Involvement of Antiapoptotic Signals in Rat PC12 Cells Proliferation by Cyclosporin A Treatment

  • Cyclosporin A (CsA) plays an important role in clinical medicine and basic biology as an immunosuppressant and a mitochondrial permeability blocker, respectively. It was reported that CsA has a protective role by preventing apoptosis and promoting the proliferation in severed neurons. However, the molecular mechanisms for CsA-induced neuronal cell proliferation are unclear. In this study, we examined the mechanisms underlying the CsA-induced proliferation of PC12 cells. CsA increased the viability of PC12 cells in a dose(over $0.1{\sim}10\;{\mu}M$)-and time-dependent manner. The level of ROS generation was decreased in the CsA-treated PC12 cells. Expression of Bcl-2, an antiapoptotic molecule that inhibits the release of cytochrome c from the mitochondria into the cytosol, was upregulated, whereas Bax, a proapototic molecule, was not changed in the CsA-treated PC12 cells. CsA downregulated the mRNA expression of VDAC 1 and VDAC 3, but VDAC 2 was not changed in the CsA-treated PC12 cells. The level of cytosolic cytochrome c released from the mitochondria and the caspase-3 activity were attenuated in the CsA-treated PC12 cells. These results suggest that the mitochondria-mediated apoptotic signal and Bcl-2 family may play an important role in CsA-induced proliferation in PC12 cells.

일부 대학생의 흡연 실태와 구강건강에 관한 인식 및 행태 조사연구 (Research on the Smoking Conditions of College Students and the Behavior and Awareness of Oral Hygiene)

  • 본 연구는 일부 대학생들의 흡연행태와 흡연자와 비흡연자 간의 흡연에 대한 인식과 구강건강관리행태를 알아보고자 경상북도, 대구광역시, 강원도에 위치한 4년제 대학교에 재학 중인 대학생 260명을 대상으로 흡연에 대한 인식과 구강건강관리행태에 대해 조사한 결과 다음과 같은 결론을 얻었다. 1. 흡연자의 하루 평균 흡연량은 반 갑 미만이, 흡연기간은 1년~3년 미만이 가장 높았으며, 흡연을 시작하게 된 동기는 호기심과 주변 권유가 가장 높았고, 흡연이유는 스트레스 해소, 습관적 순으로 나타났다. 2. 흡연유무에 따른 구강건강관리행태 중 1회 잇솔질 시간, 정기적 치과 내원, 최근 치과 내원시기, 치과 방문 목적, 스켈링 및 구강보건교육 경험에서 유의한 차이가 있었다. 3. 흡연유무에 따른 구강건강에 대한 인식 중 주관적 구강건강상태에서 유의한 차이가 있었다. 4. 흡연유무에 따른 흡연에 대한 인식 중 담배, 한 두 개피 정도의 담배, 식후의 담배 및 간접흡연이 건강에 미치는 영향에 대한 인식에서 유의한 차이가 있었다. 이상의 결과로 미루어 볼 때 흡연자는 비흡연자에 비해 구강건강관리행태 및 구강건강에 대한 인식, 흡연에 대한 인식이 부족하므로, 대학생들을 위한 금연교육과 구강보건교육 프로그램 개발의 활성화가 필요하다고 사료된다.

Qraycam을 이용한 구강보건교육의 효과 (The effect of oral health education using Qraycam)

  • 본 연구의 목적은 Qraycam을 이용한 구강환경검사, 구강보건태도 및 구강건강관리 행동점수의 전 후비교를 통하여 구강보건교육의 효과를 분석하고자 하였으며, 단일집단 사전 사후 연구설계로 진행되었다. 2018년 9월부터 12월 중순까지 실험 중재를 위한 실습대상자 33명과 구강보건교육 선호도 조사를 위한 학생 33명을 연구대상으로 하였다. 자료의 분석은 IBM SPSS 24.0 Ver을 이용하였고, 통계적 검정을 위한 유의수준은 0.05로 하였다. 그 결과, Qraycam을 사용한 중재 전 후의 구강환경 검사인 O' Leary index와 구취농도는 유의하게 감소하였다(p<.001). 중재 전 후에 따른 구강보건태도 점수와 구강건강관리 행동점수는 통계적으로 유의하게 높게 나타났다(p<.001). 학생들의 구강보건교육 선호도 중 "구강보건교육에서 유용하다"와 "치태를 인지하고 결정하는 능력이 좋다"는 항목에서 Qraycam을 사용하는 경우 만족도는 유의하게 높게 나타났다(p<.001). 결론적으로, 치과임상현장에서 Qraycam 사용을 통한 구강보건교육은 치과위생사들의 구강보건교육 효과를 증진시키기 위한 방법이 될 수 있을 것이다.

