토양유기물의 분해속도와 Microbial populaiton의 소장에 관한 연구 (On the Decay Rate of Soil Organic Matter and Changes of Soil Microbial populaiton)

  • The aim of present study is to elucidate the relationship between decay rate of soil organic matter, and the change of soil microbial population under the oak and pine forest soils in Kwang-nung plantation stand. The results obtained are as follows: 1) The correlation coefficient between decay rate and the soil bacteria is 0.84 and fungi 0.93. 2) The distribution of soil microbial population is higher in both F horizon of the oak forest soil, and F and H horizon of the pine forest soil. However, the number of soil microorganisms decreases with the depth in each forest soil. 3) The population of soil microbes is related to moisture content, total nitrogen, available phosphorus, and exchangeable calcium, except organic carbon in fungi. 4) The soil organic matter has been mainly decomposed by fungi, and the size of its population are governed by the factors such as moisture content, organic carbon, total nitrogen, available phosphorus, and exchangeable calcium.

Qualitative Evaluation of Ligninolytic Enzymes in Xylariaceous Fungi

  • Sixty-one strains representing the main genera of wood-decaying xylariaceous fungi (mainly in Daldinia, Hypoxylon, Kretzschmaria, Rosellinia, Penzigia, and Xylaria) were tested for their ability to produce ligninolytic enzymes. The phenol oxidase activity and fungal growth of the xylariaceous fungi on gallic aicid and tannic acid media showed a variation in their ability to degrade lignocellulose. A number of species showed equal 개 betterligninolytic enzyme activities than Coriolus versicolor, a known basidiomycete wood-degrader. A large variation of the enzyme activity was observed by individual strains as well as a substantial variation between the isolates of the same species. The most frequent ligninolytic enzymes were peroxidase and general oxidase. With 19% of the strains tested, peroxidase showed the strongest ligninolytic enzyme activity, while tyrosinase activity was detected only in 7% of the strains. All strains of Kretzschmaria and Rosellinia tested was positive for laccase. Xylariaceous fungi were able to degrade the macromolecule, lignin, using each specific ligninolytic enzyme in the specfic lignin degradation pathway.

신갈나무의 부후에 관여하는 곰팡이 분리 및 동정 (Isolation and Identification of Fungi Associated with Decay of Quercus mongolica)

  • 신갈나무는 국내 전역에 두루 분포되어 있는 경제, 산업적으로 활용 가치가 큰 수종이지만, 변색, 부후 등의 열화에 의한 피해가 심각하다. 이러한 이유로 신갈나무의 부후는 목재로써의 활용에 걸림돌이 되나, 부후 요인에 대한 연구는 미비한 실정이다. 본 연구에서는 신갈나무 부후에 영향하는 요인으로 곰팡이에 주목하였으며, 신갈나무의 부후 부위로부터 곰팡이를 분리, 동정하였다. 또한, 동정 된 곰팡이가 실제로 목재 열화에 영향을 미치는지 확인하기 위해 효소 활성을 평가하고, 곰팡이를 처리한 신갈나무 목재의 질량 손실을 목재 부후 실험을 통해 측정하였다. 신갈나무에서 분리된 곰팡이 5종의 genomic DNA의 ITS region을 이용한 염기서열 분석을 통해, Mucoromycota phylum에 속하는 Mucor circinelloides, Cunninghamella elegans, 그리고 Umbelopsis isabellina 3종과 Ascomycota phylum에 속하는 Ophiostoma piceae와 Aureobasidium melanogenum 2종의 곰팡이가 동정되었다. 이러한 5종의 곰팡이는 목재의 부후와 관련된 cellulase나 laccase와 같은 효소 활성이 있으며, 실제로 신갈나무의 심재와 변재의 중량을 감소시켰다. 특히, cellulase와 laccase 활성을 모두 보유한 O. piceae와 A. melanogenum는 신갈나무의 중량을 각각 6.9%와 1.5% 감소시켰다. 이러한 결과들은 본 연구에서 동정된 5종의 곰팡이가 신갈나무의 열화에 영향한다는 것을 의미하며, 신갈나무에 대한 목재 부후균으로써의 가능성을 시사한다.

Plate counts were made of fungi and bacteria from time to time. 6) Removal of the alcohol-benzol soluble substance stimulates at the beginning of the decay the growth of fungi and also of bacteria. 7) Removal of the water soluble fraction is detrimental to the growth of fungi in particular. 8) The distribution of soil microbial population is higher in both F and H horizon of the fir plantation soil in Kwangnung. However, the number of soil microorganisms decreases with the depth in forest soil.

Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

Four Unrecorded Wood Decay Fungi from Seoul in Korea

  • During the investigation of Korean indigenous fungi from Seoul, three genera-Fuscoporia, Porostereum, and Trametopsis, and four species-Fuscoporia senex, Phlebia acerina, Porostereum spadiceum, and Trametopsis cervina were found. Their morphological characteristics were examined and their identification was confirmed by molecular analysis based on internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA region sequences. These fungi are new to Korea and registered here with descriptions.

Evaluation of Two Species of Soft Wood Decay Resistance for Heat-Treated Wood Using the Catalyst (H2SO4)

  • This study was conducted to evaluate the resistance of heat-treated wood using the catalyst to decay caused by fungi for sapwood and heartwood of two tree species, Korean red pine (Pinus densiflora) and Japanese larch (Larix kaempferi), respectively. Wood samples were immersed for 10 min in sulfuric acid (7.5%) and then heat-treated at $130^{\circ}C$ for 90 min. Fomitopsis palustris, a brown-rot fungus, was used to examine the decay resistance of Korean red pine and Japanese larch wood. Weight and density of wood from the all conditions increased after heat treatment using the catalyst. Weight loss after decay resistance test was also dropped with a heat treatment. The lowest weight loss indicated at heat-treated heartwood of Japanese larch. Heat treatment using the catalyst effectively increased the resistance of wood to decay caused by fungi.

고온아마인유처리에 의한 소나무재의 특성 (Properties of Pinus densiflora Timber Wood by High Temperature Linseed Oil Treatment)

  • 본 실험은 건조되지 않은 목재를 고온의 아마인유에 침지처리함으로써 목재건조와 동시에 발수효과를 얻고자 실시하였다.함수율 90% 이상의 소나무생재를 온도 150℃의 아마인유에서 6시간 침지처리하였을 때 목재함수율은 10%까지 떨어졌으며, 목재 축방향의 깊이에 관계없이 목재단면의 20%에 해당하는 아마인유가 침투하였다. 이때 목재의 강도적 손실은 발생하지 않는 것으로 나타났다. 목재표면의 천공처리는 목재의 내부할열방지에 효과적이었고, 아마인유의 깊은 침투를 유도할 수 있었다. 흡수성 시험 결과 처리재는 무처리재에 비하여 높은 발수효과를 나타냈다. 변색균 및 부후균에 대한 항균효력시험은 처리재 자체로서 균에 대한 독성 기작이 없었으나, 백색부후균 및 갈색부후균에 대해서는 무처리재보다 낮은 중량감소율을 나타내는 등 방부효력의 상승이 있었다.

Comparative Enzyme Production by Fungi from Diverse Lignocellulosic Substrates

  • Fungi commonly encountered on monocotyledonous substrates were evaluated for their in vitro ability to produce enzymes involved in lignocellulose breakdown. Most were capable of structural polysac-charide utilization, but few produced enzymes associated with lignin breakdown. None of the mono-cotyledon-inhabiting fungi produced reactions as strongly as wood decay fungi.
