Studies on the Preparation and the Properties of Chitosan Oligosaccharide and its Derivatives (키토산 올리고사카라이드 및 그 유도체의 제조와 특성에 관한 연구)

  • Chitosan oligosaccharides having aldehyde group at reducing end were prepared by oxidative-deamination reaction of chitosan by using sodium nitrite, and the resulting aldehyde group was reduced to 2, 5-anhydro-D-mannitol group. The obtained chitosan oligosaccharides showed an average degree of polymerization 2~3 by gel permeation chromatography analysis. It was highly soluble in hydrophilic solvents and thermally stable. N, N-diacyl, O-acyl chitosan oligosaccharides were obtained from the reaction between chitosan oligosaccharides and acyl chloried under dimethylaminopyridine. From differential scanning calorimetric measurement, N, N-dilauroyl, O-lauroyl chitosan oligosaccharides showed mesophase region, which was confirmed by polarized microscope as a thermotropic liquid crystalline state. X-ray diffraction pattern revealed that N, N-dilauroyl, O-lauroyl chitosan oligosaccharedes were highly crystalline, whereas chitosan oligosaccharides were not.

Protein-protein Interaction Network Analyses for Elucidating the Roles of LOXL2-delta72 in Esophageal Squamous Cell Carcinoma

  • Lysyl oxidase-like 2 (LOXL2), a member of the lysyl oxidase (LOX) family, is a copper-dependent enzyme that catalyzes oxidative deamination of lysine residues on protein substrates. LOXL2 was found to be overexpressed in esophageal squamous cell carcinoma (ESCC) in our previous research. We later identified a LOXL2 splicing variant LOXL2-delta72 and we overexpressed LOXL2-delta72 and its wild type counterpart in ESCC cells following microarray analyses. First, the differentially expressed genes (DEGs) of LOXL2 and LOXL2-delta72 compared to empty plasmid were applied to generate protein-protein interaction (PPI) sub-networks. Comparison of these two sub-networks showed hundreds of different proteins. To reveal the potential specific roles of LOXL2- delta72 compared to its wild type, the DEGs of LOXL2-delta72 vs LOXL2 were also applied to construct a PPI sub-network which was annotated by Gene Ontology. The functional annotation map indicated the third PPI sub-network involved hundreds of GO terms, such as "cell cycle arrest", "G1/S transition of mitotic cell cycle", "interphase", "cell-matrix adhesion" and "cell-substrate adhesion", as well as significant "immunity" related terms, such as "innate immune response", "regulation of defense response" and "Toll signaling pathway". These results provide important clues for experimental identification of the specific biological roles and molecular mechanisms of LOXL2-delta72. This study also provided a work flow to test the different roles of a splicing variant with high-throughput data.

Rapid Detection Methods for Biogenic Amines in Foods (식품 내 바이오제닉아민 신속검출기술 개발 동향)

  • Biogenic amines have been used as chemical indicators to estimate bacterial spoilage of foods, particularly fish and fish products, cheese, and fermented foods. So far many chromatography methods have been developed to detect biogenic amines in foods. Although these instrumental analyses exhibit good sensitivity, they cannot be used as rapid detection methods due to the chemical treatment of the samples and the time-consuming process involved. For the rapid and simple detection of biogenic amines, enzyme linked immunosorbent assay kits are commercially available. In addition, analytical systems with enzyme-based amperometric biosensor detection have been increasingly developed. The biosensors used to detect the biogenic amines are based on the action of either amine oxidases or amine dehydrogenases that catalyzes the oxidative deamination of biogenic amines to the corresponding aldehydes and ammonia. This review mainly focused on the principle, development, and applications of the detection methods for rapid detection of biogenic amines in foods.

Metabolism of Pyrimidine Deoxyribonucleosides and Heat-resistivity of CdR-aminohydrolase in the Mouse Small Intestine (생쥐 小腸에서의 Pyrimidine Deoxyribonucleoside 代謝와 CdR-aminohydrolase의 熱抵抗性)

  • The metabolism of CdR-2-$^14 C$ and UdR-2-$^14 C$ in mouse small intestine has been studied in connection with the effect of heat treatment on the enzymes concerned in vitro. CdR-2-$^14 C$ is deaminated reaidly by CdR-aminohydrolase at nucleoside level and then degraded into U by the action of nucleosidase which is quite resistant to cleave N-pentose bond of cytosine nucleosides, CdR and CR. High inactivation temperature of $80^\\circC$ was observed for CdR-aminohydrolase, while nucleosidase has an inactivation temperature of $60^\\circC$. CdR-aminohydrolases in various tissues of mouse were inactivated at $80^\\circC$, but not one in tissues of rabbit. It might be assumed that there are correlations between order specificity and inactivation temperature of the enzyme. A physiological significance of the appearance of CdR-aminohydrolase in differentiated tissues of mammals possibly be regarded as a main function in catabolic pathways.

