Early Life Stage Toxicity of 2,3,7,8,-Tetrachlorodibenzop-Dioxin(TCDD) in Goldfish(Carassius auratus) (금붕어 수정난의 2,3,7,8-TCDD 노출에 따른 다이옥신 관련 유전자의 발현 및 형태학적 변화)

  • In this study, we obtained the fertilized eggs from goldfish(Carassius auratus) and observed normal developmental stage(from fertilized eggs to larvae) in non-exposed groups. Goldfish embryos at 3 h postfertilization(hpf) were statically exposed for 1 h to either dimethylsufoxide(DMSO, 0.1%, v/v) or TCDD($0.5{\mu}g/L$). Toxicity and morphological changes were characterized from 3 to 120 h postfertilization(hpf). Egg mortality($0{\sim}48$ hpf) and hatching ratio($72{\sim}83$ hpf) in TCDD-exposed group were significantly different from control groups. However, pericardial edema was first observed at 72 hpf, followed by the onset of yolk sac edema and mortality. In addition, goldfish embryos-larvae exposed to TCDD significantly increased TCDD-related gene such as CYP1A($24{\sim}72$ hpf) and AhR2(72 hpf). This is the first study about in-depth characterization of TCDD-induced developmental toxicity in goldfish(Carassius auratus).

  • Polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) were investigated in soil and pine needle samples taken from 16 sites of industrial and residential areas in Changwon, Korea to assess their distribution levels and to suggest the influence of industrial activities, PCDDs/DFs levels in the soil samples ranged from 0.57 to 20.79 pg I-TEQ/g dry weight with a mean value of 4.20 pg I-TEQ/g dry weight. PCDDs/DFs levels in the pine needle samples ranged from 0.39 to 8.75 pg I-TEQ/g dry weight with a mean value of 4.09 pg I-TEQ/g dry weight. In both soil and pine needle samples, the PCDDs/DFs concentrations in the industrial area sites were higher than those in the residential area sites. Homologue profiles in pine needle samples showed different patterns compared with soil samples. Based on the results of principal component analysis (PCA), it was confirmed that pine needles reflected a direct influence from local potential sources of PCDDs/DFs, showing a much higher degree of reflection than in soils. Pine needles are very useful as an indicator for monitoring or estimating the contamination of PCDDs/DFs in other areas which have been impacted by point pollution sources.

  • One prominent characteristic of2,3,7,8-tetrachlorodibenzop-dioxin (TCDD) toxicity in rats is a reduction of body weight accompanied by an altered serum lipid profile such as hyperlipidemia. A single administration of TCDD (50 ug/kg) resulted in a decrease of body weight and increase of serum cholesterol in rats. TCDD-induced weight loss and serum cholesterol elevation was reduced in rats administered with water extract (100 mg/kg) or saponin fraction (40 mg/kg) of Panax ginseng C.A.Meyer. In contrast, the administration of Panax quinquefolium did not inhibit the TCDD-induced weight loss and serum cholesterol elevation. Histological examinations of liver and testis revealed the administration of saponin fraction of Panax ginseng attenuated the TCDD-induced hispathologicallesions whereas the administration of saponin fraction of Panax quinquefolium did not. High performance liquid chromatographic analysis demonstrated high percentiles of ginsenoside Rg and ginsenoside $Rh_1$ were evident in saponin fraction of Panax ginseng. Results indicate that the protective effects of Panax ginseng, not Panax quinquefolium, on the TCDD-induced toxicity might be resulted from different compositions of saponins in Panax ginseng.

Molecular Characteristics of Pseudomonas rhodesiae Strain KK1 in Response to Phenanthrene