Normal range of facial asymmetry in spherical coordinates: a CBCT study

  • Purpose: This study aimed to measure the bilateral differences of facial lines in spherical coordinates from faces within a normal range of asymmetry utilizing cone-beam computed tomography (CBCT). Materials and Methods: CBCT scans from 22 females with normal symmetric-looking faces (mean age 24 years and 8 months) were selected for the study. The average menton deviation was $1.01{\pm}0.66$ mm. The spherical coordinates, length, and midsagittal and coronal inclination angles of the ramal and mandibular lines were calculated from CBCT. The bilateral differences in the facial lines were determined. Results: All of the study subjects had minimal bilateral differences of facial lines. The normal range of facial asymmetry of the ramal and mandibular lines was obtained in spherical coordinates. Conclusion: The normal range of facial asymmetry in the spherical coordinate system in this study should be useful as a reference for diagnosing facial asymmetry.

The Protective Effect of Chondroitin from Raja kenojei Cartilage on Collagen-induced Arthritis in DBA/1J Mice

  • In this study, we evaluated whether the oral administration of chondroitin from the cartilage of Raja kenojei is effective on the progression of rheumatoid arthritis (RA), using collagen-induced arthritic (CIA) mice. Arthritis development was delayed dose-dependently in the chondroitin-treated groups. The pre- and late-treated groups receiving 1,000 mg/kg of chondroitin had clinical scores that were reduced significantly by 56.9 (p<0.05) and 43.3% (p<0.05), respectively, compared to the vehicle-treated groups. Hematoxylin eosin staining and X-ray radiography showed that the chondroitins reduced the infiltration of inflammatory cells and prevented joint destruction of the knee and paw. Reverse transcription-polyerase chain reaction analysis revealed that chondroitin administration inhibited the expressions of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), $interlukin-1{\beta}$ ($IL-1{\beta}$), and $interferon-{\gamma}$ ($IFN-{\gamma}$) in joints more than the administration of vehicle. Chondroitin treatment also decreased the production of rheumatoid factors (RF), IgG and IgM, in the serum of CIA mice. These results indicate that chrondroitin administration has a protective effect involving the inhibition of pro-inflammatory cytokine production in CIA mice.

Intra-Oral Factors Influencing Halitosis in Young Women

  • Objectives: The aim of this research was to determine intra-oral factors that affect halitosis in young women. Methods: This study was performed between March 2014 to May 2014, and included 35 women in their 20s with good oral health. Correlation and logistic regression analyses were performed to investigate the change in halitosis immediately, and 1 hour after scaling. Results: In both oral gas (OG) and extraoral gas (EG) groups, halitosis was reduced after scaling compared to before scaling. The logistic regression analysis of oral state factors in OG showed that as oral fluid [odds ratio (OR) = 0.792, p = 0.045] and dental plaque (OR = 0.940, p = 0.016) decreased by 1 unit, the OR in the OG group decreased (> 50). In addition, as glucose levels in the oral cavity (OR = 1.245, p = 0.075) and tongue coating index (OR = 2.912, p = 0.064) increased by 1 unit, the OR in the OG group increased (> 50). Furthermore, in the EG group, as oral fluid (OR = 0.66, p = 0.01) and dental plaque (OR = 0.95, p = 0.04) decreased, the OR in the EG group decreased (> 50) significantly. Conclusion: To control halitosis, it is necessary to increase oral fluid and decrease the amount of tongue plaque. Furthermore, maintaining a healthy oral environment, aided by regular scaling and removal of dental plaque, may significantly control halitosis.

유효성분들을 배합한 치약제의 실험실적 및 임상적 치아미백유지 효과 (Tooth whitening maintenance efficacy of dentifrices containing several active ingredients in vitro and in vivo)

