Effects Of Cultured Bone Cell On The Regeneration Of Alveolar Bone (배양골세포 이식이 치조골재생에 미치는 영향)

  • This study was performed to estimate the effects of cultured bone cell inoculated on porous type hydroxyaptite for the regeneration of the artificial alveolar bone defect. In this experiment 3 beagle dogs were used, and each of them were divided into right and left mandible. Every surgical intervention were performed under the general anesthesia by using with intravenous injection of Pentobarbital sodium(30mg/Kg). To reduce the gingival bleeding during surgery, operative site was injected with Lidocaine hydrochloride(l:80,000 Epinephrine) as local anesthesia. After surgery experimental animal were feeded with soft dietl Mighty dog, Frisies Co., U.S.A.) for 1 weeks to avoid irritaion to soft tissue by food. 2 months before surgery both side of mandibular 1st premolar were extracted and bone chips from mandibular body were obtained from all animals. Bone cells were cultured from bone chips obtained from mandible with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. Porous type hydroxyapatite were immerse into the high concentrated cell suspension solution, and put 4 hours for attachin the cells on the surface of hydroxyapatite. Graft material were inserted on the artificial bone defect after 3 days of culture. Before insertion of cellinoculated graft material, scanning electronic microscopic observation were performed to confirm the attachment and spreading of cell on the hydroxyapatite surface. 3 artificial bone defects were made with bone trephine drill on the both side of mandible of the experimental animal. First defect was designed without insertion of graft material as negative control, second was filled with porous replamineform hydroxyapatite inoculated with cultured bone marrow cells as expermiental site, and third was filled with graft materials only as positive control. The size of every artificial bone defect was 3mm in diameter and 3mm in depth. After the every surgical intervention of animals, oral hygiene program were performed with 1.0% chlorhexidine digluconate. All of the animals were sacrificed at 2, 4, 6 weeks after surgery. For obtaining histological section, tissus were fixed in 10% Buffered formalin and decalcified with Planko - Rycho Solution for 72hr. Tissue embeding was performed in paraffin and cut parallel to the surface of mandibular body. Section in 8um thickness of tissue was done and stained with Hematoxylin - Eosin. All the specimens were observed under the light microscopy. The following results were obtained : 1. In the case of control site which has no graft material, less inflammatory cell infiltration and rapid new bone forming tendency were revealed compared with experimental groups. But bone surface were observed depression pattern on defect area because of soft tissue invasion into the artificial bone defect during the experimental period. 2. In the porous hydroxyapatite only group, inflammatory cell infiltration was prominet and dense connective tissue were encapsulated around grafted materials. osteoblastic activity in the early stage after surgery was low to compared with grafted with bone cells. 3. In the case of porous hydroxyapatite inoculated with bone cell, less inflammatory cell infiltration and rapid new bone formation activity was revealed than hydroxyapatite only group. Active new bone formation were observed in the early stage of control group. 4. The origin of new bone forming was revealed not from the center of defected area but from the surface of preexisting bony wall on every specimen. 5. In this experiment, osteoclastic cell was not found around grafted materials, and fibrovascular invasion into regions with no noticeable foreign body reaction. Conclusively, the cultured bone cell inoculated onto the porous hydroxyapatite may have an important role of regeneration of artificial bone defects of alveolar bone.

A Review on the Background of Takjok(濯足; Washing Feet) and the Landscape Architectual Meaning of Its Cultural Phenomenon - Focused on Takjokjiyu(濯足之遊) Shown on Poetry, Prose, and Painting - (탁족(濯足)의 배경과 그 문화현상에 담긴 조경적 의미 - 시문과 그림에 나타난 탁족지유(濯足之遊)를 중심으로 -)