  • Radiorespirometric analysis revealed that Pseudomonas sp. strain KKI isolated from a soil contaminated with petroleum hydrocarbons was able to catabolize polycyclic aromatic hydrocarbons such as phenanthrene and naphthalene. The rate and extent of phenanthrene mineralization was markedly enhanced when the cells were pregrown on either naphthalene or phenanthrene, compared to the cells grown on universal carbon sources (i.e., TSA medium). Deduced amino acid sequence of the Rieske-type iron-sulfur center of a putative phenanthrene dioxygenase (PhnAl) obtained from the strain KKI shared significant homology with DxnAl (dioxin dioxygenase) from Spingomonas sp. RW1, BphA1b (biphenyl dioxygenase) from Spingomonas aromaticivorans F199, and PhnAc (phenanthrene diokygenase) from Burkholderia sp. RP007 or Alcaligenes faecalis AFK2. Northern hybridization using the dioxygenase gene fragment cloned from KKI showed that the expression of the putative phn dioxygenase gene reached the highest level in cells grown in the minimal medium containing phenanthrene and $KNO_3$, and the expression of the phn gene was repressed in cells grown with glucose. In addition to the metabolic change, phospholipid ester-linked fatty acids (PLFA) analysis revealed that the total cellular fatty acid composition of KKI was significantly changed in response to phenanthrene. Fatty acids such as 14:0, 16:0 3OH, 17:0 cyclo, 18:1$\omega$7c, 19:0 cyclo increased in phenanthrene-exposed cells, while fatty acids such as 10:0 3OH, 12:0, 12:0 2OH, 12:0 3OH, 16:1$\omega$7c, 15:0 iso 2OH, 16:0, 18:1$\omega$6c, 18:0 decreased.

  • Purpose: Agent Orange (AO) is a herbicide and defoliant used by the United States and its military allies during the Vietnam War. Pneumonia is a common cause of death among Vietnam veterans in our hospital. There have been no previous studies researching any association between AO exposure and the prognosis for pneumonia. The primary objective of this study was to investigate associations between AO exposure and 30-day mortality due to pneumonia. The secondary objective was to examine the clinical factors associated with therapeutic outcomes in veterans with pneumonia, and to assess the prevalence of combined diseases in AO-exposed veterans. Methods: This study retrospectively included veteran patients diagnosed with pneumonia in the emergency department and hospitalized between February 2014 and March 2018. The enrolled patients were grouped according to their defoliant exposure history, and the clinical information of defoliant-exposed and non-defoliant-exposed groups were compared. Patients were divided according to 30-day mortality, and significant factors influencing mortality were evaluated by using univariate analysis and multivariate analysis. The final multivariate model revealed the effect of AO exposure on therapeutic outcomes of pneumonia. Results: A total of 1006 patients were analyzed. Of these, 276 patients had a history of AO exposure, whereas 730 patients had not been exposed. Factors positively associated with 30-day mortality were malignancy, respiratory rate, blood urea nitrogen, and albumin which was negatively associated with mortality. Conclusion: Exposure to defoliant is not associated with 30-day mortality in patients with pneumonia. However, veterans with defoliant exposure are associated with a high prevalence of diabetes mellitus, hypertension, cerebrovascular accident, malignancy, and chronic kidney disease.

  • Polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs) are ubiquitous and can contaminate the food chain. A study monitoring PCDD/Fs in imported meat was conducted at the National Veterinary Research and Quarantine Service (NVRQS, Republic of Korea) in order to maintain food safety from the bioaccumulative PCDD/Fs. Seventeen PCDD/Fs with toxic equivalency factors (TEFs) established by World Health Organization (WHO, 1998) were analyzed in imported beef, pork, and chicken by high resolution gas chromatography/mass spectrometry (HR-GC/MS). Results of the monitoring for the last 5 years are presented. The levels of PCDD/Fs were similar to other studies except two unusually high concentrations in pork and beef. Excessive levels greater than the Korean provisional maximum residue limit of PCDD/Fs were found in a sample of pork imported from Chile and a sample of beef imported from U.S, and those products were rejected and returned. There was no obvious trend or differences with respect to time or origin of meat in this study.

Effects of Formononetin on the Aryl Hydrocarbon Receptor and 7,12-Dimethylbenz[a]anthracene-induced Cytochrome P450 1A1 in MCF-7 Human Breast Carcinoma Cells