  • Objectives: The purpose of this study was to investigate tooth whitening maintenance efficacy of several dentifrices containing effective ingredients for tooth whitening. Methods: Hydroxyapatite specimens(HAPs) staining was done by using modified Stookey's methods. HAPs were treated with 2.9% hydrogen peroxide containing strip for whitening, and were shaken with several dentifrice slurry(dentifrice 1 : artificial saliva 2) for 30 minutes. The HAPs were finally dipped in staining solution for an hour. Shaking and dipping were repeated 4 times and lightness values were measured by colorimeter at each step. In clinical test, test 4 dentifrice and control dentifrice were evaluated by 21 subjects for 2 months after receiving institutional review board(IRB) approval. Organoleptic(vita shade guide) and instrumental(SHADEEYE-NCC) evaluation were performed for whiteness change of teeth. Statistical analysis was performed using repeated measures ANOVA, Tukey's post hoc test and ${\chi}^2$-test(p<0.05). Results: All dentifrices showed statistical significance in comparison with control dentifrice containing sodium fluoride and test 4 dentifrice containing sodium pyrophosphate, sodium metaphosphate, candelilla wax, and sodium fluoride showed statistical significance in comparison with other dentifrices by inhibiting staining in vitro(p<0.05). In clinical test, test 4 dentifrice showed better effects than control dentifrice in organoleptic and instrumental evaluation in tooth whitening maintenance efficacy(p<0.05). The awareness toward tooth whitening maintenance efficacy for 2 months use showed that test 4 dentifrice was much better than control dentifrice, but did not show statistically significant(p>0.05). Conclusions: Dentifrice containing sodium pyrophosphate, sodium metaphosphate, candelilla wax and sodium fluoride was more effective in keeping teeth white.

구강양치액의 최신 경향 (Contemporary Update of Mouth Rinse)

  • Introduction : Inadequate oral health control is a major risk of oral diseases. Regular home-based care is essential to maintain good oral hygiene. In particular, mouthrinses can support conventional tooth brushing in reducing accumulation of oral plaque. Effect : Antimicrobial mouthrinses are used as part of daily oral care to reduce plaque and gingivitis. Mouthrinses contains fluoride could help remineralization of enamel and dentin. The most common molecules contained in mouthrinses are chlorhexidine, essential oils, cetyl pyridinium chloride, triclosan, hyaluronic acid. Currently, chlorhexidine is the most efficacious compound, with both antiplaque and antibacterial activities. Similar results are reported for essential oils and cetyl pyridinium chloride, although with a somewhat reduced efficacy. Considering the adverse effects of chlorhexidine and its time-related characteristics, this molecule may best be indicated for acute/short term use, while essential oils and cetyl pyridinium chloride may be appropriate for long-term, maintenance treatment. Conclusion and suggestion : Antimicrobial mouthrinses are safe and effective, and when used in conjunction with brushing and flossing, they are an important method of reducing plaque and gingivitis. To improve compliance, dental health care professionals should adapt oral health care recommendations to fit patients' specific needs.

  • Purpose: This study aimed to verify the oral health behavior according to the oral health education experience and knowledge who visit public health center at Gyeonggi-do districts. Methods: The research has come into a survey from 2th on March to 27th on April 2018 on the youth and the century where is located in Gyeonggi-do public health centers. Results: There were statistically significant differences in the number of brushing, correct tooth brushing, and the use of oral aids in the presence of oral health education experience(p<.05). Oral health education has been shown to affect knowledge and behavior when oral health education experience is high. Conclusion: Periodic oral health education should be further strengthened, not once for the prevention of proper oral care and oral illness.

Four Voltage-Gated Potassium Currents in Trigeminal Root Ganglion Neurons

  • Various voltage-gated $K^+$ currents were recently described in dorsal root ganglion (DRG) neurons. However, the characterization and diversity of voltage-gated $K^+$ currents have not been well studied in trigeminal root ganglion (TRG) neurons, which are similar to the DRG neurons in terms of physiological roles and anatomy. This study was aimed to investigate the characteristics and diversity of voltage-gated $K^+$ currents in acutely isolated TRG neurons of rat using whole cell patch clamp techniques. The first type (type I) had a rapid, transient outward current ($I_A$) with the largest current size having a slow inactivation rate and a sustained delayed rectifier outward current ($I_K$) that was small in size having a fast inactivation rate. The $I_A$ currents of this type were mostly blocked by TEA and 4-AP, K channel blockers whereas the $I_K$ current was inhibited by TEA but not by 4-AP. The second type had a large $I_A$ current with a slow inactivation rate and a medium size-sustained delayed $I_K$ current with a slow inactivation rate. In this second type (type II), the sensitivities of the $I_A$ or $I_K$ current by TEA and 4-AP were similar to those of the type I. The third type (type III) had a medium sized $I_A$ current with a fast inactivation rate and a large sustained $I_K$ current with the slow inactivation rate. In type III current, TEA decreased both $I_A$ and $I_K$ but 4-AP only blocked $I_A$ current. The fourth type (type IV) had a smallest $I_A$ with a fast inactivation rate and a large $I_K$ current with a slow inactivation rate. TEA or 4-AP similarly decreased the $I_A$ but the $I_K$ was only blocked by 4-AP. These findings suggest that at least four different voltage-gated $K^+$ currents in biophysical and pharmacological properties exist in the TRG neurons of rats.
