Effects of Intraperitoneal Administration of Lactococcus lactis ssp. lactis Cellular Fraction on Immune Response

  • Cellular components of Lactococcus lactis ssp. lactis (heat-killed whole cells, cytoplasm, and cell walls) were tested for their in vivo immunopotentiating activities. Peritoneal macrophages from mice injected intraperitoneally with cell-wall fractions exhibited significantly greater phagocytic activity than groups injected with whole cells or cytoplasm fraction. Cytotoxicity of natural-killer cells was highest in cytoplasm fractions. Production of cytokines (IFN-${\gamma}$, IL-2, IL-6, and IL-12) in spleen cells was significantly higher when cellular components were injected intraperitoneally, and tended to be higher in whole-cell and cytoplasm groups than in cell-wall group. These results demonstrate lactic acid bacteria whole cells and their cytoplasm and cell-wall tractions have immunopotentiating activities.

Endogenous lipid pneumonia in a ringed seal (Pusa hispida subsp. ochotensis)

  • An adult female ringed seal died suddenly and was subsequently examined for diagnostic purposes. The animal's lungs demonstrated mild non-collapsibility and multifocal white to yellow patches. Histopathological examination revealed multifocal pulmonary histiocytosis. Alveoli were filled with numerous foamy macrophages cytoplasm and scattered multinucleated giant cells containing cholesterol clefts. The foamy cytoplasm of the macrophages stained with oil red O stain. Further, lipid droplets within the cytoplasm were detected by electron microscopy. To the author's knowledge, this is the first case report describing the histochemical staining and electron microscopic findings associated with endogenous lipid pneumonia in ringed seal.

  • The tropical tasar silkworms, Antheraea mylitta (D) produce famous silk 'Kosa' in central part of India. Due to outdoor rearing it became susceptible to viral infection including cytoplasmic polyhedrosis virus (CPV). The common mode of entry of cytoplasmic polyhedrosis virus is per os and cause gresserie disease to the larvae. Histopathological studies elucidated the insect CPV virus produces infective polyhedral inclusion bodies (PIBs) in the midgut cell cytoplasm of virus infected fifth instar larvae. The PIBs multiply enormously in the cytoplasm without invading the nucleus. Ultrastructural studies confirmed the pathological effects of CPV on in midgut cell cytoplasm. The multiplication of polyhedral inclusion bodies took place into the vacuoles and form virogenic stromata in the cytoplasm of cells. However, the encapsulations of polyhedral inclusion bodies into the polyhedrin protein occurred and polyhedra were released into the lumen. At the late stage of infection, cells showed the regressed cytoplasmic organelles with large vacuoles and elongated mitochondria. Hence, the horizontal transmission of CPV causing the midgut cells disintegration in the tasar silkworm, Antheraea mylitta (D) confirmed during infection.

A Study on the Decondensation and Pronucleus Formation of Sperm Nucleus in the Mouse Oocyte

  • To investigate the ability to decondense sperm head penetrated into cytoplasm of the oocytes and the relationship between this ability and the level of glutatione (GSH) in mouse oocyte at various maturing stages. The fertilizability of oocytes at various stages of maturation the decondensation of sperm nucleus and the formation of male pronucleus, were observed and the levels of GSH were measured in oocyte at same stages. Besides, the relation between fertilizability and level of GSH in oocyte cytoplasm treated with L-buthionine-S, R-sulfoxmine (L-BSO), the inbitor of biosynthesis of GSH, was determined. The decondensation of sperm head was not found in GV stage and L-BSO treated oocytes. In maturing oocytes (GVBD, MI), the decondensation was found, but the formation of male pronucleus was not. The levels of GSH in oocyte cytoplasm were measured; 2.2 pmol per oocyte in the ovulated and the matured in vitro each, 1.0 pmol in GV intact oocyte, 1.3 pmol in GVBD, and 1.5 pmol in MI phase oocyte. In L-BSO treated oocytes the levels of CSH were measured 0.08~o.09 pmol per oocyte, slightly lower than GV stage oocyte. In conclusion, GSH in oocyte is supposed to be synthesized and storaged in cytoplasm during maturation. The failure of decondensation in the cytoplasm of GV stage and L-BSO treated is suggested that GSH is an essential factor in decondensing the sperm head and that the a certain level of GSH, more than in GV oocyte cytoplasm, is required in decondensation.

