The patient with end-stage renal disease show several nervous complications. The factors contributing to the nervous complications are still incompletely characterized. Cyanate, known as one of the uremic toxins, is derived spontaneously from urea. To investigate the mechanism of cyanate-induced effect on C6 glioma cells, the glioma cells were treated with 0, 1, 5, 10, 20 and 40 mM cyanate. There was a dose-dependent decrease in cell viability and the decreased number of cell was observed in glioma cells by treatment with cyanate. Western blot showed the down- regulation of procaspase-3, which means up-regulation of caspase-3, and the up-regulation of caspase-8, but the down-regulation by cyanate. In addition, cDNA microarray showed 934 down-regulated genes and 165 up-regulated genes on 1,099 genes in cyanate treated group. Treatment with cyanate led to 16 down-regulated genes and 6 up-regulated genes on apoptosis category, and especially heat shock 70 kD protein 1A gene on the category of apoptosis was significantly up-regulated. These results suggest that cyanate can induce apoptosis through caspase-8 and caspase-3 in glioma cells and decrease of gene expression including apoptosis category in glioma cells. These effects of cyanate may play a role in the nervous complications of patient with end-stage renal disease.

The purpose of this study was to evaluate the effects of cyanate methacylate on the shear bond strengths to bovine dentin surfaces as a dentin primers. Seven experimental adhesives were made with different mass fraction of Isocyanatoetylme-thacrylate (IEM), 40wt% HEMA (Wako Pure Chemical Industries Osaka, Japan), 0.6% camphoroquinone, 0.4% amine and ethanol as balance dentin bonding agents (0, 2, 4, 6, 8, 10, 12%) were made and applied on the surface of bovine dentin specimens of 7 experimental groups. Shear bond strengths were measured using a universal testing machine (Instro 4466). To identify the ratio and modes of cohesive failures, microscopic examinationn was performed. The ultra-structure of resin tags were observed under scanning electron microscope. The results were as follows ; 1) A higher shear bond strengths (33.62 MPa) in group 8% of Cyanate methacrylate to dentin were found, but there were no statistically significancy between Groups (p > 0.05). 2) The higher ratio of cohesive failures mode in group 2, 6, an 10% could be seen than that in any other groups. 3) A shorter resin tags were observed in all experimental groups. This could be resulted that the preventing from the cyanate methacrylate penetrate into dentin owing to reacting it with dentin collagen. Therefore the resin tags were shorter in lengths. Whether the higher bonding strengths of dentin bonding agents can be affected was not been assured with statistic results. The results indicated that the relation between tensile strengths of the dentin adhesives to bovine dentin and resin tags formed into the dentin could not affected. The main reason of increasing the shear bond strength to bovine dentin in experimental groups could not be assured.
Carbon fiber reinforced polymer (CFRP) composites consist of carbon fibers in a polymer matrix. Recently, CFRP composites having high thermal stability and low outgassing are finding their use in high performance materials for aerospace and electronics applications under high temperature and high vacuum conditions. Cyanate ester resin is one of the most suitable matrix resins for this purpose. In this study, proper combination of cyanate ester and catalyst, curing behavior, and cure cycle were determined by chemorheology. Optimum condition was found to be catalyst content of 100 ppm and curing temperature of $150^{\circ}C$. Thermal stability and outgassing of cured resin composition were analyzed and the results showed thermal decomposition temperature of $385^{\circ}C$ and total mass loss of 0.29%. The CFRP prepregs and subsequent composites were fabricated by predetermined resin composition and the cure condition. Tensile moduli of the composites were compared with theoretical models and the results were very consistent.
We proposed a high-temperature circuit-analog radar absorbing structures (CA-RAS), and evaluated radar absorption performance and tensile properties in 350℃ and a hot-wet environment. The CA-RAS was implemented with a glass/cyanate ester composites and a square resistive pattern layer, and reflection loss was measured by 350℃ and after exposure of hot-wet condition using free space measurement. And the tensile strength at 350℃ and after exposure of hot-wet condition was measured according to the ASTM D638. The proposed CA-RAS showed a 4 GHz of -dB bandwidth and -20 dB of a peak value at 350℃. In addition, there was no deterioration in absorption performance after exposure to a hot-wet condition. The tensile strength value of more than 95% compared to the strength of the glass/cyanate ester composite was confirmed at 350℃ and after exposure of hot-wet condition. Through this, the applicability of CA-RAS proposed in this study was confirmed as a load bearing structure for stealth weapon exposed to high temperature and hot-wet environment.
