Modification of Gut Microbiota and Immune Responses via Dietary Protease in Soybean Meal-Based Protein Diets

  • Plant-based protein sources such as soybean meal have low digestibility and are generally promoted accumulation of undigested proteins into the intestine by enzymatic treatments. Moreover, potential intestinal pathogens ferment undigested proteins, producing harmful substances, such as ammonia, amines and phenols, leading to an overactive immune response and diarrhea in weaned pigs. As a solution, dietary proteases hydrolyze soybean-based antinutritive factors, which negatively affect immune responses and gut microbiota. In this study, we investigated the effects of dietary proteases (PRO) in a low-crude protein (CP) commercial diet on the immune responses and gut microbiota of weaned pigs. The experimental design consisted of three dietary treatments: a commercial diet as a positive control (PC; phase1 CP = 23.71%; phase 2 CP: 22.36%), a lower CP diet than PC as negative control (NC; 0.61% less CP than PC), and NC diet supplement with 0.02% PRO. We found that PRO tended to decrease the frequency of diarrhea in the first two weeks after weaning compared with PC and NC. In addition, pigs fed PRO showed decreased TNF-α and TGF-β1 levels compared with those fed PC and NC. The PRO group had a higher relative proportion of the genus Lactobacillus and lower levels of the genus Streptococcus than the PC and NC groups. In conclusion, the addition of PRO to a low CP commercial weaned diet attenuated inflammatory responses and modified gut microbiota in weaned pigs.

중년 남성에서 고정상혈압에 의한 고혈압발생 위험 규명을 위한 코호트내 환자-대조군 연구 (A Nested Case-Control Study on the High Normal Blood Pressure as a Risk Factor of Hypertension in Korean Middle-aged Men)

  • Objectives : High-normal blood pressure' is a factor influencing decision to initiate targeted intensive intervention strategy in westernized populations. JNC-VI offered the vigorous lifestyle modification for persons with 'high-normal blood pressure', who could be early detected. As a hypertension seems to be the result of multiple genetic factors operating in concert with associated environmental factors, it will be necessary to identify the high-normal blood pressure as a risk factor of hypertension for applying primary prevention strategy in Korean people. Methods : Although cohort study design might be adequate to recruit incidence cases, to keep time sequence of events, and to prevent information bias, nested case-control study was chosen for avoiding measurement errors because hypertension is a benign disease. Source population was the 'Seoul Cohort' participants and follow-up was done by using Korea Medical Insurance Corporation's database on the utilization of health services from 1 Jan93 to 30Jun97. Incidence cases were ascertained through the chart review, telephone contacts, and direct blood pressure measurements. Controls included the pairing of 4 individuals to each case on the basis of age. Results : As 75% of 247 incident cases had high-normal blood pressure, the crude odds ratio for hypertension was 2.04 (95% CI 1.47-2.83). Another statistically significant risk factors of hypertension were body mass index, dietary fiber, alcohol consumption, weekly activity and history of quitting smoking. The multivariate odds ratio of high-normal blood pressure adjusted for all risk factors was 1.84 (95% CI 1.31-2.56). Among high-normal blood pressure group, body mass index, weekly ethanol amounts, weekly physical activity, and dietary fiber except history of quitting smoking were still risk factors of hypertension. Conclusion : 'High-normal blood pressure' is a risk factor for hypertension in Korean middle-aged men, which represents that the vigorous lifestyle modification for persons with 'high-normal blood pressure' is need.

Modification of Hepatic Microsomal Cytochrome P450 2E1 Enzyme by Garlic Powder in Rat Hepatocarcinogenesis

