, the electron donating activity of 0.1 % water extracts from pine needle was as high as BHT (0.05%, w/v). The antioxidative activity was measured by inhibition against lipid peroxidation of rat liver microsome, and this activity was shown in the following: 67.7% at 0.1% concentration >63.1% at 0.05% concentration > 28.2% at 0.01% concentration. In antioxidative activity determined by thiocyanate method against lipid peroxidation using linoleic acid, the antioxidative activities at all concentration of 0.01 %, 0.05% and 0.1 % were much higher than control during 7 days. In TBA method, the antioxidative activity was increased with increasing concentration until 6 days. These results support that water extracts from pine needle contain antioxidative compounds.

Antioxidant Properties of Flavone-6(4')-Carboxaldehyde Oxime Ether Derivatives

  • The in vitro antioxidant properties of some flavone-6(4)-carboxaldehyde oxime ether deriva-tives (Ia-f, lIa-f) were determined by their effects on the rat liver microsomal NADPH-dependent lipid peroxidation (LP) levels by measuring the formation of 2-thiobarbituric acid reactive substances. The free radical scavenging properties of the compounds were also examined in vitro by determining their capacity to scavenge superoxide anions and interact with the stable free radical 2, 2-diphenyl-1-picrylhydrazyl (DPPH). The most active compounds, lib (Flavone-4'-carboxaldehyde-O-ethyl oxime) and Id (Flavone-6-carboxaldehyde-O-[2-(1-pyrolidino) ethyl] oxime), caused 98 and 79% inhibition of superoxide anion production and DPPH stable free radical at $10^{-3}{\;}M$, respectively.

Antioxidant Activities of Agrimonia pilosa Ledeb: In Vitro Comparative Activities of Its Different Fractions

  • The antioxidant potentials of the methanol extract and its various fractions from Agrimonia pilosa Ledeb were measured as 1,1-diphenyl-2-picrylhydrazyl (DPPH), and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities as well as the reducing power decreasing NO in vitro. The methanol extract of Agrimonia pilosa Ledeb showed significantly strong scavenging effects on free radicals of DPPH, ABTS, and NO. Hexane fraction (HF), ethyl acetate fraction (EF) and n-butyl alcohol fraction (BF) were prepared by solvent fractionation. By comparison of polyphenol contents among BF and HF, EF with high polyphenol contents showed the highest DPPH, ABTS, and NO scavenging effects with the value of 50.2%, 80.4%, and 65.7%, respectively, at the highest tested dose. HPLC analysis revealed the presence of various phenolic compounds in EF of Agrimonia pilosa Ledeb. These results suggest that EF could be considered as natural antioxidant sources and dietary nutritional supplements to prevent oxidation-related diseases.

Antioxidant Components of the Aerial Parts of Bidens frondosa L. (미국가막사리 지상부의 항산화 성분)

  • As a part of an ongoing search for natural plants with antioxidant compounds by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH), a total extract of the aerial parts of Bidens frondosa L. (Compositae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of five compounds, quercetin-3-O-${\beta}$-D-glucopyranoside (1), luteolin-7-O-${\beta}$-D-glucopyranoside (2), 7,8,3',4'-tertrahydroxy-flavanone (3), okanin-4-O-${\beta}$-D-glucopyranoside (4), and okanin (5). Their structures were elucidated by spectroscopic studies. Compounds 3-5 were isolated for the first time from this plant. Among them, compounds 3 and 5 showed the significant radical scavenging effects on DPPH, and compounds 3 and 5 also showed the potent riboflavin and xanthine originated superoxide quenching activities.

Superoxide Radical Scavengers from the Whole Plant of Veronica peregrina

  • In the course of screening for antioxidant compounds by measuring the radical scavenging effect on 1,1-diphenyl- 2-picrylhydrazyl (DPPH), a total extract from the whole plant of Veronica peregrina (Scrophulariaceae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of six phenolic compounds including chrysoeriol (1), diosmetin (2), 4-hydroxybenzoic acid (3), apigenin (4), caffeic acid methylester (5) and protocatechuic acid (6). Their structures were elucidated by spectroscopic studies. Compounds 1-5 were isolated for the first time from this plant. Compounds 5 and 6 showed significant antioxidative effects in DPPH free radical scavenging and superoxide quenching activity assays.

  • Acanthopanax species (Araliaceae) traditionally has been used as analgesics, stimulant of immune system, and replenishment of body functions. Acanthopanax divaricatus var. albeofructus is indigenous plant to Korea. The antioxidant activities of compounds from A divaricatus var. albeofructus were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and thiobarbituric acid reactive substance (TBARS) assay on human plasma low-density lipoprotein (LDL). The triterpenoid and lignan constituents from this plant showed antioxidant activities and the lignan, l-sesamin exhibited the most potent antioxidant activity in Cu$^{2+}$ -induced LDL oxidation.n.

Determination of Radical Scavenging Activity of Aster yomena (Kitam.) Honda (쑥부쟁이 추출물의 라디칼 소거활성 평가)

  • In this study, we investigated the antioxidative effects of AY by measuring 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide radical ($O_2{^-}$) scavenging activities in vitro. AY was extracted with ethanol and then partitioned with n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc) and n-butanol (n-BuOH). In the DPPH radical scavenging assay, AY at concentrations of 10 to $100{\mu}g/mL$ dose-dependently increased inhibition of DPPH oxidation, with the EtOAc fraction of AY showing the highest DPPH radical scavenging activity fractions. The $^{\cdot}OH$ radical scavenging activities of the extract and four fractions of AY increased by over 80% at a concentration of $50{\mu}g/mL$. In particular, the IC50 value of the EtOAc fraction was $0.03{\mu}g/mL$, which was the lowest value among all fractions. We also found that the EtOAc fraction of AY was better at $O_2{^-}$ radical scavenging than other fractions. Taken together, these results suggest that AY, especially the EtOAc fraction, can be used as a natural antioxidant against free radicals.

Chlorogenic Acid, an Antioxidant Principle from the Aerial Parts of Artemisia iwayomogi that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.
