Cultural Characteristics of Rhizoctonia cerealis Isolated from Diseased Wheat Fields and Evaluation of the Resistance of Korean Winter Cereal Crops (밀 잎집눈무늬병원균(Rhizoctonia cerealis)의 배양적 특성과 국내육성 맥류 품종의 저항성 평가)

  • It was identified as a sharp eyespot (Rhizoctonia cerealis) that the isolates from abnormal symptoms in wheat that showed yellowing leaves, necrotic spot on stem base and dead tillers. These isolates have slower growth property and fewer mycelia than Rhizoctonia solani AG-1(1A) (KACC 40106). They showed binuclear cell, same media cultural and DNA characteristics to R. cerealis. They caused same symptoms on leaves and stem base appeared in artificial inoculation test, comparing to diseased wheat fields and also affect to maturing of kernels. They have optimal growth temperature and acidity on the artificial media as $20{\sim}25^{\circ}C$ and pH 5~7, respectively. In the investigation of varietal resistance of Korean winter cereal crops to sharp eyespot, there was no resistant in wheat cultivars that all materials infected over 20% diseased ratio. 12 cultivars including 'Anbaekmil', however, considered to moderate resistance with 20 to 30% infection ratio. The others crops using in feeding, whole crop barley, oat, rye and triticale were resistant below 15% diseased degree except the rye that showed over 50% infection rate. It was the first evaluation to sharp eyespot resistance for the Korean feeding crop cultivars. Most tested Korean barley cultivars for malting and food were moderate and susceptible to the sharp eyespot. Only 3 hulled barley, 'Tapgolbori', 'Albori' and 'Seodunchalbori', showed resistance with less than 10% diseased ratio. All tested naked barley cultivars showed susceptible response to the disease.

  • The six barnyardgrass [Echinochloa oryzicola (vasing.) Vasing.] and seventeen barnyardgrass [Echinochloa crus-gadli (L.) Beauv.] accessions, which were collected national widely in 1986 and selected two times through 1987. To study different growth response due to herbicide, pyrazolate, bifenox, quinclorac, the experiment was conducted with herbicide agar culture without nutrients, 1. Albinistic discoloration due to pyrazolate was more sensitive in E, crus-galli than E. oryzicola and among barnyardgrass accessions, Kumi, Ichon-A, Ichon-B, Boun-B and Kwangju-B were rather susceptible than Gyongju, Ansong, Boun-A, Jongju-A and Kwangju-A. 2. Twisting and growth retardation due to bifenox was less sensitive in E. oryzicola with less intra-specific variations than in E. crus-galli. Among E. crus-galli accessions, Boun-B, Ansong, Ichon-A, Ichon-B, Wonju and Kwangju-B were particulary susceptible, and Jinyang, Jongju-B, Jongju-A, Daejon, Kurye and Kwangju-A were tolerant as much as E. oryzicola. 3. Growth retardation and withering to dead due to quinclorac was more sensitive in E. oryzicola with less intra-specific variations than in E. crus-galli. Among accessions of E. curs-galli, Boun-A, Iri, Jongju-A, Jongju-B, Kwangju-A and Kwangju-B were rather similar suseptible to E, oryzicola than kimhae, Gyongju, Kumi, Wonju, Ichon-A, Ichon-B and Ansong. 4. Most accession of E. oryzicola was tolerant to both pyrazolate and bifenox, while susceptible to quinclorac. Among other accessions of E. crus-galli, Kurye, Kimhae, and Daejon were tolerant to all experimented herbicides, and Iri, Jongju-A, Jongju-B, and Kwangju-A were only tolerant to both pyrazolate and bifenox, while Kumi, Wonju, Ichon-A, Ichon-B, Boun-B and Kwangju-B were only tolerant to quinclorac.

