Objectives : This study was conducted to observe the tip of needles which induce poor needle sensation upon injection of herbal acupuncture. Methods : Among the syringes used for herbal acupuncture, needles which cause poor sensation were collected and evaluated by SEM (scanning electron microscope) for changes in structure, existence of impure matters, physical damage, and etc. Results : 1. Various forms contributed to poor needle sensation. Bent needle body, damaged or crooked needle tip, and blunted tip 2. Poor needles could be made during the manufacturing process, during insertion of the needle into the vial(except for the lubricants), and repeated procedures with one needle at different regions. Conclusion : Poor needles induce unnecessary pain and discomfort not only to the patients but also to administrators during injection. Herbal acupuncture is a treatment technique injecting herbal extract on the meridian points, and strict quality check must be enforced to prevent occurrence of poor needles.
In this study was investigated and analyzed of damage characteristics for infrastructures by typhoon that have been many occur. The objective Structures were the road and hydraulic structure. The road structure was included the cut-slopes, retaining walls and bridges. The hydraulic structure is divided with the dike, small-scale dam, reservoir and floodgate. The analysis result of the bridge damage cause is river bottom height increase and passage ability decrease. The principal damage reasons of the cut-slope structure are weakening the ground due to the localized torrential downpour and drainage defective. Also, the principal damage reasons of the small-scale dam, reservoir, dike and the floodgate are continuous collapse of dike beside the floodgate.And we divided a typhoon damage occurrence cause with artificial and natural. As the result of analysis, the many damage occurrence cause will be removed by system improvement and technical development.
The synthesis of Ni-catalyzed multi-walled carbon nanotubes and nanofibers on a catalytic metal substrate, using an ethylene fueled inverse diffusion flame as a heat source, was investigated. When the gas temperature was varied from 1,400K to 900K, approximately, carbon nanotubes with diameters of 20∼60nm were formed on the substrate. In the regions where the gas temperature was higher than 1,400K or lower than 900K, iron nanorods or carbon nanofibers were synthesized, respectively. Based on the quantitative analyses of large amount of SEM and TEM images, the nanotubes formed closer to the flame had a tendency of having larger diameters. HR-TEM images and Raman spectra revealed that carbon nanotubes synthesized had multi-walled structures with some defective graphite layers at the wall. Based on the graphite mode of the Raman spectra, it was believed that the optimal synthesis could be obtained as the substrate was positioned at between 5.5mm and 5.0mm, from the flame axis.
Introduction of CNTs into a metal matrix has been considered to improve the mechanical properties of the metal matrix. However, the binding energy between metals and pristine CNTs wall is known to be so small that the interfacial slip between CNTs and the matrix occurs at a relatively low external stress. The interfacial strength between CNT and metal matrix is thus one of the key factors for successful development of the CNT/metal composites. Defective or functionalized CNT has been considered to enhance the interfacial strength of nanocomposites. In the present work, we design the various realistic hybrid structures of the single wall CNT/Cu complexes and characterize the interaction between single wall CNTs and Cu nano-particle and Cu13 cluster using first principle calculations. The characteristics of functionalized CNTs with various surface functional groups, such as -COOH, -OH, and -O interacting with Cu are investigated. We found that the binding energy can be enhanced by the surface functional group including oxygen since the oxygen atom can mediate and reinforce the interaction between carbon and Cu. These results strongly support the recent experimental work which suggested the oxygen on the interface playing an important role in the excellent mechanical properties of the CNT/Cu composite.
In yeast the key gluconeogenic enzyme, fructose-1,6-bisphosphatase (FBPase), is selectively targeted from the cytosol to the lysosome (vacuole) for degradation when glucose starved cells are replenished with glucose. The pathway for glucose induced FBPase degradation is unknown. To identify the receptor-mediated degradation pathway of FBPase, we investigated the presence of the FBPase receptor on the vacuolar membrane by cell fractionation experiments and binding assay using vid mutant (vacuolar import and degradation), which is defective in the glucose-induced degradation of FBPase. FBPase sedimented in the pellets from vid24-1 mutant after centrifugation at $15,000{\times}g$ for 15 min, suggesting that FBPase is associated with subcellular structures. Cell fractionation experiments revealed that FBPase is preferentially associated with the vacuole, but not with other organelles in vid24-1. FBPase enriched fractions that cofractionated with the vacuole were sensitive to proteinase K digestion, indicating that FBPase is peripherally associated with the vacuole. We developed an assay for the binding of FBPase to the vacuole. The assay revealed that FBPase bound to the vacuole with a Kd of $2.3{\times}10^6M$. The binding was saturable and specific. These results suggest that a receptor for FBPase degradation exists on the vacuolar membrane. It implies the existence of the receptor-mediated degradation pathway of FBPase by the lysosome.

