Suppressive Effect of N-Acetylcysteine on the Adriamycin-Induced Micronuclei Formation in Mouse Bone-marrow Cells (생쥐 골수세포에서 아드리아마이신의 소핵생성에 미치는 N-마세틸시스테인의 억제효과)

  • The anticlastogenic effect of N-acetylcysteine was tested in vivo in mouse bone-marrow micronucleus assay. The frequencies of micronuclei induced by adriamycin (5 mg/kg i.p.) in bonemarrow cells were decreased by the oral administration of N-acetylcysteine at 12 h before adriamycin injection. The observed suppressing effect was not a reflection of a delay in the formation of micronuclei by the cytotoxic effect of N-acetylcysteine. The anticlastogenic effects of SH compound including N-acetylcysteine, cysteine, cystine, S-carboxy methylcysteine and glutathione were also investigated by the multiple pretreatment. Each SH compound was administered orally every day for 5 days and adriamycin (5 mg/kg i.p.) was injected at 24h after the last dose of test compound. N-acetylcysteine and glutathione showed significantly the suppressive effect at dose of 10 and 25 mg/kg for N-acetylcysteine and at the dose of 25 mg/kg for glutathione. Our study suggests that N-acetylcysteine is capable of protecting the chromosomal damages in the normal cells during cancer chemotherapy by adriamycin, and may act as an anticlastogen against induction of micronuclei by superoxide generating agent such as adriamycin.

  • The purpose of this study was to investigate the characteristics of human epidermal stratum corneum as protein model soils for detergency test. The stratum corneum was collected by scraping of the skin and purified with solvent. The results obtained were as follows: 1. Purified stratum corneum contained $92.38\%$ of crude protein. 2. In the amino acid compositions, contents of glycine, glutamic acid and serine were high and methionine and cystine were low. They were similar to fibrous $\alpha$-keratin consisted of stratum corneum. Whereas the content of polar amino acids was decreased, that of nonpolar amino acids was increased after enzyme hydrolysis. 3. The hydrolysis of stratum corneum with enzyme increased muck at initial reaction time and levelled off in 4$\~$6 hours. The hydrolysis with enzyme was improved effectively at its optimum temperature and pH. 4. The hydrolysis of stratum corneum with enzyme increased by the addition of surfactants. The order of compatibility with enzyme was in the order of Triton X-100>AOS>LAS.

  • Human hair is a kind of fibrous keratin which has much cystine residues like wool fiber. It protects human head from the dangerous shock and is an organ to release heavy metals in human body. Recently, in many dyeing field there is a trend to utilize the natural dyes. Cochineal is a reddish natural mordant dye. When the cochineal is applied in human hair dyeing, it is expected that dyeing rate will be slow and wash fastness will be poor on account of large molecular weight relative to oxidative dyes. In this work, we investigated the effects of tributyl phosphate in hair dyeing with natural dye cochineal. Dyeability of hair increased significantly by adding tributyl phosphate in cochineal dyeing. Nevertheless the hair was slightly damaged during dyeing, the dyed hair showed a good wash fastness.

Evaluation on Nutritional Value of Field Crickets as a Poultry Feedstuff

  • The proximate analysis, amino acid content and true amino acid digestibility and TMEn for poultry of adult Field crickets Gryllus testaceus Walker, were investigated. The insect was also used as partial replacement of protein supplements in the broiler diet on an equal CP percentage and TMEn basis. The results indicated that the adult insect contained: crude protein 58.3%; fat 10.3%, chitin 8.7% and ash 2.96% on dry matter basis, respectively. The total amounts of methionine, cystine and lysine in the Field crickets were 1.93%, 1.01% and 4.79%, respectively, and their true digestibility coefficients, determined in cecectomized roosters, were 94.1%, 85% and 96%, respectively. The TMEn of this insect meal was 2,960 kcal/kg determined in cecectomized roosters. When cornsoybean meal diets were formulated on an equal CP percentage and TMEn basis, up to 15% Field cricket could replace control diet without any adverse affects on broiler weight gain, feed intake or gain:feed ratio from 8 to 20 d posthatching.

  • Purified diets containing five levels of methionine with 0.4% cystine were fed to growing chicks (8 days old male Arbor Acre strain) to evaluate methionine requirements for growth and maintenance. A model was developed to separate methionine requirement for maintenance from requirement for growth. From this model the daily methionine requirement for growth was 4.22 mg/g gain, and the daily methionine requirement for maintenance was 0.034 times metabolic body size ($W^{0.75}$). Based on nitrogen gain response, the methionine requirement for growth was 0.162 mg/mg N gain, and the daily maintenance requirement was 0.037 times metabolic body size. The plateau of plasma methionine concentration reached at 117.16 mg intake pre day. The total methionine requirement determined based on weight gain response was 138.29 mg/day or 0.33% of the diet and the one determined based on nitrogen gain response was 141.7 mg/day of 0.34% of the diet, respectively. As a percentage of protein, methionine was calculated to be 2.6%; the reported methionine content of carcass CP was 1.76%.

