혐기성 PCE 탈염소화 미생물 농화 배양 및 미생물 군집 해석

  • An anaerobic PCE(tetrachloroethylene) dechlorinating bacterial culture from a landfill soil was enriched and characterized. The enrichment culture could dechlorinate 60$\mu$mol/$m\ell$ of PCE during a month of incubation and cis-DCE(cis-dichloroethylene) was observed as a main product of PCE dechlorination. Microbial analysis of the dechlorinating enrichment culture by rising PCR-DGGE (Polymerase chain reaction-Denaturing gradient gel electrophoresis) method showed that at least three microorganisms were related to the anaerobic PCE dechlorination.

  • Diethyl itaconate was found to react tetrafluoroethylene, trifluorochloroethylene, and 1,1-difluoro-2,2-dichloroethylene quite rendily at $180^{\circ}C$ to form 1-carboxy-2,2,3,3,-tetrahalo-cyclobutane ethanoic acid derivatives ${C{\underline{X_2-CF_2-CH_2-}}C-(COOEt)-CH_2COOEt$, where X is Cl or F). Diethylcitraconate failed to yeild any cycloaddition products. The Physical and Chemical properties of the above esters and their dericatives are characterized and compared with other similar ring compounds.

  • 앞으로 본지를 통하여 현재 쓰이고 있는 유해물의 허용한계가 어떠한 근거로 정하여졌는가를 소개하고자 한다. 허용한계에 대하여 여러가지로 소개된 바 있으나 그 근거를 알지 못하여 그 적용에 있어서 잘못이 생길 수 있다. 소개되는 내용은 미국의 ACGIH의 TLV가 결정 됨에 있어서 이용된 여러가지 문헌을 소개하고 그 내용을 간추려 보고자한다. 1966년에는 367가지의 물질에 대한 TLV가 알려져 있었고 1971 년에는 500 가지의 물질에 대하여 결정된 바 있으나 본지에서는 우리나라에서 찾아볼 수 있는 물질과 기타 중요하다고 생각되는 물질을 선택하여 소개하고자 한다.

Analytical Potential Energy Surfaces for the Four-center Elimination Feaction of HCI from 1,1-Dechlorethylene: Translational Energy Release from Classical Trajectory Studies

  • Analytical potential energy surfaces have been constructed for the four-center elimination of HCI from 1,1-dichloroethylene.The potential functions are Morse-type functions which are modified by appropriate switching and attenuating functions with adjustable parameters. The parameters have been found by fitting the calculated vibrational frequencies, reaction endothermicity, equlibrium geometries of the reactant and products to those of experiments and ab initio calculations. The translational energy release obtained from classical trajectory calculations on this surface is in good agreement with the experiment.

DNA Single Strand Breaks of Perchloroethylene and Its Bio-degradation Products by Single Cell Gel Electrophoresis Assay in Mammalian Cell System

  • Perchloroethylene (tetrachloroethylene, PCE), a dry cleaning and degreasing solvent, can enter ground-water through accidental leak or spills. PCE can be degraded to trichloroethylene (TCE), 1, 1-dichloroethylene (DCE) and vinyl chloride (VC) as potential bio-product. These compounds have been reported that they can cause clinical diseases and cytotoxicity. However, only a little genotoxic information of these compounds has been known. In this study, we investigated DNA single strand breaks of PCE, TCE, DCE and VC by single cell gel electrophoresis assay, (comet assay) which is a sensitive, reliable and rapid method for DNA single strand breaks with mouse lymphoma L5178Y cells. From these results, $37.5\;{\mu}g/ml$ of PCE, $189\;{\mu}g/ml$ of TCE and $56.4\;{\mu}g/ml$ of DCE were revealed significant DNA damages in the absence of S-9 metabolic activation system meaning direct-acting mutagen. And in the presence of S-9 metabolic activation system, $41.5\;{\mu}g/ml$ of PCE, $328.7\;{\mu}g/ml$ of TCE and $949\;{\mu}g/ml$ of DCE were induced significant DNA damage. In the case of VC, it was revealed a significant DNA damage in the presence of S-9 metabolic activation system. Therefore, we suggest that chloroethylene compounds (PCE, TCE, DCE and VC) may be induced the DNA damage in a mammalian cell.

