A Study on Antitoxic Effects of Natural Products Against Nickel Toxicity (자연산물을 이용한 니켈 독성에 관한 연구)

  • This study was performed to find out the detoxication effects of natural products Thuja orientalis, leaf of acorn, aloe ferox mill, pine cone, pine root, lonicerae flos on nickel toxicity. The experimental rats were divided into 3 groups such as nickel alone treatment group, natural products treatment groups before and after nickel treatments. Each group was administered with difference dose of nickel such as 6.6 mg/kg/day, 13.2 mg/kg/day, 19.8 mg/kg/day respectively, for 14 days. Natural products were administered by 400 mg per kilogram body weight rat per day. The weight change of rats, nickel concentration in organs, survival rate of rats, morphology of organs were observed in the experimental groups. The results obtained by the experiment were as follows: 1. The rats in the group treated with natural products had no effect on the weight gain. 2. The mean survival rate of rats in the nickel alone treatment group was 72.5% (6.6 mg/kg/day), 68.9% (13.2 mg/kg/day), 60.2% (19.8 mg/kg/day) respectively. 3. As for the amount of nickel accumulation in organs, it was the lowest amount treated with pine cone, pine root in that order. 4. When kidney and liver tissues were observed with as optical microscope, obvious change were visible in those tissues treated groups with Lonicerae and Aloe.

Baking Characteristics of Taurine Supplemented Bread and Cookies and Its Effect on Blood Alcohol Concentrations (타우린을 첨가한 제과제빵의 베이킹 특성과 섭취가 혈중 알코올 농도에 미치는 영향)

  • This study investigated the characteristics of baking bread and cookies supplemented with taurine and the effect of taurine addition on BAC(blood alcohol concentrations) and UAC(urine alcohol concentrations). Healthy male college students were divided into two groups, the control and the taurine group. Bread was baked with the addition of 0, 2, 4 and 6% taurine and baked with the addition 0, 3 and 6% taurine. The bread containing 2% taurine showed the fastest fermentation among the 4 groups. Fermentation and oven-spring of breads baked with a taurine concentration greater than 4% of taurine disturbed. In some areas of the sensory test, the taurine supplemented bread had higher scores than the control bread. We served 6 g of taurine supplemented cookies with 1,000 $m{\ell}$ of beer to 8 students in the taurine group. After 2 hours of drinking beer, the BAC and UAC of the taurine group were found to be lower than the control group. But no difference was found in the alcohol excretion of their urine. From this study we concluded that the taurine has an effect on the detoxication of alcohol, which reduces the BAC.

Presence of Rhodanese in the Cytosolic Fraction of the Fruit Bat (Eidolon helvum) Liver

  • Rhodanese was isolated and purified from the cytosolic fraction of liver tissue homogenate of the fruit bat, Eidolon helvum, by using ammonium sulphate precipitation and CM-Sephadex C-50 ion exchange chromatography. The specific activity was increased 130-fold with a 53% recovery. The $K_m$ values for KCN and $Na_2S_2O_3$ as substrates were $13.5{\pm}2.2\;mM$ and $19.5{\pm}0.7\;mM$, respectively. The apparent molecular weight was estimated by gel filtration on a Sephadex G-100 column to be 36,000 Da. The optimal activity was found at a high pH (pH 9.0) and the temperature optimum was $35^{\circ}C$. An Arrhenius plot of the heat stability data consisted of two linear segments with a break occurring at $35^{\circ}C$. The apparent activation energy values from these slopes were 11.5 kcal/mol and 76.6 kcal/mol. Inhibition studies on the enzyme with a number of cations showed that $Mg^{2+}$, $Mn^{2+}$, $Ca^{2+}$, and $Co^{2+}$ did not affect the activity of the enzyme, but $Hg^{2+}$ and $Ba^{2+}$ inhibited the enzyme.

Effects of acupuncture on dopamine release in the nucleus accumbens in rats (백서 뇌측핵에서 도파민 분비에 대한 침의 효과)

  • Objective: Dopamine activity in thenucleus accumbens is an important neuropharmacological component of morphine reinforcement. In this nucleus a shell and core have been distinguished on the basis of anatomical and neurochemical criteria. Although acupuncture has been standard intervention in many detoxication programs worldwide, the central mechanism by which morphine acts to reinforce behavior remain elusive. The present in vivo microdialysis study was conducted, in freely moving rats, to detect the effects of acupuncture on extracellular dopamine release in the nucleus accumbens. Methods: Male Sprague-Dawley rats received acupuncture for 1 min after injection of morphine hydrochloride (5mg/kg, s.c.). The employed acupuncture needle points corresponded to bilateral Neiguan(PC6) on the pericardium channel, which has been used to treat mental and psychosomatic disorders. Extracellular dopamine and its metabolites were measured every 20 mins for 3 hrs following the subcutaneous morphine injection. Results: Results showed that acupuncture at PC6 significantly attenuated increases in dopamine levels induced by a single acute morphine injection in the nucleus accumbens shell and core, respectively. Conclusions: These results provided strong evidence for acupuncture-mediated reduction in morphine-induced dopamine release in the rat nucleus accumbens.

