Studies on the Selections, the Cross-Resistance and the Eserase Activity in the Strain of German Cockroack, Blattella germanica L.Selected with dichlorvos (바퀴에 대한 Dichlorvos(DDVP)의 누대도태, 교차저항성 및 효소활성에 관한 연구)

  • The german cockroach, Blattello germanica L, populations were successively selected with dlchlowos for 11 generations. The resulting selected shin was investigated the resistance development. the cross resistance and the esterase actim@. In the dichlowos-selected(Rd) strain, the values of LCs increased 858 times more compared to the susceptible (S) strain. In the dich!o~vos-selected (Rd) strains, the cross-res~stance to chlorpyrifos, propoxur, fenvalerate and pemethnn showed 3.35. 4.09. 283 and 2.00 times. Esterase.activity of the Rd strain showed 1.33 times higher than that of the S strain in the filter paper test. In comparison of zymogram paitems of the estemse isoryme by thin agarose gel electrophoresis against the german cockroach, the S strain was separated by 4 bands uf esterase 3, 5, 6 and 8 bands and the Rd strain was separated by 6 bands of esterase 1, 2, 4, 5, 7 and 8 bands, and the resistant mechanisms of the Rd strain were considered as the 4 bands of esterase-1.2.4 and 7 bands except the common 2 bands of esterase.6 and 8 bands.

Studies on the potential hazard of dichlorvos-resin vaporizing insecticide in human and domestic fowl (Dichlorvos 증기(蒸氣)의 지속적(持續的) 흡입(吸入)이 인체(人體) 및 가계(家鷄)에 미치는 영향)

  • Safety study of the continuous releasing dichlorvos-resin insecticide $(Mopari^{\circledR})$ was conducted in human volunteers and domestic fowls. For the purpose, the potential hazards in using the insecticide were observed in terms of the inhibition of plasma cholinesterase activity and the changes in the liver function (GOT, GPT, Alkaline phosphatase, Bilirubin, Thymol turbidity), the blood picture (RBC, WBC with differential count, Hemoglobin, Hematocrit and ESR) and the urine picture (sugar, albumin, pH and microscopic findings) in 40 healthy adult volunteers and 60 leghorn domestic fowls. In case of the human study the observation was continued for 2 months during the application of the insectiside ($1{\sim}3$ solid formulations/$30m^3$) in the living rooms of ordinary Korean dwelling houses or in the office. In the animal test, however, 1 to 5 solid formulations of the insecticide were applied in the fowl cage of $9.2m^3$ for 5 weeks. Any significant inhibition of the plasma cholinesterase activity was not observed in both the human volunteer and the fowl throughout the experimental period. And the liver function as well as the blood and urine pictures were also not changed after exposure to the vaporizing insecticide. It is considered from the result that the amount of dichlorvos released into the air by the continuous vaporizing dichlorvos-resin insecticide presents no significant hazardous effect on humans or animals in the present experimental condition.

  • Susceptibility of Blattella germanica male adults to 61 commercial insecticides was evaluated by diet dipping method. Among them three insecticides of dichlorvos, fenitrothion, and fipronil showed over $90\%$ mortality. The insect was more susceptible to dichlorvos and fenitrothion in filter paper contact method than in diet dipping one, but fipronil was vice versa. Toxicities of the three chemicals were evaluated by application parts of the such as head, thorax, abdomen, and legs. Dichlorvos and fenitrothion were more toxic when applied to head, and flpronil was thorax. $LT_{50}$ values by diet dipping method showed that fenitrothion and fipronil acted more rapidly than boric acid and hydramethylnon. The values of the former two were 1.05 and 0.98 days, and those of the latter two were 3.92 and 2.26 days, respectively.