  • Thioltransferase, also known as glutaredoxin, is an enzyme that catalyzes the reduction of a variety of disulfides, including protein disulfides, in the presence of reduced glutathione. Thioltransferase was purified from kale through ammonium sulfate fractionation, DE-52 ion-exchange chromatography, Sephadex G-75 gel filtration, and Q-Sepharose ion-exchange chromatography. Its molecular size was estimated to be about 31,000 daltons on SDS-PAGE. The purified enzyme has an optimum pH of about 8.0 with 2-hydroxyethyl disulfide as a substrate. The enzyme also utilizes L-sulfocysteine, L-cystine, bovine serum albumin, and insulin as substrates in the presence of GSH. The enzyme has $K_m$ values of 0.24-0.67 mM for these substrates. The enzyme was partly inactivated after heating at $80^{\circ}C$ or higher temperature for 30 min. The enzyme was stimulated by various thiol compounds such as reduced glutathione, dithiothreitol, L-cysteine, and $\beta$-mercaptoethanol. This is a second example of a plant thioltransferase which was purified and characterized.

  • The effect heat treatment (autoclave) on nutritional value of winged bean as compared to soybean has been investigated. The winged bean and soybean were obtained from local cultivar grown in Indonesia. The beans were autoclaved at $120^{\circ}C$ for 15, 30, 45, 60 or 90 minutes, respectively before being ground for chemical analysis. Trypsin inhibitors of winged bean and soybean decreased (p < 0.05) along with decreasing of urease activity as heating time increased from 0 to 90 minutes. Heat treatment significantly (p < 0.05) reduced protein solubility in 0.2% potassium hydroxide of winged bean as well as soybean. In vitro protein digestibility was significantly (p < 0.05) improved by heating treatment (15 to 60 min of autoclaving), however, excessive heating (90 min of autoclaving) decreased the digestibility of winged beans. Excessive heating had adverse effect on lysine, cystine and methionine contents of winged beans. The results of this study suggested that autoclaving at $120^{\circ}C$ within 45 minutes should be adequate to remove protease inhibitors and could improve protein digestibility of winged beans.

Single Hydrolysis Method for the Amino Acid Determination in Foods and Composite Dishes (식품의 아미노산 정량을 위한 단일가수분해 방법의 개발)

  • For the complete and accurate amino acid determination of protein and food samples, 3 different hydrolysis procedures have been conducted in parallel for each sample, which include the alkaline hydrolysis for tryptophan determination, performic acid oxidation prior to the acid hydrolysis for the determination of cysteine and cystine, and the 6N HCl hydrolysis for the determination of the rest of amino acids. In the present study, amino acid concentrations obtained from the modified single hydrolysis procedure were compared with the values from the conventional hydrolysis procedures in casein and nine food and composite dish samples. In most of the samples tested, the modified single hydrolysis procedure gave significantly higher values of cysteins and cystein compared to the performic acid oxidation method, but resulted in a considerable destruction of tryptophan in food and composited dish samples. There was no consistent difference in the rest of amino acid concentrations between the two hydrolysis systems. Therefore, for complete amino acid determination of various foods and composite dishes, the single hydrolysis method may replace the 6N HCl hydrolysis and performic acid oxidation methods, and thereby reduces 3 hydrolyses to 2 steps with much higher recoveries of the sulfur containing amino acids.

  • Soybean has been considered as an important source of food protein in many countries. This experiment was attempted to determine the effect of mineral nutrition on growth and amino acid composition of soybean by using the methods developed for other plants. The results showed that S was more effective than P on the growth of soybean when the amount of cation was not changed. Among cations investigated K was more effective than Mg and Ca on its growth. However, higher amounts of Ca resulted in toxic effect. In general, the content of amino acid hydrolyzed by HCl decreased in NS and NP as well as KCa and KMg groups. Amino acids, i.e., proline, cystine, aspartic acid, lysine and histidine were not detected in NS and NP in addition to KCa and KMg groups eventhough control group showed considerable amount of these amino acids.

  • The industry of amino acids has developed for the synthetic methods, zymotechnics from the methods of extraction. In the present, part of amino acids (L-cystine, L-tyrosine, etc.) is manufactured by the methods of extraction, but most of amino acids is produced by synthetic methods, zymotechnics. Among the methods, the synthetic methods use of cyanic acid, which generate a large of waste water by acid or alkali in hydrolysis. In this point of view, improved new synthetic methods are demanded for being the influence of environment. This article Introduces new synthetic methods of phenyl alnine, further more, the recent of research results are introduced to prepare the derivative ${\alpha}-keto$ acids of precursor for preparing more than particular amino acids.
