To survey the incidence of plant-parasitic nematode in strawberry nursery, 117 soil samples were collected from main producing area of strawberry nursery plants in 2016. The incidences of two major problematic nematodes such as root-lesion nematodes (Pratylenchus spp.) and root-knot nematodes (Meloidogyne spp.) were 11% and 3%, respectively. Strawberry nurseries using upland soil either in fields or in plastic-houses showed were higher infection rates of Pratylenchus spp. or Meloidogyne spp. than others. On the other hand, a strawberry nursery using pots filled with bed soil in plastic-house was the lowest infection rate with 0%. Phylogenetic relationships and distance analysis were performed for identification of Pratylenchus spp. and Meloidogyne spp. detected from strawberry nurseries. As the results, they were identified as Pratylenchus penetrans, Pratylenchus vulnus and Meloidogyne hapla. Meanwhile, when nursery plants of strawberry infested with P. vulnus transplanted to pots filled with sterilized soil, P. vulnus was detected from soil in pots as 33% frequency. Thus, P. vulnus can be spread through nursery plants of strawberry infested with that. These studies suggested that Pratylenchus spp. and Meloidogyne spp. were ploblematic nematodes in major areas of strawberry nursery plants and P. vulnus could be spread through nursery plants of strawberry.
This study was conducted to investigate the life history, morphology, and maturation of larval stages and adult worms of Fasciola gigantica in experimental mice. Lymnaea auricularia rubiginosa was used as the intermediate host, and Oryza sativa was used for encystment of the metacercariae, while Mus musculus was used as the definitive host for maturation study. Fresh eggs from the gall bladder of water buffaloes fully developed into embryonated ones and hatched out at days 11-12 after incubation at about $29^{\circ}C$. Free-swimming miracidia rapidly penetrated into the snail host, and gradually developed into the next larval stages; sporocyst, redia, and daughter redia with cercariae. Fully-developed cercariae were separated from the redia and shed from the snails on day 39 post-infection (PI). Free-swimming cercariae were immediately allowed to adhere to rice plants, and capsules were constructed to protect metacercariae on rice plants. Juvenile worms were detected in intestines of mice at days 3 and 6 PI, but they were found in the bile duct from day 9 PI. Juvenile and adult flukes were recovered from 16 mice experimentally infected with metacercariae, with the average recovery rate of 35.8%. Sexually mature adult flukes were recovered from day 42 PI. It could be confirmed that experimentally encysted metacercariae could infect and develop to maturity in the experimental host. The present study reports for the first time the complete life history of F. gigantica by an experimental study in Thailand. The obtained information can be used as a guide for prevention, elimination, and treatment of F. gigantica at environment and in other hosts.
Chromosome analysis using mitosis, meiosis and bicolor FISH were carried out in Bupleurum latissimum Nakai, which is one of the endemic plants in Ulleung island of korea. The somatic methaphase chromosomes number of this plant was 2n = 2x = 16 and the chromosome complements consisted of six pairs of metacentrics and two pairs of submetacentrics. The size of chromosomes ranged 2.40${\sim}$4.20 ${\mu}$m and NOR (nucleolus organizer region) chromosome did not observed using conventional staining. In meiosis chromosomes, metaphase-I and anaphase-I were observed. Metaphase-I anaphase-I showed 8 bivalents and chromosomes migration to make two daughter cells. Using bicolor FISH, one pair of 5S and 45S rDNA signals were detected on the centromeric region of chromosome 3 and the end of short of chromosome 2,respectively. We also observed the NOR using 45S rDNA probe.
Aspects of structural differentiation of the free -floating hydrophyte, Spirodela polyrhira, has been investigated. The hydrophyte exhibited highly reduced structures having only fronds, connective stalks, and roots as a mature plant. The fronds were major photosynthetic and vegetatively reproducing organ during the growth. Daughter fronds which developed early in the mother frond were also chlorenchymatous, but they remained within foliage sheaths of the mother frond before separation from connecting stalks. Although the stalks and roots originated from the same meristematic region of the frond, they exhibited the distinct polarity showing former lateral growth and latter axial growth. Air chambers were formed only in the fronds and roots. Cellular components were scattered throughout the diffuse cytoplasm in most of the actively growing stalk cells. Root cells protected by the root cap demonstrated relatively complex organization, showing dense cytoplasm with Golgi, rER, mitochondria, and chloroplasts in the cortical cells. Cells in the root cap were highly vacuolated within the peripheral cytoplasm. Such reduction and differentiation of the plant body in Spirodela polyrhiza most effectively contributes to the better adaptation of smaller plants to superficial aquatic environments, while also enabling the rapid growth.
