Nucleotide Sequence and Characterization of ptsG Gene Encoding Glucose-specific Enzyme II of Phosphotransferase System from Brevibacterium flavum

  • Nucleotide sequence of Brevibacterium flavum ptsG gene capable of complementing Escherichia coli ZSC113 mutations defective to glucose permease activity of phosphotransferase system was completely determined, and the gene product was compared with other glucose-specific enzyme II ($EII^{Glc}$). A ptsG gene of B. flavum consisted of open reading frame of 2,025 nucleotides putatively encoding polypeptide of 675 amino acid residues and TAA stop codon. Deduced amino acid sequence of B. flavum ($EII^{Glc}$) had high homology with ($EIIs^{Glc}$) of Corynebacterium glutamicum, C. efficiens, and B. lactofermentum. Arrangement of structural domains, IIBCA, of B. flanum ($EII^{Glc}$) protein was identical to that of EIIs belonging to glucose-phosphotransferase system.

대두 $\beta$- Amylase의 N-말단 아미노산 배열 (N-Terminal Sequence of Soybean $\beta$- Amylase)

  • The blocked N-terminus and N-terminal sequence of soybean B-amylase were aetermined by analyzing the acidic peptides derived on peptic digestion of the enzyme. The acidic peptides were separated from the digest on a Dowex 50$\times$2 column(1X5cm) and purified by reversed phase-high performance liquid chromatography(RP-HPLC). The major acidic peptide, PEP-1, was a heptapeptlde. The N-terminal 7 amino acid sequence of soybean B-amylase was deduced to be acetyl-Ala-Thf-Ser-Asp-Ser-Asn-Met- from the results of sequence analysis of PEP-1 and amino acid analysis of other acidic peptides.

Molecular Cloning and Expression of a Thermostable Xylose (Glucose) Isomerase Gene, xylA, from Streptomyces chibaensis J-59

  • In the present study, the xylA gene encoding a thermostable xylose (glucose) isomerase was cloned from Streptomyces chibaensis J-59. The open reading frame of xylA (1167 bp) encoded a protein of 388 amino acids with a calculated molecular mass of about 43 kDa. The XylA showed high sequence homology (92% identity) with that of S. olivochromogenes. The xylose (glucose) isomerase was expressed in Escherichia coli and purified. The purified recombinant XylA had an apparent molecular mass of 45 kDa, which corresponds to the molecular mass calculated from the deduced amino acid and that of the purified wild-type enzyme. The N-terminal sequences (14 amino acid residues) of the purified protein revealed that the sequences were identical to that deduced from the DNA sequence of the xylA gene. The optimum temperature of the purified enzyme was $85^{\circ}C$ and the enzyme exhibited a high level of heat stability.

Physical Analysis of nahQ tnpA Genes from Pseudomonas fluorescens

  • Pseudomonas fluorescens SM11 is a naphthalene-degrading strain whose dissimilatory genes are cho-mosomally encoded. We have cloned the 2.9 kb Sal I fragment harboring genes for the naphthalene-degrading upper pathway. The nucleotide sequences were determined to be nahQ, napA, and partial regions of nahE genes. The nahQ encods a protein of 188 amino acid residues with a deduced molec-ular wight of 20.8kDa. The high homology with other proteins suggests that NAhQ may be an active and useful protein whtich gives as selective advantage to naphthalene degradatin. Transposase(TnpA)encodes a polypetide chain with a molecular mass of 41.8kDa consisting of 376 amino acid residues. The deduced anino acid sequence of tnpA revealed 96% idenitity with putative transposase of P. stutzeri OX1,. It was assumed that transposase plays an important role in the evloution of the catabloic-path way in the regulation of nah expression.