Development of SCAR Markers for Early Identification of Cytoplasmic Male Sterility Genotype in Chili Pepper (Capsicum annuum L.)

  • We previously used Southern blot analysis to detect restriction-length polymorphisms between male fertile and cytoplasmic male sterile (CMS) cytoplasms at the coxII and atp6 loci of the mtDNA of Capsicum annuum L. Two copies of atp6 were found in each male fertile and CMS pepper lines. Interestingly, one of the copies of atp6 in CMS pepper was a 3'-truncated pseudogene. The open reading frame of the coxII gene was the same in the fertile (N-) and CMS (S-) lines. However, the nucleotide sequence in the S-cytoplasm diverged from that in the N-cytoplasm 41 bp downstream of the stop codon. To develop CMS-specific sequence-characterized amplified region (SCAR) markers, inverse PCR was performed to characterize the nucleotide sequences of the 5' and 3' flanking regions of mitochondrial atp6 and coxII from the cytoplasms of male fertile (N-) and CMS (S-) pepper plants. Based on these data, two CMS-specific SCAR markers, 607 and 708 bp long, were developed to distinguish N-cytoplasm from S-cytoplasm by PCR. The CMS-specific PCR bands were verified for 20 cultivars containing either N- or S-cytoplasm. PCR amplification of CMS-specific mitochondrial nucleotide sequences will allow quick and reliable identification of the cytoplasmic types of individual plants at the seedling stage, and assessment of the purity of $F_1$ seed lots. The strategy used in this report for identifying CMS-specific markers could be adopted for many other crops where CMS is used for F1 seed production.

중격측좌핵과 선조체 기저핵의 비교연구 I. 세포형의 구분 (Comparative Study on the Nucleus accumbens septi and the Nucleus fundus striati I. Classification of Neuronal Cell Types)