RNA Editing Enzyme ADAR1 Suppresses the Mobility of Cancer Cells via ARPIN

  • Deamination of adenine or cytosine in RNA, called RNA editing, is a constitutively active and common modification. The primary role of RNA editing is tagging RNA right after its synthesis so that the endogenous RNA is recognized as self and distinguished from exogenous RNA, such as viral RNA. In addition to this primary function, the direct or indirect effects on gene expression can be utilized in cancer where a high level of RNA editing activity persists. This report identified actin-related protein 2/3 complex inhibitor (ARPIN) as a target of ADAR1 in breast cancer cells. Our comparative RNA sequencing analysis in MCF7 cells revealed that the expression of ARPIN was decreased upon ADAR1 depletion with altered editing on its 3'UTR. However, the expression changes of ARPIN were not dependent on 3'UTR editing but relied on three microRNAs acting on ARPIN. As a result, we found that the migration and invasion of cancer cells were profoundly increased by ADAR1 depletion, and this cellular phenotype was reversed by the exogenous ARPIN expression. Altogether, our data suggest that ADAR1 suppresses breast cancer cell mobility via the upregulation of ARPIN.

Frequency of Meals and Hyperlipogenesis of Rat (쥐의 급식회수(給食回數)와 체지방과잉합성(體脂肪過剩合成))

  • This experiment was performed to investigate the effect of the frequency of meals on the metatolism and the body composition of rats when equal amount of purified diet was ingested. Thirty approximately days old rats weighing 290 g and thirty-two about 40 days old rats weighing 180 g were employed for the period of 34 days. Rats fed ad libitum (10 to 15 meals per day) and two-meal per day were pair-fed and equal amount of diet was fed to each rat in pair. The experimental results obtained are summarized as follows: 1. Frequency of meal did not exert any effect on the body weight gain. However, rats fed two-meal per· day gained significantly (p <0.005) more fat and energy than ad libitum group. The rate of gain of protein in ad libitum group was higher than that of two-meal group. No difference was observed for the mineral deposition of rat body. 2. From the preperation of rat liver it was found that the activity of glucose-6-phosphate dehydrogenase was much higher for the rats fed two-meals per day than those fed ad libitum. Therefore, it is suggested that the metabolic pathway of carbohydrate for two-meal group has been shifted from glycolysis to Hexose Monophosphate Shunt and produced more NADPH which would be the essential cofactor of fatty acids synthesis. 3. The rate of excretion of urinary nitrogen for two-meal group was significantly (p<0.005) higher than that of ad libitum group. It is apparent that considerable amount of over-loaded amino acids by feeding two-big-meal daily· could not be used for the protein biosynthesis all at once and excreted following deamination through urine. The residual carbon chain could be served as a precursor of fatty acids synthesis. 4. The heat production rate of rats fed two-meal group was significantly (p<0.005) lower than that of ad libitum group. It seems possible that the activity of thyroid gland (and consequently BMR) can be depressed by the frequency of meal.

Protective Effect of Glycoprotein Isolated from Cudrania tricuspidata on Liver in $CCl_4$-treated A/J Mice (생쥐에 있어서 꾸지뽕 당단백질의 간보호 효과)

  • This study aimed to determine whether or not glycoprotein isolated from Cudrania tricuspidata Bureau fruit(CTB glycoprotein) exerts a hepatoprotective effect on liver injury induced by the administration of carbon tetrachloride($CCl_4$, 1.0mL/kg) to A/J mice. Following the administration of CTB glycoprotein(0-20mg/kg), the activities of antioxidant enzymes (superoxide dismutase(SOD), catalase(CAT), and glutathione peroxidase(GPx)), and the quantities of measured thiobarbituric acid reactive substances(TBARS), lactate dehydrogenase(LDH), and nitric oxide(NO) were evaluated from the murine liver tissues and plasma. Additionally, the activity of nuclear factor-kappa B(NF-${\kappa}B$) was assessed after pretreatment with $CCl_4$. When the mice were treated with $CCl_4$ alone, the activities of antioxidative enzymes reduced but amounts of TBARS, LDH, and NO increased. However, the results of treatment with CTB glycoprotein(10 and 20 mg/kg) revealed significantly increased activities of antioxidant enzymes(SOD, CAT, and GPx), as compared with $CCl_4$ alone. On the other hand, the result showed significant diminutions of the quantities of TBARS, LDH, and NO after treatment with CTB glycoprotein(10 and 20 mg/kg), as compared to $CCl_4$ alone. The activity of NF-${\kappa}B$ also declined after pretreatment with CTB glycoprotein, as compared with $CCl_4$ treatment alone. Thus, it is suggested that the CTB glycoprotein exerts a protective effect against $CCl_4$-induced liver injury in A/J mice.