  • This study was designed to investigate the effects of dietary garlic powder on cytochrome P450 enzymes and membrane stability in murine hepatocarcinogenesis initiated by diethylnitrosamine (DEN). Male Sprague-Dawley rats received a single intraperitoneal injection of DEN (200 mg/kg body wt) dissolved in saline. After 2 weeks on a basal diet, animals were fed diets containing 0. 0.5. 2.0. or 5.0% garlic powder for 6 weeks, and were subjected to two-thirds partial hepatectomy. The areas of placental glutathione S-transferase (GST-P) positive foci were inhibited in rats fed with garlic diets. GST-P is the most effective marker for DEN-initiated lesions. Hepatic microsomal lipid peroxidation was significantly decreased in rats fed with 2.0 and 5.0% garlic powder diets compared with that observed in the control animals and hepatic microsomal glucose 6-phosphatase (G6Pase) activity was found to increase significantly in rats fed 0.5 and 2.0% garlic powder diets. Thus as little as 0.5% garlic powder has a positive effect on the stability of hepatic microsomal membranes. p-Nitrophenol hydroxylase (PNPH) activity and the level of cytochrome P450 2E1 protein in the hepatic microsomes from rats fed diets containing 2.0 and 5.0% garlic powder were much lower than those of control microsomes. Rats fed 5.0% garlic powder diets exhibited the lowest P450 2E1 activity and protein levels among groups. Pentoxyresorufin O-dealkylase activity and immunoblot (cytochrome P450 2B1) analyses were not different between groups. However, the levels of cytochrome P450 1A1/2 protein in rats fed 0.5 and 2.0% garlic powder were significantly induced compared to controls. These results suggest that 2.0% garlic powder is effective in inhibiting the areas of GST-P positive foci, modulating certain isoforms of cytochrome P450 enzymes and stabilizing the hepatic microsomal membrane. Thus, the selective modification of cytochrome P450 enzymes and membrane stability by dietary garlic powder may influence areas of GST-P positive foci and chemoprevention of post-initiation of rat hepatocarcinogenesis.

Proteomic Approach to Aging Research

  • The aging process is multifactorial and results from the combined effects of inherited(genetic) and acquired factors including life style, food habits, physical activity, and diseases. That give rise to the various approaches in aging. We are trying to study biological changes with aging, In detail we are focused on gene and protein function accompanied by normal or abnormal aging process, especially our efforts are aimed at revealing the functional relationship of proteins in aging as a final product of gene. We expect that proteomic approach to the study of protein function involved in aging should give us variety of integrated data to understand biological changes of long lived lives, We have applied expression proteomics to rat liver bred in dietary restriction or in at libitum to elucidate the effects of food habit on aging. Expression proteomics shows us protein profile in a selected tissue or cells as a whole and gives us the information about protein expression level, posttranslational modification and degenerative modification of expressed proteins. Comparative analysis of young and old rat liver by two dimensional gels shows that gene expression of several proteins was down regulated in old rats and some protein expression level is increased with aging. Dietary restriction slows down these changes of gene expression and in some proteins there's no difference in protein expression level at same ages in comparison with rats bred in at libitum. About forty protein was identified by peptide mass fingerprint with MALDI-TOF and rest of the protein of interest is in the course of identification, Also we are trying to make mitochondrial and cytosolic proteom reference map. These suborganelle proteom map will gives us the information about low abundance proteins and cellular localization of proteins. Proteomics is a growing methodology to study biological system. High throughput qualitative and qualitative aspect of this approach will gives us large amount of integrated information and speed up our understanding about biological system

스마트폰 앱을 이용한 체중감량 프로그램이 고교생의 비만도와 신체조성에 미치는 효과 (Effects of a Smart Phone Weight Loss Program on Obesity and Body Composition in Adolescents)

  • Purpose: The purpose of this study was to determine the effects of a weight loss program administered through a smart phone application on body composition and obesity in adolescents. Methods: This study applied a 6-week weight loss program with dietary education, exercise therapy, and behavior modification therapy from 4 June 2012 to 20 July 2012. The collected data were analyzed by descriptive statistics and the Wilcoxon signed rank test using the SPSS WIN 18.0 program. Results: After the 6-week program, the average weight of the subjects was reduced from 75.96kg to 74.41kg (Z=$-3.994^p$, p<.001), while the body mass index (BMI) decreased from 29.23 to 28.60 (Z=$-3.980^p$, p<.001), and the obesity index decreased from 38.45% to 35.59% (Z=$-3.977^p$, p<.001). The percentage of body fat decreased from 32.45 to 31.92 (Z=$-3.947^p$, p<.001), and the rate of skeletal muscle growth increased from 26.06% to 26.58% (Z=$-3.822^p$, p<.001). Conclusion: A weight loss program using a SNS (social network service) application, with dietary education, exercise therapy, and behavior modification therapy can reduce obesity and impact body composition in a short-term. Therefore, the utilization of the SNS application can be used to effectively enhance the management of adolescent obesity.