Susceptibility commercially of North American planthopper, Metcalfa pruinosa to commercially registered insecticides in Korea (국내시판 살충제에 대한 미국선녀벌레의 감수성)

  • Insecticidal activity of 31 registered insecticides was tested against Metcalfa pruinosa adults. All experiments were conducted at the recommended concentration (ppm) of each insecticide. Among them, 16 insecticides from organophosphates (dichlorvos, fenitrothion, fenthion, methidathion, phenthoate), carbamates (methomyl), pyrethroids (${\alpha}$-cypemethrin, deltamethrin, fenpropathrin, ${\gamma}$-cyhalothrin), neonicotinoids (acetamiprid, clothianidin, dinotefuran, imidacloprid, thiamethoxam), and other (endosulfan) showed 100% mortality by spraying on the body of M. pruinosa adults. Dichlorvos, fenitrothion, fenthion, methidathion, phenthoate and endosulfan showed 100% mortality by plant-dipping method. The residual effect was showed 100% mortality in four insecticides (fenitrothion, fenthion, methidathion, phenthoate) at one day after treatment, and three insecticides (fenitrothion, methidathion, phenthoate) were showed the mortality of 90% at three days after treatment.

  • This study was conducted to investigate the resistance development and cross-res~stance of house fly (Musco dornestica L.) selected with chlorpyrifos, dichlorvos and permethrin for 11 generations to various p u p s of insectiodes. The resistance ratio (RR) of the chlorpyrifos-selected (Q), the d~chlorvos- selected (&) a d the permethrin-selected (R,) stmlns were 42 0. 38 and 187 tlrnes in female. and 42.0, 4 1 and 16.4 time; in male from the susceptible strain, respectively. The Rc strain showed highest cross-resistance to permethlin among the insectic~des tested: RR=7.5 and 9.6 tunes in female and male, respectively, whereas negatively correlated cross-resistance to propoxur was observed. High cross-res~atance to chlorpyrifos were produced for female (RR= 13.3) and male (RR=15.9) of Rd strain, and female (RR=8.7) and male (RR= 9 7) of R, strain. respectively

Determination of Trichlorfon Pesticide Residues in Milk via Gas Chromatography with μ-Electron Capture Detection and GC-MS

  • The pesticide trichlorfon is readily degraded under experimental conditions to dichlorvos. A method has therefore been developed by which residues of trichlorfon in milk are determined as dichlorvos, using gas chromatography with ${\mu}$-electron capture detection. The identification of dichlorvos was confirmed by mass spectrometry. Milk was extracted with acetonitrile followed by centrifugation, freezing lipid filtration, and partitioning into dichloromethane. The residue after partitioning of dichloromethane was dissolved in ethyl acetate for gas chromatography. Recovery concentration was determined at 0.5, 1.0, and 2.0 of times the maximum permitted residue limits (MRLs) for trichlorfon in milk. The average recoveries (n = 6) ranged from 92.4 to 103.6%. The repeatability of the measurements was expressed as relative standard deviations (RSDs) ranging from 3.6%, to 6.7%. Limit of detection (LOD) and limit of quantification (LOQ) were 3.7 and $11.1{\mu}g/l$, respectively. The accuracy and precision (expressed as RSD) were estimated at concentrations from 25 to $250{\mu}g/l$. The intra- and inter-day accuracy (n = 6) ranged from 89.2% to 91% and 91.3% to 96.3%, respectively. The intra- and inter-day precisions were lower than 8%. The developed method was applied to determine trichlorfon in real samples collected from the seven major cities in the Republic of Korea. No residual trichlorfon was detected in any samples.

Monitoring of the residues of some pesticides in mulberry leaves and their safety evaluation (뽕잎중 잔류농약의 monitoring 및 안전성 평가)

