Dark-chilling Pretreatment Protects PSI from Light-chilling Damage

  • In chilling-sensitive plants, the donor side of Photosystem II is inhibited by the chilling treatment in the dark, while the acceptor side of Photosystem I is inhibited by the chilling under the moderate light. Since the addition of inhibitors of electron transfer from Photosystem II protects Photosystem I from chilling induced photoinhibition of Photosystem I, inhibition or down-regulation of Photosystem II activity in vivo may also protect Photosystem I from photoinhibition. It was revealed that dark-chilling pretreatment actually protected Photosystem I from photoinhibition. The results imply that down-regulation of Photosystem II under stress conditions may have a role to protect Photosystem I from photoinhibition.

High Frequency Plant Regeneration from Leaf Explant Cultures of Domestic Cultivated Strawberry (Fragaria x ananassa Duch) (국내 딸기 재배품종의 잎절편 배양으로부터 고빈도 식물체 재생)

  • To develop a high efficiency plant regeneration system from in vitro cultures of strawberry, cv. Yeobong, petiole and leaf explants were cultured on MS basal medium containing a combination of 0.5 mg/L IBA and 3.2 mg/L kinetin or zeatin or benzyl amino purine (BAP) for 6 weeks, and leaf explants with dark pretreatment for a week ($T_1$), 2 weeks ($T_2$), and 4 weeks ($T_3$) were cultured on medium supplemented with 0.5 mg/L IBA and 3.2 mg/L zeatin under 16 hr photoperiod for 6 weeks. Shoot organogenesis was observed from the greenish calli containing minimal anthocyanin formed at proximal cutting edges of the leaf explant (57%) when cultured adaxial side on the medium, whereas was directly formed from a cutting edges of petiole explant (6.3%). Frequency of callus formation and shoot organogenesis at large size of leaf explant ($1.0{\sim}1.5\;cm^2$) was higher than small size ($0.5{\sim}1.0\;cm^2$), and dark pretreatment significantly improved the frequency of leaf explants that produced calli and shoots. The maximum frequency (87%) for shoot organogenesis was obtained from the leaf explants that transferred to a 16 hr photoperiod condition after the initial 4 weeks dark period. The improved frequency (87%) in comparision with control without dark pretreatment (27%). When the shoots were transferred to 1/2 MS basal medium, formed roots with 20 d of culture. The rooted plants were subsequently transferred to the pots and to the field.

Influences of Cold Pretreatment, Dark Condition, and Ficoll on the Callus Formation and Organ Differentiation in Anther Culture of Carnation (카네이션 약배양에 있어서 저온전처리, 암처리 및 Ficoll처리가 캘러스 형성 및 기관분화에 미치는 영향)

  • These studies were carried out to investigate the effects of cold pretreatment, dark condition, and Ficoll on the callus formation and organ differentiation in anther culture of carnation for the purpose of settling the techniques of anther culture of 'Rony' carnation. Diameter of flower buds containing anthers with microspores before or after uniuncleated stage was between 4 and 6 mm. A high percentage of callus formation and responding anthers were achieved by low temperature pretreatment at $4^{\circ}C$ for 7 days. Callus formation was promoted under dark condition. Roots were differentiated from calli formed under darkness. Ficoll didn't promote callus formation but promoted differentiation of shoots and roots.

  • Effect of various physical and chemical treatments on dark germination of Oenothera lamarckiana seeds were primarily investigated to find out factors affecting germination. Germination of seeds which did not germinate in the constant temperature and darkness was induced by several physical treatments such as sonification, wetting and drying, freezing and thawing, and removal of seed coat. Pretreatment of chilling ($4^{\circ}C$), freezing ($-10^{\circ}C$) and incubation at high temperature ($80^{\circ}C$) induced dark germination of seeds which did not germinate in the constant temperature ranges of 15 to $40^{\circ}C$ under darkness. Alternating temperature also had a stimulatory effect on dark germination of Oenothera lamarckiana seeds. Sensing of seeds to alternating temperature appeared to be completed during the first two days after imbibition. The minimum difference of temperature required for dark germination was $5^{\circ}C$ in the range of $15-25^{\circ}C$. A thiourea (1.0%) treatment induced dark germination, but GA, IAA, BA and Ethrel failed to do so.