Effectiveness of Multi Interventional Package on Selected Parameters of Metabolic Syndrome among Women - A Pilot Study

  • Purpose: The purpose of the study was to compare the effectiveness of multi interventional package (MIP) and lifestyle interventions (LI) on physiological parameters of women with metabolic syndrome, to compare the effectiveness of MIP and LI on biochemical parameters of women with metabolic syndrome and to compare the effectiveness of MIP and LI on socio-psychological parameters of women with metabolic syndrome. Methods: A quasi experimental nonequivalent control group design with two experimental groups and one control group was used to collect data from 60 self-help group women. Samples were selected by multistage sampling. Reflexology foot massage, dietary modification, moderate intensity exercise and structured education were given to MIP group and dietary modification, moderate intensity exercise and structured education were given to LI group for 12 weeks. Control group received routine care. Demographic and clinical data sheets were used to collect basic information. Knowledge was assessed by a knowledge questionnaire. Physiological (weight, body mass index, waist circumference and blood pressure) and biochemical parameters (HDL, triglycerides and FBS) were assessed before and after the intervention. Results: The study found significant change in the physiological and biochemical parameters of metabolic syndrome as well as knowledge among the MIP group and LI group compared to the control group (p <.001). Conclusion: MIP and LI are effective in controlling the parameters of metabolic syndrome. Hence the guidance may be provided to women with metabolic syndrome for adopting necessary lifestyle changes as well as reflexology foot massage to control the physiological and biochemical parameters of metabolic syndrome.

Inverse association of improved adherence to dietary guidelines with metabolic syndrome: the Seoul Metabolic Syndrome Management program

  • BACKGROUND/OBJECTIVES: The Seoul Metabolic Syndrome Management (SMESY) program is a 1-yr lifestyle modification program targeting metabolic syndrome (MetS) in Seoul residents. This study investigated the associations between adherence to dietary guidelines and MetS among the SMESY program participants. SUBJECTS/METHODS: Data of 54,385 participants aged 20-64 yrs who completed the SMESY program in 2015, had information on adherence to dietary guidelines, and were not medicated for diabetes, hypertension, or dyslipidemia were analyzed. Participants underwent MetS screening and completed a lifestyle questionnaire including adherence to 10 dietary guidelines before and after participation. Participants were classified according to the number of MetS risk factors at baseline (MetS group, ≥ 3; risk group, 1-2; healthy group, none). Adherence to dietary guidelines was determined from the number of "yes" responses regarding the fulfillment of each guideline on ≥ 5 days/week. Multiple logistic regression was used to evaluate associations between newly diagnosed MetS and changes in adherence to dietary guidelines. RESULTS: In the MetS group, MetS prevalence decreased after the SMESY program (men, -41.9%p; women, -48.7%p), and all risk factors were significantly improved (P < 0.0001). All groups exhibited improved adherence to all dietary guidelines after participation (P < 0.0001). In the MetS group with positively changed adherence scores, the MetS prevalence decreased by -44.1%p for men and -49.5%p for women, whereas the prevalence in those with negative changes decreased by -38.1%p for men and -48.6%p for women. In the risk group, those with positively changed adherence scores had significantly decreased odds ratios (ORs) for newly diagnosed MetS compared with those with negative changes (OR, 0.70; 95% confidence interval [CI], 0.61-0.80 for men; OR, 0.88; 95% CI, 0.79-0.99 for women). CONCLUSIONS: The SMESY program may effectively reduce the risk of MetS among adults with risk factors by improving adherence to dietary guidelines.

Linolenic acid/linoleic acid 비율이 다른 식이가 연령이 다른 흰쥐의 뇌구조지방 조성과 Acetylcholinesterase 활성에 미치는 영향 (Influence of Dietary Linolenic Acid/linoleic Acid Ratio on Brain Lipid Composition and Acetylcholinestease Activity in Different Aged Rats)

