식이섬유 첨가식이 Streptozotocin-유도 당뇨쥐의 장기능과 지질 및 당질대사에 미치는 영향 (The Effects of Dietary Fiber Feeding on Gastrointestinal Functions and Lipid and Glucose Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study was conducted to compare the effects of four types of dietary fiber supplementations (cellulose, pectin, guar gum, and polydextrose) on gastrointestinal function, diabetic symptom amelioration and lipid & glucose metabolism in streptozotocin-induced diabetic rats. Six groups of male rats were fed ad libitum dietary fiber-free control diet or one of experimental diets containing 5% dietary fiber for four weeks. All types of dietary fiber supplementation seemed to protect the diabetic animals from the loss of body weight. The primary diabetic symptoms such as polydipsia, polyphasia, polyuria and urinary glucose excretion were ameliorated by cellulose, pectin, and guar gum, but not by polydextrose. Gastrointestinal transit time was significantly shortened and fecal dry weight was significantly increased in all the dietary fiber-supplemented groups except the polydextrose group. Large intestine was significantly lengthened by dietary fiber feeding. The serum triglyceride and total cholesterol levels were effectively lowered by pectin, guar gum and polydextrose. Regardless of their types, the fiber supplementation had no effect on serum HDL-cholesterol. Whereas fasting blood glucose level was significantly lowered by all types of fiber supplementations, glucose tolerance was more effectively improved by pectin and guar gum.

철분과 셀레늄의 섭취수준이 흰쥐의 망간 이용에 미치는 영향 (Effects of Iron and Selenium Intakes on Utilization of Manganese in Rats)

  • The purpose of this study was to investigate the effect of iron and selenium intakes on utilization of manganese in rats fed adequate, 2-fold, 4-fold iron and adequate, high selenium for 6 weeks. There was no difference feed intake across iron and selenium containing diet groups. Body weight gain in 2-fold iron and high selenium group(MFeHSe) was significantly higher than those in other groups. Serum iron level was increased with iron increment, and liver iron content was decreased with selenium supplementation. Selenium and manganese contents in tissues were decreased with iron increment. In the case of manganese balance, manganese excretion through feces was significantly increased as iron intake was increased. However, retention and apparent absorption of manganese were not significantly affected by dietary iron. From these results, it could be suggested that the supplementations of iron and selenium affected the manganese utilization. Therefore, it must be considered interaction with various minerals in micro-nutrient supplementations.

골다공증 모델 흰쥐에서 고수준의 단백질의 칼슘 섭취가 칼슘대사 및 신장기능에 미치는 영향 (Effects of High Protein and Calcium Intakes on Calcium Metabolism and Renal Function in Ovariectomized Osteporosis Rat Model)

  • This study was to investigate interaction between dietary protein and Ca levels in Ca metabolism and renal function in osteporosis rats. Five week-old female rats were fed a low Ca diet for 4 weeks after ovariectomy operation to establish rat models of osteoporosis. The ovariectomized osteoporosis rats were divided into six groups and were fed experimental diets which contained two levels of protein, normal (20%) and high(40%) , and three levels of Ca, low (0.06%), normal (0.47%) and high(0.94%) for 4 weeks , respectively. The ovaricetmized rat model of osteoporosis showed a remarkable decrease in serum Ca concentration, fresh weight and breaking force of femur, Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca at the dietary levels of normal and high levels significantly enhanced Ca bioavailability shown in the above experimental rat models of osteoporosis, regardless of dietary protein levels ; whereas the rats which were fed the low Ca diet demonstrated rather a decrease in its bioavailability. Irrespectively of the dietary Ca levels, the rats which were fed high protein diet exhibited an increase in kidney weight, urinary Ca, volume and hydroxyproline, and glomerular filtration ratio(GFR). The results show that dietary protein and calcium levels affect the renal function and Ca metabolism independently, while the interaction between protein and calcium have not been shown.

두류의 첨가가 당뇨병 쥐의 장기능과 당뇨증세에 미치는 영향 (Effects of Legume Supplementation on the Gastrointestinal Function and Diabetic Symptoms in Streptozotocin-Induced Diabetic Rats)

  • The present study was conducted to evaluate the usefulness of four kinds of legumes(black soybean: BS, yellow soybean: YS, green peas: GP, soybean curd residue: SCR) as a high-fiber supplement in the therapeutic diet for diabetic patients. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of legumes for 6 weeks. The effects of legumes on the gastrointestinal function and diabetic symptoms in diabetic rats were also examined and compared with the effects of cellulose or pectin diet. Legume supplementations had no significant effects on alleviating typical diabetic symptoms such as polydipsia, polyphasia, polyuria, and urinary glucose excretion when compared with cellulose or pectin supplementations. The supplementations of legumes all resulted in remarkable changes in gastrointestinal functions: shortening of GI transit time and increases in fecal volume, fecal water and crude fat contents. Legume supplementations appeared to supress the damages of the small intestine villi and the enteritis symptoms in diabetic rats. GP led to regular villus patterns closer to normal villus. BS, YS and GP led to the enlargement of cecum in diabetic rats. In conclusion, the present studies have demonstrated that legumes may possess a significant physiological actions in the control of various diabetic conditions.

