New Record of Three Isopod Species (Crustacea: Malacostraca: Isopoda) from South Korea

  • Three marine isopods, Symmius yamaguchiensis Shimomura, 2008, Colanthura pigmentata Kensley, 1980 and Synidotea nipponensis Nunomura, 1985 are reported for the first time in South Korea. The family Chaetiliidae, and genera Symmius and Colanthura are new to South Korea. We provide descriptions of the diagnostic characteristics, illustrations of three species and their partial sequences of the mitochondrial cytochrome c oxidase subunit 1 (CO1) for molecular characteristics.

A New Record of Sea Star Genus Nearchaster (Asteroidea: Notomyotida: Benthopectinidae) from East Sea, Korea

  • An asteroid specimen was collected in adjacent water of Gisamun, Gangwon-do in the East Sea, Korea at a depth of 170 m by fishing net at May 2013. It was identified as Nearchaster (Nearchaster) pedicellaris (Fisher, 1910) belonging to family Benthopectinidae of order Notomyotida, which was new to the Korean fauna. The genus, family, and order of this species were also new to Korean waters. The partial sequence of mitochodrial cytochrome c oxidase subunit 1 (CO1) was determined for the first time and registered at GenBank.

  • Two feather mites, Bychovskiata hypoleuci Mironov and Ddabert, 1997 and Phyllochaeta interifolia (Mégnin and Trouessart, 1884) are reported for the first time in South Korea. Specimens of these two species were collected from the common sandpiper Actitis hypoleucos. The genera Bychovskiata Dubinin, 1951 and Phyllochaeta Dubinin, 1951 are also new reports for South Korea. Here, we provide morphological descriptions and illustrations of these two species. Additionally, we provide partial sequences of the mitochondrial cytochrome c oxidase subunit I(COI) as DNA barcodes.

Three feather mites(Acari: Sarcoptiformes: Astigmata) isolated from Tringa glareola in South Korea

  • We describe three feather mites recovered from a wood sandpiper Tringa glareola that was stored in a $-20^{\circ}C$ freezer at the Chungnam Wild Animal Rescue Center. These feather mites are reported for the first time in South Korea: Avenzoaria totani (Canestrini, 1978), Ingrassia veligera Oudemans, 1904 and Montchadskiana glareolae Dabert and Ehrnsberger, 1999. In this study, we provide morphological diagnoses and illustrations. Additionally, we provide partial sequences of the mitochondrial cytochrome c oxidase subunit I(COI) gene as molecular characteristics of three species.

Molecular Characterization of Taenia multiceps Isolates from Gansu Province, China by Sequencing of Mitochondrial Cytochrome C Oxidase Subunit 1

  • A total of 16 Taenia multiceps isolates collected from naturally infected sheep or goats in Gansu Province, China were characterized by sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene. The complete cox1 gene was amplified for individual T. multiceps isolates by PCR, ligated to pMD18T vector, and sequenced. Sequence analysis indicated that out of 16 T. multiceps isolates 10 unique cox1 gene sequences of 1,623 bp were obtained with sequence variation of 0.12-0.68%. The results showed that the cox1 gene sequences were highly conserved among the examined T. multiceps isolates. However, they were quite different from those of the other Taenia species. Phylogenetic analysis based on complete cox1 gene sequences revealed that T. multiceps isolates were composed of 3 genotypes and distinguished from the other Taenia species.

한국 섬진강산 누치(Hemibarbus labeo)의 분자 계통유전학적 연구 (Molecular Phylogenetic Study of the Barbel Steed (Hemibarbus labeo) in Seomjin River of Korea)