  • In order to evaluate the safety of mulberry leaves for sericultural purpose to the contamination by pesticide residues, the analyses of dichlorvos, acephate, phenthoate, pyrazophos, deltamethrin, and cypermethrin were done for 24 samples collected in spring and fall over two consecutive years (1996 and 1997), each, from 8 provinces across the country, by selecting three representative farmhouses in each province. In spring samples of 1996, the residual amounts of dichlorvos, acephate, and phenthoate were $0.018{\sim}0.032$, $0.013{\sim}0.072$, and 0.051 ppm, respectively, whereas, pyrazophos was not detected, and the detection frequencies were 29, 58, and 4%, respectively. In fall samples, on the other hand, those of the above pesticides were $0.012{\sim}0.048$, $0.020{\sim}0.156$, and $0.018{\sim}0.050$ ppm, respectively, and pyrazophos was not detected either and the detection frequencies were 42, 17, and 13%, respectively. While in spring samples of 1997, the residual amounts of dichlorvos, acephate, and cypermethrin were $0.014{\sim}0.064$, $0.033{\sim}0.061$, and $0.019{\sim}0.068$ ppm, respectively, and deltamethrin was not detected. The detection frequencies were 75, 13, and 8%, respectively. In fall samples, on the other hand, those of the above pesticides were $0.013{\sim}0.062$, 0.015, and $0.009{\sim}0.013$ ppm, respectively, and deltamethrin was not detected either. The detection frequencies were 88, 4, and 17%, respectively. Almost all of the samples turned out to be non-toxic to silkworms, except that one sample (No. 10, spring 1997) contaminated by a high concentration of cypermethrin (0.068 ppm) was judged to do harm to silkworms. In the GC analyses, the selection of columns, the change in column temperature, and the use of MSD made the separation and identification of the vague compounds possible.

  • Effect of Synethrin on the Toxicity of Dichlorvos in Rats. Hematological, biological and enzymatic effects were investigated in rats treated with DDVP and synethrin. Mutagenicity was also examined. In serological analysis, LDH and ALP were more significantly increased in rats treated with the mixture of DDVP (10mg/kg) and synethrin (250mg/kg) than with either DDVP or synethrin. DDVP alone slightly increased cytochrome P-450 in the liver while synethrin or the mixture decreased it. Lipid peroxidation was significantly increased in the liver when rats were treated with both DDVP and the mixture. Cholinesterase activity was significantly decreased in the liver and serum when treated with DDVP as well as with the mixture. In mutagenicity test, DDVP was shown to be weakly positive to TA 97a, TA 100 and TA 102, but negative to TA 1537. Synethrin showed negative in all strains tested. These results suggest that the mixture of DDVP and synethrin increased the toxicities but not the mutagenicity.

  • The present study was aimed to investigate persistence of dichlorvos EC, fenitrothion EC, fenthion EC, and phenthoate EC in and on mulberry leaves with special reference to silkworm mortality and cocoon production under greenhouse condition. The halflives of fenitrothion, fenthion, and phenthoate in and on mulberry leaves were ranged from two to three days, while that of dichlorvos was less than 9 hours. The insecticide residues in and on mulberry leaves persisted longer in spring cropping season than in autumn cropping season. Elapsed periods from last application of each insecticide to leaf harvest for silkworm feed and maximum residue limits for safe cocoon production as well as relations between insecticide residues in and on mulberry leaves and silkworm mortality were produced.

  • This study was conducted to investigate the effect of ozone treatment on the growth and pesticide-residual contents of soybean sprouts during cultivation. Total weight and length of the soybean sprouts treated by 0.3 ppm ozone water for 30min during soaking of soybean increased 27% and 19% higher than those of control group, respectively. But 27% of root weight decreased. No major differences in growth state were observed between the treatments during soaking and watering with ozone water, and watering with ozone water. Pesticide residues in soybeans treated with carbendazim, captain, diazinon, fenthim, dichlorvos and chlorpyrifos ranged from 4.75 to 8.35 ppm. The pesticides in soybean sprouts by the treatment of soaking and watering with water for 5 days, those by 0.3 ppm ozone-water watering, and those by soaking and watering with 0.3 ppm ozone water were destroyed to $85{\sim}99$, $89{\sim}100$ and $94{\sim}100%$, respectively. The order of degradation ratio in the pesticides was captan>dichlorvos>fenthion>carbendazin>diazinon>chlorpyrifos.