This study was carried out to determine agronomic and genetic characteristics of the production process in tropical yams(Dioscorea atata L.). To make clear the possibility of cultivation of tropical yams in the Suwon area, the growth of the aboveground parts and the enlargement of tuber of Dioscorea alata, were investigated. The aboveground parts of the plants whose sprouted mother tuber were planted in late April were grown slowly until the 70-days after planting and grown rapidly after. from about 150-days after plantation, total length of vines slowly decreased due to the burning of the branch vines of lower modes. With regard to the branching progressed to 4th for the solo yam and to 6th for purple yam. The secondary branches tended to grow better. When the formation of daughter tubers was observed at the 50-days after planting, their enlargement was very slow. The rapid growth of tubers began at the 140-days and continued to the 190-days. General components such as protein, fiber, and lipid were higher in D. opposita rather than in D. alata. Hardness of D. alata was 2696.2 while that of D. opposita was 4946.9. Lightness of D. alata was 73.99, being higher than that of D. opposita.

Experiments were conducted to evaluate the effect of low temperature-darkness treatment on floral initiation in four kinds of Korean strawberry cultivars. Mother plants were planted on March 29 and daughter plants were raised for the experiment. Temperature treatment was done for 7, 14, and 21 days keeping $13^{\circ}C$ in cooling storage from August 29, September 5 and September 14 to September 21. After the treatment, flower bud initiation was examined by a microscope, and the seedlings were transplanted on hydroponic system with Yamazaki's strawberry solution of EC $0.8dS{\cdot}m^{-1}$ to check the flowering. 'Ssanta' and 'Maehyang' showed early flower bud initiation in 7 days treatment than 'Seolhyang' by microscope check. 'Ssanta', 'Daewang', and 'Maehyang' showed early flower bud initiation in 14 days treatment than 'Seolhyang'. There were no differences among the treatments in 21 days treatment. Percentage of flowering of 'Seolhyang' and 'Maehyang' by low temperature-darkness treatment didn't show any difference but 'Daewang' and 'Ssanta' showed high percentage of flowering. It suggested that low temperature-darkness treatment technique can be use for improving early flowering and yield of 'Ssanta' and 'Daewang' cultivars.
This study was conducted to evaluate the influence of crown diameter on plant growth after planting and fruit yield. All nursery plants were classified into three classes: A, > 9 mm; B, 8~9 mm; C, < 8 mm. The A-class nursery plant plot showed better root and plant growth after planting compared to the B-class and C-class nursery plants. The A-class nursery plant plot was 10 days faster in first flowering time then those of the B-class and the C-class. Also, the A-class nursery plant plot was 2~3 days faster in the mean flowering time. The accumulative yield of the A-class nursery plant plot was 481 g per plant, that of the B-class was 445 g, and the C 422 g, which the nursery plant plot of A-class yielded more 8.1% and 13.9% respectively than B-class plot and C-class plot per plant. In early yield of to February; A-class nursery plant plot showed 23.3% higher yield than B-class nursery plant plot, and 39.1% higher the C-class nursery plant.

The objective of this research was to investigate the influence of application rates of dolomitic lime in the acid substrate on the mitigation of high bicarbonate damage in 'Seolhyang' strawberry. For this purpose, an acid substrate was formulated by mixing of sphagnum peat moss and pine bark with the ratio of 5:5 (v/v). The pH, EC and CEC of the substrate analyzed before application of dolomite were 4.07, $0.46dS{\cdot}m^{-1}$, and $91.3cmol+/kg^{-1}$, respectively. To adjust the pH of substrate, various amounts of dolomitic lime [$CaMg(CO_3)_2$] such as 0 (control), 1, 2, 3, and $4g{\cdot}L^{-1}$ were added during substrate formulation. Then, seedlings with 3 leaf stage were transplanted as mother plants and those were fed with Hoagland solution containing $240mg{\cdot}L^{-1}$ of the $HCO_3{^-}$. The growth parameters of mother plants 140 days after transplanting, such as plant height, chlorophyll content, and fresh weight were the highest in the treatments of 2 and $3g{\cdot}L^{-1}$ of dolomitic lime. The physiological disorders in mother plants were not observed in the 1, 2 and $3g{\cdot}L^{-1}$ treatments, but the symptoms of Ca, K and B deficiencies were observed in the 0 and $4g{\cdot}L^{-1}$ treatments. During the propagation period, the number of daughter plants derived from each mother plant were 21.0, 29.5, 35.8, 27.3 and 16.0 in the treatments of 0, 1, 2, 3 and $4g{\cdot}L^{-1}$, respectively. The substrate pH during cultivation of mother plants were maintained at appropriate levels for the 1 and $2g{\cdot}L^{-1}$ treatments, whereas it was the highest in $4g{\cdot}L^{-1}$ treatment. The contents of macro- and micro-elements in the above ground tissue were the highest in $2g{\cdot}L^{-1}$ and the lowest in $4g{\cdot}L^{-1}$ lime treatments. Above results suggest that the bicarbonate injury originated from ground water can be mitigated by adjusting the amount of dolomitic lime incorporated into the acid substrate.