  • Formononetin is an isoflavonoid phytoestrogen found in certain foodstuffs such as soy and red clover. In this study, we examined the action of formononetin with the carcinogen activation pathway mediated through the aryl hydrocarbon receptor (AhR) in MCF-7 breast carcinoma cells. Treating the cells with formononetin alone caused the accumulation of CYP1A1 mRNA as well as elevation in CYP1A1-specific 7-ethoxyresorufin O-deethylase (EROD) activity in a dose dependent manner. However, a concomitant treatment with 7,12-dimethylbenz[a]anthracene (DMBA) and formononetin markedly reduced both the DMBA-inducible EROD activity and CYP1A1 mRNA level. Under the same conditions, formononetin inhibited the DMBA-induced AhR transactivation, as shown by reporter gene analysis using a xenobiotic responsive element (XRE). Additionally, formononetin inhibited both DMBA-inducible nuclear localization of the aryl hydrocarbon receptor (AhR) and metabolic activation of DMBA, as measured by the formation of the DMBA-DNA adducts. Furthermore, formononetin competed with the prototypical AhR ligand, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), for binding to the AhR in an isolated rat cytosol. These results suggest that formononetin might be considered as a natural ligand to bind on AhR and consequently produces a potent protective effect against DMBA-induced genotoxicity. Therefore, that's the potential to act as a chemopreventive agent that is related to its effect on AhR pathway as antagonist/agonist.

Effect of Genistein on the Aryl Hydrocarbon Receptor and Cytochrome P450 1A1 in MCF-7 Human Breast Carcinoma Cells (인체유방암 세포주 MCF-7 세포에서 genistein의 Aryl Hydrocarbon Receptor와 Cytochrome P450 1A1에 대한 영향)

  • 화학적 예방효과가 있는 식물성 에스트로젠은 다양한 환성을 나타내며 여러 세포 수용체와 상호작용한다. Genistein은 isoflavone의 주요물질 중의 하나로 콩류에 존재하며 대표적인 식물성 에스트로젠이다. 본 논문에서는 유방암 세포주인 MCF-7에서 aryl hydrocarbon receptor(AhR)에 의해 매개되는 발암물질 활성화 경로에 대한 genistein의 영향을 살펴보았다. 세포에 genistein을 처리할 경우 cytochrome P450 1A1(CYP1A1) 약물대사효소의 특이적인 효소반응인 7-ethoxyresorufin O-deethylase (EROD) 활성도와 CYP1A1의 유전자 발현이 genistein의 농도 의존적으로 증가하였다. Genistein과 발암물질인 방향족탄화 수소 7, 12-dimethylbenz[a]anthracene(DMBA)를 동시 처리하였을 경우 DMBA에 의해 유도되어 증가된 EROD활성도와 CYP1A1의 유전자 발현이 genistein에 의해 감소하였다. 랫트의 간에서 분리한 세포질을 이용하여 genistein과 AhR의 대표적인 ligand인 2,3,7,8-tetrachlorodibenzo-p-dioxin과 경쟁적 결합에 대한 영향을 조사한 결과 genistein이 AhR에 경쟁적으로 결합함을 알 수 있었다. 이러한 결과들은 genistein이 천연 AhR ligand임을 암시한다. 따라서, 식물성 에스트로젠인 genistein은 AhR경로의 길항제/항진제로 작용할 수 있을 것으로 사료된다.

  • Objectives: The concentration distributions of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), dioxin-like polychlorinated biphenlys (dl-PCBs), and polycyclic aromatic hydrocarbons (PAHs) in fine particles were investigated to provide basic data on POP behavior and composition analysis. Methods: The concentrations of PCDD/Fs, dl-PCBs, and PAHs by particle size were evaluated for TSP, PM10, and PM2.5. Also, fine dust component analysis and factor analysis were performed to identify the source of PCDD/Fs. Results: The particle size distribution was found to account for 24.3% of >10 ㎛, 14.5% of 2.5-10 ㎛, and 61.2% of <2.5 ㎛. The average contributions of coarse particles (>2.5 ㎛) and fine particles (<2.5 ㎛) were PCDD/Fs 67%, dl-PCBs 66%, benzo (a) pyrene 83% and PAHs 84%, and the contributions of fine particles (<2.5 ㎛) were higher than coarse particles (>2.5 ㎛). However, the contributions of coarse particles increased in April to September with higher temperatures, while those of fine particles increased in February to March with lower temperatures. Conclusions: Low chlorinated (4Cl-5Cl) PCDD/Fs were more adsorbed compared to coarse particles due to the influence of pollutant migration from particulate to gas phase according to temperature rise, whereas high chlorinated (6Cl-8Cl) PCDD/Fs were more adsorbed compared to fine particles. PCDD/Fs sources were assessed to be major sources of emissions, such as incineration facilities and/or open burning.