Radiation is one of the major therapy for the removal of cancer cells. The results of the radiation therapy depend on the radio-resistance of cancer cells. For the effective treatment in these radio-resistant cancers, the use of chemicals that act on cancer cells is known to enhance the cytotoxic effects of radiation therapy. In this study, I investigated the effect of potassium cyanate (KCN) on the irradiated-colorectal cancer cell line, HCT 116 cells. KCN induces the carbamylation of proteins and can change the biological activity of various human cells. To understand the effect of KCN on the radiosensitivity of HCT 116 cells, I examined alteration of the cell cycle, generation of reactive oxygen species (ROS), cell viability, apoptosis and intracellular signaling proteins in the irradiated cells with/without KCN treatment. Combination treatment caused significant increase in sub $G_0/G_1$ and ROS generation in HCT 116 cells. KCN inhibited the proliferation and cell viability in irradiated HCT 116 cells. KCN-induced apoptosis of irradiated cells was processed via the activation of caspase 3 and caspase 9. Apoptosis-associated signal proteins, including Bax and Bcl-2 were regulated by irradiation with KCN treatment. Taken together, these results may indicate that KCN enhances the radiosensitivity of radio-resistant cell and then has a synergistic effect on radiation therapy in colorectal cancer.
Potassium cyanate (KOCN) is an inorganic compound and induces the carbamylation of proteins with cytotoxic effects on human cells. Although there is a potential cytotoxic molecule, the role of KOCN on the apoptosis of cancer cell is not well understood. The present study investigated the effects of KOCN on the human colorectal cancer cell line, HCT 116 cells. To understand the anti-cancer effect of KOCN on HCT 116 cells, we examined alteration of apoptosis, the intracellular $Ca^{2+}$ concentration, the intracellular signaling pathway and generation of reactive oxygen species (ROS) in these cells treated with KOCN. The apoptosis of HCT 116 cells was induced by KOCN in a dose-dependent manner at 24 hours and 48 hours, respectively. The apoptosis was processed via the cleavage of poly ADP-ribose polymerase (PARP) and activation of caspase 3 in HCT 116 cells. KOCN induced the elevation of intracellular $Ca^{2+}$ concentration and changed the expressions of Bcl-2 family proteins. The pro-apoptotic Bax was continuously up-regulated, and the anti-apoptotic Bcl-2 was down-regulated by KOCN. KOCN also induced the hyperpolarization of mitochondria and the generation of ROS in HCT 116 cells. Taken together, these results indicate that KOCN induces the apoptosis of HCT 116 cells by disruption of $Ca^{2+}$ homeostasis and via mitochondrial pathway. This study provides the compound that may be used as a potent agent for the treatment of colorectal cancer.
Potassium cyanate (KOCN) that is known as an inducer of the protein carbamylation is an inorganic compound and is the conjugate based of cyanic acid (HOCN). Based on these studies, we confirmed that KOCN induces the apoptosis of the human colorectal cancer cell line, HCT 116 cells, by various mitochondrial pathways. To investigate other mechanisms of KOCN-mediated apoptosis, in the present study, we examined KOCN-induced cytokines production in HCT 116 cells and identified the intracellular signaling pathway in these processes. We first demonstrated that KOCN considerably increased the cell apoptosis via intracellular $Ca^{2+}$ signaling, mitochondrial dysfunction and ROS production. And then we examined TNF-${\alpha}$ and IL-$1{\beta}$ levels mediated by KOCN in HCT 116 cells. Although IL-$1{\beta}$ was not involved in KOCN-mediated HCT 116 cell apoptosis, the release of TNF-${\alpha}$ was mediated by KOCN in HCT 116 cells via NF-${\kappa}B$ activation. Apoptosis was also enhanced by incubation with supernatants from HCT 116 cells after KOCN treatment and this effect was partially reduced by BAY 11-7085 pre-treated supernatant. Taken together, our results indicate that KOCN-induced apoptosis in HCT 116 cells is dependent on the releases of TNF-${\alpha}$ and the increased factors and that the mechanism involves the activation of NF-${\kappa}B$.
Potassium cyanate (KCN) is an inorganic reagent and can induce the post-translation carbamylation of proteins. The carbamylated reaction in the body is involved in cell death in various diseases. According the results in our previous study, KCN enhances the radiosensitivity of human colorectal cancer cell line, HCT 116 cells. However, it was not enough to confirm the mechanism that KCN works in these cells. To determinated the mechanisms of KCN in the cells with increased radiosensitivity, HCT 116 cells were treated KCN with low-dose gamma-radiation. And then, we examined alteration of the cell cycle, cell proliferation, cytokine level and the activation of cell signaling protein. As a result, cell cycle arrest and cell death were induced by the activation of caspase-3 and PARP in the irradiated cells with KCN treatment. These changes of the irradiated cell with KCN treatment were induced by the release of $TNF-{\alpha}$ via $NF-{\kappa}B$ activation. In conclusions, enhanced radio-sensitivity mediated by KCN induced cell death and it occurs by $NF-{\kappa}B$-dependent $TNF-{\alpha}$ production.

The optical bench was designed using composite material, M40J/Cyanate Ester. Mechanical tests, thermal tests were carried out for M40J and structural vibration FEM analysis was performed. From the experimental results, the material properties of M40J/Cyanate Ester were obtained in longitudinal and transverse directions. By applying the properties into FEM vibration analysis, a proper stacking sequence was proposed for the laminated facesheets, so that natural frequency of the platform structure satisfy the design specification.