  • This study suggests the necessity of landscaping alternatives for the succession of Takjok(濯足) culture by considering the background and meaning of Takjok's cultural phenomenon shown in old literatures and paintings and its result is as follows. An old idiom, 'Takyoung Takjok(濯纓濯足)' implying a disinterested living attitude from the mundane world and an attitude complying with nature, has been sublimated to 'Takjokjiyu(濯足之遊)' which means living in comfortable retirement through life in seclusion(隱逸). Classical scholars immerse their feet in soft-flowing(柔軟) water for 'Mulailche(物我一體; synchronized with nature)' which is a unified condition with 'Do(道; truth)' and connected to the stage of 'Yu(遊)', a free mental state, and its nature. The cultural phenomenon of Takjok appeared in the inherited landscape paintings in the Joseon dynasty period after the late stage of Koryo. Takjok shown in 'Pal Kyung Shi'(八景詩; poetry singing for the eight scenary) was described as not a transcendent scene, but as a scenery of daily life. Dense forest and water, such as a stream with clean water, rocks, and pine trees shown in Takjok paintings have been symbolized as a seclusion space for classical scholars with higher thinking and their mental states have been more emerged. Mental pleasures called as seclusion and Takjokjiyu have been relatively emphasized in the Takjok paintings of the Joseon Dynasty period contrary to the Chinese Takjok paintings emphasizing Chung Gye(淸溪; clean stream) and Chang Rang(滄浪; high and clean wave) and strongly representing the image of 'Chung Ryu'(淸流; clean flowing water) and the veneration for antiquity. The view of nature described in the Takjok paintings represents the provision of nature as a situation and attitudes of classical scholars and implies a Taoism perspective which describes the 'do' of nature. This view of nature itself remained intact(無爲自然) with the love of mountains and water, showing a side of the zeitgeist and aesthetic consciousness of China and Joseon. The 'Takjokjiyu' of both countries has be interpreted as a symbol of personality development, behavior, life in seclusion, or transcending the mundane world and has also been accepted as a method of summer vacation in the real world. It should be considered that Takjok includes ordinary people's wisdom to resist the hot weather, as well as the classical scholar's ideal and the veneration of antiquity. From this perspective, water space, Takjok rocks, and the use of water based on the environmental supportability should be newly focused as a recreational space and it reminds us that the spirit of Takjok is a classical mental healing method.

A Study of Removal Property of Harmful Algal Blooms by Hwangto and Oriental Mineral Medicines (황토와 광물성 한약재의 적조구제 특성에 관한 연구)

  • This study was carried out to find a new material having high removal efficiency for the harmful red tide. C. polykrikoides grow very fast and accumulate into dense and visible patches near the surface of the seawater ('Water bloom'). Some mineral medicines and Hwangto (reddish soil consist of clay minerals and Fe-oxides) were used in this study to remove C. polykrikoides. The pre-determined sprinkling ratio of mineral vs. seawater which contains approximately 5,000 cells/mL of C. polykrikoides was 10 g/L. In order to quantify the removal efficiency, the density of living cells was measured by counting with the Intervals of 0, 10, 30, and 60 minutes after sprinkling. Five Hwangtos feom different localities were examined in this study. It is found that a material with a high concentration of Fe and Al was the most effective to remove C. polykrikoides. After the sprinkling of the Hwangto showing the best removal efficiency in the test, 99% of total algaes were found to be eliminated within 60 minutes. Jeokeokji showed the highest removal efficiency among clay mineral medicines(92% removal efficiency after 60 minutes), and the rests in decreasing order are as follows: Gamto (91%) > Baekseokji (89%) > Hydromica (81%). In addition, Fe-oxide mineral medicine similarly looking as fine-grained earthy Daejaseok showed 100% removal efficiencyafter 30 minutes, and Wooyoeryang, 95% after 60 minutes. It is noted that even little addition (1 g/L) of Daejaseok, 10% of Hwangto concentration into seawater showed the removal efficiency of 100% after 60 minutes. From the results, it could be concluded that the fine-grained earthy Daejaseok was the most effective natural mineral medicine to remove the C. polykrikoides from seawater. Under the microscope the removal mechanism was found to be activated in the following order: adsorption, swelling of chain colony, chain colony crisis and algaecide.

