환원제 첨가가 쌀당화액의 Bifidobacterium발효에 미치는 영향 (Effect of Reducing Agents on Bifidobacterium Fermentation of Saccharified Rice Solution)

  • This study was intended to develop a new rice product by the fermentation of saccharified rice solution using Bifidobacterium and to select an appropriate reducing agent to provide the anaerobic condition for the growth of Bifidobacterium during fermentation. The enhancement of the growth of Bifidobacterium in saccharified rice solution was achieved by the treatment of reducing agents such as ascorbic acid and cysteine. The physical and chemical properties of the fermented product were evaluated, and the effect of the reducing agents were compared between ascorbic acid and cysteine. The fermented product with the addition of ascorbic acid shows the lower pH and the higher titratable acidity comparing the product with the addition of cysteine. This indicated that ascorbic acid was more appropriate reducing agent than cysteine for the fermentation of the saccharified rice solution. The number of viable Bifidobacterium in the fermented product with the addition of ascorbic acid(2.2$\times$108~3.4$\times$108CFU/ml) was greater than that with the addition of cysteine (8$\times$107~2.8$\times$108CFU/ml). Ascorbic acid supplement also contributed better sensory properties, such as flavor, taste and overall acceptibility than cysteine supplement did.

식이내의 단백질과 타우린 함량이 Cysteine 대사에 미치는 영향 (Effect of Dietary Protein and Taurine on Cysteine Catabolism in Cat Liver)

  • Activieties of hepatic cysteine desulfhydration was assessed in cats fed one of the following diets for 5 weeks : 20% protein, 0% taurine diet(LPOT) ; 20% protein, 0.15% taurine diet (LPNT) ; 60% protein, 0% taurine diet(HPOT) ; and 60% protein, 0.15% taurine diet(HPNT). Cats fed LPOT and HPOT had been maintained on a taurine-free diet for 6 weeks prior to the experiment in order to deplete body taurine. Activities of cysteine desulfhydration were determined by measuring the production of H235S from 35S-cysteine in the presence and absence of $\alpha$-ketoglutarate ($\alpha$-KG) in the incubation medium. The direct pathway via cysteine desulfhydrase appears to account for the major route of cysteine desulfhydration in the cat liver since the values obtained in the absence of $\alpha$-KG were between 81 and 88% of those obtained in the presence of $\alpha$-KG. Mean$\pm$SEM of the hepatic total desulfhydration activities(umol H2S.min-1.kg body wt-1)in cats fed LPOT, LPNT, HPOT and HPNT were 117$\pm$6, 135$\pm$10, 137$\pm$10, and 190$\pm$9, respectively. The capacity of hepatic cysteine desulfhydration (UA/kg body wt) was positively cerrelated not only with the dietary concentration of taurine but also with the concentration of protein.

The Effects of Different Concentrations of Glycine and Cysteine on the Freezability of Moghani Ram Spermatozoa

  • Two experiments were designed to evaluate the effects of the amino acids glycine and cysteine on cryopreservation of ram spermatozoa. After primary evaluation of collected ejaculates, the semen samples were pooled and diluted 1:4 before cooling (experiment 1) and freezing (experiment 2) with Tris-Citrate-Fructose-Yolk (TCFY) extender supplemented with different concentrations of glycine and cysteine (5, 10, 15 and 20 mM). As the control, semen was diluted and frozen in the extender without amino acids. Motility, viability and membrane integrity were assessed as the parameters for semen quality in the first experiment. In the second experiment, motility, progressive motility, viability, membranes and acrosome integrity were evaluated after the freezing-thawing process. The results of the first experiment indicated that the addition of 10 and 15 mM cysteine compared to the control (basic) extender significantly increased (p<0.01) the motility, viability and membrane integrity of spermatozoa after cooling. However, further increasing these amino acids up to 20 mM had a significant negative effect (p<0.05). Our results showed no significant differences (p>0.05) between 5 mM glycine compared to 5 mM cysteine and between 20 mM glycine and 20 mM cysteine. The results of experiment 2 showed that the amino acids significantly improved post-thaw motility, progressive motility, viability, membranes and acrosome integrity of ram spermatozoa. These positive effects were observed at concentrations between 5 to 15 mM of glycine and cysteine, with the best results at 15 mM. Further increasing of amino acid concentrations significantly decreased the post-thaw characteristics of spermatozoa, but the results showed that cysteine was better than glycine and control extenders. The data indicated that addition of glycine or cysteine to the freezing extender can be recommended for cryopreservation of ram spermatozoa. However, further studies are still needed to determine the effect of such addition on fertility in farm animals.

