This study was designed to confirm the effect of dietary protein level and oral administration of tryptophan on brain serotonin metabolism. Two animal experiments were conducted. The objectives and results of research were as follows : In the first experiment, it was investigated whether administration of reserpine to Sprague-Dawley rats fed 6% or 20% casein diet induced decrease in serum tryptophan and large neutral amino acid(LNAA) concentrations, tryptophan/LNAA concentration ratio, brain tryptophan, serotonin and 5-hydroxyindoleacetic acid(5-HIAA) contents. Brain serotonin content of 6% casein diet group was lower than those of 20% casein diet group. Both 6% and 20% casein diet groups administered with reserpine to induce the analogous depression, showed the notable decrease in brain serotonin content when they were compared with 20% casein diet group not administered with reserpine. Serum tryptophan/LNAA ration and brain 5-HIAA content showed a tendency similar to the change of serotonin content, but the mean difference among all groups was not significant. From these results, it could be said that when the dietary protein level was low, brain serotonin content was decrease. The second experimnt was to see the change in serum tryptophan concentration and tryptophan/LNAA ratio and brain tryptophan, serotonin and 5-HIAA content when tryptophan was administered orally to the animals treated with reserpine. Serum tryptophan concentration tended to increase in both reserpine-treated 6% and 20% casein diet groups administered with tryptophan, especially in the 6% casein diet group. Serum tryptophan/LNAA concentration ratio tended to incrase in reserpine-tteated 6% casein diet group, while decrease in reserpine-treated 20% casein diet group. Brain tryptophan content was increased in both reserpine-treated 6% and 20% casein diet groups. However, brain serotonin content of reserpine-treated 6% casein diet group showed a tendency to decrease, while that of reserpine-treated 20% casein group increase. Consequently, the effect of tryptophan administration on increase of brain tryptophan and serotonin content in animals treated with reserpine was far more excellent in 20% casein diet groups. It was concluded that dietary protein intake and tryptophan administration increase brain serotonin level. Accordingly, it was possible to confirm that brain function, particularly in aspect of behavior related to the serotonin, was changed with manipulation of dietary composition.
Objectives : The aim of this study was to investigate the relationship between the contributions of macronutrients to the total energy intakes and Body Mass Indices (BMI, $kg/m^2$) of Korean women. Methods : We used dietary records data from 115 healthy Korean women, ages 20 and over, who completed four 3-day dietary records between February 2003 and March 2004. For the calculation of nutrients we used a dietary assessment program developed by the Korean Nutritional Society. Macronutrient intakes were estimated by averaging individual total daily intakes in four 3-day dietary records. Subjects were categorized into three Body Mass Index (BMI) groups : underweight (BMI < 20), normal (20 $\leq$ BMI < 25), and overweight (BMI $\geq$ 25) group. Results : The total energy intakes were not different among the three BMI groups. Similarly, other macronutrient intakes such as protein, fat, carbohydrate, vegetable protein, animal protein, saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids were not different. From the multivariate nutrient density model, substituting polyunsaturated fatty acid for carbohydrate was positively associated with BMI in women aged 20 to 49 ($\beta=2.31$, p<0.01). In women aged 50 and over, substituting animal protein for carbohydrate was positively associated with BMI ($\beta=0.549$, p=0.04). Conclusions : No single macronutrient was associated with BMI when all subjects were combined. However, when stratified by age, polyunsaturated fatty acid intake in younger women, and animal protein intake in older women, were positively associated with BMI. In the future, we recommend a larger study to confirm these results.
This study was designed to observe the effects of both control and atherogenic diets on the cholesterol and triglyceride (TG) in serum and liver of adult rats fed diets supplying two levels of dietary fat and two different sources of dietary protein in early life. For the first experimental period, the rats were assigned into the four diet groups: High fat, casein (HC); High fat, gluten (HG); Low fat, casein (LC): Low fat, gluten (LG). Each group was subdivided into control and atherogenic groups for the second experimental period. Cholesterol and TG were determined in serum and liver after 7 hr fasting. The body weight gain was greater in the rats of the casein groups than those of the gluten groups tut not influenced by the level of the dietary fat. The difference in body weight from the quality of dietary protein in the first period was not disappeared even after the second period. After the first period, higher serum cholesterol was observed in the rats fed either casein or high fat diets. With the second experimental diet, rats fed atherogenic diet showed higher serum cholesterol concentration but lower serum TG levels compared to those fed control diet, regardless the diets fed in the first period. Serum cholesterol level of the rats of both groups which had been fed high fat diets in early life was increased compared to those of the low fat diet groups. This effect was more pronounced with the atherogenic diet groups than control groups. However, no differences were found in serum cholesterol levels resulted from the different types of dietary protein fed in the first period. Serum TG concentration was not influenced by the quality of protein and level of fat in the diet but seemed to be mere affected by the amount of carbohyrates in the diet. Liver cholesterol per unit weight was greater in the gluten diet groups than in the casein groups but total cholesterol was higher in casein fed rats. There were no differences in liver TG among the groups.