Freeze-thaw Resistance Estimation of Concrete using Surface Roughness and Image Analysis (콘크리트의 동결융해 저항성 추정을 위한 표면 거칠기 및 이미지 분석의 적용성)

  • As part of a research dedicated to the field evaluation of the durability of concrete subjected to freezing-thawing, this study analyzes the relationship between the surface roughness and the relative dynamic elastic modulus through image analysis. Four mix compositions with water-to-binder ratios (W/B) of 40%, 50%, 60% and 70% and without AE agent were considered to provoke early freezing. The basic physical properties of the mixes including the relative dynamic elastic modulus and the compressive strength were first evaluated experimentally according to W/B. Then, tests were performed to measure the surface roughness followed by photographs and SEM image analysis. The measured surface roughness tended to increase with larger number of freezing-thawing cycles regardless of W/B. The relative dynamic elastic modulus appeared to increase gradually with the number of cycles for the relatively denser mixes with W/B of 40% and 50%. Besides, the surface roughness increased only at rupture for the mixes with W/B of 60% and 70%. Moreover, the analysis of the photographs of the surface of the mixes with W/B of 40% and 50% revealed that the degradation progressed gradually from the surface with the freezing-thawing cycles. However, for the mixes with W/B of 60% and 70%, apparent change of the surface remained very insignificant until rupture at which damage like cracking could be observed. Consequently, the analysis of surface photograph or the measurement of the surface roughness presented some limitation in assessing the degree of freezing-thawing-induced degradation in case of relatively porous specimens. On the other hand, the photograph and surface roughness appeared to be sufficient for assessing such degradation for the mixes with W/B of 40% and 50%. Accordingly, the image of the surface and the surface roughness are potentially applicable on site for the assessment of freezing-thawing damages in relatively dense mixes.

  • The study describes on the identification and morphological characteristics of each species, ecological characteristics and propagation techniques, and developing utilization measures of the Parthenocissus plants for environment conservation and revegetation of the babe rock-slopes. The following species and varieties are disscussed in this study; Parthenocissus tricuspidata (S. et Z.) Planch. var. veitchii Rehd. var. lowii Rehd. var. pupurea Hort. Parthenocissus quiquefolia (L.) Planch. var. engelmanni Rehd. var. saint-pauli Rehd. var. hirsuta Planch. Parthenocissus henryana Diels et Gilg. Parthenocissus thomsoni Planch. Parthenocissus heptaphylla Small. Parthenocissus inserta (Kern.) K. Fritsch. Parthenocissus laetevirens Rehd. Parthenocissus himalayana Planch. These are, in general, all vigorous self-clinging climbers that will quickly cover a wall and bare rock surfaces with a dense network of branch growths and beautiful green leaves which change to shades of scarlet and crimson before they fall in Autumn. Parthenocissus tricuspidata out of 8 species in the genus Parthenocissus is the most useful plant for the environment conservation including the bare rock-slope revegetation and for the production of food and shelter for wildlifes. This native of Korea clings by means of small rootlike holdfasts (adhesive discs) and holds (tendrils) to stone work or any other solid support, tenaciously.

A Case Study of Geometrical Fracture Model for Groundwater Well Placement, Eastern Munsan, Gyeonggido, Korea (지하수개발을 위한 단열모델 연구사례(경기도 문산 동쪽지역))