This study was carried out to evaluate the yield of cuttings according to the planting and runner releasing times of mother plants in order to raise the cutting seedlings of raising seedling period 75 days or more needed for forcing culture of strawberries to be transplanted into the field around the 15th of September. Three domestic cultivars of 'Maehyang', 'Jukhyang', and 'Kuemsil' were tested. For experiment 1 to determine the yield of cuttings with the change of transplanting time, the mother plant were planted on February 28, March 20, and April 9 in 20 days intervals, and the cuttings were collected two to three times from June 4 to July 1. Experiment 2 was conducted to investigate the yield of cuttings depending on the runner releasing time, the runners were released in three intervals of 20 days, 40 days, and 60 days after planting the mother plant on March 5, and the cutting were collected once to three times from May 29 to June 26. From the comparisons of cutting yield according to the transplanting time of mother plants, February 28 treatment was more 9~25% and 114~165% for each cultivar than March 20 and April 9, respectively (Experiment 1). The yield of cuttings with releasing time 20 days after planting the mother plants had higher by 60~77% and 104~176% for each cultivar than 40 days and 60 days, respectively (Experiment 2). From these results, in case of propagating the seedlings from cuttings needed for field planting around September 15, early planting around in the latter part of February is the best for cuttings yield. In addition, releasing after the removal of the runners produced from mother plants by 20 days after planting gives an advantage over higher yield of cuttings. Consequently, this study suggest to apply an efficient raising seedling system for labor saving and quality improvement in raising seedlings of three strawberry cultivars in Korea.
In 1997 when cloned sheep Dolly and soon after Polly were born, it had become head-line news because in the former the nucleus that gave rise to the lamb came from cells of six-year-old adult sheep and in the latter case a foreign gene was inserted into the donor nucleus to make the cloned sheep produce human protein, factor IX, in e milk. In the last few years, once the realm of science fiction, cloned mammals especially in livestock have become almost commonplace. What the press accounts often fail to convey, however, is that behind every success lie hundreds of failures. Many of the nuclear-transferred egg cells fail to undergo normal cell divisions. Even when an embryo does successfully implant in the womb, pregnancy often ends in miscarriage. A significant fraction of the animals that are born die shortly after birth and some of those that survived have serious developmental abnormalities. Efficiency remains at less than one % out of some hundred attempts to clone an animal. These facts show that something is fundamentally wrong and enormous hurdles must be overcome before cloning becomes practical. Cloning researchers now tent to put aside their effort to create live animals in order to probe the fundamental questions on cell biology including stem cells, the questions of whether the hereditary material in the nucleus of each cell remains intact throughout development, and how transferred nucleus is reprogrammed exactly like the zygotic nucleus. Stem cells are defined as those cells which can divide to produce a daughter cell like themselves (self-renewal) as well as a daughter cell that will give rise to specific differentiated cells (cell-differentiation). Multicellular organisms are formed from a single totipotent stem cell commonly called fertilized egg or zygote. As this cell and its progeny undergo cell divisions the potency of the stem cells in each tissue and organ become gradually restricted in the order of totipotent, pluripotent, and multipotent. The differentiation potential of multipotent stem cells in each tissue has been thought to be limited to cell lineages present in the organ from which they were derived. Recent studies, however, revealed that multipotent stem cells derived from adult tissues have much wider differentiation potential than was previously thought. These cells can differentiate into developmentally unrelated cell types, such as nerve stem cell into blood cells or muscle stem cell into brain cells. Neural stem cells isolated from the adult forebrain were recently shown to be capable of repopulating the hematopoietic system and produce blood cells in irradiated condition. In plants although the term$\boxDr$ stem cell$\boxUl$is not used, some cells in the second layer of tunica at the apical meristem of shoot, some nucellar cells surrounding the embryo sac, and initial cells of adventive buds are considered to be equivalent to the totipotent stem cells of mammals. The telomere ends of linear eukaryotic chromosomes cannot be replicated because the RNA primer at the end of a completed lagging strand cannot be replaced with DNA, causing 5' end gap. A chromosome would be shortened by the length of RNA primer with every cycle of DNA replication and cell division. Essential genes located near the ends of chromosomes would inevitably be deleted by end-shortening, thereby killing the descendants of the original cells. Telomeric DNA has an unusual sequence consisting of up to 1,000 or more tandem repeat of a simple sequence. For example, chromosome of mammal including human has the repeating telomeric sequence of TTAGGG and that of higher plant is TTTAGGG. This non-genic tandem repeat prevents the death of cell despite the continued shortening of chromosome length. In contrast with the somatic cells germ line cells have the mechanism to fill-up the 5' end gap of telomere, thus maintaining the original length of chromosome. Cem line cells exhibit active enzyme telomerase which functions to maintain the stable length of telomere. Some of the cloned animals are reported prematurely getting old. It has to be ascertained whether the multipotent stem cells in the tissues of adult mammals have the original telomeres or shortened telomeres.