This study was designed to evaluate the economic analysis of channel catfish production in 1998 based on fish value and total feed cost. Catfish received higher protein feeds with lesser amount based on the dietary protein levels, but received the constant total protein input for all treatments. Weight gain per pond for treatment 1 $(28\%\;protein,\;100\%\;of\;satiation)$ was higher (P<0.10) than for treatment 3 $(36\%\;protein,\;77.8\%\;of\;satiation)$, but not significantly higher than for treatment 2 $(32\%\;protein,\;87.5\%\;of\;satiation)$ at constant DE. At constant DE/P (treatments 4, 2 and 5), weight gain per pond for treatment 5 $(36\%\;protein,\;77.8\%\;of\;satiation)$ was lower (P<0.10) than for treatment 2, but not significantly lower than for treatment 4 $(28\%\;protein,\;100\%\;of\;satiation)$. At constant DE, feed conversion slightly improved as dietary protein level increased from $28\%\;to\;32\%$ and feed allowance decreased by $12.5\%$, but did not improve further as dietary protein level increased from $28\%\;to\;36\%$ and feed allowance decreased by $22.2\%$. At constant DE/P, feed conversion improved as dietary protein level increased from $28\%\;to\;32\%$ increased and feed allowance decreased by $12.5\%$, but did not improve as dietary protein level increased from $28\%\;to\;36\%$ and feed allowance decreased by $22.2\%$ Total feed cost for treatment 1 was slightly, but not significantly higher than for treatments 2 and 3 at constant DE. At constant DE/P, total feed cost for treatment 5 was higher (P<0.05) than for treatment 2, but not significantly higher than for treatment 4. Total value of fish ($ /ha) produced for treatment 1 was highest and lowest was for treatment 5. Return above feed cost was highest for treatment 1 and nearly the same as treatment 2. Return over feed cost for treatments 3 and 4 were slightly lower than for treatments 1 and 2. Economic analysis showed that feeding fish the diet containing $28\%$ protein and 3.08 kcal/g DE to satiation and the diet containing $32\%$ protein and 3.08 kcal/g DE to $87.5\%$ of satiation produced the highest profit to farmer.
The effect of dietary protein on growth and lipid levels of plasma and liver was studied in weanling male rats fed diets differing protein sources and amino acid balance. Rats were devided into 9 experimental diets which were grouped into 3 categories ; 1) Simple protein category includes gluten-, soy protein isolate-, and casein-containing diet groups, 2) Supplemented category includes casein supplemented with methionine, soy protein isolate supplemented with methionine, and gluten supplemented with lysine and methionine, 3) Mixed protein category includes diet groups containing gluten (2/3), casein (1/3), soy protein isolate (2/3) and casein (1/3), and casein (1/3), soy protein isolate (1/3) and gluten (1/3). The experimental diets composed of 15% protein, 65.8% carbohydrate, 10% fat and 1% cholesterol. The body wt. gain and P.E.R. were greater in rats of supplemented and mixed protein groups than simple protein groups. No statistical differences were found in plasma cholesterol among gluten, soy protein isolate and casein groups. Consumption of diets supplemented with limiting amino acid to gluten or soy protein isolate reduced the plasma cholesterol level by 23.2% and 34.2% respectively. However there was no difference between casein and the supplemented casein groups. The mixed protein groups shows relatively high plasma cholesterol concentration and low liver cholesterol levels. On the other hand gluten group showed low plasma cholesterol and high liver cholesterol levels, which means body cholesterol pool may not have been changed by the dietary protein. Feeding soy protein meal and the supplemented soy protein isolate resulted in lower plasma cholesterol, plasma triglycerides, liver cholesterol and liver triglycerides levels. This hypolipidemic effect is considered to see unique to soy protein isolate. Rats in gluten and the supplemented gluten groups showed lower plasma protein levels and a tendency of fatty liver.

The present study examined the effects of excessive dietary protein and energy on growth response to clenbuterol in broilers. The chicks were allocated into 6 groups at 14d old, and used for a $3{\times}2$ factorial experiment. Birds were fed six diets, the control diet containing 21% crude protein (CP) and 3,100 kcal of metabolizable energy ME/kg, a high protein (30% CP) or a high energy (3,500 kcal/ ME/kg) diet, with or without 1 ppm clenbuterol, for 18 d. Clenbuterol feeding markedly decreased (p < 0.05) body weight gain by 23% in the high energy group. Feed intake was also decreased (p < 0.05) by clenbuterol administration across diet treatments. Abdominal fat weight was reduced (p < 0.05) by clenbuterol only when chickens were fed the high energy diet. Clenbuterol increased (p < 0.05) leg muscle weight in the control diet group, but decreased (p < 0.05) it in the high energy group. Muscle protein concentration was increased by 11 % in leg muscle only of the birds at the high energy level. In leg muscle, clenbuterol enhanced the protein/DNA ratio by 18%, except for the high protein group. These results indicate that feeding a diet containing excessive amounts of protein and more energy than normal did not necessarily improve growth response to clenbuterol.