  • This study is the case of groundwater development based on the geometrical fracture model of target area established only through geological fracture mapping technique. A fracture mapping of $9km^2$, eastern Munsan, has been conducted to determine geological and hydrological factors for new water well placement in the Gyeonggi gneiss complex. Geophysical exploration was not applicable because of small restricted area and dense underground utilities at the site. Form line mapping on the basis of foliation orientation and rock type revealed a synform of NS fold axis bearing to the south. An EW geological cross-section passed through the site area shows a F2 synform as a double-wall ice cream spoon shape. Three regional faults of $N20^{\circ}E,\;N30^{\circ}W$, and NS have been dragged into the site to help understand extensional fault paths. The $N20^{\circ}E$ fault with dextral sense is geometrically interpreted as a western fault of two flexural conjugate type-P shear faults in the F2 synformal fold. The NE cross-section reveals that a possible groundwater belt in the western limb of super-posed fold area is formed as a trigonal prism within 100 m depth of the intersectional space between the $N20^{\circ}E$ fault plane and the weakly sheared plane of transposed foliation. Another possible fault for water resource strikes $N40^{\circ}E$. Recommended sites for new water well placement are along the $N20^{\circ}E\;and\;N40^{\circ}E$ faults. As a result of fracture mapping, 145 ton/day of water can be produced at one well along the $N20^{\circ}E$ fault line. Exploration of groundwater in the area is succeeded only using with geological fracture mapping and interpretation of geological cross-section, without any geophysical survey. Intersection of fault generated with the F2 synformal fold and foliation supply space of groundwater reserver.

  • We investigated the phytoplankton community from June to September 1997 in the waters off Kohung, Korea where red tides dominated by harmful dinoflagellates had occurred from August to September or October since 1995. We took water samples five times from 5 depths at 6 or less stations in this study period. The most dominant harmful dinoflagellate during the red tide which had outbroken on August 24, 1997 was Gyrodinium impudicum, not Cochlodinium polykrikoides. On August 21 just before the harmful red tide occurred the abundance of G. impudicum at the inner bay station, 90cells $ml^{-1}$, was higher than that at the outer bay station. However, on August 27 just after the red tide had outbroken, the abundance of G. impudicum at the inner bay station did not increase, whereas that at the outer bay increased rapidly and reached to the maximum of 30,000 cells $ml^{-1}$. Instead, diatoms such as Skeleltonema costatum, Chaetoceros pseudocurvisetus, Pseudonitzschia pungens rapidly increased at the inner bay station where fresh water from lands has reached. The high abundance of diatoms might have inhibited the growth of red tide dinoflagellates at this station. The transport of already formed red tide patches from offshore areas, aggregation of scattered cells driven by physical forces, and/or competition between diatom and dinoflagellates might be responsible for this appearance of dense red tide patches at the outer bay station.

Changes of Fruit Structure and Sugar Contents during the Fruit Growth and Development in 'Yumyeong' Peach [Prunus persica (L.) Batsch] (과실생육시기별 '유명' 복숭아의 과실조직 발달과 당 변화)

  • 'Yumyeong' is one of the most popular peach varieties in Korea. This study was conducted to monitor the developments of cells and tissues, and the changes in sugar contents during fruit growth and development. At bloom, there were two rows of vascular tissues, and the number and the position of internal vascular bundles were consistent during the fruit growth, however, the number of vascular tissues were increased and the distribution was irregular in the flesh tissues. The tissues between the inner integument and the internal vascular bundles showed different development characteristics from other parenchyma cells which were consisted of small and dense cells containing tannins. Such observation suggested that the stone of peach was consisted of inner epidermis and cells in the internal vascular tissues. The outer epidermis consisted of single layer cells at bloom differrentiated into 1-2 layers by horizontal cell divisions on 14 days after full bloom. On 30 days after full bloom, the epidermis was consisted of 5-6 layers by vertical cell divisions. The cell layers consisting the outer epidermis were gradually decreased to 1-2 layers at maturity. The observations on the changes in the epidermis confirmed that some of the cells consisting the hypodermis of peach fruit were originated from the cells of outer epidermis. Tylosis was observed from 35 days after full bloom and the size and number of tylosis were increased until full fruit maturity. The sucrose content showed the tendency of sharp increase from 50 days to 120 days after full bloom then decrease slightly. After when stone hardening ended, other solids showed a gradual decrease tendency from 80 days after full bloom.