Arabidopsis thaliana로부터 지방산 불포화효소 유전자의 분석 (Characterization of a fad3 cDNA Encoding Microsomal Fatty Acid Desaturase from Arabidopsis thaliana)

  • Linoleic acid를 linolenic acid로 전환시키는 지방산불포화효소의 유전자(fad3)를 유채의 fad3 DNA probe를 이용한 plaque hybridization방법으로 $\lambda$ZAPII Arabidopsis thaliana cDNA expression library로부터 분리하였으며 1.8 kb-EcoRI DNA조각을 지니는 lambda clone을 pGEM7으로 subcloning하여 염기서열을 분석하였다. 그 결과로부터의 아미노산서열분석에 의하면 fad3 유전자는 open reading frame이 386개의 아미노산으로 이루어졌으며 44,075 Da의 분자량이 예측되고 있다. 엽록체의 $\omega$-3 지방산불포화효소(fad7)와 endoplasmic reticulum의 지방산불포화효소(fad2)와의 비교시 각각 70%와 58%의 유사성이 나타났다. 특히 82-151의 아미노산서열과 276-333의 아미노산지역은 보존성이 높았으며 이는 불포화지방산효소의 기능에 필수적인 지역으로 보여진다. 한편 대장균을 이용한 IPTG에 의한 Fad3단백질의 유도생성은 대장균의 생육에 독성효과를 나타내는 것으로 나타났다.

Identification and molecular characterization of a rabbit hemorrhagic disease virus variant (KV0801) isolated in Korea

  • Rabbit hemorrhagic disease (RHD) is caused by RHD virus (RHDV) and is one of the most fatal diseases of rabbits. Acute death of rabbits occurred in a farm located in the Gyeonggi province of South Korea. The virus was isolated and confirmed as RHDV based on reverse transcription polymerase chain reaction and hemagglutination assay (HA), and the isolate was designated as KV0801. The nucleotide sequence of the complete VP60 gene of KV0801 was determined and the corresponding amino acid sequence was deduced. Molecular analysis showed that the KV0801 isolate can be classified as a pandemic antigenic variant strain, RHDVa. The VP60 nucleotide sequence and deduced amino acid homology between KV0801 and other Korean isolate, RHF89, which was isolated in 1988, were 92.1 and 94.3%, respectively. The pathogenicity of the KV0801 isolate at an HA titer ranging from 16,384 to 0.16 HA units was evaluated in five-month-old SFP rabbits. The rabbits inoculated with KV0801 isolate containing more than 1.63 HA units died within six days of inoculation. These results suggest that a highly pathogenic RHDVa is circulating in the rabbit populations of Korea.

Molecular Cloning, Purification, and Characterization of a Cold-Adapted Esterase from Photobacterium sp. MA1-3

  • The gene encoding an esterase from Photobacterium sp. MA1-3 was cloned in Escherichia coli using the shotgun method. The amino acid sequence deduced from the nucleotide sequence (948 bp) corresponded to a protein of 315 amino acid residues with a molecular weight of 35 kDa and a pI of 6.06. The deduced protein showed 74% and 68% amino acid sequence identities with the putative esterases from Photobacterium profundum SS9 and Photobacterium damselae, respectively. Absence of a signal peptide indicated that it was a cell-bound protein. Sequence analysis showed that the protein contained the signature G-X-S-X-G included in most serine-esterases and lipases. The MA1-3 esterase was produced in both soluble and insoluble forms when E. coli cells harboring the gene were cultured at $18^{\circ}C$. The enzyme was a serine-esterase and was active against $C_2$, $C_4$, $C_8$ and $C_{10}$ p-nitrophenyl esters. The optimum pH and temperature for enzyme activity were pH 8.0 and $30^{\circ}C$, respectively. Relative activity remained up to 45% even at $5^{\circ}C$ with an activation energy of 7.69 kcal/mol, which indicated that it was a cold-adapted enzyme. Enzyme activity was inhibited by $Cd^{2+}$, $Cu^{2+}$, $Zn^{2+}$, and $Hg^{2+}$ ions.