Three dietary ME levels of 3,200, 2,900 and 2,600 kcal /kg in the same 13% single-stage low protein diet were compared to evaluate the effect of ME levels of grower diets on egg-type pullet growth and subsequent laying performance. As the ME levels of grower diets decreased, cumulative feed and protein consumptions increased(P<0.05), however, the ME intake and body weight at 18 wk of age decreased(P<0.05). Grower feed cost decreased as the dietary ME level was decreased, but no significant difference was found among dietary ME levels of grower diets. During the laying period, sexual maturity, hen-day egg production and average egg weight were not significantly affected by the ME levels of grower diet, however, daily feed intake and feed required per egg decreased as the dietary ME level of grower diet was reduced(P<0.05). Results of this study indicate that pullets can be reared on the low ME diet of 2,600 kcal /kg and it would he economical to utilize the low energy diet during the growing period when they are reared on a 13% single-stage low protein diet.
In order to study the interrelationships of calcium (0.45 vs. 0.90%), phosphorus (0.40 vs. 0.70%) and protein (17, 20, 23%), $2{\times}2{\times}3$ factorial design was employed. A total of 480 broilers (Hisex-Hibro) aged 3 days were fed the experimental diets for a period of 28 days. Body weight gain, daily feed intake and feed efficiency were investigated for the simple effects, first order interaction and second order interaction of the dietary factors. These effects were also applied to bone ash retention, percent Ca in bone & ash, percent P in bone & ash, and protein utilizability. Results were as follows. 1) For body weight gain, simple effects of dietary levels of Ca, P, CP were found to be significant (p<0.05). Body weight gain at 0.90% Ca level was improved as the dietary CP levels increased. For the feed intake, single effects of dietary levels of both P and CP were found (p<0.05). Feed efficiency was improved as the dietary CP and P levels increased. Ca $\times$ P interaction was found to be significant for body weight gain, feed intake and feed efficiency (p<0.05), however, Ca $\times$ P $\times$ CP interaction effect was not found. 2) Protein utilizability decreased as the dietary CP level increased (p<0.01). 3) 0.90% Ca in diet showed less bone ash retention than 0.45% Ca level. And, increasing the dietary P level resulted in increased bone ash retention. Increasing the dietary P level resulted in increased bone Ca retention (p<0.01) and increased bone P retention (p<0.05). Dietary CP levels had significant (p<0.01) effect on bone Ca retention except for 23% CP level. Increasing the dietary Ca level resulted in wider Ca:P ratio of bone, but increasing the dietary P level resulted in narrower Ca:P ratio of bone. 4. Ca $\times$ P interaction effects were found to be significant (p<0.01) for bone ash, bone Ca & P, ash P content, and bone Ca:P ratio. Ca $\times$ P $\times$ CP interaction effects were found for bone ash (p<0.01), bone Ca (p<0.05) and bone P content (p<0.01).
To investigate the effect of levels of dietary protein and age on bone metabolism 40% and 5% casein were fed to the rats of 2 & 13 months of age for 12 weeks. High protein groups showed higher bone weight and Ca content than low protein groups and urinary Ca loss was increased in high protein groups but the difference disappeared gradually. A significant increase in urinary hydroxyproline excretion was noted in high protein groups of both age. Another short term study was undertaken to study if the above effect was related with renal function or PTH. Extremely high and low protein diets(60%, 6%) were fed to the rats of different ages(6wks, 6mos.) for 2 weeks, Urinary Ca excretion was significantly increased in high protein groups of young and aged rats and GFR was increased as well. There was no difference in serum iPTH levels between low and high protein groups, but it was elevated in aged rats. Alkaline phosphatase activity was higher in young rats, reflecting faster bone formation. The observed hypercalciuria in high protein groups, especially in aged rats, seems to be related to higher GFR, and PTH dose not appear to be a major mediator.
Objective: The aim of this experiment was to evaluate the effects of different dietary ratio of metabolizable glucose (MG) to metabolizable protein (MP) on growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of 8 to 10-month-old heifers. Methods: A total of 24 Holstein heifers weighing an average of 282.90 kg (8 month of age) were randomly assigned to four groups of six. The heifers were fed one of four diets of different dietary MG/MP (0.97, 1.07, 1.13, and 1.26). Results: The results showed that the ratio of MG/MP affected the growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of heifers. The average daily gain of heifers was enhanced by increasing the ratio of MG/MP (p<0.05). The concentration of blood urea nitrogen, cholesterol, and low density lipoprotein cholesterol as well as the concentration of total volatile fatty acid in the rumen fluid of heifers decreased with the improvement in the ratio of dietary MG/MP (p<0.05). However, the relative amount of Ruminococcus albus and Butyrivibrio fibrisolvens in the rumen of heifers was increased significantly (p<0.05) when the dietary MG/MP increased. At the same time, with the improvement in dietary MG/MP, the amount of Fibrobacter succinogenes increased (p = 0.08). Conclusion: A diet with an optimal ratio (1.13) of MG/MP was beneficial for the improvement of growth, rumen fermentation, dietary protein and energy utilization of 8 to 10-month-old dairy heifers in this experiment.