Effect of Monensin and Live Yeast Supplementation on Growth Performance, Nutrient Digestibility, Carcass Characteristics and Ruminal Fermentation Parameters in Lambs Fed Steam-flaked Corn-based Diets

  • In performance, digestibility and slaughter trials, a total of forty five male weaned lambs were used to examine the effects of monensin and live yeast supplementations on growth performance, nutrient digestibility, carcass characteristics and ruminal fermentation parameters when the lambs were fed steam-flaked corn-based diets. Animals were allotted to one of three treatment diets in a completely randomized design. The three treatment diets were: (1) basal diet (CON) with steam-flaked corn as a sole grain source, (2) basal diet supplemented with monensin (MO), and (3) basal diet supplemented with live yeast (LY). Total average daily intake (ADI) was unaffected by MO and LY supplementations. LY supplementation increased (p<0.05) average daily gain (ADG) by 13.1% compared with the CON diet. Both MO and LY supplementations resulted in a significant improvement (p<0.05) of feed efficiency over the CON diet (4.47, 4.68 vs. 5.05). Hemicellulose digestibility was higher (p<0.05) for lambs in the LY supplementation group (62.4%) as compared with the CON group (55.7%), but no differences were observed in digestibilities of dry matter (DM), organic matter (OM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF). All carcass traits were not influenced by dietary supplementations. Ruminal pH in lambs fed the LY supplemental diet was more stable than that with the CON diet (6.57 vs. 6.17). Neither MO nor LY supplementation influenced the concentration of ruminal ammonia-N and total volatile fatty acid (VFA), and molar percentages of individual VFA. Plasma urea-N concentration was decreased (p<0.05) by MO and LY supplementations, while plasma ${\beta}$-hydroxybutyrate (BHBA), glucose and other blood parameters were unaffected. In conclusion, while both MO and LY supplementations had a positive impact on feed efficiency and LY supplementation stabilized ruminal pH and improved fiber utilization, none of the supplements had the capacity to significantly enhance the carcass characteristics.

식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 항산화계에 미치는 영향 (Effects of Vitamin E on Antioxidative Defense System of Liver in Acute Cadmium-Poisoned Rats)

  • The purpose of this study was to investigate the effects of vitamin E on antioxidative defense system of liver in acute cadmium poisoned rats. Sprague-Dawley male rats weighing 100$\pm$10gm were randomly assigned to one control and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(OE-Cd group), 40mg vitamin E per kg diet(40E-Cd group) or 400 mg vitamin E per kg diet(400E-Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd$^2$$\^$+//kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4weeks. Activities of superoxide dismutase(SOD), glutathione peroxidase(GSHPx) and glutathione S-transferase(GST) were decreased in cadmium injected groups but those were significantly improved by dietary vitamin I supplementations. Vitamin E contents reduced glutathione(GSH) in the live were decreased in cadmium injected groups, but we., not significantly different among three groups with different levels of vitamin E supplementations. Contents of liver thiobarbituric acid reactive substance (TBARS) of 0E-Cd group were higher than those of 400E-Cd and 400E-Cd groups, but those were markedly alleviated according to vitamin E supplementations. These results indicate that cadmium poisoning in rats causes decreasing antioxidative defense system and increasing peroxidative damage in liver, however can be restored by vitamin E supplements. The Effects of Dietary Sulfur and Vitamin E Supplementation on the Quality of Beef from the Longissimus Muscle of Hanwoo Bulls

  • This study was carried out to investigate the effects of dietary sulfur (S) and vitamin E (Vit E) supplementation on the quality of beef from longissimus muscle of Hanwoo bulls. Eleven, 29 months-aged Hanwoo bulls were randomly divided into three feed supplementation groups (S (n = 4), Vit E (n = 4) and S+Vit E (n = 3)). S was given as much as 12 g/head/d and Vit E was given as much as 1,200 IU/head/d; both supplements were given for 3 months prior to slaughter. At 24 h post-slaughter, the carcasses were weighed and evaluated by official grader for carcass traits. At 48 h post-slaughter, the M. longissimus from each carcass was collected and stored at $4{\pm}0.2^{\circ}C$ for 10 days. There were no significant differences in yield and quality grades of carcass and proximate composition, physical properties and aroma pattern of meat among all groups. At 5 days of storage, the TBARS value of meat from cattle with S+Vit E supplementation was significantly lower (p<0.05) than other groups; and at 10 days of storage, the TBARS value of meat from cattle with Vit E and S+Vit E supplementations was significantly lower (p<0.05) than from cattle with S supplementation. At 5 days of storage, MetMb concentration of meat from cattle with S+Vit E supplementation was significantly lower (p<0.05) than from cattle with Vit E supplementation; and at 10 days of storage, MetMb concentration of meat from cattle with S+Vit E supplementation was significantly lower (p<0.05) than from other groups. At 10 days of storage, the redness value of meat from cattle with S supplementation was significantly higher (p<0.05) than from cattle with Vit E supplementation and the hue-angle value of meat from cattle with S and S+Vit E supplementations was significantly lower (p<0.05) than from cattle with Vit E supplementation. Dietary Vit E supplementation had a better effect on lipid stability whereas dietary S supplementation had a better effect on OxyMb stability. The dietary combination of S and Vit E created the highest protection for beef from myoglobin oxidation and thus improved the color stability of meat.