N-acetyl-L-cysteine and cysteine increase intracellular calcium concentration in human neutrophils

  • N-acetyl-L-cysteine (NAC) and cysteine have been implicated in a number of human neutrophils' functional responses. However, though $Ca^{2+}$ signaling is one of the key signalings contributing to the functional responses of human neutrophils, effects of NAC and cysteine on intracellular calcium concentration ($[Ca^{2+}]_i$) in human neutrophils have not been investigated yet. Thus, this study was carried out with an objective to investigate the effects of NAC and cysteine on $[Ca^{2+}]_i$ in human neutrophils. We observed that NAC ($1{\mu}M{\sim}1mM$) and cysteine ($10{\mu}M{\sim}1mM$) increased $[Ca^{2+}]_i$ in human neutrophils in a concentration-dependent manner. In NAC pre-supplmented buffer, an additive effect on N-formyl-methionine-leucine-phenylalanine (fMLP)-induced increase in $[Ca^{2+}]_i$ in human neutrophils was observed. In $Ca^{2+}$-free buffer, NAC- and cysteine-induced $[Ca^{2+}]_i$ increase in human neutrophils completely disappeared, suggesting that NAC- and cysteine-mediated increase in $[Ca^{2+}]_i$ in human neutrophils occur through $Ca^{2+}$ influx. NAC- and cysteine-induced $[Ca^{2+}]_i$ increase was effectively inhibited by calcium channel inhibitors SKF96365 ($10{\mu}m$) and ruthenium red ($20{\mu}m$). In $Na^+$-free HEPES, both NAC and cysteine induced a marked increase in $[Ca^{2+}]_i$ in human neutrophils, arguing against the possibility that $Na^+$-dependent intracellular uptake of NAC and cysteine is necessary for their $[Ca^{2+}]_i$ increasing activity. Our results show that NAC and cysteine induce $[Ca^{2+}]_i$ increase through $Ca^{2+}$ influx in human neutrophils via SKF96365- and ruthenium red-dependent way.

함유황아미노산이 지질대사에 미치는 영향 (Effects of Sulfur-Containing Amino Acids on Lipid Metabolism in Rats)

  • The effect of sulfur-containing amino acids on lipid metabolism was studied in rats fed casein as a protein source. Plasma cholesterol, HDL-cholesterol and atherosclerotic index decreased in the cysteine group compared to the methionine group. Plasma triglyceride and phospholipid level were not affected by the supplementation of the sulfur-containing amino acids. The levels of cholesterol and triglyceride in liver decreased by both methionine and cysteine. Cysteine increased the fecal excretion of coprostanol, total neutral steroid and bile acid. The results suggest that plasma cholesterol level is affected by dietary ratio of cysteine/methionine and that the hypocholesterolemic effects of cysteine is, at least in part, through reducing cholesterol absorption from small intestine and through enhancing fecal excretion of bile acids.

백출 추출물과 L-cysteine의 병용처리가 양송이버섯의 갈변억제에 미치는 영향 (Browning Inhibition Effect of the Atractylodis Rhizoma Alba Extract and L-cysteine Combination on Agaricus bisporus)

