Insecticidal Effect of Neem Cake Extracts on Cabbage Pests, Aphis gossypii and Pluetella xylostella

  • In organic agriculture, choose of effective and cheap bio-pesticide is very important. The authors developed an insecticidal extract from neem cake, waste of neem oil from kernel, and applied as a bio-pesticide. Bio-pesticide neem cake extracts experiment on cabbage pest was carried out at Wonju Agricultural Technology and Extension Center from 11 March to 30 May 2003. There were six treatments with three replications, using completely randomized design. Treatments involved three and six sprays of synthetic pyrethroid pesticide cypermethrin 10 EC at the dilution rate of 2.2 mL $L^{-1}$ of distilled water and four, five and six sprays of bio-pesticide neem at the dilution rate of 13.3 mL $L^{-1}$ of distilled water, and untreated control. For each treatment, designated sprayings were done at 7 days interval. Pre-spray data showed that the plants in all the experimental plots were already infested with aphid (Aphis gossypii), and diamondback moth (Pluetella xylostella). The results indicated that all neem pesticide treatments were more effective in insecticidal activity than the untreated control and the chemical treatments in controlling aphids and diamondback moth. Among the three neem treatments, there were no significant differences between them.

Enhanced Production of Phaeodactylum tricornutum (Marine Diatoms) Cultured on a New Medium with Swine Wastewater Fermented by Soil Bacteria

  • There have been a number of studies of methods for recycling animal wastewater to provide new bioresources. In the present work, a marine algal culture medium, designated KEP II, was prepared by adding swine waste (3% v/v) fermented by soil bacteria to a dilution of f/2 culture medium (CT). When Phaeodactylum tricornutum was grown in batch culture in KEP II, the cells lasted long at the exponential phase producing the specific growth rate and biomass; the production of total amino acids and secondary metabolites rose up to 5-fold. It also substantially enhanced the maximum quantum yield of photo system (PS) II of P. tricornutum, greatly increased the level of thylakoid membranes containing PS, and stimulated the production of pyrenoids, including enzymes for $CO_2$ fixation in chloroplasts. KEP II should improve the cost efficiency of industrial mass batch cultures and the value of microalgae for long-term preservation of fresh aquaculture feed as well as production of anticancer and antioxidant agents. Specifically, a low-cost medium for growing the diatoms of aquaculture feed will be economically advantageous.

Characterization of auxin production plant growth promotion by a bacterium isolated from button mushroom compost

  • An auxin-producing bacterial strain, designated 4-3, was isolated from waste button mushroom compost in Boryeong-si, Chungnam. The strain 4-3 was classified as a novel strain of Leucobacter tardus, based on chemotaxonomic and phylogenetic analyses. TLC and HPLC the isolated L. tardus strain 4-3 produced indole-3-acetic acid (IAA), the auxin. Maximum IAA productionof $94.3mg\;L^{-1}$ was detected for bacteria cultured in R2A medium with 0.1% l-tryptophan, incubated for 24 h at $35^{\circ}C$. Negative correlationwas observed between IAA production and pH of the culture medium, indicating that the increase inIAA caused acidification ofthe medium. The effect of supplementation with varying concentrations of l-tryptophan, a known precursor of IAA, was also assessed. production was maximal at 0.1% l, but decreased at lconcentrations above 0.2%. To investigate the plant growth-promoting effects of the bacterium, L. tardus strain 4-3 culture broth was used to inoculate water cultures and seed pots of mung bean. We found thatadventitious root induction and root growth were 2.2-times higher in thethan in the non-inoculated plants.

A Synergistic Effect of Chitosan and Lactic Acid Bacteria on the Control of Cruciferous Vegetable Diseases

  • Two lactic acid bacteria (LAB) designated J02 and J13 were recovered from fermented vegetables based on their ability to suppress soft rot disease caused by Pectobacterium carotovorum subsp. carotovorum (Pcc) on radish. J02 and J13 were identified as Lactobacillus pentosus and Leuconostoc fallax, respectively. The ability of J02 and J13 to suppress plant diseases is highly dependent on chitosan. LAB alone has no effect and chitosan alone has only a moderate effect on disease reduction. However, J02 or J13 broth cultures plus chitosan display a strong inhibitory effect against plant pathogens and significantly reduces disease severity. LAB strains after being cultured in fish surimi (agricultural waste) and glycerol or sucrose-containing medium and mixed with chitosan, reduce three cruciferous vegetable diseases, including cabbage black spot caused by Alternaria brassicicola, black rot caused by Xanthomonas campestris pv. campestris, and soft rot caused by Pcc. Experimental trials reveal that multiple applications are more effective than a single application. In-vitro assays also reveal the J02/chitosan mixture is antagonistic against Colletotrichum higginsianum, Sclerotium rolfsii, and Fusarium oxysporum f. sp. rapae, indicating a broad-spectrum activity of LAB/chitosan. Overall, our results indicate that a synergistic combination of LAB and chitosan offers a promising approach to biocontrol.

