Effect of Priming and Preimbibition on Germination of Zoysiagrass Seeds (Priming과 침지처리가 들잔디의 종자발아에 미치는 영향)

  • Studies were conducted to evaluate the effect of priming and preimbibition on gerraination of unscarified and KOH-scarified zoysiagrass (Zoysia japonica Steud.) seeds, respectively. Germi-nation percentages at 35˚C of seeds primed in $KNO_3$ and Ca($NO_3$)$_2$ were significantly higher than that of untreated seeds. The optimum conditions for priming unscarified seeds were in aerated $KNO_3$ solution at O4 M for 4 days at 20˚C. No additional benefit of $GA_3$ on increasing germination was not found in seed priming with solution of $KNO_3$ + $GA_3$ or in seeds preimbibed with $GA_3$ after priming. Scarified seeds preimbibed with distilled water(DW) for 4 days at 20˚C germinated higher and with fewer days to 50% of total germination(T 50) than untreated seeds. Preimbibition with $GA_3$ solution at 50 ppm for 4 days at 5, 10, 15 and 20˚C enabled up to 97% of seeds to germinate and reduced days to T 50 about 1.1 to 1.2 days at 35˚C Especially, seeds preimbibed with $GA^2$˚$_3_2_1$$\times$ had greatly higher germination and lower T 50 than untreated or DW-preimbibibed seeds at 30˚C, which is inadequate to germination in general. Preimbibed seed performance was unchanged after storage at room temperature for 50 days. Seeds preimbibed with GA $_3$ had higher rate of seedling emergence and faster growth of shoot than untreated seeds after sowing at 30˚C in growing medium in growth chamber.

  • Growth properties of carrot hairy roots transformed by Agrobacterium rhizogenes were compared in flask and bioreactor. Oxygen transfer coefficient KLa was measured during the cultivation in bioreactor. In flask cultures, initially sucrose 30g/l was nearly exhausted after 20days. pH was dropped from initially 5.8 to 4.79 after 4 days, but it is stable after that time. Finally, after 28 days, hairy roots were grown about twelve times. In view of the results studied optimum conditions, hairy roots were maintained high growth rates in sucrose 50g/l, pH 5.8, total nitrogen 60mM. Also in bioreactor cultures, fixed stainless sieve in bottom and aerated 0.31 vvm, the results of cultivation by the use of sucrose 50g/l had grown about twenty-eight times and pH variations were liked in flask. As a results, growth rate of 1.756g fresh weight/day/g inoculum in bioreactor were higher about three times than 0.57g fresh weight/day/g inoculum in flask culture. KLa values were showed a tendency to decrease from 0.209 min-1 to 0.068 min-1.

  • The ultimate aim of periodontal treatment is periodontal regeneration, which necessiates the regeneration of bone tissues. To evaluate the effects of Dex growth and differentiation of MC3T3-E1 cells, cells were seeded in alpha-modified eagle medium containing 10% fetal bovine serum, 10mM beta-glycerophosphate , $50{\mu}g/ml$ of ascorbic acid, with or without $10^{-7}M$ Dex and examined cell proliferation activities, alkaline phosphatase activities, and bone nodule formation until 25days. The results were as follows : 1. In Dex group, cell proliferation activities were lower until 15 days compared to control group. Bone nodules formation were showed at 10 days. 2. In the time-response effect, ALP activities were increased until the 10 days in control groups thereafter decreased and ALP activities of Dex group were lower aspect than control group until the 10 days In this study, bone nodule formation of osteoblast-like cells were accelerated by Dex and cell proliferation activities, ALP activity of Dex group showed lower than control group. Dex was considered that it did suppress initial growth, but accerelate mineralization of osteoblast-like cells.