Characterization of the Membrane Potential Relevant to Permeability Changes in the Plasmalemma of Lemna gibba G3 (좀개구리밥 (Lenma gibba G3)의 원형질막의 투과성 변화와 관련된 막전위의 특성)

  • The membrane potential in the subepidermal cells of Lemna gibba G3 fronds was measured in the dark with glass capillary microelectrodes. At pH 7, the membrane potential, approximately-215 mV, could be depolarized to -82∼-88 mV by 0.1 mM dicyclohexylcarbodiimide (DCCD) or by KCN at 0.3 mM or higher concentrations. When the pH of the medium was altered the potential showed reversible changes, while it revealed no response to the external pH changes when energy transduction across the membrane was being blocked by 0.1 mM DCCD. The results support an assumption that the active component of the membrane potential of Lemna subepidermal cells is generated by electrogenic H+ -pump. By the addition of 0.10∼5.00 mM salicylic acid(SA) to the bathing medium the membrane potential was depolarized to a great extent, and the removal of SA from the medium repolarized the potential showing almost complete recovery, 92.3∼97.6% to the initial levels. Although the potential was greatly depolarized by 5.0% or higher concentrations of dimethylsulfoxide (DMSO), the recovery rate by DMSO removal was decreased as the pretreatment concentration had increased. Twenty percent DMSO pretreatment limited the recovery at only 47.1%. The presence of SA in the bathing medium could reversibly increase the permeability of the plasmalemma. DMSO at its concentration of 5.0% or higher increased the permeability of the membrane by irrevesibly impairing the membrane component involved in the membrane permeability.

Adsorption of Heavy Metal Ions on Bark(I) (수피에 의한 중금속 흡착(I))

  • The pretreatment of bark powder with sodium hydroxide and formalin showed the most excellent adsorption ratio, but this method could not practically be used because of the occurrence of dark-colored pigments in filtrates during pretreatment. Instead, acid and formalin were the most affirmative and effective among the pretreatment methods tested, and could be used for this purpose. Among tested species, Quercus acutissima and Robinia pseudo-accacia showed the largest amount of metal adsorption, and $Pb^{2+}$ was the best(83 to 96%) among the four heavy metals tested. The order of adsorption ratios other metals was as follows; $Cu^{2+}$ > $Zn^{2+}$ > $Cd^{2+}$, and the ratio was approximately 45 to 55%. In addition, as the substrate amount increased, the amount of adsorbed heavy metals in subtrates gradually increased, but the adsorbed amount was not proportional to the substrate amount. The order of heavy metal adsorption was as follows; $Pb^{2+}$ > $Cu^{2+}$ > $Cd^{2+}$ > $Zn^{2+}$. Depending on flow rate and column size, pine bark power adsorbed more heavy metals in the 5ml/min flow rate and 3.5cm column size rather than the 10ml/min and 2.0cm. However, oak bark power showed contrary results compared with pine bark powder. The adsorption of $Pb^{2+}$ occurred rapidly in the incipient stagte. Even though bark powders were repeatedly used three times, there was no change in the adsorption ratio(45%), but after four times, the adsorption ratio was significantly reduced to 35%.

Effect of Seed Pretreatment with Chilling, $GA_3$ and Light on Bupleurum falcatum Germination (파종 전 저온, $GA_3$ 및 광 처리가 시호의 발아에 미치는 영향)

  • In the cultivation of Bupleurum falcatum, one of the problems to surmount is long-term germination period and unstable germination. This experiment was done to examine the effect of $GA_3$ concentration [0(water), 0.01, 0.lmM], chilling, their treatment period [2, 4, 8 days ($GA_3$) : 0, 2, 4 weeks (chilling)] and light quality (red, white, dark) given during the period as pretreatment before sowing on the seed germination of its two cultivars (cv. Jaerae, cv. Jangsu). Light treatment was given during all the periods of $GA_3$ treatment or for 0, 2, 4 days at the end of the chilling treatment. There was no difference in the mean germination rate between the levels of all the treatments except the $GA_3$ concentration meaning that water imbibition and $GA_3$ treatment had the same effect. As light quality treatment during the water imbibition was forced, the mean germination rate of Jaerae, 2 to 4 days imbibition period or red light was more increased or accelerated compared to the other levels of the same treatment, respectively. The rate of Jaerae not affected by the light quality was the greatest in the 2 days water imbibition while the rate of Jangsu was the greatest when water-imbibed for 4 days or treated by red light. No chilling before sowing showed the highest rate due to the light quality and white light forced after sowing had greater rate than the dark treatment. Although there was no difference between the rates of light quality treatment levels in the condition of no chilling before sowing and white light treatment after sowing, the rate of Jangsu was enhanced or accelerated only under illumination during 2 days water imbibition before sowing.