Fabrication of a solid catalyst using coal fly ash and its utilization for producing biodiesel

  • To recycle raw fly ash (RFA), a waste from thermal power plants, it was used to prepare solid catalysts which have many advantages compared with homogenous catalysts. When biodiesel was produced from soybean oil using RFA, only 1.2% of biodiesel conversion was obtained. A metal hydroxide, NaOH, KOH or $Ca(OH)_2$, was mixed with the acid-treated fly ash (ATFA), and the mixture was calcined at $700^{\circ}C$ for 3 h to prepare the solid catalyst. The solid catalyst prepared by mixing ATFA with NaOH, designated as SC-Na, showed a better performance than those prepared by mixing ATFA with KOH or $Ca(OH)_2$, respectively. The optimal mass ratio of ATFA with NaOH was 1:3, at which the proportion of $Na_2O$ increased to 60.2% in SC-Na, and 97.8% of biodiesel conversion was achieved under optimal reaction conditions (2 w% SC-Na relative to oil and 5 mL-methanol/g-oil at $50^{\circ}C$ for 4 h). Finally, a batch operation was repeatedly carried out to test the feasibility of reusing the solid catalyst, and more than 96% biodiesel conversion was stably achieved for the third round of operations. This study shows that RFA was successfully recycled to solid catalysts through a simple preparation method, and the solid catalyst was reused for the production of biodiesel with high conversion.

Development and Application of Streamline Analysis Method (유선 분석법의 개발 및 적용)

  • In order to properly evaluate the spatio-temporal variations of groundwater flow, the data obtained in field experiments should be corroborated into numerical simulations. Particle tracking method is a simple simulation tool often employed in groundwater simulation to predict groundwater flow paths or solute transport paths. Particle tracking simulations visually show overall the particle flow path along the entire aquifer, but no previous simulation studies has yet described the parameter values at grid nodes around the particle path. Therefore, in this study, a new technical approach was proposed that enables acquisition of parameters associated with particle transport in grid nodes distributed in the center of the particle path in groundwater. Since the particle tracking path is commonly referred to as streamline, the algorithm and codes developed in this works designated streamline analysis method. The streamline analysis method can be applied in two-dimensional and three-dimensional finite element or finite difference grid networks, and can be utilized not only in the groundwater field but also in all fields that perform numerical modeling.

Development for Improvement Methodology of Radiation Shielding Evaluation Efficiency about PWR SNF Interim Storage Facility (PWR 사용후핵연료 중간저장시설의 몬테칼로 차폐해석 방법에 대한 계산효율성 개선방안 연구)

  • For the purpose of improving the efficiency of the radiation impact assessment of dry interim storage facilities for the spent nuclear fuel of pressurized water reactors (PWRs), radiation impact assessment was performed after the application of sensitivity assessment according to the radiation source term designation method, development of a 2-step calculation technique, and cooling time credit. The present study successively designated radiation source terms in accordance with the cask arrangement order in the shielding building, assessed sensitivity, which affects direct dose, and confirmed that the radiation dosage of the external walls of the shielding building was dominantly affected by the two columns closest to the internal walls. In addition, in the case in which shielding buildings were introduced into storage facilities, the present study established and assessed the 2-step calculation technique, which can reduce the immense computational analysis time. Consequently, results similar to those from existing calculations were derived in approximately half the analysis time. Finally, when radiation source terms were established by adding the storage period of the storage casks successively stored in the storage facilities and the cooling period of the spent nuclear fuel, the radiation dose of the external walls of the buildings was confirmed to be approximately 40% lower than the calculated values; the cooling period was established as being identical. The present study was conducted to improve the efficiency of the Monte Carlo shielding analysis method for radiation impact assessment of interim storage facilities. If reliability is improved through the assessment of more diverse cases, the results of the present study can be used for the design of storage facilities and the establishment of site boundary standards.