  • Chlorinated aliphatic hydrocarbons (CAHs) especially perchlorethylene (PCE) and trichlooethylene (TCE) are common groundwater contaminants in Korea. PCE and TCE were often reductively dechiorinated in an aquifer. Several isolates dechlorinate PCE to TCE or cis-1,2 dichloroethylene (c-DCE) were obtained from contaminated and pristine sites in USA and Europe. However in Korea, no information on indigenous microorganism being involved in reductive dechlorination of PCE and TCE is available and different dechlorinating microorganisms might be reside in Korea, since geochemical, and hydrogeological conditions are different, compared to those in the other sites. So we evaluate that: 1) if reductive dechlorinating microorganisms are present in PCE-contaminated site in Korea, 2) if so, what kinds of microorganisms are present; 3) to what extent PCE is reductively dechlorinated. As a results in some PCE-contaminated aquifers in Korea other dechlorinating microorganisms but Dehalococcoides ethenogenes may be responsible for PCE dechlorination. More detailed molecular works are required to evaluate that different dechlorinating microorganisms would reside in Korea.

전기화학적 합성 Ferrate(VI)를 이용한 수중 Trichloroethylene 분해특성 연구 (Degradation of Trichloroethylene in Aqueous Phase by Electrochemical Ferrate(VI))

  • The degradation characteristics of TCE by Ferrate(VI) oxidation have been studied. Ferrate(VI) were prepared by electrochemical method. The degradation efficiency of TCE in aqueous solution was investigated at various pH values, Ferrate(VI) doses and aqueous solution temperature values. GC-ECD was used to analyze TCE. TCE was degraded rapidly by ferrate(VI) in aqueous solution, Also, the experimental results showed that TCE removal efficiency increased with the increase of Ferrate(VI) doses. The effect of pH was investigated and the maximum degradation efficiency was obtained at pH 7. And intermediate products were identified by GC-MS techniques. Ethyl Chloride, Dichloroethylene, Chloroform, 1,1-dichloropropene, Trichloroacetic acid and Trichloroethane were identified as a reaction intermediate, and $Cl^-$ was identified as an end product.

Separation of Dichlorinated Bydrocarbons by Pervaporntion Using ZSM-5 Zeolite Membrane

  • Pervaporation with a membrane is one of the economic technologies for separation of liquid mixtures including organic/water mixtures. The ZSM-5 membrane was used fur pervaporation of dichloromethane, 1,2-dichloroethane and trans-1,2-dichloroethylene from their aqueous solutions since its physical property shows very hydrophobic. ZSM-5 crystals were hydrothermally grown and deposited on the inside of a porous sintered stainless steel tube by the secondary growth method. Fluxes of dichlorinated organic compounds were observed to be $50{\~}429\;g/m^2/h$ while separation factors were $15{\~}320$ depending on a mole fraction of a dichlorinated organic compound in a feed solution ranged from 0.0001 to 0.001 mole fraction and the operation temperature between $25^{\circ}C\;and\;35^{\circ}C$.

  • The transformation capacity (T$\_$c/) of Methylosinus trichosporium OB3b in the degradation of ethylene chlorides was determined by measuring the decrease of soluble methane monooxygenase (sMMO) activity of resting cells in batch experiments. All measurements of sMMO activity were taken in the presence of 20 mM formate to avoid the deficiency of reducing power, and within 2 hrs to avoid the effect of natural inactivation from instability of the resting cells. The constant T$\_$c/ values of 0.58 ${\pm}$ 0.132 and 0.80 ${\pm}$ 0.17 ${\mu}$mol/mg cell were obtained for trichloroethylene (TCE) and 1,2-dichloroethylene (cis and trans-1,2-DCE), respectively, regardless of their concentrations. The transformation capacity measured by this method can be used to predict the amount of cells that should be stimulated in in-situ bioremediation.

Photoreaction of 1,6-Disubstituted-1,3,5-hexatriynes with Some Olefins

  • When the two conjugated poly-ynes, 1-phenyl-6-methyl- and 1,6-diphenyl-1,3,5-hexatriynes, were irradiated with UVA in deaerated 2,3-dimethyl-2-butene solution, 1:2 photoadducts, 1-(1'-phenylethynyl-2',2',3',3'-tetramethylcyclopr opyl)-2-(l",2",2",3",3"-pentamethylcyclopropyl) acetylene and 1-(1'-phenylethynyl-2',2',3',3'-tetramethylcyclopr opyl)-2-(1"-phenyl-2",2",3",3"-tetramethylcyclopropyl) acetylene, were obtained, respectively. No photoadduct was formed with aerated 2,3-dimethyl-2-butene, or deaerated solutions of dimethyl fumarate, methyl crotonate, dimethyl maleate, and trans-1,2-dichloroethylene. The results suggest that the reactions proceed from the triplet state only with electron rich olefins such as 2,3-dimethyl-2-butene.