Molecular Biological Studies on Korean Garlic Viruses

  • To understand the molecular structure and pathogenesis mechanism of Korean garlic viruses, we have isolate cDNA clones for garlic viruses. The partial nucleotide sequences of 24 cDNA clones were determined and that of six clones containing poly (A) tail were compared with those of other plant viruses. One of those clones, V9 has 81.8% similarity in nucleotide sequence and 93.0% in deduced amino acid sequence, respectively, to the coat protein gene for garlic mosaic virus (GMV). Northern blot analysis with the clone V9 demonstrated that the genome of GMV is 7.8 kb long and has poly (A) tail. The anti-coat protein antibody for GMV recognizes 35 kDa polypeptide which could be the coat protein of GMV from infected garlic leaf extract or virus preparation. Clone G7 has about 62% of deduced amino acid sequence identity with the members of potyvirus group. Northern blot analysis with the clone G7 demonstrated that the genome of the potyvirus I garlic is 9.0 kb long and has poly (A) tail. The third clone, S81, shows 42% amino acid identity to the potexvirus. The other clones are under the characterization. To test the possibility of producing garlic virus resistant plant, we have designed a hairpin type ribozyme to cleave V9 RNA at the middle of the coat protein gene. From the cleavage reactions in vitro with two different sizes of RNA substrates, V9SUB (144 nucleotides) and V9 RNA (1,361 nucleotides), the ribozyme can cleave V9 sequence effectively at the predicted site. To study the activity of the ribozyme in vivo, plant transformation is in progress. Further possibilities to produce garlic virus resistant plant will be discussed.

영지버섯 Laccase 유전자의 구리결합부위 I과 IV사이 지역의 클로닝 (Cloning of a Laccase Gene Fragment from Ganoderma lucidum)

  • 백색부후균류 laccase의 아미노 말단과 카복시 말단에 잘 보존된 구리결합부위를 암 호화하는 DNA 염기서열과 상보적인 primer를 이용하여 백색부후균의 일종인 영지버섯 Gandoderma lucidum에서 laccase 유전자 단편을 분리하였다. PCR 로 증폭하여 클로닝한 1.6 Kb DNA 절편의 염기 서열을 결정하여 분석하였다. 이 DNA에는 7개의 인트론이 존재 하였으며 엑손의 염기서열과 이로부터 추정된 아미노산 서열은 Tranmetes villosa laccase(lccl)와 각각 47%, 79% 동일하였다. 기타 다른 백색부후균류의 laccase 아미노산 서 열과는 66~78% 동일하였다.

Thermus aquaticus YT-1의 내열성 프로테아제 aqualysin I의 구조와 특징 (Characterization of aqualysin I structure(a thermophilic alkaline Serine protease) of Thermus aquaticus YT-1)

  • Aqualysin I is an alkaline serine protease which is secretet into the culture medium by Thermus aquaticus YT-1, an extreme thermophile. Aqualysin I was purified, and its partial amino acid sequence was determined. The gene encoding aqualysin I was cloned into E. coli using synthetic oligodeoxyribonucleotides as hybridization probes. The nucleotide sequence of the cloned DNA was determined. The primary structure of aqualysin I, deduced from the nucleotide sequenc, agreed with the determid amino acid sequences, including the $NH_2-$ and COOH terminal sequence of the tryptides derived from aqualysin I. Aqualysin I comprised 281 amino acid residues and its molecular mass was determined to be 28350. On alignment of the whole amino acid sequence, aqualysin I showed high sequence homology with the subtilisin type serine protease, and 43% identity with proteinase K, 37-30% with subtilisins and 34% with thermitase. Extremely high sequence identity was observed in the regions containing the active-site residues, corresponding to Asp32, His64 and Ser221 of subtilisin BPN'. Aqualysin I contains two disulfide bonds, Cys67-Cys99 and Cys163-Cys194, and these disulfide bonds seem to contribute to the heat stability of the enzyme. The determined positions of the twe disulfide bonds of aqualysin I agreed with those predicted previously on the basis of computer graphics of the crystallographic data for subtilisin BPN'. Therefore, these findings sugests that the three-dimensional structure of aqualysin I is similar to that of subtilisin BPN' Aqualysin I is produced as a lage precursor, which contains $NH_2-$ and COOH- terminal portions besides the mature protease sequence.