  • Purpose :. To investigate the effect of aqueous extract of Ganoderma lucidum(G.I.) on the surival of tumor cells in vitro and on the growth of tumors in vivo. Materials and Methods : Dried G.I. was made into powder, extracted with distilled water, filtered and diluted from a maximum concentration of 100 mg/ml in sequence. The cytotoxicity of G.I, in vitro was evaluated from its ability to reduce the clonogenicity of SCK tumor cells. For the tumor growth delay study, about $2{\times}10^5$$ of SCK tumor cells were subcutaneously inoculated in the legs of A/J mice. The first experimental group of mice were injected i.p. with 0.2ml of 250 mg/kg of G.I. from the first day after tomor inoculation for 10 days. The second experimental group of mice were injected i.p. with 0.2ml of 250 mg/kg of G.I. either once a day for 10 days or twice a day for 5 days beginining from the 7th day after tumor inoculation Results : 1. Cytotoxicity in vitro;survival fraction, as judged from the curve, at G.I. concentration of 0.5, 1,5, 10, 25, 50 and 100 mg/ml were 1.0, $0.74{\pm}0.03$, $0.18{\pm}0.03$, $0.15{\pm}0.02$, $0.006{\pm}0.002$, 0.015 and 0.0015, respectively. 2. Tumor growth delay in vivo; a) the time required for the mean tumor volume to grow to $1,000mm^3$ was 11 days in the control group and 14 days in the experimental group. b) the time required for tumor volume to increase 4 times was 11 days in the control group while it was 10.5 and 12 days in the groups injected with G.I. once a day and twice a day from the 7th day after tumor inoculation respectively. Conclusion : Aqueous extracts of G.I. showed a marked cytotoxicity on the SCK mammary cells in vitro. Tumor growth delay was statistically signiricant when G.I. in-jection was started soon after tumor inoculation, but it was not significant when injection was started after the tumors were firmly established.

  • This study was carried out to determine agronomic and genetic characteristics of the production process in tropical yams(Dioscorea atata L.). To make clear the possibility of cultivation of tropical yams in the Suwon area, the growth of the aboveground parts and the enlargement of tuber of Dioscorea alata, were investigated. The aboveground parts of the plants whose sprouted mother tuber were planted in late April were grown slowly until the 70-days after planting and grown rapidly after. from about 150-days after plantation, total length of vines slowly decreased due to the burning of the branch vines of lower modes. With regard to the branching progressed to 4th for the solo yam and to 6th for purple yam. The secondary branches tended to grow better. When the formation of daughter tubers was observed at the 50-days after planting, their enlargement was very slow. The rapid growth of tubers began at the 140-days and continued to the 190-days. General components such as protein, fiber, and lipid were higher in D. opposita rather than in D. alata. Hardness of D. alata was 2696.2 while that of D. opposita was 4946.9. Lightness of D. alata was 73.99, being higher than that of D. opposita.

Growth and Optimum Harvesting Time of Pod-edible Peas (Pisum sativum L.)

  • The present study was performed to obtain the basic information about growth and quality related characteristics and optimum harvesting time for podedible pea which is a new crop in Korea but believed to have a great deal of potentials for both domestic and overseas markets. They can be consumed either as a fresh succulent vegetable or as tender green pods. The daily green pod yield of pod-edible peas started to increase from ten days after flowering and the maximum yield was recorded on 26 days after flowering. Ninety percent of pod yields could be harvested from 16 to 36 days after flowering. Mean green pod yield for the tested varieties was approximately 8.0 t/ha. Total vitamin C content of pod-edible peas showed continuously decreasing trends from five days after flowering. The highest sucrose content was obtained at ten days after flowering. The highest panel score based on sweetness, chewiness, and hardness for the processed green pods was shown at 10-15 days after flowering in all varieties tested, indicating that the optimum harvesting time for pod-edible peas was considered to be 10-15 days after flowering.

Effect of Dry Matter Production and Growth Construction of Zoysia japonica on a photoperiod (일조시간 조절이 잔디(Zoysia japonica)의 물질생산과 생장에 미치는 영향)

  • This study was designed to estimate on the interaction of the first productivity, light condition and to examine the ecophysiological characteristics of Zoysia japonica. rtificial community of Zoysia japonica was analyzed effect of matter production and growth construction under various shading condition. The results summarized this experiment were as follows; 1. The relative growth rate (RGR) in all experimental plots was high during the growth stage of 20 days after transplanting, and then decreased. The maxium RGR value appeared in the control plot (2.13g/g. 10days) during the growth stage from 10 to 20 days after transplating. RGR value in server shading of short day condition was remarkably decreased from the early growth stage. The main factor to lower RGR value considered as a short day condition than that of shading. 2. The net assimilation rate (NAR) in all experimental plots except 3hour photoperiod plot was high during the early growth stage after transplanting, and then decreased early growth stage after transplanting, and then decreased as the growth proceeded. The maxium value appeared in the control plot (35g/g. 10days). NAR value in the severe shading of short day condition plots was low. Especially, NAR value in the three hours photoperiod plot was remarkably low from the early growth, there was no great difference by growth stage. NAR had negative correlation with LAI and positive correlation with RGR. 3. The increasing rate of leaf area ratio(LAR) was high during the early growth stage after transplanting in the control plot and in the photoperiod plot, the shorter the exposure time to sunlight, the higher the LAR value from the early growth stage. Especially, its value in the 3 hour photoperiod plot was remarkably high but its increasing rate was lower as the growth proceeded. LAR hade negative correlation on RGR and NAR. 4. The crop growth rate(CGR) in each experimental plot was increased until 50 days after transplanting, and then decreased. After that time, the maxium CGR value appeared in the control plot (1.56g/$\textrm{cm}^2$. 10-days), 60 days after transplanting, CGR had positive correlation with LAI. The optimum LAI in the control, 9 hour, 7 hour, 5 hour, and 3 hour photoperiod plots appeared as 1.87, 1.12, 0.83, and 0.18.