  • 섬진강 수계에 서식하는 누치(Hemibarbus labeo)의 미토콘드리아 DNA에서 cytochrome c oxidase subunit I(COI) 유전자를 발굴하고 누치속 Hemibarbus 및 잉어과(Cyprinidae) 내 계통유전학적인 위치를 확인하는 연구를 수행하였다. 발굴된 577 bp COI 시컨스의 다중배열 결과 섬진강산 누치들의 높은 염기서열 상동성을 보였다(99~100%). 우리나라에서 발견되는 누치속 3종에서 누치(H. labeo: HD1)와 어름치(H. mylodon)의 염기서열 유사성은 88.91%, 참마자 (H. longirostis)와는 88.81%이었다. 또한, H. maculatus, H. meditus, H. umbrifer, H. barbus는 각각 누치와의 염기서열 유사성이 98.97%, 97.20%, 96.87%, 98.85% 등으로 나타났다. 누치속 7종의 계통유전학적 분석결과 섬진강산 누치(H. labeo)들은 두개의 단계통 (clade)을 형성하는데 하나는 하동, 임실, 강진, 순창의 섬진강 누치들로만 이루어진 단계통, 다른 하나는 하동의 HD2, HD8, HD9와 국내 부산, 아산, 서울 등에서 채집 보고된 누치들과 단계통을 형성하였다. 잉어과 내 섬진강산누치(HD1)의 계통진화적 위치는 피라미(Zacco platypus)와는 진화적 거리가 0.143, 누치속 H. maculatus와는 0.006으로 나타났다. 또한 잉어과 28종 내 계통유전학적 위치는 모래무지아과(Gobioninae) 어류들이 포함된 그룹 I에 섬진강산 누치가 위치함을 확인하였다. 본 연구의 결과는 누치를 포함하는 잉어과 내 어류의 계통유전학적 비교와 환경오염에 따른 담수환경 모니터링을 위한 모델어류의 발굴연구에 주요한 유전적 정보를 제공할 것이다.

DNA Barcoding of Fish, Insects, and Shellfish in Korea

  • DNA barcoding has been widely used in species identification and biodiversity research. A short fragment of the mitochondrial cytochrome c oxidase subunit I (COI) sequence serves as a DNA bio-barcode. We collected DNA barcodes, based on COI sequences from 156 species (529 sequences) of fish, insects, and shellfish. We present results on phylogenetic relationships to assess biodiversity the in the Korean peninsula. Average GC% contents of the 68 fish species (46.9%), the 59 shellfish species (38.0%), and the 29 insect species (33.2%) are reported. Using the Kimura 2 parameter in all possible pairwise comparisons, the average interspecific distances were compared with the average intraspecific distances in fish (3.22 vs. 0.41), insects (2.06 vs. 0.25), and shellfish (3.58 vs. 0.14). Our results confirm that distance-based DNA barcoding provides sufficient information to identify and delineate fish, insect, and shellfish species by means of all possible pairwise comparisons. These results also confirm that the development of an effective molecular barcode identification system is possible. All DNA barcode sequences collected from our study will be useful for the interpretation of species-level identification and community-level patterns in fish, insects, and shellfish in Korea, although at the species level, the rate of correct identification in a diversified environment might be low.

  • 본 연구를 통해 한국의 양식 가숭어에서 최초로 분리된 등각류는 Nerocila japonica와 가장 근연한 종으로 확인되었으나, 향후 형태학적 분류 등 명확한 종 동정이 추가로 필요할 것으로 판단되며, 약물의 직접 노출에 의한 구충 효과를 확인한 결과 trichlorfon이 가장 효과가 있는 것으로 확인되었다.

First Report of Echinococcus equinus in a Donkey in Turkey

  • A 2-year-old female donkey (Equus asinus) was euthanized in the Pathology Department of Firat University, Elazig, Turkey. Necropsy disclosed the presence of 7 hydatid cysts distributed throughout the lung parenchyma. One of those cysts represented the parasite material of the present study and was molecularly identified through sequencing of a fragment of cytochrome c oxidase subunit 1 (CO1) and nicotinamide adenine dinucleotide dehydrogenase subunit 1 (NADH1) gene, as Echinococcus equinus. The generated CO1 sequence supports the presence of the dominant haplotype as has been described in Europe and Africa. The NADH1 sequence was found similar to sequences reported in equids in Egypt and the United Kingdom. The molecular identification of E. equinus in a donkey is being reported for the first time in Turkey.

DNA Barcoding for Diophrys quadrinucleata (Ciliophora: Euplotia) from South Korea

  • One marine ciliate, Diophrys quadrinucleata Zhang et al., 2020 was newly recorded from South Korea in this study. We provided morphological diagnosis and images of the Korean D. quadrinucleata population. We determined the small subunit ribosomal DNA (SSU rDNA) and cytochrome oxidase subunit I (CO1) sequence data of D. quadrinucleata, and then the sequences were compared with other Diophrys species. Intra-specific variation between the Korean and type (Chinese) populations was identical in the SSU rDNA, while the inter-specific variations between seven Diophrys species were 0.3-3.8% in the SSU rDNA and 12.6-18.2% in the CO1. In this study, we obtained 18S and CO1 data from species with identified morphology. As the importance of securing 18S and CO1 based on morphology increases in current studies, this study will contribute to ciliate studies.