Histomorphologic Change of Radiation Pneumonitis in Rat Lungs : Captopril Reduces Rat Lung Injury Induced by Irradiation (X-선 조사로 생긴 흰쥐 폐장 상해의 형태학적 변화: Captopril에 의한 폐장 상해의 경감 효과)

  • Purpose : To assess the histomorphologic changes in the rat lung injury induced by radiation, to determine whether captopril reduces the rat lung injury and to evaluate change in TNF-${\alpha}$ and TGF-${\beta}$ in rat lung damage by radiation and captopril Methods and material : Right lungs in male Sprague-Dawley rats were divided irradiation alone (10, 20, 30 Gy) or radiation (same dose with radiation alone group) with captopril (500 mg/L). Radiation alone group were sacrificed at twelve hours and eleven weeks after radiation and radiation with captopril group (captopril group) were sacrificed at eleven weeks after radiation with captopril. We examined the light microscope and electron microscopic features in the groups. Results : In radiation alone group, there were patch parenchymal collapse and consolidation at twelve hours after radiation. The increase of radiation dose shows more prominent the severity and broader the affected areas. Eleven weeks after radiation, the severity and areas of fibrosis had increased in proportion to radiation dose given in the radiation alone group. There was notable decrease of lung fibrosis in captopril group than in radiation alone group. The number of mast cells rapidly increased with increase of radiation dose in radiation alone group and the degree of increase of mast cell number and severity of collagen accumulation more decreased in captopril group than in radiation alone group. In radiation alone group, expression of TNF-${\alpha}$ and TGF-${\beta}$ increased according to increase of radiation dose at twelve hours after radiation in both group. At eleven weeks after radiation, expression of TGF-${\beta}$ increased according to increase of radiation dose in radiation group but somewhat decreased in captopril group. In the captopril group the collagen deposition increased but less dense than those of radiation alone group. The severity of perivascular thickening, capillary change, the number and degranulation of mast cells more decreased in the captopril group than in the radiation alone group. Conclusion : It is concluded that the effect of captopril in the rat lungs after radiation was considered to be due to its effect on inhibition of mast cells and reduction of collagen deposition, and captopril may be protect in lung damage after radiation. We observed expression of TNF-${\alpha}$ and TGF-${\beta}$ increased at the early phase after radiation and expression of TGF-${\beta}$ increased in proportion to increase of radiation dose at the chronic phase after radiation. This results will contribute to future investigation in reduction mechanism of captopril in lung damage after radiation.

Crustal Characteristics and Structure of the Ulleung Basin, the East Sea (Japan Sea), Inferred from Seismic, Gravity and Magnetic Data (탄성파 및 중자력자료에 의한 울릉분지의 지각특성 및 구조 연구)

  • Depths to four seismic sequence boundaries and the thickness of each sequence were estimated and mapped based on multi-channel seismic data in the Ulleung Basin. These depth-structure and isopach maps were incorporated into the interpretation of gravity and magnetic anomaly maps. The sediment thickness ranges from 3,000 m to 4,000 m in the central basin, while it reaches 6,000 m locally along the southwestern, western, and southeastern margins. The acoustic basement forms a northeast-southwest elongated depression deeper than 5000 m, and locally deepens up to 7,500 m in the southwestern and western margins. Low gravity anomalies along the western and southern margins are associated with basement depressions with thick sediment as well as the transitional crust between the continental and oceanic crusts. Higher gravity anomalies, dominant in the central Ulleung basin, broaden from southwest toward northeast, are likely due to the shallow mantle and a dense crust. A pair of magnetic elongations in the southeastern and northwestern margins appear to separate the central Ulleung basin from its margin. These magnetic elongations are largely dominated by intrusive or extrusive volcanics which occurred along the rifted margin of the Ulleung basin formed during the basin opening. The crust in the central Ulleung Basin, surrounded by the magnetic elongations, is possibly oceanic as inferred from the seismic velocity. The oceanic crust can be mapped in the central zone where it widens to 120 km from the southwest toward northeast. Bending of the crustal boundary in the southern part of the Ulleung Basin suggests that the Ulleung Basin has been deformed by a collision of the Phillipine plate into the Japan arc.