Standardized Sod Production Using Box Seeding (상자파종에 의한 규격화된 잔디의 생산)

  • The objective of this research was to produce sod by box seeding for zoysiagrass or by vegetative propagation for zoysiagrass and manilagrass.1 Various ratio of peatmoss to sand(v /v) were prepared to find idea[ medium for fast and light weight sod production. Then, the days required for sod formation, the effect of growth regulators on the growth of turfgrass, and the various storage methods for winter keeping of sods were also investigated. 1.The mixed medium of sand and peatmoss(v /v, 1 : 2) showed more biomass production than that of sand. 2.In comparison of seeding rate of zoysiagrass, the amount of log /$m^2$ was most effective in the fast and dense sod formation. The amount of 20g /$m^2$ also showed fast sod formation. But, it resulted in weak plant and less tillering. During April to June, about 100 days were required to form sod with seeding rate of 5g /$m^2$ regardless of seeding time. Whereas 80 days were required to form sod in the rate log /$m^2$, which was 20 days shorter than that of 5g /$m^2$. 3.More than 85% of shoots in sod stored in field or plastic house during the winter time resumed the growth in good appearance after transplanting. The whole covering of ground with sod resulted in less weeds and faster formation of lawn. 4.Vegetative propagation of manilagrass showed about 7 to 15 days faster formation of sod than that of zoysiagrass. Application of GA increased shoot growth and BA increased the total number of tillering. However, the effects of the combined application of GA and BA were negligable.

Effect of the supplementation of persimmon peels on mycelial growth of Pleurotus eryngii (감과피 첨가가 큰느타리버섯 자실체 생육에 미치는 영향)

  • Five to thirty percent of persimmon peels were added to mushroom medium to investigate mycelial growth. Mycelial growth on the medium with persimmon peels was 82~96 mm (28 days) and was slower than that of the control without persimmon peels as, 100 mm (28 days). Mycelial growth time on medium with 5 to 10% persimmon peels was similar to the control without persimmon peels as 28 days but that on medium with 30% persimmon peels was delayed for 4 days. The time of pinhead formations in medium with 10 to 20% persimmon peels was 8 days, the growth time to harvest was 10 days. These results were similar to those of the control without persimmon peels. The sizes of pileus of treatments with 10 to 30% persimmon peels were tend to be smaller compared with the control. The length of stipe of 10% persimmon peels treatment was 89 mm which was shorter than that of the control with 90 mm. The thickness of stipes of 10 to 20% persimmon peels treatments were 36 to 42 mm which were tend to be thicker than that of the control with 42 mm.

Effects of Maturation Periods of Pig Manure Composts on Growth of Leaf lettuce(Lactuca safiva L.) (돈분발효퇴비의 부숙기간이 상추 생육에 미치는 영향)

  • To clarify the proper and safe duration of maturation periods for sawdust-pig manure composts, leaf lettuces were applied with pig manure composts fermented for 15, 30, 45, 60, 75 days and 1 year (control) and cultivated in a plastic house with or without additional PE film tunnel. The changes in physiochemical properties of soil and leaf lettuce growth were measured. Shorter duration of maturation periods enhanced the generation of N $H_3$ gas from the composts and resulted in significant decrease in seed germination, growth and yield of leaf lettuce. Under with and without PE tunnel conditions the concentration of N $H_3$ from compost over 3.8 and 2.1 mg/100g composts, respectively induced growth reduction. In proportion to the increase of maturation duration saw-dust containing pig manure exhibited decrease in C/N ratio, N $H_4^{+}$-N, N $H_4^{+}$N $O_3^{-}$ ratio but increase in N $O_3^{-}$-N contents. In case of applying pig manure compost without PE film tunnel condition the minimum maturation period of pig manure composts for safe leaf growth was 60 days, while minimum 75 days of maturation was required when applied with PE film tunnel. tunnel.