  • There's been arguments on the different morphological status between the nucleus accumbens septi and nucleus fundus striati of ventral striatum. Authors carried out the comparative study on the neuronal cell types of these nuclei, in the chick and the rat. Results are summarized as follows: In the nucleus accumbens septi of the chick, there found 3 main cell types. Type I cells are oval or spindle-shaped. They are the most abundant cell types, comprising more than 80% of neurons. The pale nucleus is usually indented. The cytoplasm is also pale and contains small amount of mitochondria, rough r-ER and Golgi complexes. This cell has a few symmetric synapses on the cell membrane. Type II cells are pale large cells. They are polygonal or irregularly-shaped. They contain pale spherical nucleus, and the pale cytoplasm with relatively large amount of mitochondria, free ribosomes and well-developed Golgi complexes. Some axo-somatic synapes are found on the cell. Type III cells are oval or spherical-shaped. The nucleus is relatively pale and large, In the dense cytoplasm, well developed. r-ER formed typical Nissl's body, and there found many mitochondria, ribosomes and lysosomes. In the chick fundus striati nucleus, there also found 3 main cell types. Type I cells are small and spindle-shaped. This type is the most abundant one and constitutes more the 80% of the neurons. Morphological features other than it's shape, is generally similar with that of Type I cell in the nucleus accumbens. Type II cells are irregularly shaped large cells. Dense cytoplasm contains large amount of cell organelles. Some axo-somatic synapses are found. Type III cells are small dense cells. This oval cell contains the oval nucleus, and the plentiful cytoplasm with well developed r-ER, ribosomes and mitochondria. In the nucleus accumbens septi of the rat, there found 4 main cell types. Type I cells are small, oval or spherical cells, comprising more than 90% of all the neurons. Spherical nucleus shows typical chromatin rim along the nuclear membrane. Dense cytoplasm contains many ribosomes and mitochondria. Type II cells are large oval cells. The eccentric nucleus is deeply invaginated. Pale cytoplasm contains large amount of ribosomes, Golgi complexes, mitochondria, and dense bodies. Type III cells are pale, large, oval cells. They contain moderate amount of ribosomes and mitochondria, and some scattered stacks of r-ER. Type IV cells are small pale cells. Small oval nucleus is indented and shows chromatin rim. Only small amount of ribosomes and mitochondria can be found. In the nucleus fundus striati of the rat, there also found 4 main cell types. Type I cells are spherical or oval cells, comprising more than 90% of the neurons. The chromatin rim of the spherical nucleus is not so prominent as compared to the rim of type I cell in the nucleus accumbens septi. The cytoplasm contains moderate amount of mitochondria, ribosomes and some scattered r-ER. A few axo-somatic synapses were found. Type II cells are small round or polygonal cells. Golgi complexes are especially well-developed in this cell type. The cytoplasm also contains moderate amount of mitochondria, ribosomes, and dense bodies. Type III cells are small cells. The large nucleus shows prominent chromatin rim. The cytoplasm contains many ribosomes and mitochondria. Type IV cells are large, spheircal or oval cells. The nucleus is deeply indented. The plentiful cytoplasm contains large amount of ribosomes, mitochondria, Golgi complexes, neurotubules, but not r-ER. In the present study, it is clear that the nucleus accumbens septi and the nucleus fundus striati are independant cell groups, according to their cytoarchitectonics and the ultrastructural features of their cell types.

당근 웅성부임성(雄性不稔性)의 형태별(形態別) 이용현황(利用現況)과 핵내(核內) 웅성부임(雄性不稔) 유전자(遺傳子)의 분포(分布) (Utilization of Male Sterility Type and Frequency of Male Sterile Genes in Carrots)

  • 세계 각 종묘회사에서 수집된 교배종 당근의 웅성부임(雄性不稔) 형태와 그 분리비(分離比)를 조사한 결과 지역별, 회사별, 품종그룹별로 사용된 웅성부임(雄性不稔) 형태에 차이가 있었다. 즉, 미국의 종묘회사는 Imperater계 품종이 대부분이며 petaloid형의 웅성부임(雄性不稔)을 주로 이용하였고 Nantes계 품종이 대부분인 유럽의 종묘회사는 brown anther형의 웅성부임(雄性不稔)을 주로 이용하였지만 이들 중 일부 회사에서 Nantes계의 특성에서 벗어나는 품종에서 petaloid형도 이용하고 있었다. Chantaney, Kuroda계 품종이 대부분인 일본(日本), 한국의 경우에는 petaloid형, brown anther형 두 가지 모두 이용하지만 각 회사별로는 한가지 형태의 웅성부임(雄性不稔)을 주로 이용하고 있었다. 한 품종에서는 한가지 형태의 웅성부임(雄性不稔)만 나타났는데 이는 웅성불임의 형태가 세포질의 종류에 따라 결정된다는 다른 연구자의 결과와 일치하는 것으로 나타났다. 한편 국내에서 이용되는 당근 계통들 중 서로 다른 두 가지 세포질(Sa, Sp)에 대해 유지친으로서의 사용 가능성이 어느 정도되는지 알아보기 위하여 여교잡하였을 때 petaloid형 세포질(Sp) 에서는 20계통 중 75%인 15계통에서 유지친을 찾을 수 있어 유지친의 분포빈도가 높은 것을 알 수 있었으며 brown anther형 세포질(Sa)에서는 4계통 중 3 계통에서 유지친을 찾을 수 있었지만 여교잡 된 소재(素材)가 매우 한정되어 분포빈도를 말하기는 어렵다고 판단되었다.