  • This study was undertaken to investigate the influence of age and dietary linolenic acid content and the linolenic acid/linoleic acid (LAN/LA) ratio on the brain lipid composition and membrane-bound enzyme, acetylcholinesterase(AchE) activities. AchE was selected as a test case for the relationship between cell lipid composition and cell membrane function. The male rats were fed diets with 0.2, 0.4, 0.6 of LNA/LA ratio within 8% LNA(H-LNA) or 4% LNA(L-LAN) of total fatty acid content for different feeding period(1, 4, 12 month). The fats used s source were sesame oil, perilla oil, soybean oil and beef tallow. The AchE activity of brain crude synaptosomal fraction was reduced with advancing age, showing 20-30% reduction in 12M compared with 1 M, and the P/C ratio was reduced in old rats. In 1 and 4 monthed rats, AchE activites was higher in H-LAN-0.2 and L-LNA-0.2 and 0.4 group. In accordance with rising of AchE activities was higher in H-LNA-0.2 and L-LNA-0.2 and 0.4 group. In accordance with rising of AchE activities, the PC/PE ratio increasedin those groups. Paricularly in L-LNA, the PC/PE ratio increased as the AchE activites for decline of membrane fluidity with increasing cholesterol and decreasing P/C ratio when rats were old. Also, AchE activity increaed with increasing PC/PE ratio which depended on the dietary LNA/LA ratio within each LNA content. Therefore, it is concluded that the lipid composition of cell membrane influenced the AchE activiteis, which was mediated by aging and the modification of dietary LNA/LA ratio.

Effects of Dietary Modification on Plasma Glucose and Insulin Sensitivity in Streptozotocin-induced Diabetic Rats

  • There is substantial evidence that insulin sensitivity can be enhanced through appropriate dietary management . In this study, insulin sensitivity was evaluated using and insulin suppression test. Male Sprague-Dawley rats, were caused to be in a diabetic condition by the injection of streptozotocin, and divided into four groups. They were fed one of the following diets for 2 weeks : (group 1) a high-carbohydrate(CHO) low-fat low-fiber diet, (group 2) a high CHO low-fat and high-fiber diet, (group 3) a low-CHO high-fat and low-fiber diet, and (group 4) a low-CHO low-fat diet groups (as comparison between group 1 and group 2 shows). In the low-CHO high-fat diet groups, dietary fiber tended to decrease plasma glucose levels at the end of the experiment, but not significantly (as comparison between group 3 and group 4 shows). The average steady state plasma glucose level in rats on the group 3 diet was the highest among all four groups(p<0.05), indicating the poorest insulin sensitivity . However, high fiber increased insulin sensitivity in rats on the low-CHO high-fat diets(as shows by a comparison between group 3 and group 4). On the other hand , the high-CHO low-fat enhanced insulin sensitivity in rats on the low fiber diet(group 1 and group 3). The degree of enhancement of insulin sensitivity depends on the combination of CHO, fat , and fiber in the diet. In conclusion, this study demonstrates that a low-CHO high-fat low -fiber diet may be deleterious to diabetic rats. In view of insulin sensitivity enhancement , dietary fiber level is irrelevant, as long as the diet has a high-CHO and low-fat level.

플라보노이드에 의한 사람 Low Density Lipoprotein 변형에 대한 항산화 효과 (Antioxidative Effects of Flavonoids toward Modification of Human Low Density Lipoprotein)

  • The flavonoids are one of the most numberous and widespread groups of natural consituents. The low molecular weight of benzo-${\gamma}$-pyrone derivative are ubiquitous in plants and are vegetables nuts, seeds, leaves, flowers, and bark. The flavonoids constitute of a large class of compounds ubiquitous in plants containing a number of phenolic hydroxyl groups attached to ring structures conferring the antioxidant activity. Epidemiologic studies suggest that the dietary intake of antioxidants constitutes a risk factor for vasclar disease indicating that oxidation may be important in the pathogenesis of human athero-sclerosis. Elevated plasma low density lipoprotein (LDL) cholesterol concentration are associated with accelerated atherosclerosis, LDL is oxidized by smooth muscle cells resulting in several chemicals and physical changes of LDL. Oxidized LDL is responsible for cholesterol loading of macrophages foam cells formation and atherogenesis. There have been insulficient tests of the protective effects of flavonoids against LDL oxidation to make definitive statements about their structure activity relationships. How-ever hydroxylation of the flavone uncleas can appears to be advantageous because polyhydroxylated aglycone flavonoids are potent inhibitor of LDL modification. This identification may lead to new and more effective antioxidant strategies for abrogating the atherosclerotic process the leading cause of death and disability in industrialized societies.