Selection of Legume Crop by Intercropping with Sorghum $\times$ Sudangrass Hybrid (Sorghum $\times$ Sudangrass 교잡종과 간작에 의한 청예 두과작물 선발)

  • A field experiment was conducted in Chungju and Jungwon to evaluate growth characteristics, dry matter yield, protein yield and palatability of intercropping comparing with monocropping forage crops by the use of sorghum $\times$ sudangrass hybrid(S.S.H.), five forage soybeans and two forage cowpeas of superior to selected experiment of varieties. All eight treatment plots were replicated three times and cutting date were cut July 6 and September 9. 1. The mean leaf number of S.S.H. in intercropping T2 treatment showed high compared to T1 treatment in all treatment except for Hwangkeum treatment, but T3 treatment was lower than the other treatments. In the leaf number of soybean, Jangbaek treatment was the highest as 47 leaves per plant, cowpea treatments showed high above 50 leaves. The stem diameter of S.S.H. in Jangyeob treatment of T2 treatment was 9.6mm. In legume, cowpea of T3 treatment was high as ranged from 7.9mm to 8.2mm. In stem hardiness of S.S.H., TI treatment was the lower than the other treatments, while Jangbaek treatment of T2 treatment was the highest as $2.0kg/cm^2$. In legume crops, Jangbaek and Baekun treatment were the highest as $1.6kg/cm^2$, but T3 treatment was very low as $0.3~0.6kg/cm^2$. 2. In the mean leaf ratio of S.S.H., T1 treatment was higher than T2 and T3 treatment as 34.9%. In the legume crops, Togyu treatment was the highest as 40.9%. In the mean tiller number of S.S.H., T2 treatment showed high compared to TI treatment, but T3 treatment of was rather decrease than The tiller number of S.S.H. in Jangyeob treatment of T2 treatment was the highest as 4.6 per plant. The dead stubble of S.S.H. was high in order to T3>T1>T2 treatment>, T3 treatment was highly about 2 times compared to T2 treatment. 3. S.S.H. as T3 treatment was resistant to lodging, but T3 and T1 treatment showed highly lodging. In the legums, T3 treatment was resistant to lodging, but T2 treatment showed highly lodging. In the S.S.H. of T2 treatment, the leaf of summer depression was not occurrence, but T3 treatment was higher than the other treatments. 4. The palatability of domestic animals was high in order of T2>Tl>T3, especially Togyu treatment of T2 treatment was the highest at the holstein and deer, and Jangyeob treatment of T2 treatment was the highest in the Korean native cattle. But T3 treatment was lower than other treatments in the holstein and Korean native cattle. 5. Fresh yield of monocropping(T1) was the highest as 94, 650kg/ha, while dry matter yield in Jangyeob treatment of T2 treatment was the highest as 15, 575/ha But fresh yield and dry matter yield of T3 treatment were the lowest(P<0.05). 6. Protein yield in T2 treatment was high, especially Jangyeob treatment of T2 treatment was the highest as 1, 605.5 kg/ha T3 treatment was low in spite of high protein content, because dry matter yield was lower than the other treatments. In conclusion, among T1, T2 and T3 treatment, Jangyeob, Togyu and Baekun treatments of T2 treatment showed optimum varieties for intercropping with S.S.H., because they showed high Dry matter yield, protein yield and palatability.

Health Assessment by Hematological Studies and Blood chemistries in cultured carps (혈액성상(血液性狀)에 따르는 양식어(養殖魚)의 건강진단(健康診斷))

  • The purpose of this study was to obtain the fundamental data for health assessment of cultured fishes. A series of hematological studies and blood chemistries were made on cultured israeli carps from May, 1988 to May, 1989. The results of this study were as followings : 1. The blood constituents of healthy carps were Hct $32{\pm}3.4%$, Hb $8.3{\pm}0.9g/l$, RBCs $132{\pm}27.6({\times}10^4cm^3)$), Total protein $2.7{\pm}0.7g/dl$, GOT $143{\pm}19.5U$, GPT $50{\pm}14.2U$, Total glucose $70{\pm}12.6mg/dl$ and BUN $7.5{\pm}1.9mg/dl$, etc. 2. The blood constituents by change of water temperature with the control of $23^{\circ}C$ showed the decrease in Hct and RBCs at $18^{\circ}C$, and the increase in Hct, RBCs and glucose at $28^{\circ}C$. 3. The blood constituents by change of dissolved oxigen with the control of 4.5ppm showed the increase in Hct and RBCs at 3ppm, and the decrease in Hct and RBCs at 7ppm. 4. In the case of intramuscular injection of Streptococcus sp. with $10^6cells$/fish infection dose, there showed marked decrease in Hct, RBCs, glucose and BUN with inflammatory reaction, and the fishes were recovered in 16 days. 5. In the case on intraperitoneal injection of Streptococcus sp. with $10^4cells$/fish infection dose, there showed decrease in Hct, RBCs and BUN with inflammatory reaction, but there were no dead cases. 6. In the case of intramuscular injection of Edwardsilla tarda with $1.2{\times}10^8cells$/fish infection dose, there showed decrease in Hct and RBCs, and increase in TCHO, GOT, GPT and BUN with marked inflammatory reaction, and 60% of inoculated fishes were died within 3 or 4 days.