Effect of Ginseng Saponin on the Circadian Rhythm of Pentobarbital-induced Sleep in Mouse (인삼 Saponiol이 Mouse의 Pentobarbital수면 Circadian Rhythm에 미치는 영향)

  • Circadian susceptibility of sleeping induced by pentobarbital was observrd in male DDO mouse treated with phenobarbital and ginseng saponin. The pentobarbital elimination rate was also measured in the same animal. The mouse had been maintained for one week under 12 hours of artificial illumination extending from 06:00 to 18:00 hours alternating with 12 hours of darkness. During the period the animals were administered intraperitoneally with 100mg/kg of phenobarbital for three days or 10mg/kg and 100mg/kg of ginseng saponin for seven days. At 24 hours after last injection pentobarbital sleeping time and elimination rate were measured following intraperitoneal administration of 50mg/kg of pentobarbital sodium. In a control group treated with saline, the duration of pentobarbital-induced sleep varied with circadian rhythmicity, which had a trough at 02:00 hours of light phase and a crest at 14:00 hours of dark phase. And the elimination rate measured at 02:00 hours was faster than that at 14:00 hours. Pretreatment with phenobarbital markedly shortened the pentobarbital steeping time and abolished the circadian rhythmicity. Those were correlated with the increased pentobartital elimination by phenobarbital throughout light and dark phases examined. Ginseng saponin, given for seven days in a dose of 10mg/kg or 100mg/kg, did not affect the circadian rhythmicity of sleeping and the elimination rate. Sleeping time during light phase, however, was somewhat shortened in ginseng treated animals, which was not matched with the finding of unaltered elimination rate. It seemed that the central nervous system stimulating effect of ginseng saponin might be involved in the findings observed.

Effect of Light Quality,$GA_3$ and Temperature as Treatments Before or During Germination on Tobacco Seed Germinability (파종전후 종자에 가해지는 광질, $GA_3$ 및 온도에 따른 담배의 발아율)

  • It failed occasionally to take a reasonable emergence rate since tobacco (Nicotiana tabacum L.) seeds were planted on late Jan. showing lowest temperature. This experiment was done to measure the effect of GA3 (concentration ; period) , light quality (red ; white ; dark) during or after its treatment, daily irradiation hours( 0 ; 8 ; 12 ;16) and germination termperature (20 or 10 $^{\circ}C$ ocnstant ; 20/1$0^{\circ}C$ alternating) on the germination rate. Red and white light given during grmination showed no differences between the other daily irradiation hours except that 8 hours red light delayed germination although their 12 hours irradiation had the gratest rate. The rate was increased with increased concentration to GA3 0.01 mM or increased imbibition period to 3 days although the rate of cv. NC 82 was less than that of cv. Burley 21 in the case of dark imbibition of GA3 but daily 12 hours irradiation during germination. Light quality forced during GA3 imbibition eliminated such effect of GA3 shown in the darkness so that only light quality pretreatment and termperature during germinition were affected on the rate. The germination rate of thecultivars was decreased in the order of red, white light, darkness meaning that it was highly influenced by the light quality during GA3 treatment. Regardless of GA3 or light quality treatment,on the other hand, the rate was greater in 20 $^{\circ}C$ constat than 1$0^{\circ}C$ constant and 20/1$0^{\circ}C$ alternating germination temperature having similar germinative patterns.

Study on Bio-H2 Production from Synthetic Food Waste and Activated Sludge from Industrial Waste Water Processes using Dark-fermentation (산업공정의 폐수처리에서 발생된 폐활성슬러지 및 인공음식폐기물을 이용한 생물학적 수소생성에 관한 연구)

  • This study performed to extract operation factors of major organic wastes, which were food wastes and waste activated sludge generated in industries in order to use them as a substrate for bio-H2 production. According to the results of experimental analysis for hydrogen production capacity by various organic concentrations, the hydrogen production yield was the highest at 80 g/L, and the efficiency was improved by the pretreatment of waste activated sludge (acid treatment, alkali treatment). Hydrogen production efficiency was improved by mixing food wastes and waste activated sludge if waste activated sludge was below than 30%, however, it was decreased when it was more than 50%. The impacts of heavy metals on the hydrogen production shows that the inhibition level depends on the concentration of Cr, Zn, and Cu, Fe was able to enhance the hydrogen production.
