This study is intended to give information on agricultural nonpoint source pollution and transport related to fertilizer application and irrigation practice. Field-simulated soil columns were set up and leaching studies on fertilizer components such as nitrogen and phosphorus were performed. Nitrogen and phosphorus in the leachate showed different trends in each column and nonpoint source pollution in agricultural areas may be expected to depend on planted crops, soil conditions, and climate as well as irrigation and fertilizing management.
The possibility of microalgal nitrogen treatment was tested in wastewaters with a low carbon/nitrogen (C/N) ratio. Chlorella kessleri was cultured in the two different artificial wastewaters with nitrate as a nitrogen source: one contained glucose for an organic carbon source and the other without organic carbon sources. The growth rates of the two cultures were almost identical when the aeration rate was over 1 vvm. These results suggest that microalgae could successfully remove nitrogen from wastewater, as far as the mass transfer of $CO_2$, was not limited. Nitrate was successfully reduced to below 2 mg $NO_3^-$-N/ml from the initial nitrate concentration of 140 mg $NO_3^-$-N/ml in 10 days, even in the wastewater with no organic carbon source. Similar results were obtained when ammonium was used as the sole nitrogen source instead of nitrate. Higher concentrations of nitrogen of 140, 280, 560 and 1,400 mg/ml were also tested and similar amounts of nitrogen were removed by algal cultures without showing any substrate inhibition.
Isolates of Ophiocordyceps heteropoda (Kobayasi) collected from Mt. Halla on Jeju-do, Korea were tested for mycelial growth on different agar media and in the presence of different carbon and nitrogen sources. Similarly, isolates were also incubated at different temperatures as well as under continuous light and dark conditions. Growth was better on Hamada agar, basal medium, and malt-yeast agar, but poor on Czapek-Dox agar. Different carbon sources such as dextrin, saccharose, starch, lactose, maltose, fructose, and dextrose resulted in better growth. Complex organic nitrogen sources such as yeast extract and peptone revealed the most effective growth. Mycelial growth was best at $25^{\circ}C$. The growth rate was faster in the dark than the light, but mycelial density was less compact in the dark.
Cultural conditions for mass production of the antagonistic bacteria, Bacillus subtills CAP134 against pathogens causing major airborne diseases to apple tree, effect of temperature, pH, carbon and nitrogen source in the culture broth were investigated. The bacterial growth was most vigorous when the temperature and pH of the culture broth was 30~$35^{\circ}$C, and 7, respectively. As for carbon source, dextrose was best followed in order by dextrose(monosaccharides)>sucrose(disaccharides)$\geq$saccharose(di-saccharides)>starch (polysaccharides). Among different sugars, bacterial growth was favored by in the order of brown, black and white sugars, indicating that the bacterial growth might be promoted by the minor elements presented as impurities in the less purified sugars. As for nitrogen source, organic forms were better to bacterial growth than inorganic forms, that is polypeptone was best followed in order by soy sauce, soybeen milk and inoganic nitrogens. Differences in bacterial growth among different forms of inorganic nitrogen were negligible.

Possibility of biological nitrogen treatment was tested in wastewaters with low C/N ratio. Chlorella kessleri was inoculated at $10^6\;cell/mL$ of initial density in two different artificial wastewaters: one that contained glucose for organic carbon source and the other without carbon source. Nitrate could be successfully reduced below 10 mg $NO_3/mL$ from initial nitrate concentration of 560 mg $NO_3/mL$ in 10 days even in the wastewater without carbon source, This 98% removal of nitrate without extra organic carbon source lights up the future of biological wastewater treatment, where the insufficient ability of nitrogen removal is a major problem.
The carotenoid biosynthesis of a red oleaginous yeast, Rhodotorula glutinis was significantly changed when the yeast was grown on different carbon substrates. The highest carotenoid production was obtained on culture medium containing glucose when the carbon to nitrogen ratio (C/N ratio) was adjusted to 25.7. Galactose stimulated the biosynthetic rate of torularhodin, a xanthophyll component of the yeast. With decreasing C/N ratio of the medium, significant changes of $\gamma$-carotene and torularhodin were observed such that increase in the torularhodin concentration was nearly equal to the decrease in $\gamma$-carotene. It was speculated that the nature of carbon substrate affected the metabolic rate of the cell, and accompanied by the different pattern of carotenoid accumulation in the cell.

Increasing demands on fossil fuel have led to the unprecedented attraction to microalgal biofuel as an alternative energy. In this study, we investigated growth and lipid productions of microalga Nannochloris oculata under various carbon dioxide or nitrogen source concentrations and irradiance conditions. Biomass production of N. oculata was highest under 2% $CO_2$ with 0.3 flow rate (vvm). In addition, biomass productivities were proportional to the concentration of nitrogen source, whereas lipid biosynthesis was suppressed under higher nitrogen concentration (up to 50 mg/L). High irradiation ($160{\sim}180\;{\mu}mol/m^2{\cdot}s$) enhanced growth rate and lipid production of N. oculata.
Natural astaxanthin mainly derives from a microalgae producer, Haematococcus pluvialis. The induction of nitrogen starvation and high light intensity is particularly significant for boosting astaxanthin production. However, the different responses to light intensity and nitrogen starvation needed to be analyzed for biomass growth and astaxanthin accumulation. The results showed that the highest level of astaxanthin production was achieved in nitrogen starvation, and was 1.64 times higher than the control group at 11 days. With regard to the optimization of light intensity utilization, it was at $200{\mu}mo/m^2/s$ under nitrogen starvation that the highest astaxanthin productivity per light intensity was achieved. In addition, both high light intensity and a nitrogen source had significant effects on multiple indicators. For example, high light intensity had a greater significant effect than a nitrogen source on biomass dry weight, astaxanthin yield and astaxanthin productivity; in contrast, nitrogen starvation was more beneficial for enhancing astaxanthin content per dry weight biomass. The data indicate that high light intensity synergizes with nitrogen starvation to stimulate the biosynthesis of astaxanthin.
Saccharomyces cerevisiae is one of the most employed cell factories for the production of bioproducts. Although monomeric hexose sugars constitute the preferential carbon source, this yeast can grow on a wide variety of nitrogen sources that are catabolized through central nitrogen metabolism (CNM). To evaluate the effects of internal perturbations on nitrogen utilization, we characterized strains deleted or overexpressed in GLT1, encoding for one of the key enzymes of the CNM node, the glutamate synthase. These strains, together with the parental strain as control, have been cultivated in minimal medium formulated with ammonium sulfate, glutamate, or glutamine as nitrogen source. Growth kinetics, together with the determination of protein content, viability, and reactive oxygen species (ROS) accumulation at the single cell level, revealed that GLT1 modulations do not significantly influence the cellular physiology, whereas the nitrogen source does. As important exceptions, GLT1 deletion negatively affected the scavenging activity of glutamate against ROS accumulation, when cells were treated with H2O2, whereas Glt1p overproduction led to lower viability in glutamine medium. Overall, this confirms the robustness of the CNM node against internal perturbations, but, at the same time, highlights its plasticity in respect to the environment. Considering that side-stream protein-rich waste materials are emerging as substrates to be used in an integrated biorefinery, these results underline the importance of preliminarily evaluating the best nitrogen source not only for media formulation, but also for the overall economics of the process.