The Evaluation of Radiation Therapy and Combined-modality Therapy for Non-small-cell Lung Cancer in Elderly (고령의 비소세포성폐암 환자의 방사선 및 병용치료에 대한 효과 평가)

  • [ $\underline{Purpose}$ ]: To compare radiation therapy alone to combined modality therapy about survival rate and tolerance of elderly patients ($70=or{\geq}$) with non-small-cell lung cancer (NSCLC). $\underline{Materials\;and\;Methods}$: Between 1998 and 2002, 57 patients given radiation therapy due to NSCLC (Stage III) were analysed retrospectively. Radiation therapy alone (RT), concurrent chemoradiation (CRT), and sequential chemoradiation (SCRT) was done to 33, 16 and 8 patients, respectively. Patients' median age was 74 (range $70{\sim}85$). Male and female are 51 patients and 6 patients, respectively. 23 patients were stage IIIa and 34 were stage IIIb. Patients' characteristic distribution of RT and CRT was not significantly different except mass size that RT has a bigger than CRT. The fraction size of radiation therapy was 1.8 Gy in CRT and $1.8{\sim}3\;Gy$ in other groups. Total radiation dose was $51{\sim}63\;Gy$ according to the fraction size. If the prescribed total radiation dose was successfully irradiated, we stated that it was completion of radiation therapy. $\underline{Results}$: 52 patients were dead. Median period of radiation therapy was as follow: RT, 35 days, CRT, 60.5 days and SCRT, 35 days. Overall median survival time (MST) was 10.1 months. The 1 yr- and 2 yr-overall survival rate was 39.8% and 17.6%, respectively. MST of RT, CRT and SCRT was 8.9, 8.2 and 11.7 months, respectively. The 1 yr survival rate of RT, CRT and SCRT was 38.4%, 37.5% and 50% (not significant). Patients given incomplete radiation therapy were 12 (RT, 5 CRT, 6 SCRT, 1). N stage (p=0.081) and the difference of treatment methods (p=0.079) were the factors affecting incompletion of radiation therapy, but it was not significant. In case of combined-agents chemotherapy, 4 of 8 ceased radiation therapy. T stage ($T{\geq}3$), mass size (${\geq}5\;cm$), Karnofsky performance scale (${\leq}70$) and completion of radiation therapy were the prognostic factors in uni- and multi-variate analysis. $\underline{Conclusion}$: In elderly patients with NSCLC, radiation therapy alone was a treatment method with similar survival period compared with other methods. Generally, patients given radiation therapy alone was tolerable to a treatment. Before planning concurrent chemoirradiation in elderly patients with NSCLC, physicians pay attention to a selection of patients and chemotherapy agents considering general condition and toxicity.

Studios on the cell-mediated immunity in experimental Nnegleria spp. infections (실험적 Naegleria 감염에 있어서 세포매개성 면역에 관한 연구)

  • Observations were made on the differences in cell-mediated immune responses in the mice infected with strongly pathogenic Naegleria fewleyi ITMAP 359, weakly pathogenic Naegzeria jadini 0400, or non.pathogenic Naegleria gruberi EGB, respectively. Variations in cell-mediated responses and changes in antibody titers according to the duration after infection wore noted. Infections were done by dropping $5{\;}{\mu}l$ saline suspension containing $10{\times}10^4$ trophozoites cultured Bxenically in the CGVS medium into the right nasal cavity of ICR mice aging about 6~7 weeks, under the anesthesia by intraperitoneal injection of'secobarbital. Following infection, delayed type hypersensitivity(DTH) iesponses in the footpad and blastogenic responses of the mouse spleen cells using [$^3H$]-thymidine were observed on the day 1, 4, 7, 10 and 14 after infection. For the preparation of amoeba Iysates, each of cultured trophosoites were homogenized with an ultrasonicator, and centrifugated at 20,000 g. The supernatants of amoeba Iysates were used as the mitogen'and antigen for ELISA. Confanavalin A(Con. A) and lipopolysaccharide(LPS) were also used as mitogens in the blastogenic response. 1. The mice infected with N, fowleri showed the mortality rate of 75.7%. The rate was 6.2% for the N. jadini infected group, while no dead mouse was observed for N. gruberi infections. 2. In regard to DTH responses in the H. fewleri infected mice, the level increased in com- parison to the control group but declined after 7 days. An increase was also noted for the JV. jadini group after 1 day, but gradual decreases were observed through the infection period. In addition, no difference was noted between the N. gruberi infected and control groups. 3. Concerning the blastogenic response of the splenocytes, it increased after 10 days in the experimental group of N, fcwleri infection, but the differences ware not statistically significant compared with control group. It was evident that N. jadini group was not different from control group either, while there was a tendency of decrease in SV. gruberi infected group. In regard to the blastogenic response of the splenocytes by LPS, it was found that the N. fowlgri, N. jadini and N. gruberi infected groups had no differences from the control group. 4. The serum antibody titer of N. fcwleri and N. jadini infected mice increased from the day 7 and 14 after infection respectively, while the N. gruberi infected mice showed no increase. In summary of the results, it was observed that there were differences in the cell-mediated immune responses and serum antibody titers in the mice infected with strongly pathogenic JV. fowleri, weakly pathogenic N. jadini, or non.pathogenic N. gruberi, respectively.