Effect of the Addition of Binders on the Fuel Characteristics of Wood Pellets (바인더의 첨가가 목재 펠릿의 연료적 특성에 미치는 영향)

  • This work was conducted to investigate the effect of the addition of binders, such as rapeseed flour, coffee waste, bark, pine cone and lignin powder, on the fuel characteristics of the pellets fabricated with larch and tulip tree sawdust. Moisture content, bulk density and higher heating value of most pellets fabricated with the binders exceeded the 1st-grade pellet standard designated by Korea Forest Research Institute, but ash content of the pellets fabricated with rapeseed flour or bark of 10 wt% on the dry weight basis of sawdust was satisfied with just the 2nd- or 3rd-grade standard. The durability of tulip tree-pellets was positively influenced by the addition of rapeseed flour, coffee waste or lignin powder and increased with increasing the amount of the binders. For larch-pellets, the increase of binders did not greatly affect the durability, and even the durability reduced with the increase of bark or pine cone. From the microscopic observation, the obvious feature of pellet surfaces was not identified by the type of binder but by the addition amount of the binder. In summary, the addition of binders contributed to the fuel characteristics of wood pellets, and particularly the characteristics of wood pellets fabricated with coffee waste improved greatly. Therefore, if the binders are secured sufficiently with a reasonable cost, it might be possible to commercialize wood/binder pellets, which have better fuel characteristics than conventional wood pellets.

Construction of Information System for Management of Cultural Heritage on the Web Using a Pilotless Helicopter Photogrammetry System (무인 헬기 사진측량시스템을 이용한 Web 상에서의 문화재 관리 정보시스템 구축)

  • Structure-typed cultural heritage, objects of preservation are positioned as one of the very important heritage in the nation, and the preservation of prototypical structures become influential in national development and against natural disaster. For this reason, Digital Close Range Photogrammetry has recently been diversely used. Despite its popular use, the measurement has limits that make it unsuitable for photographing precise cultural heritage situated at high mountainous terrain or where people can not approach easily. These high gigantic stone statues are among the preserved structure-typed cultural heritage. In order to supplement the limits, when using the measurement, a camera tripod with +30m, a ladder truck and a shore should be equipped, which means additional equipment leads to it being a waste of cost and time. In this vein, a device was developed in detail, using a RC Helicopter installed with a CCD video camera with ease of control, safety, equipment, carrying, movement and approach, then checked image shot by a wireless modem at real time and considered the economical efficiency without re-photographing. Next, the author digitized the images of the nationally designated structure-typed cultural heritage, used materials on their restoration as the third dimension in order to construct the integrated management-information system for cultural heritage. Through the above processes, this study can provide specific information on 3D images and 3D CAD sections of structured-typed cultural heritage for both the public and specialists on the web. Moreover, it suggests the foundation to restore the damaged cultural heritage in the future by aiming for their effective management and preservation.

Experimental & computational study on fly ash and kaolin based synthetic lightweight aggregate

  • The objective of this study is to manufacture environmentally-friendly synthetic lightweight aggregates that may be used in the structural lightweight concrete production. The cold-bonding pelletization process has been used in the agglomeration of the pozzolanic materials to achieve these synthetic lightweight aggregates. In this context, it was aimed to recycle the waste fly ash by employing it in the manufacturing process as the major cementitious component. According to the well-known facts reported in the literature, it is stated that the main disadvantage of the synthetic lightweight aggregate produced by applying the cold-bonding pelletization technique to the pozzolanic materials is that it has a lower strength in comparison with the natural aggregate. Therefore, in this study, the metakaolin made of high purity kaolin and calcined kaolin obtained from impure kaolin have been employed at particular contents in the synthetic lightweight aggregate manufacturing as a cementitious material to enhance the particle crushing strength. Additionally, to propose a curing condition for practical attempts, different curing conditions were designated and their influences on the characteristics of the synthetic lightweight aggregates were investigated. Three substantial features of the aggregates, specific gravity, water absorption capacity, and particle crushing strength, were measured at the end of 28-day adopted curing conditions. Observed that the incorporation of thermally treated kaolin significantly influenced the crushing strength and water absorption of the aggregates. The statistical evaluation indicated that the investigated properties of the synthetic lightweight aggregate were affected by the thermally treated kaolin content more than the kaoline type and curing regime. Utilizing the thermally treated kaolin in the synthetic aggregate manufacturing lead to a more than 40% increase in the crushing strength of the pellets in all curing regimes. Moreover, two numerical formulations having high estimation capacity have been developed to predict the crushing strength of such types of aggregates by using soft-computing techniques: gene expression programming and artificial neural networks. The R-squared values, indicating the estimation performance of the models, of approximately 0.97 and 0.98 were achieved for the numerical formulations generated by using gene expression programming and artificial neural networks techniques, respectively.