Effects of Chunggansan on Acute Damages of Liver Induced by Ethanol in Rats (청간산(淸肝散)이 급성(急性) Ethanol 중독(中毒)에 의한 흰쥐의 간손상(肝損傷)에 미치는 영향(影響))

  • Chunggansan(淸肝散) was tested for the effects on detoxication mechanism of alcohol. Chunggansan was treated firstly into samples, and then ethanol intoxicated animal models were set with them. In vitro, the level of lipid peroxide in tissue of liver proportionally decreased with the level of concentration of extract prepared from Chunggansan. In vivo, after the extract was administered to the animal model for seven days, the level of lipid peroxide in liver dependently decreased in proportion of concentration. The glutathione percentage purposely induced acute ethanol intoxicated samples, followed by Chunggansan pre-medicating administration, increased in compare with the group treated with only ethanol. Also, the enzyme activities of ALT, AST, and ${gamma}-GTP$ in liver decreased.

Inhibitory Effects of Leachianone A from Sophora fIavescens Ait. against Cadmium Induced Cytotoxicity (고삼으로부터 분리된 Leachianone A의 카드뮴에 대한 독성억제 효과)

  • In this study, cytotoxicity of cadmium on NIH 3T fibroblasts was utilized in order to discover antitoxic compound in methanol extract of Sophora fIavascens Ait. There were treatment groups; control (medium only), $MTT_{50}$ group and five experimental groups. MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H- tetrazoliumbromide} assay was performed to evaluate the cytotoxicity of cell organelles and $IC_{50}$ was also measured. Accordingly we have examined the detoxification effects of methanol extract of S. flavescens Ait. and leachianone A (LA) on cadmium-treated NIH 3T3 fibroblasts ($IC_{50}=\;12.5{\mu}M$) to observe morphological changes by the light microscopy. Both S. flavescens Ait. methanol extract and LA showed inhibitory effects on cadmium-induced cytotoxicity. Furthermore, LA showed dose-dependency in detoxication. From these results, it is conceivable to suggest that LA from S. flavescens Ait. methanol extract is a potential antitoxic agent.

The Protective Effects of the Extract of Saururus chinesis against Cadmium Induced Cytotoxicity(II) (삼백초 추출물의 카드뮴독성에 대한 방어효과(II))

  • This study was conducted to investigate the antitoxic component in ethanol extract of Saururus chinesis (S. chinesis). The results were as follows: Generally, detoxication effects by s. chinesis extract increased in proportion to the extract concentration. non 8 $\mu\textrm{g}$/g dosage of S. chinesis extract was administered, it showed the highest antitoxic effects in metallothionein induction. After the extract treatment, body weights generally increased In proportion to the extract concentrations. from the above results, S. chinesis extract Increased Metallothionein concentration and decreased the toxicity of cadmium In rats. In vitro the antitoxic activity of ethanol extract of S. chinesis on NIH3T3 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide) and SRB (sulforhodamine B protein) assays. The light microscopic study was carried out to observe morphological changes of the treeated cells. $10^{-2}$mg/ml Concentrations of S. chinesis extract was shown significant antitoxic activity. The number of NIH 3T3 fibroblasts were increased and tend to regenerate. These result suggest that S. chinesis extract retains a potential antitoxic activity.

  • This report describes the studies as follows through the research into the data of Cimicifuga heraclefolia Komarov blended prescriptions from Donguibogam. Cimicifuga heracleifola Komarov blended prescriptions are utilized for 39 therapeutic ranges. such as in dentistry, onclology, and 16 therapeutic ranges occupy over 84% totally, from dentistry to nasal disease. Cimicifuga heracleifola Komarov blended prescriptions use in the symptpms resulting from dentistry, intestine, internal disease, and they are used 22 disease also. The main pathologies of Cimicifuga heracleifola Komarov blended prescriptions found weakness in the digestive system, heat of stomach, wind of stomach in the splanchnology, and pathologies of wind-heat, wind-moisture, cold, excluding they are using for energy-blood, weakness, internal disease. Treatment groups were chosen as follows; control (medium only), $MTT_{50}$ group and five experimental groups. MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide} method was performed to evaluate the cytotoxicity of cell organelles and $IC_{50}$ was also measured. Accordingly we have examined the detoxification effects of Galgeuntang extract on cadmium-treated NIH 3T3 fibroblasts to observe morphological changes by the light microscopy. Galgeuntang extract showed cytoprotective effects on cadmium-induced cytotoxicity. Furthermore, Galgeuntang showed a dose-dependency in detoxication. The phenolic content of Galgeuntang ethanol extract was higher than that of water content. These results suggest that Galgeuntang extract may be used as a cytoprotective agent against cadmium (II)-mediated cytotoxicity.

Catalytic Detoxication of Coal Combustion Gases (연탄 연소가스의 촉매제독에 관한 연구)

  • A catalytic conversion of carbon monoxide has been studied in a coal combustion system. Three different catalysts were prepared by impregnating 0.2% platinum on three different types of supports, ${\gamma}-Al_2O_3$ pellets, ceramic foam and honeycomb. These catalysts have shown an excellent initial activities in the coal combustion system, but they were rapidly deactivated in repeated uses. Among these catalysts ceramic foam has shown to be better than others both in activities and durabilities. The main cause of deactivation seems to be ascribed to poisoning by zinc metal and sulfur compounds and to decrease in platinum surface area by sintering.