Therapeutic Potential of Membrane Fatty Acid Modifiaction in Tumor Cells

  • The membrane fatty acid composition of tumor cell can be modified either in cell by altering the lipid composition of the medium of during growth in animals by changing the dietaty fat composition. These modifications are associated with changes in membrane physical properties and certain cellular functions, including carrier-mediated transport and enzyme contained within the membrane. Such effects influence the transport of nutrients and chemotherapeutic agents in cancer cells .Fatty acid modification also can enhance the sensitivity of the neoplastic cell to chemotherapy. The alteration in plasma membrane composition will be affected through dietary supplementations and the potential value to cancer patients could be a better understanding of the effects of diet on responsiveness of neoplasms to chemotherapy, i.e. cancer patients' chances for a "cure" can be improved by diet changes prior to treatment.

사료 내 타우린 첨가가 육계의 복강지방 무게 및 혈청과 간의 콜레스테롤 농도에 미치는 영향 (Effects of Dietary Taurine on the Abdominal Fat Weight and Serum and Liver Concentrations of Cholesterol in Broiler Chicks)

  • 타우린 첨가가 복강지방 및 간과 혈청 중 콜레스테롤 농도에 미치는 영향을 육계 수컷병아리에서 조사하였다. 1일령 병아리를 사료에 타우린을 첨가하지 않은 대조구와 1% 그리고 2%를 첨가한 3가지 시험구에 균일하게 분포시켰다. 2% 타우린 첨가 시 육계의 증체량은 6% 감소하였으나, 사료효율은 처리구간 차이가 없었다. 간 중량과 체중대비 간 중량도 처리구간 차이가 없었으나, 복강지방은 1%와 2% 타우린 첨가구가 대조구에 비하여 각각 14%와 20% 낮았다. 혈청 중 중성지방, glucose, GOT, 및 GPT 농도는 처리구간 차이가 없었으나, 혈청 중 총 콜레스테롤 농도는 1%와 2% 타우린 첨가구가 대조구에 비하여 각각 10%와 12% 증가하였다. HDL-콜레스테롤 역시 1%와 2% 첨가구가 대조구에 비하여 각각 20%와 34% 높았다. 간의 총 콜레스테롤 농도는 1%와 2% 첨가구가 대조구에 비하여 각각 9%와 13% 낮았으며, 간의 HDL-콜레스테롤 농도 역시 1%와 2% 첨가구가 대조구에 비하여 각각 20%와 38% 낮았다. 본 연구의 결과는 타우린 첨가가 육계의 복강지방의 축적을 억제시키며 콜레스테롤 대사에 영향을 미친다는 것을 보여준다.

Alpha-Tocopherol Transfer Protein (${\alpha}$-TTP): Insights from Alpha-Tocopherol Transfer Protein Knockout Mice

  • Alpha-tocopherol transfer protein (${\alpha}$-TTP) is a liver cytosolic transport protein that faciliates ${\alpha}$-tocopherol (${\alpha}$-T) transfer into liver secreted plasma lipoproteins. Genetic defects in ${\alpha}$-TTP, like dietary vitamin E deficiency, are associated with infertility, muscular weakness and neurological disorders. Both human and ${\alpha}$-TTP deficient (${\alpha}-TTP^{-/-}$) mice exhibit severe plasma and tissue vitamin E deficiency that can be attenuated by sufficient dietary ${\alpha}$-T supplementations. In this review, we summarize the literature concerning studies utilizing the ${\alpha}-TTP^{-/-}$ mice. Levels of vitamin E in the ${\alpha}-TTP^{-/-}$ mice do not appear to be directly related to the amounts of dietary ${\alpha}$-T or to the levels of ${\alpha}$-TTP protein in tissues. The ${\alpha}-TTP^{-/-}$ mice appear to present a good model for investigating the specific role of ${\alpha}$-T in tissue vitamin E metabolism. Furthermore, ${\alpha}-TTP^{-/-}$ mice appear to be useful to elucidate functions of ${\alpha}$-TTP beyond its well recognized functions of transferring ${\alpha}$-T from liver to plasma lipoprotein fractions.