The effects of selenium on fetal growth and development in CD-1 mice exposed with mercury for the gestation period (임신 중 수은을 섭취한 CD-1 마우스 태아의 성장발육과 기형발생에 미친 셀레늄의 효과)

  • Teratogenic and embryotoxic effects of mercury have been reported, however, there is little information about possible antidotes against mercury exposure during gestation. In order to evaluate therapeutic effects of selenium as an antidote against mercury poisoning, pregnant CD-1 mice were exposed to methylmercury chloride(20ppm) through the drinking water with treatment of sodium selenite (1.0mg, 2.0mg or 3.0mg/kg b.w., subcutaneously) or BAL(5.0mg/kg b.w., subcutaneously) under the single or combination base as the therapeutic agents from day 6 to 15 of gestation. Fetal growth parameters such as body weight and crown-rump length in the mice exposed to mercury, were reduced as was placental weight compared to those in the control. Treatment of selenium(alone, combination with BAL) reduced the harmful effects induced by mercury on the fetal growth parameters even though no specific relationship between dose and therapeutic effect. The incidence of dead fetuses/resorptions and malformed fetuses(especially cleft palate) was also increased in the mercury only treated group. Selenium treatment demonostrated reduced the incidence of abnormal fetuses under the exposure of mercury. Relative maternal organ weights(liver, kidney, spleen) were increased significantly but relative brain weight was decreased as evidenced by decreased in the mercury treated mice compared to that in the control. A subtle indication of maternal mercury toxicity evidenced by changes of relative maternal organ weights, decreased water and feed consumption were also prevented efficiently by selenium treatment. The present study suggests that methylmercuric chloride is embrytoxic and teratogenic in CD-1 mice when exposured during organogenesis and that selenium administration may have therapeutic application for the treatment of mercury poisoning although more applicable study in human should be performed with caution in the future.

Effects of Various Physical and Chemical Factors on the Death of Trouble Seaweed Ulva australis (구멍갈파래(Ulva australis) 해조류 사멸에 미치는 여러 물리화학적 요인들의 영향)

  • Green tides, which was mainly caused by Ulva spp., have been increasing in severity and frequency globally, and have negatively affected on marine ecosystems. This study was conducted to investigate effects of various physical and chemical factors on the death of Ulva australis (ULAUS) and to consider a practical measures useful for alleviating Ulva bloom. Soaking of ULAUS thalli in pure water for 8 hr didn't induce a death, but incubation in 1.0-1.5% salinity for 7 d inhibited sporulation by about 70%. Desiccation gave rise to a serious damage when more than 40-50% of initial fresh weight was lost. ULAUS growth was sensitive to temperature and seriously inhibited from more than $30^{\circ}C$. At $35^{\circ}C$, $40^{\circ}C$, $45^{\circ}C$ and $50^{\circ}C$, treatment time required for 90-95% death of ULAUS thalli was 1 d, 10 min, 30 sec, and 1 sec, repectively. ULAUS growth was seriously inhibited at lower than pH 6.0 and completely dead at pH 4.0. Several compounds for ULAUS control was selected and the chemcals causing a rapid death were oxidants such as hydrogen peroxide and sodium percarbonate. Taken together, our results suggest that low salinities, dryness, pH, high temp. and compounds could be selected for Ulva bloom control, and high temperature and compounds seems to be useful for a development of practical control methods.