Biogeochemical Study of Dissolved Organic and Inorganic Compounds under Oxic/Anoxic Environment in Lake Shihwa (시화호 산화-환원 환경하의 용존 유, 무기 화합물의 생지화학적 연구)

  • Lake Shihwa, artificially constructed since 1988, shows a typical two-layered system depending on strong haline density stratification. Sill of the water gate at 6 m depth greatly restricts physical mixing with outer seawater and circulation in the lake, and contributes to the enhancement of anoxic environment in the deeper layer. With this enclosed physical environment, Lake Shihwa receives enormous amounts of organics, ammonia, and other pollutants from the neighboring municipal and industrial complexes through six major streams, thus developing biogeochemical differentiation of anoxic to suboxic environment in the high saline bottom water and highly eutrophicated brackish surface water. This study investigated vertical structures, biogeochemical behaviors and processes of various organic and inorganic compounds around oxic-anoxic interface. Nitrite and nitrate rapidly decreased below the pycnocline where about $1{\times}10^8$ tons of hypoxic bottom water exist. In this bottom layer, ammonium ranged from 75 to 360 ${\mu}M$ mainly resulting from deamination of dissolved organic nitrogen and ammonification of precipitated organic particles. Despite large amounts of surface water discharge and dilution by outer seawater inflow about $3{\times}10^8$ tons from April to August, 1996, bottom layer did not show any improvement of water quality and maintained highly reduced environment. The main reason seems to be imbalance between ineffectiveness of dilution due to shallow depth and large surface area, overloaded POC influx from the eutrophicated surface biological activity, and poor replenishment of oxygen in this artificial lake system. Therefore, as long as current salinity dependent two-layered system maintains with its physical limitations, any improvement of water quality cannot be foreseen in Lake Shihwa.

Changes in Components and Peptides during Fermentation of Cheonggookjang (청국장 발효시의 성분 변화 및 펩티드의 생성)

  • We analyzed content and peptides in order to investigate the productivity from Cheonggookjang(fast-fermented soybean paste), fermenting it for 180 hours at $40^{\circ}C$. Results showed that pH was 7.07 at the start and became 7.41 in 24 hours, it eventually increased to 8.63 after 180 hours. Acidity was 0.2 in 12 hours, 0.5 in 12 hours, and then remained on 0.1 thereafter. Total sugar was 1.54 mg/$m\ell$ at the start, but it gradually decreased to 0.76 mg/$m\ell$ after the lapse of 48 hours, and 1.0 mg/$m\ell$ in 120 hours, and finally 0.8 mg/$m\ell$ in 180 hours. Reducing sugar was 0.14 mg/$m\ell$ at the start, and 0.88 mg/$m\ell$ after the lapse of 24 hours, 0.64 mg/$m\ell$ in 48 hours, 0.26 mg/$m\ell$ in 72 hours, and showed no definite change untill 180 hours. The amount of free amino acid was $0.19\;{\mu}M/\ell$ at the start, and $4.88\;{\mu}M/\ell$ after the lapse of 72 hours, $4.5\;{\mu}M/\ell$ in 120 hours, and then it rapidly decreased to $0.23\;{\mu}M/\ell$ after180 hours. Absorbance of soluble protein and peptide at 280 nm was 12.4 in 48 hours, 31.12 in 120 hours, and 31.12 in 180 hours. HPLC revealed that in the fermentation process, large molecular proteins are hydrolyzed into small peptides and amino acids, and after the lapse of 48 hours the pattern became almost the same. The protease activity of Cheonggookjang was 0.011 unit/$m\ell$ after the lapse of 36 hours and then it decreased. The result shows as Cheonggookjang started its deamination of amino acid in 100 hours, it is desirable to produce peptide within 100 hours of its fermentation.