  • Ultrastructures of the large and elongated cells (idioblasts) in the haustorium of a parasitic angiosperm, Cuscuta australis R. Brown growing on the host plant, Trifolium repens L. were investigated by the electron microscopy. The idioblasts were characterized by the presence of a large nucleus, small vacuoles, and dense cytoplasm including a number of various cell organelles such as ribosome, rough endoplasmic reticulum(r-ER), mitochondrion, dictyosome, proplastid, multilamellar structure(MLS), microfilament bundle(MFB), and cytosegresome. Therefore, it is suggested that the idioblasts are metabolical1y very active. Particularly, MLS, MFB, and cytosegresome observed in this study did not appear in the haustorial cells of the other parasitic angiosperms. MLS was transformed into vacuole and also incorporated with cell wall. MFB composed of microfilaments, about each 7.5 nm in diameter, was observed in nucleus and also cytoplasm. Some types of MFB were distinguished on the basis of arrangement of microfilaments. A part of cytoplasm sequestered by stacked cisternae of smooth ER(s-ER), cytosegresome, was altered into a vacuole which was formed by digestion of the sequestered cytoplasm and of cisternae of s-ER. Cell organelles such as MLS, MFB, and cytosegresome were discussed in relation to the metabolic control of the idioblasts.

Relationship between PGCs Settle and Gonad Development in the Early Chicken Embryo

  • Chick embryos from stage 14 to stage 31 were studied by means of serial section and light microscopy in order to learn the relationship between the settlement sites of the primordial germ cells (PGCs) and the forming genital ridge. The results showed that: when embryo hatched for 53-56 h, the PGCs reached the coelomic epithelial tissue where gonad would be formed, meanwhile the epithelial tissue began thicker before the PGCs reached. Before stage 19, the final region the PGCs arrived was the thickened portion of the coelomic epithelium, the glycogen in the PGCs cytoplasm maintenance remained unchanged. However at the 3.5-5th hatching day, the glycogen in the PGCs cytoplasm reduced gradually. On the 6th hatching day, the gonad of the embryo appeared the feature of ovary, and the glycogen in the PGCs cytoplasm reduced further. On the 7th hatching day, the differentiation of ovary or testis was obvious and the glycogen in the PGCs cytoplasm later disappeared.

임신월수(姙娠月數)에 따르는 유우(乳牛)의 질점액(膣粘液)에 함유된 세포성분(細胞成分)의 변화상(變化像) (Studies on the Cell Picture of Vaginal Smears Stained by Papanicolaou's Method in Pregnant Dairy Cows)

  • Every month of pregnancy, vaginal smears were obtained from 42 dairy cows, and an attempt was made to correlate the cell picture with the stage of pregnancy. The results obtained in this work were summerized as follows: 1. At early pregnancy, most of the cells in the smears were the cells with normal nucleus and cytoplasm, and leucocytes. After the first month of pregnancy there was a progressive decrease in the rate of appearance of the cells with normal nucleus and cytoplasm, which dropped to the lowest value at the third month of pregnancy, thereafter, showing steady state. 2. After the first month of prgnancy the rate of appearance of the cells with karyolysis began to increase gradually, and reached a maximum at the fifth month of pregnancy, returning towards initial levels. 3. The rate of appearance of the cells with karyopyknosis was much lower at early pregnancy, but at the third month of pregnancy there was a rapid increase, thereafter, showing steady state (ranging from 20 to 50 percent). 4. The non-nucleated cornified cells, the cells with vacuolized cytoplasm, and the cells with swollen nucleus and cytoplasm were appeared much fewer or not, and significant changes were not recognized during pregnancy. 5. At early pregnancy there appeared many of leucocytes but dropped intensively at the third month of pregnancy, and, thereafter, appeared much fewer or not.