  • 본 연구는 0.1% 백출 추출물과 0.05% L-cysteine이 양송이버섯의 저장 및 유통 시 갈변억제에 미치는 영향을 조사하기 위하여 각각의 갈변저해제에 양송이버섯을 3분 간 침지 시킨 후 실온에서 1시간 동안 건조시켜 6개씩 PS tray에 담아 PVC랩으로 포장 한 후 저장 기간 별 갈변억제능을 측정하였다. 실험결과 저장 마지막 날 0.1% 백출+0.05% L-cysteine 처리군의 Hunter L 값이 87.24로 가장 높게 유지되었으며, ${\Delta}E$ 값은 5.56으로 색 변화가 적었다. Tyrosinase 저해활성 측정결과 0.1% 백출+0.05% L-cysteine 처리군의 효소저해활성이 높게 측정되어 0.1% 백출 추출물과 0.05% L-cysteine 처리가 갈변억제에 효과적이었다. 또한 0.1% 백출 추출물과 0.05% L-cysteine 병용처리가 양송이버섯의 갈변억제 이외에 품질유지에 어떠한 영향을 끼치는지 조사하기 위하여 품질평가를 시행하였다. 중량감소율과 경도측정 결과 모든 저장일 수에서 대조군보다 0.1% 백출+0.05% L-cysteine 처리군이 중량감소가 적었으며 높은 경도를 유지하였다. 관능평가 결과 전체 기호도 항목에서 대조군이 저장 11일, 0.1% 백출+0.05% L-cysteine 처리군은 14일에 상품성이 상실되어 대조군에 비해 약 3일 가량 상품성이 오래 유지되었다. 본 실험결과를 바탕으로 0.1% 백출 추출물과 0.05% L-cysteine 병용처리는 양송이버섯의 갈변억제에 효과적이며, 중량 및 경도 감소, 이취 발생 등 품질감소를 지연시켜 선도연장에 도움을 줄 것으로 사료된다.

랫트에 있어서 captafol의 독성에 대한 cysteine의 방어 작용 1. 혈액학 및 혈청 생화학적 성상에 미치는 영향 (Cysteine antagonism of captafol induced toxicities in rats 1. Effects on hematological and serum biochemical values)

  • 과일 및 채소류의 곰팡이 억제제로 개발된 captafol은 인간 및 동물에서의 독성작용은 많이 보고된 바 있으나 이들의 독작용을 완화시키거나 치료할 수 있는 제제에 대한 연구는 별로 알려진 바 없다. captafol에 의한 혈액학 및 혈청 생화화적 변화에 따른 cysteine의 방어 작용을 확인할 목적으로 랫트에 5mg/kg 용량의 captafol을 1회 복강 투여하고 동시에 5, 58, 290 및 580mg/kg cysteine을 각각 투여하고 체중의 변화, 부검소견, 절대장기중량, 혈액학 및 혈청 생화학적 소견을 비교 분석 하였다. captafol 단독 투여군에서는 체중 및 간 장기중량 감소, 복강내 장기들의 섬유소에 의한 유착 및 복수 등이 현저하였으며 적혈구 수, hemoglobin 농도, 혈청 AST의 증가가 인정되었으며 또한 혈청 인지질 함량 감소가 확인되었다. 한편, cysteine 58mg/kg 이상의 용량군에서는 복강내 장기들의 유착 및 복수가 확인되지 않았으며 장기중량, 혈액학 및 혈청 생화학적 소견에서도 정상을 나타내었다. 이 연구 결과는 captafol의 중독증상에 대해 cysteine 및 sulfhydryl군을 가진 물질은 그 증세를 감소시키거나 완화시킬 수 있는 효과가 있음을 보여주고 있다.

숫컷 생쥐에서 타우린 투여에 의한 간내 글루타치온의 감소 (Reduction of Hepatic Glutathione by Acute Taurine Treatment in Male Mice)

  • Effect of taurine treatment on metabolism of glutathione (GSH) was studied in adult male ICR mice. An acute injection of taurine (250 mg/kg, ip) resulted in a significant decline of hepatic GSH level at t = 6 hr, but plasma GSH level was not altered. The activity of GSH-related enzyme in liver, such as GSH peroxidase, GSSG reductase, GSH S-transferases, ${\gamma}$-glutamylcysteine synthetase or ${\gamma}$-glutamyltranspeptidase, was not affected by taurine at t = 2.5 or 6 hr. Plasma cysteine and cystine levels were elevated rapidly following taurine treatment. Hepatic cysteine level was decreased by taurine, reaching a level approximately 70% of control at t = 4 and 6 hr. In conclusion, the results indicate that an acute dose of taurine decreases hepatic GSH level by reducing the availability of cysteine, an essential substrate for synthesis of this tripeptide in liver. It is also suggested that taurine may decrease the cysteine uptake by competing with this S-amino acid for a non-specific amino acid transporter.