Geometric imperfections may be created during the production process or setting borders of single-layer graphene sheets (SLGSs). Vacancy defects are an instance of geometric imperfection, so investigating the effect of these vacancies on the mechanical properties of single-layer graphene is extremely important. Since very few studies have been conducted on the structure of imperfect graphene (with the vacancy defect) as an anisotropic structure, further study of this defective structure seems imperative. Due to the vacancy defects and for the proper assessment of mechanical properties, the graphene structure should be considered anisotropic in certain states. The present study investigates the effects of site and size of vacancy defects on the mechanical properties of graphene as an anisotropic structure using the lekhnitskii interaction coefficients and Molecular Dynamic approach. The effect of temperature on the severity of the SLGS becoming anisotropic is also investigated in this study. The results reveal that the amount of temperature has a big effect on the severity of the structure getting anisotropic even for a graphene without any defects. The effect of aspect ratio, temperature and also size and site of vacancy defects on the material properties of the graphene are studied in this research work. According to the present study, using material properties of flawless graphene for imperfect structure can lead to inaccurate results.

In this paper, the effects of inevitable out-of-plane defects on the postbuckling behavior of single-layered graphene sheets (SLGSs) under in-plane loadings are investigated based on nonlocal first order shear deformation theory (FSDT) and von-Karman nonlinear model. A generic imperfection function, which takes the form of the products of hyperbolic and trigonometric functions, is employed to model out-of-plane defects as initial geometrical imperfections of SLGSs. Nonlinear equilibrium equations are derived from the principle of virtual work and variational formulation. The postbuckling equilibrium paths of imperfect graphene sheets (GSs) are presented by solving the governing equations via isogeometric analysis (IGA) and Newton-Raphson iterative method. Finally, the sensitivity of the postbuckling behavior of GS to shape, amplitude, extension on the surface, and location of initial imperfection is studied. Results showed that the small scale and initial imperfection effects on the postbuckling behavior of defective SLGS are important and cannot be ignored.
The degradation of wheel tread may result in serious hazards in the railway operation system. Therefore, timely wheel defect diagnosis of in-service trains to avoid tragic events is of particular importance. The focus of this study is to develop a novel wheel defect detection approach based on the relevance vector machine (RVM) which enables online detection of potentially defective wheels with trackside monitoring data acquired under different running-speed conditions. With the dynamic strain responses collected by a trackside monitoring system, the cumulative Fourier amplitudes (CFA) characterizing the effect of individual wheels are extracted to formulate multiple probabilistic regression models (MPRMs) in terms of multi-kernel RVM, which accommodate both variables of vibration frequency and running speed. Compared with the general single-kernel RVM-based model, the proposed multi-kernel MPRM approach bears better local and global representation ability and generalization performance, which are prerequisite for reliable wheel defect detection by means of data acquired under different running-speed conditions. After formulating the MPRMs, we adopt a Bayesian null hypothesis indicator for wheel defect identification and quantification, and the proposed method is demonstrated by utilizing real-world monitoring data acquired by an FBG-based trackside monitoring system deployed on a high-speed trial railway. The results testify the validity of the proposed method for wheel defect detection under different running-speed conditions.
The Wnt/β-catenin signaling plays crucial roles in early development, tissue homeostasis, stem cells, and cancers. Here, we show that RNF152, an E3 ligase localized to lysosomes, acts as a negative regulator of the Wnt/β-catenin pathway during Xenopus early embryogenesis. Overexpression of wild-type (WT) RNF152 inhibited XWnt8-induced stabilization of β-catenin, ectopic expression of target genes, and activity of a Wnt-responsive promoter. Likewise, an E3 ligase-defective RNF152 had repressive effects on the Wnt-dependent gene responses but not its truncation mutant lacking the transmembrane domain. Conversely, knockdown of RNF152 further enhanced the transcriptional responses induced by XWnt8. RNF152 morphants exhibited defects in craniofacial structures and pigmentation. In line with this, the gain-of-RNF152 function interfered with the expression of neural crest (NC) markers, whereas its depletion up-regulated NC formation in the early embryo. Mechanistically, RNF152 inhibits the polymerization of Dishevelled, which is key to Wnt signaling, in an E3 ligase-independent manner. Together, these results suggest that RNF152 controls negatively Wnt/β-catenin signaling to fine-tune its activity for NC formation in Xenopus embryo.
The rapid development of mRNA vaccines has contributed to the management of the current coronavirus disease 2019 (COVID-19) pandemic, suggesting that this technology may be used to manage future outbreaks of infectious diseases. Because the antigens targeted by mRNA vaccines can be easily altered by simply changing the sequence present in the coding region of mRNA structures, it is more appropriate to develop vaccines, especially during rapidly developing outbreaks of infectious diseases. In addition to allowing rapid development, mRNA vaccines have great potential in inducing successful antigen-specific immunity by expressing target antigens in cells and simultaneously triggering immune responses. Indeed, the two COVID-19 mRNA vaccines approved by the U.S. Food and Drug Administration have shown significant efficacy in preventing infections. The ability of mRNAs to produce target proteins that are defective in specific diseases has enabled the development of options to treat intractable diseases. Clinical applications of mRNA vaccines/therapeutics require strategies to safely deliver the RNA molecules into targeted cells. The present review summarizes current knowledge about mRNA vaccines/ therapeutics, their clinical applications, and their delivery strategies.