Performance Measurements of Positron Emission Tomography: An Investigation Using General Electric $Advance^{TM}$ (양전자방출단층촬영기의 표준 성능평가 방법: GE $Advance^{TM}$에 적용한 예)

  • A series of performance measurements of positron emission tomography (PET) were performed following the recommendations of the Computer and Instrumentation Council of the Society of Nuclear Medicine and the National Electrical Manufacturers Association. We investigated the performance of the General Electric $Advance^{TM}$ PET. The measurements include the basic intrinsic tests of spatial resolution, scatter fraction, sensitivity, and count rate losses and randoms. They also include the tests of the accuracy of corrections: count rate linearity correction, uniformity correction, scatter correction and attenuation correction. GE $Advance^{TM}$ PET has bismuth germanate oxide crystals (4.0mm transaxial ${\times}$ 8.1mm axial ${\times}$ 30.0mm radial) in 18 rings, which form 35 imaging planes spaced by 4.25mm. The system has retractable tungsten septa 1mm thick and 12cm long. Transaxial resolution was 4.92mm FWHM in 2D and 5.14mm FWHM in 3D at the center. Average axial resolution in 2D decreased from 3.91mm FWHM at the center to 6.49mm FWHM at R=20cm. Average scatter fraction of direct and cross slices was 9.57%. Dead-time losses of 50% corresponded to a radioactivity concentration of $4.86{\mu}Ci/cc$ and a true count rate of 519 kcps in 2D. The accuracy of count rate linearity correction was 1.84% at the activity of $4.50{\mu}Ci/cc$. Non-uniformity was 2.06% in 2D and 2.93% in 3D. Remnant errors after scatter correction were 0.55% in 2D and 4.12% in 3D. The errors of attenuation correction were 6.21% (air), 0.20% (water), -6.32% (teflon) in 2D and 5.00% (air), 6.94% (water), 3.01% (teflon) in 3D. The results indicate the performance of GE $Advance^{TM}$ PET scanner to be well suited for clinical and research applications.

Characterization of Catechol l,2-Dioxygenase Purified from the Benzoate Degrading Bacterium, Pseudomonas sp. NFQ-l Isolated from Dead Coal Pit Areas (폐광지역에서 분리한 Benzoate 분해세균 Pseudomonas sp. NEQ-1에서 정제된 Catechol 1,2-Dioxygenase의 특성)

  • Our previous research has demonstrated that the bacterium, Pseudomonas sp. NFQ-l capable of utilizing quin­oline (2,3-benzopyridine) as the sole source of carbon, nitrogen, and energy was isolated and characterized [Yoon et ai. (2003) Kor. J. Biotechnol. Bioeng. 18(3):174-179]. In this study, we have found that Pseudomonas sp. NFQ-l could degrade quinoline as well as benzoate, and extended this work to characterize the catechol 1,2­dioxygenase (C1,2O) purified from the bacterium cultured in benzoate media. Initially, C1,2O has been purified by ammonium sulfate precipitation, gel permeation chromatography, and Source 15Q. After Source 15Q, puri­fication fold was increased to approximately 14.21 unit/mg. Molecular weight of C1,2O was about 33 kDa. Physicochemical characteristics (e.g., substrate specificity, Km, Vmax, pH, temperature and effect of inhibitors) of purified C1,2O were examined. C1,2O demonstrated the activity for catechol, 4-methylcatechol and 3-meth­ylcatechol as a substrate, respectively. The Km and Vmax value of C1,2O for catechol was 38.54 ${\mu}M$ and $25.10\;{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}.$ The optimal temperature of C1,2O was $30^{\circ}C$ and the optimal pH was approximately 8.5. Metal ions such as $Ag^+,\;Hg^+,\;Ca^{2+},\;and\;Cu^{2+}$ show the inhibitory effect on the activity of C1,2O. N-terminal amino sequence of C1,2O was analyzed as ^1TVKISQSASIQKFFEEA^{17}.$ In this work, we found that the amino acid sequence of NFQ-l showed the sequence homology of 82, 71, 59 and $53\%$ compared with C1,2O from Pseudomonas aeruginosa PA0l, Pseudomonas arvilla C-1., P. putida KT2440 and Pseudomonas sp. CA10, respectively.