대두(Glycine max) trypsin 억제제의 불활성화 및 품종별 억제활성 (Inactivation of trypsin inhibitor and inhibitory activity of soybean(Glycine max) cultivars)

  • 대두 STI의 열처리에 의한 불활성화에 L-cysteine 및 sodium sulfite의 첨가 효과를 조사하고, 대두 품종별 STI활성을 측정하였으며 활성도와 단백질 및 cysteine함량 그러고 소화율과의 관계를 비교하였다. 열처리에 의한 STI의 불활성화에 L-cysteine과 sodium sulfite의 첨가는 불활성화 효과를 크게 증가시켰으며 대두 품종간 불활성화 정도는 차이를 보이지 않았으며 L-cysteine과 sodium sulfite의 첨가는 불활성화된 STI의 재활생화를 크게 억제하였다. 품종별 STI의 활성도는 대두분말 g당 $64.7{\sim}86.47\;TIU$의 범위에 있었으며, 장백>힐>장엽, 광교>단엽>백운>단경>팔달, 새알, 덕유>황금 이었으며, 단백질 함량과 STI의 활성도와는 상관관계(r=-0.192)가 없는 것으로 나타났다. 품종별 cysteine 함량은 힐, 장백, 단경, 단엽, 황금, 백운, 장엽, 새알, 덕유, 광교, 활달의 순서이었으며, cysteine의 함량은 $73.5{\sim}40.0\;{\mu}mole/g$ 대두분 이었다. 또한 cysteine함량과 STI 활성도 사이에 정의 상관(r=0.6568)을 나타냈다. 품종별 소화율은 광교, 백운, 팔달, 단경, 새알, 힐, 장엽, 덕유, 황금, 장백, 단엽의 순서이었으며 $81.9{\sim}76.7%$ 정도이었다. 또한 소화율과 STI활성도 사이에는 부의 상관(r=-0.7695)을 나타내었다.

마늘의 마이크로웨이브 추출 특성과 갈변억제 효과 (Extraction Characteristics and Browning Inhibitory Effects of Fresh Garlic by Microwave-assisted Extraction)

  • 마늘을 MAE방법에 의하여 물과 에탄올로 추출한 후 기능성과 갈변억제 효과를 비교하였다. 에탄올, 물 추출물의 수율은 5분 동안 추출한 경우 각각 12.9% 및 12.7%이었고 추출시간이 증가할수록 감소하였으며 비교적 낮은 값을 나타내었다. Polyphenol 함량은 5분 추출물이 각각 574.3mg% 및 569.0mg%로 가장 높았다. 에탄올과 물 추출물의 전자공여작용은 20분 추출물의 경우 각자 17.0% 및 20.7% 수준으로 가장 높은 효과를 나타내었다. 아질산염 소거작용은 pH 1.2일 때 5분 동안 추출한 추출물이 각각 98.9% 및 99.7%로 가장 높았으며 pH가 증가할수록 효과가 현저하게 감소하였다. Angiotensin I-converting enzyme 저해 효과는 물 추출물의 경우 5분 추출물이 83.1%로 가장 높은 것에 비해 에탄올 추출물은 87.1%로 20분 동안 추출 하였을 때 그 값이 제일 높았다. Tyrosinase 저해효과는 물 및 에탄올 추출물 모두 30%이하로 매우 낮은 효과를 나타내었으나 15분 에탄올 추출물은 60%이상의 값을 나타내었다. 이들 물과 에탄올 추출물에 대해 0.1% ascorbic acid, citric acid 및 cysteine으로 10일 동안 갈색 억제 효과를 측정한 결과 모든 처리구에 비해 0.1% cysteine이 가장 높은 갈변 억제 효과를 나타내었다. 0.1% ascorbic acid는 cysteine보다는 낮은 갈변 억제 효과를 보였으며 0.1% citric acid는 대조구와 비슷한 갈변화 진행을 나타내었다. 갈색화 억제 상승 효과는 0.1% cysteine과 ascorbic acid를 첨가한 실험구가 citric acid를 첨가한 실험구에 비해 효과적인 갈색화 억제를 나타내었다.
